The effect of linolenic acid (18:3) on release of the 43 kDa polypeptide and manganese from photosystem II ( PS II ) membranes depleted of extrinsic polypeptides was studied. In both control and NaCl-washed particles which were depleted of the extrinsic 23 and 16 kDa polypeptides, the 18:3 treatment caused a 20% release of the 33 and 43 kDa polypeptides. In CaCl2 , (or urea + NaCl)-washed particles, which were depleted of the 33 kDa polypeptide in addition to the 23 and 16 kDa polypeptides, the release of the 43 kDa polypeptide increased to 70%, whereas only 25% of the 47 kDa polypeptide was removed. These findings suggest (i) that the 33 and the 43 kDa polypeptides are neighbows in the photosynthetic membrane and (ii) that the 33 kDa polypeptide shields the 43 kDa polypeptide against the action of 18:3. Incubation of CaCl2 , or (urea + NaCI)-treated PSII particles in the presence or absence of 18:3 resulted in the loss of only 2 of the 4 Mn atoms present per reaction center. this indicates that the 2 Mn atoms more firmly associated with PSII are not affected by the removal of the extrinsic 16, 23 and 33 kDa polypeptides, and the intrinsic 43 kDa polypeptide. nor by the treatment with linolenic acid. 相似文献

High rate anaerobic digestion of a petrochemical wastewater using biomass support particles 总被引:6,自引:0,他引:6
Anaerobic digestion of wastewater from a dimethyl terephthalate plant was studied in continuously stirred tank reactors with plastic net biomass support particles (BSP) at a level of 20% (v/v). The experimental results showed that the BSP system could treat the wastewater at a hydraulic retention time as low as 1.5 d, organic loading as high as 20 kg COD/m3/d and at acidic feed pH as low as 4.5 with 95% COD reduction and biogas production of about 8l/l/d, while the control system without support particles could not treat the wastewater above a 5-d hydraulic retention time, 5 kg COD/m3/d organic loading and a feed pH of 6.0. Thus, augmentation of BSP upgraded the performance of the conventional suspended growth system to an equivalent level to advanced reactors. 相似文献

A complete synthesis of 1-O-hexadecyl-2-O-N-(heptadec-8-cis-enyl)carbamyl-sn-glycero-3-Phosphocholine, a novel analog of phosphatidylcholine, has been described. Each step is simple to perform and gives the desired products in high yield. Also, some of the intermediates formed during the synthesis have been efficiently utilized to prepare 1-O-hexadecyl-2-O-oleyl-sn-glycero-3-phosphocholine, 1-O-hexadecyl-2-oleoyl-sn-glycero-3-phosphochloine and 3-O-hexadecyl-2-oeloyl-sn-glycero-1-phosphocholine. A successful approach has been developed for the sequencing of apolipoprotein B based upon the procedure of Cleveland et al. [(1977) J. Biol. Chem. 252, 1102-1106] involving limited proteolysis in the presence of sodium dodecyl sulfate. Staphylococcus aureus protease was employed to produce large peptides which were isolated in relatively pure form by preparative gel electrophoresis. Two peptides were partially sequenced using spinning-cup microsequencing techniques. The sequences are: Peptide R2-5, -Ala-Leu-Val-Gly-Ile-Asn- Gly-Glu-Ala-Asn-Leu-Asp-Phe-Leu-Asn-Ile-Pro-Leu-Arg-Ile-Pro-Pro- Met-Arg-(Arg)-; Peptide R3-1, -Leu-Val-Ala-Lys-Pro-Ser-Val-Ser-Val-Glu- Phe-Val-Thr-Asn-Met-Gly-Ile-Ile-Pro-Lys-Phe-Ala-Arg-. Summary The cortical cell membranes of maize and marrow roots grown at normal, or chilling, temperatures have been studied by freeze-fracture electron microscopy. Using computer-assisted methods to analyse intramembraneous particle (IMP) frequencies, diameters and distribution, no significant trends in differences between normal and chilled roots were found. While this result does not correspond with the findings from similar experiments on microorganisms, it is compatible with contemporary ideas concerning temperature-induced phase transitions in the lipids of higher plant cell membranes. The cortical cell membranes of barley roots that had been subjected to cold osmotic shock also showed no differences from untreated roots as demonstrable in freeze-fracture replicas.IMPs were found to cluster around plasmodesmata after chilling but the physiological significance of this, if any, remains to be investigated further.While these negative results only indirectly help towards understanding how cell membranes react to chilling, the techniques described open the way for more detailed analyses of IMP characteristics in plant cell membranes. 相似文献

Nuclear proteins have been fractionated into five distinct classes according to their extractability from rat liver nuclei at different pH and salt concentrations. The fractions have been analyzed for their amino acid composition which shows the presence of NG, NG-dimethylarginine, in sizable amount, in non-histone nuclear proteins (NHNP). Direct evidence for the presence of a rotenone-resistant NADH dehydrogenase on the inner surface of the inner membrane of plant mitochondria 总被引:2,自引:0,他引:2
Submitochondrial particles (SMP) were produced from Jerusalem artichoke (Helianthus tuberosus L.) mitochondria by sonication and differential centrifugation. The SMP were about 50% inside-out as measured by the access of reduced cytochrome c to cytochrome c oxidase. Uncoupled NADH oxidation (1 mM NADH) by the SMP was 120 nmol O2 min?1mg?1, which was reduced to 98 nmol O2 min?1 (mg mitochondrial protein)?1 in the presence of EGTA. In contrast, the oxidation of NADH by intact mitochondria was completely inhibited by EGTA (from 182 to 14 nmol O2 min?1mg?1). The EGTA-resistant NADH oxidation by the SMP is ascribed to the NADH dehydrogenase(s) on the inside of the inner membrane and exposed to the medium in the inside-out SMP. In the presence of EGTA it could be shown that two NADH dehydrogenase activities were present in the SMP. One had an apparent Km of 7 μM for NADH, a Vmax of 80 nmol NADH min?1mg?1, and was rotenone-sensitive. This dehydrogenase is equivalent to the mammalian Complex I NADH dehydrogenase. The other dehydrogenase, which was rotenone-resistant, had a Km of 80 μM and a Vmax of 131 nmol NADH min?1mg?1; it is probably responsible for the rotenone-resistant oxidation of organic acids often observed in plant mitochondria. The redox poise of the pyridine nucleotides had only a small effect on the relative rates of the two internal dehydrogenases. Electron flow through these dehydrogenases appears, therefore, to be regulated mainly by the concentration of NADH in the matrix of the mitochondria. 相似文献
