Lignin-carbohydrate complexes were extracted from grass cell walls by a variety of solvents. The yield of complexes was greatly enhanced if the sample was finely milled in a ball mill; dimethyl sulphoxide and N alkali extractions gave the highest yields. Hydrolysis showed that the carbohydrate fraction of the alkali-extracted complex contained mainly xylose (ca. 70%) and arabinose (ca. 20 %) whereas the dimethyl sulphoxide extracted complex contained glucose (ca. 50 %), xylose (ca. 30%), arabinose (ca. 12 %) and galactose (ca. 5 %). The UV spectrum of the dimethyl sulphoxide extracted complex showed lignin absorbance at 280 nm, but, in addition, ester bonding was also observed by the presence of a secondary absorbing region near 325 nm. This secondary absorbing region was absent from the spectrum of the alkali-extracted complexes. Fractionation of the complexes by ethanol precipitation gave a major component which appeared homogeneous by molecular sieve chromatography and had a MW of ? 150,000.  相似文献

Extended sieve element salivation (E1 waveform in the electrical penetration graph) is a characteristic activity during early sieve element punctures, particularly in resistant plants. In order to explore a chemically-mediated mechanism of resistance associated with sieve element salivation, we compared the pattern of feeding behaviour of the aphid, Sitobion fragariae (Walker), on two cultivars of the wheat Triticum aestivum L., with different concentrations of hydroxamic acids (Hx). During 24 h of electronic monitoring, aphids dedicated over 50% of the total time to phloem ingestion from the sieve elements. Total time allocated to E1 in the experiment, time to first E1 within the experiment, time allocated to E1 before a sustained phloem ingestion (E2) and the contribution of sieve element salivation to the phloem phase (E1/[E1+E2]) were significantly higher in the high-Hx cultivar. The increased salivation in plants with higher contents of Hx suggests the existence, at least in this system, of a chemically-mediated sieve element constraint.  相似文献

It has been observed that water, which is absolutely essential for enzyme activity, can induce the agglomeration of enzyme particles in organic media. Although enzyme agglomeration is significant in that it usually reduces enzyme activity and stability, little attention has been paid to the quantitative analysis of enzyme agglomeration behavior in nonaqueous bioactalytic systems. In this study, the effects of water and silica gel on enzyme agglomeration were investigated usingCandida rugosa lipase and cyclohexane as a model enzyme and an organic medium. The extent of enzyme agglomeration was quantified by sieve analysis of freeze-dried agglomerates. Increasing the water content of the medium increased the size of the enzyme agglomerates, and it was found that water produced during the esterification reaction could also promote the agglomeration of enzyme particles suspended in organic media. On the other hand, the size of the enzyme agglomerates was remarkably reduced in the presence of silica gel at the same water content. We also show that this increase in the size of enzyme agglomerates results in lower reaction rates in organic solvents.  相似文献

为了增加芦丁的脂溶性从而使其具有更优秀的抗氧化活性,以硬脂酸和月桂酸为酰基供体,在脂肪酶Novozym 435催化下对芦丁选择性酯化.经色谱柱提纯,得到两种带不同长度烃基的芦丁脂肪酸酯.用红外光谱和核磁共振波谱对芦丁硬脂酸进行了结构鉴定,表明该类酯化物的酯化反应位为鼠李糖的C4′″位羟基.以高效液相色谱监测酯化反应进程,分子筛添加时间对酯化率的研究结果显示,分子筛对酯化率和反应速率有提高的作用.分子筛添加时间对酯化率有影响.对于硬脂酸为酰基供体的情况,反应24h后添加分子筛的酯化反应可以得到最大的酯化转化率46%.以月桂酸为酰基供体的酯化反应,反应11 h后添加分子筛可以得到最大的酯化转化率64.5%.  相似文献

