DNA polymerase X (pol X) from African swine fever virus (ASFV) is the smallest naturally ocurring DNA-directed DNA polymerase (174 amino acid residues) described so far. Previous biochemical analysis has shown that ASFV pol X is a highly distributive, monomeric enzyme, lacking a proofreading 3'-5' exonuclease. Also, ASFV pol X binds intermediates of the single-nucleotide base excision repair (BER) process, and is able to efficiently repair single-nucleotide gapped DNA. In this work, we perform an extensive kinetic analysis of single correct and incorrect nucleotide insertions by ASFV pol X using different DNA substrates: (i) a primer/template DNA; (ii) a 1nt gapped DNA; (iii) a 5'-phosphorylated 1nt gapped DNA. The results obtained indicate that ASFV pol X exhibits a general preference for insertion of purine deoxynucleotides, especially dGTP opposite template C. Moreover, ASFV pol X shows higher catalytic efficiencies when filling in gapped substrates, which are increased when a phosphate group is present at the 5'-margin of the gap. Interestingly, ASFV pol X misinserts nucleotides with frequencies from 10(-4) to 10(-5), and the insertion fidelity varies depending on the substrate, being more faithful on a phosphorylated 1nt gapped substrate. We have analyzed the capacity of ASFV pol X to act on intermediates of BER repair. Although no lyase activity could be detected on preincised 5'-deoxyribose phosphate termini, ASFV pol X has lyase activity on unincised abasic sites. Altogether, the results support a role for ASFV pol X in reparative BER of damaged viral DNA during ASFV infection.  相似文献

African swine fever (ASF) has recently made its appearance in Madagascar. Ticks of the Ornithodoros moubata group, considered to be O. porcinus Walton, 1962 were formerly known to occur in western Madagascar, but seem to have disappeared from that region. However, three new sites where they occur were found in the humid and cool central highlands of Anatananarivo province. These ticks are known to be efficient reservoirs and vectors of ASF and constitute a considerable complication to the control of the disease. The authors also discuss another potentially complicating factor, the presence of a species of African bushpig, Potamochoerus larvatus.  相似文献

Trends in phenological phases in Europe between 1951 and 1996   总被引:15,自引:0,他引:15  
Increases in air temperature due to the anthropogenic greenhouse effect can be detected easily in the phenological data of Europe within the last four decades because spring phenological events are particularly sensitive to temperature. Our new analysis of observational data from the International Phenological Gardens in Europe for the 1959–1996 period revealed that spring events, such as leaf unfolding, have advanced on average by 6.3 days (–0.21 day/year), whereas autumn events, such as leaf colouring, have been delayed on average by 4.5 days (+0.15 day/year). Thus, the average annual growing season has lengthened on average by 10.8 days since the early 1960s. For autumn events, differences between mean trends of species could not be detected, but for spring events there were differences between species, with the higher trends for leaf unfolding and flowering of shrubs indicating that changes in events occurring in the early spring are more distinct. These observed trends in plant phenological events in the International Phenological Gardens and results of other phenological studies in Europe, summarised in this study, are consistent with AVHRR satellite measurements of the normalized difference vegetation index from 1981 to 1991 and with an analysis of long-term measurements of the annual cycle of CO2 concentration in Hawaii and Alaska, also indicating a global lengthening of the growing season. 为构建具有凝集性、免疫反应性的双功能融合蛋白,本研究采用重叠延伸PCR方法将2E8ScFv(抗人红细胞H抗原单链抗体基因)和mE2(猪瘟病毒E2蛋白主要抗原编码区基因)拼接成融合基因2E8mE2,并插入原核表达载体pET-DsbA,将重组表达质粒pET-DsbA-2E8mE2转化入大肠杆菌Escherichia coli BL21(DE3)PlysS中进行IPTG诱导表达,表达的融合蛋白经SDS-PAGE和Western blotting分析鉴定,结果表明:2E8mE2融合基因在大肠杆菌中获得了表达,表达产物以包涵体形式存在,分子量约为65kDa,与预期的大小一致。分别采用亲和层析法和谷胱甘肽再氧化法对融合蛋白进行纯化和复性,红细胞凝集试验证实:2E8mE2融合蛋白复性效果良好,既能够与人红细胞结合,又能够与猪瘟病毒抗体反应,具有双功能特性。  相似文献

Endothelial cells are the main target of classical swine fever virus during infection, and extensive hemorrhage is the most typical clinical sign of classical swine fever. To investigate the molecular mechanism of hemorrhagic pathogenesis, two-dimensional difference gel electrophoresis with fluorescent dyes (2D-DIGE) was used to analyze the proteomic profile of primary porcine umbilical vein endothelial cells (PUVECs) following CSFV infection. Of 15 protein spots with differential expression, 8 were characterized by MALDI-TOF-MS/MS in infected PUVECs at 48 h p.i.: moesin, peroxiredoxin 6, stathmin-1, a protein similar to nascent polypeptide-associated complex alpha subunit isoform 2, phosphoglycerate kinase 1, glucosidase II, transketolase and α-tubulin. These could be sorted into 5 functional groups: glycometabolism, cell proliferation, anti-oxidative stress, inflammatory response and cytoskeleton. Western blot and real-time RT-PCR analysis confirmed the down-regulation of phosphoglycerate kinase 1 (PGK1) and up-regulation of moesin identified by 2D-DIGE. Pathway analysis of these 15 differentially expressed proteins showed that CSFV infection altered the metabolism, cytoskeleton and cell proliferation of PUVECs, and that consequently an inflammatory response was induced.  相似文献

