Cottonwoods, riparian poplars, are facultative phreatophytes and can obtain water from shallow soil moisture originating from rainfall, or from the deeper capillary fringe above the alluvial water table that is recharged by river water infiltration. The correspondence between cottonwood growth and river flows should reveal the dependency upon alluvial groundwater and subsequently, the vulnerability to reduced river flows. To explore this association, we analyzed historic growth patterns of plains cottonwoods (Populus deltoides) along the Red Deer River (RDR), which is at the northwestern limit of the North American Great Plains. We developed chronologies of yearly radial increments (RI) and basal area increments (BAI) and explored correspondences with the environmental records from the past century. In this semi-arid region, the RI or BAI were not correlated with local precipitation while negative correlation with growth season temperature (T) (r = −0.37, p < 0.01) could reflect reduced growth with hot summers. There was correlation between growth and annual river discharge (Q, and particularly log Q that approximates river stage) and this increased with two year averaging (r = 0.51, p < 0.01), reflecting carry-over in the watershed hydrology and in the ecophysiological response. There was correspondence with the Pacific Decadal Oscillation index (PDO, r = −0.45, p < 0.01), which provides multi-decade transitions that influence Rocky Mountain headwater precipitation and other weather characteristics, and river flows. The combination of Q, PDO and T provided the strongest multiple regression model, accounting for 44% of the historic growth variation (52% correspondence for 1953–2013). The RDR was dammed in 1983, enabling winter flow augmentation, but summer flows were sustained and cottonwood growth and the streamflow correspondence persisted. This indicates that it is the pattern of dam operation and not damming per se that determines the fate of established riparian cottonwoods downstream. This study revealed that these cottonwoods are phreatophytic and dependent upon alluvial groundwater that is recharged from the river. This provides a research strategy to determine whether riparian woodlands along other regulated rivers are similarly groundwater-dependent and could be vulnerable to river flow reductions from excessive water withdrawal for irrigation or other uses, or with climate change. 相似文献

Clostridium tetani produce tetanospasmin, a potent exotoxin; that causes tetanus or lockjaw disease. Scientists developed an anti-tetanus toxoid to protect the body from the spasm's neurotoxic effect. In Pakistan recently, 478 cases of neonatal tetanus were reported. The study was carried out at The National Control Laboratory for Biologicals Islamabad, aiming to decipher the effectiveness of the most frequently used tetanus toxoid vaccine adsorbed in Pakistan in comparison to standard reference vaccine having earlier known consistent values. The vaccines included domestic public sector, domestic private sector, imported private sector I, and imported private sector II. The triplicate experiments on purebred Swiss albino mice were performed by immunizing with Tetanus toxoid and then tested parallel with standard reference vaccine. Various analytical tests were performed on the test organism that included flocculation test/identity test, antibody quantification using enzyme-linked immunosorbent assay (ELISA), potency test, abnormal toxicity test, osmolality, pH test, liquid sub-visible particle test, and sterility test. Results of all the vaccines were compared in comparison with the standard reference vaccine. Absorbances of test vaccines were recorded at the lowest dilution by ELISA. The domestic private sector, imported private sector I, imported private sector II and standard reference vaccine were flocculated at mean dilution (Mean: 0.24, 95% CI: 0.1903–0.2897), and the domestic public sector was flocculated at mean dilution (Mean: 0.23, 95% CI: 0.2052–0.2548). All the products were found within the normal ranges where it was concluded that the maximum average titer of 2.81 was observed at dilution 10?1.6, indicating that these vaccines were adequate/suitable for the prevention of tetanus. 相似文献

A fundamental property of tumor cells is to defy anoikis, cell death caused by a lack of cell-matrix interaction, and grow in an anchorage-independent manner. How tumor cells organize signaling molecules at the plasma membrane to sustain oncogenic signals in the absence of cell-matrix interactions remains poorly understood. Here, we describe a role for phosphatidylinositol 4-phosphate 5-kinase (PIPK) Iγi2 in controlling anchorage-independent growth of tumor cells in coordination with the proto-oncogene Src. PIPKIγi2 regulated Src activation downstream of growth factor receptors and integrins. PIPKIγi2 directly interacted with the C-terminal tail of Src and regulated its subcellular localization in concert with talin, a cytoskeletal protein targeted to focal adhesions. Co-expression of PIPKIγi2 and Src synergistically induced the anchorage-independent growth of nonmalignant cells. This study uncovers a novel mechanism where a phosphoinositide-synthesizing enzyme, PIPKIγi2, functions with the proto-oncogene Src, to regulate oncogenic signaling. 相似文献

