A vessel was constructed for growth of photosynthetic bacteria at defined light intensity, temperature and partial pressure of oxygen.Under growth conditions at light intensities larger than 1,000 lx, the particles exposed by freeze-fracturing of thylakoids are unordered.Under growth conditions at light intensities lower than 30 lx, the particles seen are hexagonally arranged. If the oxygen partial pressure is increased from 0 to 30 mm Hg while keeping the light intensity at 30 lx, the particles seen in the thylakoids are found to be unregularly arranged.The protein pattern of thylakoids isolated from bacteria grown either at 2,000 lx or at 30 lx revealed a constant ratio of reaction centre polypeptide to either of the membrane polypeptides of 8 kdalton apparent Mr and 12 kdalton apparent Mr.Dedicated to Prof. Dr. G. A rich and varied meiofauna inhabits a Cornish mudflat near the mouth of the Tamar River in southwestern England. Population densities range from 117 to 943 individuals · g–1 (wet) sediment (1.4–11.4 × 106 individuals · m–2), with foraminifera, harpacticoid copepods and nematodes appearing in nearly equal numbers and comprising most of the meiofauna. Seasonally, meiofaunal numbers rise and fall with solar radiation and vary inversely with river discharge. Two species, the atestate allogromiid A and the calcareous Haynesina germanica (Ehrenberg), far outnumber other foraminifera; their population densities and growth rates reach maxima in spring and summer.Monthly rates of sediment respiration are locally variable, but clearly increase from winter (4.13 ml O2 · m–2 · h–1 in December) to spring (38.87 ml O2 · m–2 · h–1 in April). Experiments and calculations ascribe approximately 30% of this total to the meiofauna (including microfauna and microflora), 50% to bacteria and less than 20% to chemical oxidation. A tentative energy budget for the mudflat suggests that secondary production by meiofauna is small as compared with coastal environments elsewhere, and that meiofaunal production (426 Kcal · m–2 · y–1) is nearly twice meiofaunal respiration (252 Kcal · m–2 · yr–1).  相似文献

The present view on the regulation of respiration and ATP synthesis in higher organisms implies only Michaelis-Menten type kinetics and respiratory control as regulatory principles. Recent experimental observations, suggesting further regulatory mechanisms at respiratory chain complexes, are reviewed. A new hypothesis is presented implying regulation of respiration and ATP synthesis in higher organisms mainly via allosteric modification of respiratory chain complexes, in particular of cytochromec oxidase. The allosteric effectors, e.g., metabolites, cofactors, ions, hormones, and the membrane potential are suggested to change the activity and the coupling degree of cytochromec oxidase by binding to specific sites at nuclear coded subunits. Recent results on the structure and activity of cytochromec oxidase, supporting the hypothesis, are reviewed.Dedicated to Professor Dr. Carl Martius on the occasion of his 80th birthday.  相似文献

Summary A mast-cell activator, compound 48/80, causes proliferation of mesenchymal cells in the mesentery of rats. Its effect on W/W vmice deficient in mast cells was tested to determine whether the proliferation is mediated in the degranulation of mast cells. Incorporation of [3H]thymidine into mesenchymal cells in the mesentery of these mice with or without compound 48/80 was very small compared to their normal litter mates. However, bone marrow transplantation markedly enhanced the effect of compound 48/80, and resulted in an incorporation of [3H]thymidine almost comparable to that observed in normal mice. Thiobacillus tepidarius, isolated from the hot springs at Bath, Avon, UK, grew optimally at 43–45°C and pH 6.0–7.5 on thiosulphate or tetrathionate. In batch culture, thiosulphate was oxidized stoichiometrically to tetrathionate, with a rise in pH. The tetrathionate was then oxidized to sulphate, supporting growth and producing a fall in pH to a minimum of ph 4.8. The organism contained high levels of thiosulphate-oxidizing enzyme, rhodanese and ribulose bisphosphate carboxylase. It was obligately chemolithotrophic and autotrophic. In chemostat culture, T. tepidarius grew autotrophically with the following sole energy-substrates: sulphide, thiosulphate, trithionate, tetrathionate, hexathionate or heptathionate. Thiocyanate, dithionate and sulphite were not used as sole substrates, although sulphite enhanced growth yields in the presence of thiosulphate. Maximum specific growth rate on tetrathionate was 0.44 h-1. True growth yields (Y max) and maintenance coefficients (m) were calculated for sulphide, thiosulphate, trithionate and tetrathionate and observed yields at a single fixed dilution rate compared with those on hexathionate and heptathionate. Mean values for Y max, determined from measurements of absorbance, dry wt, total organic carbon and cell protein, were similar for sulphide, thiosulphate and trithionate (10.9 g dry wt/mol substrate) as expected from their equivalent oxygen consumption for oxidation. Y max for tetrathionate (20.5) and the relative Y o values (as g dry wt/g atom oxygen consumed) for thiosulphate and all four polythionates indicated that substrate level phosphorylation did not contribute significantly to energy conservation. These Y max values were 40–70% higher than any of those previously reported for obligately aerobic thiobacilli. Mean values for m were 6.7 mmol substrate oxidized/g dry wt·h for sulphide, thiosulphate and trithionate, and 2.6 for tetrathionate.Abbreviation PIPES Piperazine-N,N-bis(ethane sulphonic acid)  相似文献