Classical swine fever virus (CSFV) outer surface E2 glycoprotein represents an important target to induce protective immunization during infection but the influence of N-glycosylation pattern in antigenicity is yet unclear. In the present work, the N-glycosylation of the E2-CSFV extracellular domain expressed in goat milk was determined. Enzymatic N-glycans releasing, 2-aminobenzamide (2AB) labeling, weak anion-exchange and normal-phase HPLC combined with exoglycosidase digestions and mass spectrometry of 2AB-labeled and unlabeled N-glycans showed a heterogenic population of oligomannoside, hybrid and complex-type structures. The detection of two Man8GlcNAc2 isomers indicates an alternative active pathway in addition to the classical endoplasmic reticulum processing. N-acetyl or N-glycolyl monosialylated species predominate over neutral complex-type N-glycans. Asn207 site-specific micro-heterogeneity of the E2 most relevant antigenic and virulence site was determined by HPLC-mass spectrometry of glycopeptides. The differences in N-glycosylation with respect to the native E2 may not disturb the main antigenic domains when expressed in goat milk.  相似文献

非洲猪瘟病毒微滴数字PCR (ddPCR)方法的建立及应用   总被引:3,自引:1,他引:3  
【目的】非洲猪瘟病毒(African swine fever virus,ASFV)能导致猪群高死亡率,从而造成严重的经济损失。因此,建立严密、高效的防控系统,包括灵敏、准确的诊断方法以及高效的预报预警机制等,以避免ASF传入我国。本文旨在建立一种新型而高灵敏度的ASFV微滴数字PCR(Droplet digital PCR,dd PCR)检测方法。【方法】针对ASFV K205R基因设计一对特异性引物和Taq Man探针,通过反应条件优化,建立了ASFV实时荧光定量PCR(q PCR)和dd PCR检测方法;并对两种方法的线性关系、灵敏性、特异性和重复性进行了评估,利用建立的ASFV检测方法对163份国内和进境的组织样本及血清样本进行检测,血清样本经商品化ASFV ELISA试剂盒复检,评估不同方法检测结果的符合率。【结果】建立的ASFV q PCR和dd PCR检测方法线性关系较好(R2≥0.998),dd PCR的最低检测限度可达到0.36拷贝,在20μL反应体系中约为10拷贝/反应,检测灵敏度是q PCR的10倍。ASFV dd PCR检测方法具有很高的特异性和重复性,不与其他猪常见病毒发生交叉反应。【结论】该方法灵敏度高、特异性强,可作为一种有效的分子生物学方法来诊断ASFV,为防止该病传入我国以及ASFV的实时监测提供新的技术储备,同时促进dd PCR技术在我国的发展和应用。  相似文献

African swine fever (ASF) is a highly contagious and acute hemorrhagic viral disease with a high mortality approaching 100% in domestic pigs. ASF is an endemic in countries in sub-Saharan Africa. Now, it has been spreading to many countries, especially in Asia and Europe. Due to the fact that there is no commercial vaccine available for ASF to provide sustainable prevention, the disease has spread rapidly worldwide and caused great economic losses in swine industry. The knowledge gap of ASF virus (ASFV) pathogenesis and immune evasion is the main factor to limit the development of safe and effective ASF vaccines. Here, we will summarize the molecular mechanisms of how ASFV interferes with the host innate and adaptive immune responses. An in-depth understanding of ASFV immune evasion strategies will provide us with rational design of ASF vaccines.  相似文献

猪瘟是由猪瘟病毒引起猪的一种急性、热性和高度接触性传染病.该病呈世界性分布,给世界养猪业造成了巨大的经济损失.目前,疫苗接种仍然是防控猪瘟的主要手段.虽然传统的猪瘟弱毒疫苗(如C株)安全有效,但猪瘟的临床表现发生了很大变化,呈现典型猪瘟和非典型猪瘟共存、隐性感染和持续感染并现,免疫失败的现象时有报道,且不能区分野毒感染和免疫接种.因此,研制安全、高效、能区分野毒感染和疫苗免疫动物(DIVA)的新型猪瘟疫苗极为必要.文中就近年来开发的核酸疫苗、病毒活载体疫苗、基于蛋白/肽的疫苗、基因缺失疫苗、嵌合瘟病毒疫苗等新型DIVA猪瘟疫苗作一综述.  相似文献
