Gum specimens from Acacia tortilis ssp. spirocarpa, ssp. raddiana var. pubescens (two specimens) and ssp. heteracantha (three specimens) have been analysed. The results are of chemotaxonomic interest. Although the gum from ssp. raddiana var. pubescens appears to be intermediate between those from ssp. spirocarpa and ssp. heteracantha in terms of some of the analytical parameters, the overall impression is that ssp. raddiana is more similar chemically to ssp. spirocarpa than to ssp. heterocantha. The latter yields a viscous, proteinaceous polysaccharide that differs from those from both ssp. spirocarpa and ssp. raddiana by having a much higher ratio of arabinose to galactose, higher nitrogen and methoxyl contents, and much higher intrinsic viscosity and molecular weight; preliminary experiments have shown this gum to consist of two components.  相似文献

The gum polysaccharides from Terminalia sericea and T. superba have been analysed. They have a complex sugar composition, containing galacturonic, glucuronic, and 4-O-methylglucuronic acids as well as galactose, arabinose, rhamnose, mannose and xylose. The exudates from T. sericea and T. superba are remarkably similar in composition, particularly with respect to their proportions of neutral sugars and total uronic acid content, although T. sericea gum contains considerably more 4-O-methylglucuronic acid than T. superba. Both gums are very viscous and dissolve readily to give solutions of good colour.!!  相似文献

Summary Inoculation of pearl millet (Pennisetum americanum (L.) Leeke) with Azospirillum significantly increased the numbers of this organism in the rhizosphere, rhizoplane, washed and crushed roots and surface sterilized and crushed roots. The maximum number of organisms plant–1 were localized in the rhizosphere. The numbers of Azospirillum on the roots of inoculated plants grown under sterilized conditions were much higher than in the field grown plants. In both cases populations outside the roots were higher than in the surface sterilized roots. The highest numbers per unit root weight were recorded between 60–75 days of growth. N2-ase activity throughout the growth cycle was very low and was not related to the populations of Azospirillum on the roots. Root exudates and extracts of pearl millet showed a stimulatory effect on the growth of Azospirillum suggesting their possible involvement in the colonization of this organism on the roots of inoculated plants.  相似文献

Summary Young stems ofCitrus plants were infected with the fungusPhytophthora citrophthora. The effect of the infection on gum duct development was studied. The following sequence of structural changes was observed in the cambial zone: 1. The middle lamellae between layers of xylem mother cells dissolve forming duct cavities. 2. The cells around the duct cavities differentiate into epithelial cells rich in cytoplasm. 3. The amount of Golgi bodies and associated vesicles increases. The vesicles and small vacuoles, some of which seem to originate from the fusion of Golgi vesicles, contain fibrillar material that stains for polysaccharides. Vesicles and vacuoles appear to fuse with the plasmalemma. Material staining positively for polysaccharides accumulates between the plasmalemma and cell wall, and penetrates the latter. 4. The protoplast shrinks and the space below the cell wall, which contains polysaccharides, increases in volume. 5. After a period of 10 days or more the gum ducts become embedded in the xylem, and the activity of the epithelial cells ceases. The cell walls of many of them break, and the gum still present in the cells is released.  相似文献

