Impatiens glandulifera, or Himalayan balsam, is a prolific invader of riverine habitats. Introduced from the Himalayas for ornamental purposes in 1839, this annual species has naturalised across Great Britain (GB) forming dense monocultures with negative affects across whole ecosystems. In 2006 a programme exploring biocontrol as an alternative control method was initiated and to date, two strains of the rust fungus Puccinia komarovii var. glanduliferae have been released. To better understand the observed differences in susceptibility of GB Himalayan balsam stands to the two rust strains, inoculation studies were conducted using urediniospores and basidiospores. Experiments revealed large variation in the susceptibility of stands to urediniospores of the two rust strains, with some resistant to both. Furthermore, the infectivity of basidiospores was found to differ, with some stands fully susceptible to the urediniospore stage, being immune to basidiospore infection. Therefore, before further rust releases at new sites, it is necessary to ensure complete compatibility of the invasive stands with both urediniospores and basidiospores. However, for successful control across GB it is essential that plant biotypes are matched to the most virulent rust strains. This will involve additional strains from the native range to tackle those biotypes resistant to the strains currently released.  相似文献

Recent tree decline was hypothesized to be connected to root damage caused by soil acidification and increased frequency of pathogenic root colonizing fungi. The rhizoplane is constituted by the mycorrhizal sheath and a high diversity of microfungi, some of which are known to behave antagonistically against pathogens. Disturbance of the balance between pathogens and antagonists by soil acidification may endanger the health of tree roots. Liming may stabilize the interactions. The microfungal populations connected to the mycorrhizoplane of Norway spruce (Picea abies) and beech (Fagus sylvatica) were, therefore, investigated on experimental Norway spruce plots that had been treated with acidified water or were limed. Beech presented the original forest and was left untreated. Eight microfungal species known as either pathogenic or antagonistic, Trichoderma viride, T. hamatum, T. polysporum, Cylindrocarpon destructans, Sesquicillium candelabrum, Mycelium radicis atrovirens, Tolyplocladium geodes and Oidiodendron maius, were isolated from the mycorrhizoplanes and their abundance in the five different plots compared. Acidification enhanced the frequency of Mycelium radicis atrovirens and Oidiodendron maius but reduced Trichoderma viride. Liming promoted Sesquicillium candelabrum and Cylindrocarpon destructans. Detailed analysis of the population patterns indicated that changes in the frequency of a particular fungal species may not only be caused by shift of chemical soil factors but also by antagonistic interactions between the microfungi, thus reducing pathogenic attacks on rootlets.  相似文献

The antifungal activities of the known antimicrobial peptide, P-113, as well as a new type of Trp-rich peptide, Ac-KWRRWVRWI-NH2, Pac-525, and its modified peptide, d -Nal-Pac-525, were determined using the broth microdilution method in three different media. All peptides had similar activities against yeast pathogens in low-salt LYM media. However, only d -Nal-Pac-525 retained its antifungal activity in the media containing high concentrations of salt. Hence, d -Nal-Pac-525 has the potential of becoming a promising antifungal agent, especially for fungal pathogens with intrinsic resistance to fluconazole.  相似文献

Termites exploit environments that make them susceptible to infection and rapid disease transmission. Gram-negative bacteria binding proteins (GNBPs) signal the presence of microbes and in some insects directly damage fungal pathogens with β-1,3-glucanase activity. The subterranean termites Reticulitermes flavipes and Reticulitermes virginicus encounter soil entomopathogenic fungi such as Metarhizium anisopliae, which can evade host immune responses after penetrating the cuticle. An external defense that prevents invasion of fungal pathogens could be crucial in termites, allowing them to thrive under high pathogenic pressures. We investigated the role of secreted β-1,3-glucanases in Reticulitermes defenses against M. anisopliae. Our results show that these termites secrete antifungal β-1,3-glucanases on the cuticle, and the specific inhibition of GNBP associated β-1,3-glucanase activity with d-δ-gluconolactone (GDL) reduces this activity and can cause significant increases in mortality after exposure to M. anisopliae. Secreted β-1,3-glucanases appear to be essential in preventing infection by breaking down fungi externally.  相似文献

The human enteropathogen Campylobacter jejuni, like many bacteria, employs siderophores such as enterobactin for cellular uptake of ferric iron. This transport process has been shown to be essential for virulence and presents an attractive opportunity for further study of the permissiveness of this pathway to small-molecule intervention and as inspiration for the development of synthetic carriers that may effectively transport cargo into Gram-negative bacteria. In this work, we have developed a facile and robust microscale assay to measure growth recovery of C. jejuni NCTC 11168 in liquid culture as a result of ferric iron uptake. In parallel, we have established the solid-phase synthesis of catecholamide compounds modeled on enterobactin fragments. Applying these methodological developments, we show that small synthetic iron chelators of minimal dimensions provide ferric iron to C. jejuni with equal or greater efficiency than enterobactin.  相似文献

Background

We profiled the expression of circulating microRNAs (miRNAs) in mice using Illumina small RNA deep sequencing in order to identify the miRNAs that may potentially be used as biomarkers to distinguish between gram-negative and gram-positive bacterial infections.