应用DEAE52-cellulose、SephacrylS-500HR柱层析从自培养的富硒木耳中分离纯化得到4种木耳硒多糖,SeAPⅠ-1、SeAPⅠ-2、SeAPⅡ和SeAPⅢ,紫外扫描显示在280nm和260nm均无吸收,表明不含蛋白质和氨基酸;高效凝胶渗透色谱为单一对称峰,表明4种硒多糖均为均一多糖。对S180细胞膜的游离脂肪酸和膜脂流动性的研究显示,4种硒多糖具有较好的活性,且SeAPⅠ-2、SeAPⅡ的活性显著强于SeAPⅠ-1、SeAPⅢ(p0.05)。  相似文献

Patterns of seedling recruitment may have persistent effects on population and community processes. Assuming seed availability is not limiting, the environmental sieve (i.e., the suite of factors influencing seed germination and seedling emergence and survival) determines how many seedlings establish and, most importantly, where they do so. In this study, we identify the spatial structure of some resources and abiotic conditions known to be significant for tree seedling emergence and survival and determine how these environmental factors influence the establishment of Fagus grandifolia, Acer saccharum, Fraxinus americana, and Ostrya virginiana in a deciduous forest of southern Québec (Canada). We expect an increase from Fagus, through Acer and Fraxinus, to Ostrya in the control of environmental variables on seedling emergence and survival, because of differences in the seed size of these species. Density of newly-emerged seedlings of all four species showed positive spatial autocorrelation at distances of up to ca. 10 m. Environmental variables were also structured at the same spatial scale, except for soil moisture. Acer seedling emergence pattern was positively correlated to photosynthetic photon flux density (PPFD), and the pattern of Fraxinus to soil N and moisture. Seedling survival was not spatially autocorrelated for any of the four species, although it was positively density-dependent in Acer and Fagus. In only Ostrya was seedling survival correlated (positively) to one of the environmental variables studied, i.e., PPFD. Overall, environmental variables were spatially less heterogeneous than seedling emergence and survival. Either seed availability was not saturating or factors not considered here, such as competition and predation (the intensity of which often varies with resources and/or abiotic conditions), modified the influence that the physical environment had on patterns of seedling establishment. Our prediction of a greater environmental control on seedling emergence and survival in small-seed species was not totally confirmed.  相似文献

A marked salinity-related decrease in gillraker number in sea bass samples during growth was documented. All specimens share the same genetic stock and rearing conditions were constant during early ontogeny. Variation in gillraker number could be related to the ecophenotypism of this character but selection can not be excluded. Results obtained from reared specimens were compared with those reported in the literature and with data collected from wild stocks. Attention is focused on the pitfalls that the use of this character may have on the taxonomy of fish species.  相似文献

Cyphonautes larvae of a bryozoan, Membranipora membranacea, used several ciliary mechanisms to capture algal cells upstream from the lateral band of cilia that produces a feeding current. (1) Lateral cilia changed beat and a backcurrent occurred at the time and place that particles were retained. (2) Algal cells were sieved and held stationary at the upstream (frontal) side of a row of laterofrontal cilia that were not beating. (3) Localized extension of cilia toward the inhalant chamber, coincident with particle captures, indicated that laterofrontal cilia flick toward the inhalant chamber. These flicks may aid transport of captured particles toward the mouth. Thus my earlier report that larvae only sieve, in contrast to the adults (which have an active ciliary response) was in error. The similar ciliary bands in adult and larval bryozoans and in other lophophorates (brachiopods, and phoronids) suggest that these animals share a core repertoire of ciliary behaviours in the capture and concentration of suspended food particles.  相似文献

Somatic embryos were produced in seven cultivars of Exacum affine Balf. using flower buds and peduncles as explants. Flowering plants were produced from five of the cultivars, and no visible mutations were detected. The best medium for callus induction and growth was MS supplemented with 9.0 M 2,4-dichlorophenoxyacetic acid and either zero or 0.089 M BA. Callus suspensions were made by passing the callus through a 100 m sieve. The best embryo regeneration was achieved on growth regulator-free medium.