The polysaccharide isolated from the gum exudate of palm Scheelea phalerata (SPN) was water-insoluble and composed of Fuc, Ara, Xyl, and uronic acid moieties in a 5:34:54:7 molar ratio: 12% of phenolics were also present. A soluble polysaccharide (SPNa) was obtained after alkaline treatment, which contained Fuc, Ara, Xyl and uronic acid in a 7:44:42:7 molar ratio, with only 2% phenolics. SPNa had an M(W) approximately 1.04 x 10(5) g mol(-1) and was almost monodisperse (M(W)/M(N) : 1.25 +/-0.22). It had a branched structure with side chains of 2-O-substituted Xylp (approximately 8%) and 3-O-substituted Araf (12%) units, and a large proportion of nonreducing end-units of Araf (15%), Fucp (10%), Xylp (4%), and Arap (6%). The (1 --> 4)-linked beta-Xylp main-chain units were 3-O- (9%), 2-O- (13%), and 2,3-di-O- (13%) substituted. Its (13)C NMR spectrum contained at least 9 C-1 signals, those at delta 108.6 and 107.7 arising from alpha-Araf units. Others were present at delta 175.4 from C-6 of alpha-GlcpA and delta 15.6 from C-6 of Fucp units. The main chain of SPNa was confirmed by analysis of a Smith-degraded polysaccharide (SPDS): methylation analysis provided a 2,3-Me(2)-Xyl (65%) derivative and its (13)C NMR spectrum showed five main signals typical of a (1 --> 4)-linked beta-Xylp units. Methylation analysis of a carboxy-reduced polysaccharide (SPN-CR) revealed a 2,3,4,6-Me(4)-Glc derivative (4%) arising from nonreducing end-units of GlcpA. Alpha-GlcpA-(1 --> 2)-alphabeta-Xy1p and alpha-GlcpA-(1 --> 2)-beta-Xylp-(1 --> 4)-alphabeta-Xylp were obtained via partial acid hydrolysis of SPN, showing the structure of side-chain substituents on O-2 of the main-chain units.  相似文献

Acid phosphatase activity in phosphorus-deficient white lupin roots   总被引:15,自引:0,他引:15  
White lupin ( Lupinus albus L.) develops proteoid roots when grown in phosphorus (P)-deficient conditions. These short, lateral, densely clustered roots are adapted to increase P availability. Previous studies from our laboratory have shown proteoid roots have higher rates of non-photosynthetic carbon fixation than normal roots and altered metabolism to support organic acid exudation, which serves to solubilize P in the rhizosphere. The present work indicates that proteoid roots possess additional adaptations for increasing P availability and possibly for conserving P in the plant. Roots from P-deficient (–P) plants had significantly greater acid phosphatase activity in both root extracts and root exudates than comparable samples from P-sufficient (+P) plants beginning 10 d after emergence. The increase in activity in –P plants was most pronounced in the proteoid regions. In contrast, no induction of phytase activity was found in –P plants compared to +P plants. The number of proteoid roots present was not affected by the source of phosphorus supplied, whether organic or inorganic forms. Adding molybdate to the roots increased the number of proteoid roots in plants supplied with organic P, but not inorganic P. Increased acid phosphatase activity was detected in root exudates in the presence of organic P sources. Native-polyacrylamide gel electrophoresis demonstrated that under P-deficient conditions, a unique isoform of acid phosphatase was induced between 10 and 12 d after emergence. This isoform was found not only within the root, but it comprised the major form exuded from proteoid roots of –P plants. The fact that exudation of proteoid-root-specific acid phosphatase coincides with proteoid root development and increased exudation of organic acids indicates that white lupin has several coordinated adaptive strategies to P-deficient conditions.  相似文献