Results

Recombinant-specific gram-negative pathogen Escherichia coli (Xen14) and gram-positive pathogen Staphylococcus aureus (Xen29) were used to induce bacterial infection in mice at a concentration of 1 × 108 bacteria/100 μL of phosphate buffered saline (PBS). Small RNA libraries generated from the serum of mice after exposure to PBS, Xen14, Xen29, and Xen14 + Xen29 via the routes of subcutaneous injection (I), cut wound (C), or under grafted skin (S) were analyzed using an Illumina HiSeq2000 Sequencer. Following exposure to gram-negative bacteria alone, no differentially expressed miRNA was found in the injection, cut, or skin graft models. Exposure to mixed bacteria induced a similar expression pattern of the circulating miRNAs to that induced by gram-positive bacterial infection. Upon gram-positive bacterial infection, 9 miRNAs (mir-193b-3p, mir-133a-1-3p, mir-133a-2-3p, mir-133a-1-5p, mir-133b-3p, mir-434-3p, mir-127-3p, mir-676-3p, mir-215-5p) showed upregulation greater than 4-fold with a p-value < 0.01. Among them, mir-193b-3p, mir-133a-1-3p, and mir-133a-2-3p presented the most common miRNA targets expressed in the mice exposed to gram-positive bacterial infection.

Conclusions

This study identified mir-193b-3p, mir-133a-1-3p, and mir-133a-2-3p as potential circulating miRNAs for gram-positive bacterial infections.

Tropospheric ozone has been proven to trigger biochemical plant responses that are similar to the ones induced by an attack of fungal pathogens, i.e. it resembles fungal elicitors. This suggests that ozone can represent a valid tool for the study of stress responses and induction of resistance to pathogens. This review provides an overview of the implications of such a phenomenon for basic and applied research. After an introduction about the environmental implications of tropospheric ozone and plant responses to biotic stresses, the biochemistry of ozone stress is analysed, pointing out its similarities with plant responses to pathogens and its possible applications.  相似文献

1. Aggregations of fine sediments are a suitable proxy for the presence and abundance of Tubifex tubifex, one of the obligate hosts in the parasitic life cycle that causes salmonid whirling disease (Myxobolus cerebralis). 2. To determine and evaluate practical approaches to predict fine sediments (<2 mm diameter) that could support Tubifex spp. aggregations, we measured habitat features in a catchment with field measures and metrics derived from digital data sets and geospatial tools at three different spatial extents (m2) within a hierarchical structure. 3. We used linear mixed models to test plausible candidate models that best explained the presence of fine sediments measured in stream surveys with metrics from several spatial extents. 4. The percent slow water habitat measured at the finest extent provided the best model to predict the likely presence of fine sediments. The most influential models to predict fine sediments using landscape metrics measured at broader extents included variables that measure the percentage land cover in conifer or agriculture, specifically, decreases in conifer cover and increases in agriculture. 5. The overall best‐fitting model of the presence of fine sediments in a stream reach combined variables measured and operating at different spatial extents. 6. Landscape features modelled within a hierarchical framework may be useful tools to evaluate and prioritise areas with fine sediments that may be at risk of infection by Myxobolus cerebralis.  相似文献

Aims:  To evaluate the effectiveness of various antimicrobial mortar formulations in inhibiting the growth of a selection of pathogens of environmental and hygienic concern.
Methods and Results:  Mortar prisms containing triclosan-incorporated fibres or different concentrations of silver copper zeolites were incubated with Escherichia coli , Listeria monocytogenes , Salmonella enterica or Staphylococcus aureus at 4 or 20°C for 24 h. From plate counting, a substantial bactericidal effect (>4 log units) could only be observed for the mortar specimens containing more than 3% zeolites on cement weight base, the effect being more pronounced at 20°C compared to 4°C. No inhibitory effect could be observed for mortar specimens containing antimicrobial fibres. Adenosinetriphosphate (ATP) measurements allowed for a rapid indication of the occurrence of antimicrobial activity.
Conclusions:  In order to obtain a bactericidal effect on mortar surfaces, concentrations of silver copper zeolites of more then 3% are required.
Significance and Impact of the Study:  To our knowledge, this is the first study in which the effectiveness of various antimicrobial mortar mixtures towards the inhibition of pathogens has been evaluated in a quantitative way. Antimicrobial concrete mixtures can be used for the improvement of the hygienic conditions in a variety of environments.  相似文献
