Oncogenes,protein tyrosine kinases,and signal transduction   总被引:1,自引:0,他引:1  
Many oncogenes encode protein tyrosine kinases (PTKs). Oncogenic mutations of these genes invariably result in constitutive activation of these PTKs. Autophosphorylation of the PTKs and tyrosine phosphorylation of their cellular substrates are essential events for transmission of the mitogenic signal into cells. The recent discovery of the characteristic amino acid sequences, of thesrc homology domains 2 and 3 (SH2 and SH3), and extensive studies on proteins containing the SH2 and SH3 domains have revealed that protein tyrosine-phosphorylation of PTKs provides phosphotyrosine sites for SH2 binding and allows extracellular signals to be relayed into the nucleus through a chain of protein-protein interactions mediated by the SH2 and SH3 domains. Studies on oncogenes, PTKs and SH2/SH3-containing proteins have made a tremendous contribution to our understanding of the mechanisms for the control of cell growth, oncogenesis, and signal transduction. This review is intended to provide an outline of the most recent progress in the study of signal transduction by PTKs.  相似文献

透明颤菌血红蛋白基因vgb在多种研究和工业发酵菌中异源表达很好的解决了高密度发酵中的溶氧率问题。酿酒酵母是经典的真核模型,且在发酵工业中具有重要的应用价值,但vgb在酿酒酵母中异源表达对细胞生长的影响并不清楚。以ADH1为启动子构建了含透明颤菌(Vistreoscilla)血红蛋白基因vgb的异源表达质粒YEplac195-ADH1pr-vgb,并转化至酿酒酵母BY4741。通过生长敏感性实验,发现在发酵碳源和非发酵碳源中,vgb的异源表达均抑制了菌株生长。接着,通过2',7'-二氯荧光黄双乙酸盐和PI染色和脂质过氧化产物检测分析,发现过表达vgb的酿酒酵母细胞中活性氧(ROS)的积累、细胞膜通透性改变以及脂质过氧化。结果表明,酿酒酵母中过表达vgb改变细胞的氧化状态促进活性氧的累积,氧化应激导致菌株的生长抑制。  相似文献

传统的土壤水分模拟研究难以从土壤水分变化的时空双向出发表达其连续演变的过程,存在时空尺度效应问题。借助SWAT模型模拟的长时间序列优势,结合高分辨率卫星影像和遥感技术,力图在时空尺度效应问题上取得突破。并利用长时间序列的模拟结果分析流域土壤水分的空间格局和不同维度时空异质性。结果表明:(1)2008-2014年间艾比湖流域土壤水分主要受气温、降水及人类活动影响,呈波动变化,总体偏低且具有逐年减小趋势。(2)受降水、地形及土地覆被影响,土壤水分分布呈现出由山区向两侧平原减少的特点,且林地 > 农用地 > 草地 > 稀疏植被。(3)近10年间土壤水分低值区由原来的北部山区及平原向东部、东南部平原区及南部山区迁移,东部减少最为明显。(4)流域四季土壤水分变化差异显著。其中,春季主要受融雪影响;夏季、秋季主要受降雨量和气温影响;冬季主要受固态降雪和气温影响;且不同年份、相同季节、相同子流域土壤水分变化趋势表现一致。  相似文献

A shift in the growth temperature of Streptococcus faecalis from 37 to 10°C resulted in an 18% increase in the proportion of unsaturated fatty acids. Electron spin resonance spectra of spin-labeled membranes and extracted phospholipids indicated viscosity changes consistent with the alterations in fatty acid composition. Growth temperature had no significant effect on the active transport of leucine and alanine; uptake rates assayed at 10 or 35°C were essentially the same in cells grown at either 10 or 37°C. The effects of enhanced UV-B radiation on three examples of European shrub-dominated vegetation were studied in situ. The experiments were in High Arctic Greenland, northern Sweden and Greece, and at all sites investigated the interaction of enhanced UV-B radiation (simulating a 15% reduction in the ozone layer) with artificially increased precipitation. The Swedish experiment also involved a study of the interaction between enhanced UV-B radiation and elevated CO2 (600 ppm). These field studies were supported by an outdoor controlled environment study in the United Kingdom involving modulated enhancement of UV-B radiation in combination with elevated CO2 (700 ppm). Effects of the treatments on plant growth, morphology, phenology and physiology were measured. The effects observed were species specific, and included both positive and negative responses to the treatments. In general the negative responses to UV-B treatments of up to three growing seasons were small, but included reductions in shoot growth and premature leaf senescence. Positive responses included a marked increase in flowering in some species and a stimulation of some photosynthetic processes. UV-B treatment enhanced the drought tolerance of Pinus pinea and Pinus halepensis by increasing leaf cuticle thickness. In general, there were few interactions between the elevated CO2 and enhanced UV-B treatments. There was evidence to suggest that although the negative responses to the treatments were small, damage may be increasing with time in some long-lived woody perennials. There was also evidence in the third year of treatments for effects of UV-B on insect herbivory in Vaccinium species. The experiments point to the necessity for long-term field investigations to predict the likely ecological consequences of increasing UV-B radiation.  相似文献