Batch studies were conducted with Mn oxides (birnessite-hausmannite mixture, BHM) and samples of four soil series from the Mid-Atlantic region of the USA to determine effects of reducing organic acids, similar to those found in the rhizosphere, on the SeO3/SeO4 distribution. Jackland (Typic Hapludalf), Myersville (Ultic Hapludalf), Christiana (Aeric Paleaquult), and Evesboro (Typic Quartizipsamment) A and B horizon soil samples with and without prior Mn oxide reduction were incubated aerobically for 10 d with 0.1 mmol kg-1 SeO3 and 0 or 25 mmol kg-1 of ascorbic acid, gallic acid, oxalic acid, or citric acid. Selenite was also added to BHM (10 mmol kg-1) with 0 or 0.1 mmol kg-1 ascorbic acid. The availability of Se for plant uptake as a result of root-soil interactions was examined using growth chamber studies with barley (Hordeum vulgare L.) and wheat (Triticum aestivum L.) seedlings grown in 150-mL cone-shaped containers to maximize root-soil surface interactions and to create rhizosphere soil throughout the root zone. In the BHM system ascorbic acid increased oxidation of SeO3 to SeO4 to 33% of added SeO3. In the presence of ascorbic and gallic acids and Mn oxides, oxidation of SeO3 to SeO4 occurred in the B horizons of all the soils and in the A horizons of Jackland and Myersville soils. Removal of Mn oxides decreased the oxidation in some samples. Wheat and barley plants were able to accumulate up to 20 mol Se kg-1 from the Jackland soil when soluble Se was not measurable. The root-soil interactions in the Jackland soil with barley and wheat provided the plant with Se from insoluble sources. The results also indicate that Mn oxides coming in contact with reducing root exudates have a greater ability to oxidize SeO3 to SeO4. This study was conducted to determine if plants can assimilate indole-3-acetic acid (IAA) from rooting media and if exogenous L-tryptophan (L-TRP) can be assimilated and converted by plants into auxins. The addition of 2-14C-IAA (3.7 kBq plant-1) to wheat (Triticum aestivum L.) seedlings of three varieties grown in nutrient solution resulted in the uptake (avg.=7.6%) of labelled IAA. Most of the label IAA was recovered in the shoot (avg.=7.2%) with little accumulation in the root (avg.=0.43%). A portion of the assimilated IAA-label in the plant was identified by co-chromatography and UV spectral confirmation as IAA-glycine and IAA-aspartic acid conjugates. Little of the assimilated IAA label was found as free IAA in the wheat plants. These same assimilation patterns were observed when 2-14C-IAA was added to wheat plants grown in sterile and nonsterile soil. In contrast, the wheat varieties assimilated considerably less (avg.=1.3%) of the added microbial IAA precursor, 3-14C-L-TRP (3.7 kBq plant-1) and thus much lower amounts of IAA conjugates were detected. Glasshouse soil experiments revealed that 2 out of 3 wheat varieties had increased growth rates and increased yields when L-TRP (10-5 and 10-7 M) was added to the root zone. It is surmised that this positive response is a result of microbial auxin production within the rhizosphere upon the addition of the precursor, L-TRP. The amino acid composition of the root exudates plays a critical role in microbial production of auxins in the rhizosphere. This study showed that wheat roots can assimilate IAA from their rooting media, which will supplement the endogenous IAA levels in the shoot tissue and may positively influence plant growth and subsequent yield.  相似文献

Proteoid roots are bottlebrush-like clusters of rootlets which form along lateral roots. They are characteristic of most species of the Proteaceae, which are mainly distributed in Australia and South Africa. Homologous root clusters are present in species of the Casuarinaceae, Mimosaceae, Fabaceae, Myricaceae and Moraceae. Many similarities exist between these species in relation to morphology and function of root clusters. Many are non-mycorrhizal and are highly efficient in phosphorus (P) acquisition. In these species, proteoid roots and proteoid-like root clusters are abundant when grown on infertile soils. Their formation is predominantly affected by the P status of the plants, being induced at low P levels and repressed at high P levels. Proteoid roots and proteoid-like root clusters play an important role in acquisition of P and other mineral nutrients. Although increase in root surface area may be a contributing factor, in many species these roots excrete large amounts of organic acids and phenolics. The excretion of these compounds in a small soil volume gives rise to extensive nutrient mobilization by acidification, reduction and chelation of sparingly soluble forms of P and micronutrients such as Fe and Mn.  相似文献

A series of pot and laboratory experiments was carried out to assess the effects of N status of sorghum roots and timing of N application (as NH4NO3) on the germination of Striga hermonthica seeds. Root N concentrations varied from 10 to 26 mg N g?1. The cut root and the root exudates technique used in assaying S. hermonthica seed germination gave similar results. However, the cut root technique was easier to handle and more discriminating at low germination levels. S. hermonthica seed germination per unit sorghum root mass followed a broken‐stick model. It decreased with increasing root N concentrations, reaching lowest levels at a root N concentration of 19.5 mg N g?1, after which no further reduction occurred. It was not possible to reduce S. hermonthica seed germination to a zero level. Timing of N application influenced the time a higher N concentration is reached, not the S. hermonthica seed germination. Both timing and rate of N application are important in maintaining root N concentrations above 19.5 mg N g?1, thereby potentially reducing S. hermonthica germination in the field. Translation of results to reductions in infection levels and yield losses is hampered by density‐dependent relations after the S. hermonthica germination stage.  相似文献