Summary The distribution of the vitamin-D dependent calcium-binding protein (Calbindin-D 28K) (CaBP-28K) in the tibial growth plate cartilage of the rat has been studied immunohistochemically using an antibody raised against rat renal CaBP-28K. The protein was detected mainly in the nuclei of chondrocytes and occasionally in the juxta-nuclear cytoplasm. The distribution was not uniform throughout the growth plate, but concentrated in the proliferatively active chondrocytes of the resting and proliferative zones. These findings raise the possibility that CaBP-28K may be involved in the mitotic activity of the chondrocytes, acting as a regulator of the proliferative process, perhaps via intranuclear calcium.  相似文献

向颈总动脉头端注入枸橼酸钠能使大多数家兔发生呼气性呼吸暂停和呼吸频率变慢。此呼吸抑制效应可被地卡因麻醉颈动脉窦区所消除。切断窦神经不能阻断枸橼酸钠对呼吸的抑制。切断迷走神经窦支则使半数以上家兔的呼吸抑制减弱或消失,而在结状神经节上方切断迷走神经能阻断大多数家兔的呼吸抑制。结果提示,迷走神经窦支是颈动脉窦区感受器传入通路之一,向颈动脉窦区注入枸橼酸钠对呼吸的抑制主要是通过迷走神经传入引起的。  相似文献

实验在62只家兔上进行。结果观察到,中缝大核(NRM)区562个单位中,有118个单位的自发放电频率低,放电比较规则,动作电位时程长,易被微电泳5-羟色胺所阻遏,称为A 组单位。其余444个单位的自发放电频率高,动作电位时程短,称为B 组单位。大多数 B组单位对微电泳5-羟色胺不起反应。脑桥臂旁内侧核(NPBM)区微量注射吗啡(200μg/2μl)或静脉注射吗啡(3mg/kg)后,20个A 组单位中有19个发生兴奋效应,而49个B 组单位中仅有29个发生兴奋效应,而且A组单位发生兴奋的程度也比B组单位的高。这些结果提示,NRM区的A 组单位可能是5-羟色胺神经元,吗啡对这些神经元有相对选择性的兴奋作用。 在另外11只家兔上,应用辣根过氧化物酶(HRP)逆行追踪技术观察到,NPBM 区与NRM 区有纤维联系。 本实验结果提示,静脉注射吗啡所致的呼吸抑制,可能与吗啡作用于 NPBM,通过纤维联系,引起NRM 5-羟色胺神经元兴奋有关。  相似文献

Synopsis The brown trout in Llyn Frongoch, a mature upland reservoir, and its nursery stream was sampled during 1983. The stream stock consisted largely of the 1983 and 1982 year classes, with fish reaching mean lengths of 7.0 and 11.6 cm at one and two years of age. The size and biomass of the stream stock at the beginning of 1983 and 1984 were estimated to be 120 and 125 (1.20 and 1.25 fish m–2) and 1.41 and 0.69 kg (14.1 g m–2 and 6.9 g m–2) respectively. Annual stream production ranged from an estimated minimum of 2.49 kg (24.9 g m–2) to an estimated maximum of 4.59 kg (45.9 g m–2). Both downstream and upstream movements of 0+ juveniles were recorded. The adult spawning stock was estimated at 79 males and 32 females, a sex ratio of 2.5:1, with most spawners belonging to the 1980 yearclass. The average size of the lake stock over the year was estimated to be 1 650 (229 fish ha–1) or 250.8 kg (34.8 kg ha–1). The 1980 yearclass was predominant; there were few fish older than five years. Seasonal variations in netting catches suggested movements to and from the littoral region. Growth in the lake was moderately fast, with fish reaching mean lengths of 21.7 and 27.2 cm by three and four years of age. Fish entering the lake after one year appeared to grow faster than fish which remained in the stream for two years. Annual production in the lake was estimated at 136.7 kg (19.0 kg ha–1). The total angling catch for the season was estimated to be 62.6 kg (8.7 kg ha–1).  相似文献