Effects of different N/P ratios on several root parameters and on net P uptake were studied in winter wheat, Triticum aestivum cv. Starke II, grown in water culture. In the First experiment N/P ratios of (0/4, 2/3, 4/2, 6/1 and 8/0) were used, and plants were harvested at age 3, 5, 8, 11 and 14 days. In the second experiment N/P ratios of 6/1, 10/1, 15/1, 17/1, 20/1 and 25/1 were applied at two different N,P levels. Root length and number were determined using a digitizer connected to a computer. In the first experiment. the 6/1 N/P ratio gave the largest plants at day 14, and growth decreased with decreasing N/P ratio, The same pattern was found fur lateral root length and root number (seminal and lateral). In the second experiment the root weights decreased with increasing N/P ratio within each level. Lateral root number and overall length decreased with increasing N/P ratio at both levels as did the average lateral root length at the high N,P level. At the low N.P level, average lateral root length was about the same at all N/P ratios. Increasing the N/P ratio increased net uptake of P at the low N,P level, but decreased net P uptake at the high N,P level. Net P uptake increased with increasing P concentration in the roots and then decreased with further increase in P concentration. Net P uptake based on calculated root length [m (g root)−1 ] showed no significant deviation from weight-based uptake plots. The effect of N and P on root structure is discussed as well as the interaction of N and P in P uptake. The relevance of a proper basis for expressing root activity is stressed. 相似文献

We have found that a murine hepatoma displays a considerable phenotypic diversification in culture, which depends upon the substratum utilized, and is manifested by the formation of multicellular structures of differing geometry: Monolayer on glass and plastic, thick multilayer pads on Gelfilm, and spheroids on agar and agarose. These multicellular morphological phenotypes were assayed without disruption to ascertain their antigenicity in vitro and their tumorigenicity in vivo and to obtain quantitative information on the effect of the spatial arrangement of the hepatoma cells upon the ability of each multicellular structure to interact, as a whole, with molecules and cells in its surroundings. The antigenicity of the multicellular structures was determined with calibrated probes and a methodology that measures the total antigenicity, as well as antigenicity per unit of surface area. Antigenicity was found to differ in the following decreasing order: Monolayer on plastic > spheroids on agarose > spheroids on agar > multilayer on Gelfilm. At least part of these antigenic variants arise from different degrees of masking of the structures' surface determinants by a trypsin-sensitive material. The multicellular phenotypes also differed in tumorigenicity. When assayed in syngeneic hosts under comparable conditions, agar-grown spheroids produced the fewest tumors, whereas Gelfilm-grown multilayers produced the most. These two independent sets of data show that the various geometries that a tumor tissue is induced to acquire by the culture substratum are accompanied by a distinctive combination of surface and biological properties. 相似文献

The submerged vegetation of Lake Kariba is described in relation to degree of slope (lake morphometry), depth and light transparency. The direct gradient analysis technique — canonical correspondence analysis and the TWINSPAN classification programs were used to analyse the data set. The western end of the lake with low transparency has a low species diversity (with Vallisneria aethiopica dominating). Species diversity increases with increased transparency in the other parts of the lake. The classification revealed monospecific communities for all species as well as mixed communities with Lagarosiphon as the associate species with the broadest distribution. The ordination revealed a first axis strongly related to depth and transparency gradients and the second axis related to slope. Vallisneria aethiopica has a growth form adapted to grow in shallow areas subjected to wave action. Potamogeton octandrus also growing in shallow areas is restricted to sheltered areas. Lagarosiphon ilicifolius precluded from the shallow exposed areas grows at medium depth but has a growth form adapted to tap light at the surface. Lactobacillus gasseri ATCC33323T has seven putative phospho-β-glycosidase genes. Using column chromatography, we found that this strain cultured in lactose medium expresses five phospho-β-glycosidases (LacG1, LacG2, Pbg1, Pbg2, and Pbg3), where these gene expressions can be suppressed by glucose. Abstract Mesophilic Aeromonas spp. strains (serotype O:34) showed sensitivity to complement-mediated killing when they were cultivated at 37°C (serum-sensitive) but not when they were cultivated at 20°C (serum-resistant). These strains produced smooth lipopolysaccharide when they were grown at 20°C and rough lipolysaccharide when cultivated at 37°C. The reason for the resistance to complement-mediated killing could be that C3b is rapidly degraded (possibly because it is bound far from the cell membrane), consequently the lytic complex (C5b-9) is not formed. 相似文献

Three groups of children, those of European parentage, those of Guatemalan parentage, and those of mixed European-Guatemalan parentage were measured for height, weight, and skeletal maturity. The children were born between 1945 and 1965, they were all of high socioeconomic status, and they all attended the same private school in Guatemala City. At 7 years, the boys of the European group were significantly taller than boys of the Guatemalan group. European and mixed European-Guatemalan girls were significantly taller than Guatemalan girls. These results are maturity independent. The influence of skeletal age was removed statistically by analysis of covariance. Girls of the mixed group were significantly heavier than girls of European and Guatemalan groups. Mixed group girls also had more significantly advanced skeletal ages than European girls. When the patterns of size and maturity status are analyzed by sex, there is evidence for a relatively greater environmental influence on the boys and a relatively greater genetic influence on the girls. Dividing the data into two birth year cohorts, 1945 to 1955, and 1956 to 1965, does not provide evidence for secular trends in growth or maturation. These results are similar to those from studies in developed nations that report an end to the secular trend for the “well off” population of those countries. 相似文献