Elevated atmospheric CO2 is known to stimulate photosynthesis and growth of plants with the C3 pathway but less of plants with the C4 pathway. An increase in the CO2 concentration can therefore be expected to change the competitive interactions between C3 and C4 species. The effect of long term exposure to elevated CO2 (ambient CO2 concentration +340 µmol CO2 mol-1) on a salt marsh vegetation with both C3 and C4 species was investigated. Elevated CO2 increased the biomass of the C3 sedgeScirpus olneyi growing in a pure stand, while the biomass of the C4 grassSpartina patens in a monospecific community was not affected. In the mixed C3/C4 community the C3 sedge showed a very large relative increase in biomass in elevated CO2 while the biomass of the C4 species declined.The C4 grassSpartina patens dominated the higher areas of the salt marsh, while the C3 sedgeScirpus olneyi was most abundant at the lower elevations, and the mixed community occupied intermediate elevations.Scirpus growth may have been restricted by drought and salt stress at the higher elevations, whileSpartina growth at the lower elevations may be affected by the higher frequency of flooding. Elevated CO2 may affect the species distribution in the salt marsh if it allowsScirpus to grow at higher elevations where it in turn may affect the growth ofSpartina.  相似文献

Abstract: Casein kinase II (CKII) is a protein kinase acting in the intracellular cascade of reactions activated by growth factor receptors, and that has a profound influence on cell proliferation and survival. In this investigation, we studied the changes in the activity and levels of CKII in the rat brain exposed to 10. 15 and 20 min of transient forebrain ischemia followed by variable periods of reperfusion. The cytosolic CKII activity decreased during reperfusion by ∼ 30 and ∼ 50% in the selectively vulnerable areas, striatum and the CA1 region of the hippocampus, respectively. In the resistant CA3 region of hippocampus and neocortex, the activity increased by ∼ 20 and ∼ 60%, respectively. The postischemic changes in CKII activity were dependent on the duration of the ischemic insult. The levels of CKII did not change after ischemia, suggesting that the enzyme is modulated by covalent modification or is interacting with an endogenous inhibitor/activator. Treatment of the cytosolic fraction from cortex of rats exposed to ischemia and 1 h of reperfusion with agarose-bound phosphatase decreased the activity of CKII to control levels, suggesting that CKII activation after ischemia involves a phosphorylation of the enzyme. The correlation between postischemic CKII activity and neuronal survival implies that preservation or activation of CKII activity may be important for neuronal survival after cerebral ischemia.  相似文献

Summary The aim of this study was to assess the effects of developmental events, occurring in fish during the first weeks after hatching, on the quantity and quality of the ingested food and on growth. The investigation was carried out with the larvae and juveniles of Rutilus rutilus, the single cyprinid species occurring in an oligotrophic subalpine lake in Tirol, Austria. Comparison between availability of prey in the water and gut contents suggests that the selection of food by the young fish is strongly influenced by developmental processes. For example, the prevalence of indigestible phytoplankton in the gut of young larvae can be taken as a sign of the not yet fully developed sensory and locomotory capacities of the young fish (El-Fiky et al. 1987). Furthermore, quantitative and qualitative changes in the gut contents correlate strongly with changes in the form and relative length of the gut, but reflect only weakly the availability of prey in the water. In the Seefelder See population of R. rutilus the switch from a phytoplankton to a cladoceran dominated diet is accompanied by an increase in relative growth rate by nearly one order of magnitude (Wieser et al. 1988).  相似文献

The biomass concentration extant in potassiumlimited cultures of either Klebsiella pneumoniae or Bacillus stearothermophilus (when growing at a fixed temperature and dilution rate in a glucose/ammonium salts medium) increased progressively as the medium pH value was raised step-wise from 7.0 to 8.5. Because the macromolecular composition of the organisms did not vary significantly, this increase in biomass could not be attributed to an accumulation of storage-type polymers but appeared to reflect a pH-dependent decrease in the cells' minimum K+ requirement. Significantly, this effect of pH was not eviden with cultures in which no ammonium salts were present and in which either glutamate or nitrate was added as the sole nitrogen source; however, it was again manifest when various concentrations of NH4Cl were added to the glutamate-containing medium. This suggested a functional replacement of K+ by NH 4 + , a proposition consistent with the close similarity of the ionic radii of the potassium ion (1.33 Å) and the ammonium ion (1.43 Å). At pH 8.0, and with a medium containing both glutamate (30 mM) and NH4Cl (100 mM), cultures of B. stearothermophilus would grow without added potassium at a maximum rate of 0.7 h-1. Under these conditions the cells contained maximally 0.1% (w/w) potassium (derived from contaminating amounts of this element in the medium constituents), a value which should be compared with one of 1.4% (w/w) for cells growing in a potassiumlimited medium containing initially 0.5 mM K+. Qualitatively similar findings were made with cultures of K. pneumoniae; and whereas one may not conclude that NH 4 + can totally replace K+ in the growth of these bacteria, it can clearly do so very extensively.  相似文献
