  1. The young leaves are the main source of nucleic acids for population genetic studies in palm‐trees; however, the access to this tissue may be limited by specific features of each species. Using root tissues as an alternative source of nucleic acids could facilitate the sampling in large populations.
  2. This study tests root tissue viability as an alternative nucleic acid source (root versus. leaf) and explores different protocols (tissue storage and DNA extraction methods) to obtain high‐quality DNA samples.

The results showed no significant differences in DNA concentration (603.7 vs. 599.1 ng/μl) and quality ratios (A260/280:2.1 vs. 1.9, and A260/230:2.1 vs. 2.0) for the comparisons of tissue source (leaf vs. root) and DNA extraction method (manual vs. kit). For tissue storage method, DNA concentration was significantly higher for root tissues stored in 70% and 90% alcohol solutions (692.8 and 822.6 ng/μl, respectively) versus those obtained from leaf tissue (603.7 ng/μl); however, for the quality parameters, no differences were found.

Results showed the effective potential of using root tissue as an alternative source for nucleic acids, which could facilitate population sampling of palm‐tree species for future studies, and this methodological alternative could be applied to other plant systems with similar sampling challenges.

Summary Leaves of olive trees growing in the vicinity of the Aluminium Factory of Greece were ultrastructurally investigated in order to determine any malformations caused by environmental air pollutants, especially hydrogen fluoride, in comparison with control samples and normal seasonal senescence. Estimation of some elements accumulated by these leaves showed that they contained high amounts of F and Al attributable to the operation of the nearby factory. The most seriously effected cell components were found to be the mesophyll chloroplasts that show a dilation of the intrathylakoid space, increase of the number of plastoglobuli, discoloration of plastoglobuli, accumulation of large starch grains and an overall disorganized appearance of the organelle. The nuclear crystalloid inclusions have unusual shapes, while the vacuoles contain a fibrillar/granular material that increases their electron density. It is concluded that the ultrastructural malformations are caused by a combination of environmental stresses and air pollutants.This paper is dedicated to Professor Dr. Eberhard Schnepf on the occasion of his 60th birthday.  相似文献

In the ectomycorrhizal caesalpiniaceous groves of southern Korup National Park, the dominant tree species, Microberlinia bisulcata, displays very poor in situ recruitment compared with its codominant, Tetraberlinia bifoliolata. The reported ex situ experiment tested whether availabilities of soil potassium and magnesium play a role. Seedlings of the two species received applications of K and Mg fertilizer in potted native soil in a local shade house, and their responses in terms of growth and nutrient concentrations were recorded over 2 years. Amended soil concentrations were also determined. Microberlinia responded strongly and positively in its growth to Mg, but less to K; Tetraberlinia responded weakly to both. Added Mg led to strongly increased Mg concentration for Microberlinia while added K changed that concentration only slightly; Tetraberlinia strongly increased its concentration of K with added K, but only somewhat its Mg concentration with added Mg. Additions of Mg and K had small but important antagonistic effects. Microberlinia is Mg‐demanding and apparently Mg‐limited in Korup soil; Tetraberlinia, whilst K‐demanding, appeared not to be K‐limited (for growth). Added K enhanced plant P concentrations of both species. Extra applied Mg may also be alleviating soil aluminum toxicity, and hence improving growth indirectly and especially to the benefit of Microberlinia. Mg appears to be essential for Microberlinia seedling growth and its low soil availability in grove soils at Korup may be an important contributing factor to its poor recruitment. Microberlinia is highly shade‐intolerant and strongly light‐responding, whilst Tetraberlinia is more shade‐tolerant and moderately light‐responding, which affords an interesting contrast with respect to their differing responses to Mg supply. The study revealed novel aspects of functional traits and likely niche‐partitioning among ectomycorrhizal caesalps in African rain forests. Identifying the direct and interacting indirect effects of essential elements on tropical tree seedling growth presents a considerable challenge due the complex nexus of causes involved.  相似文献

The origin of cultivated tree peonies, known as the ‘king of flowers'' in China for more than 1000 years, has attracted considerable interest, but remained unsolved. Here, we conducted phylogenetic analyses of explicitly sampled traditional cultivars of tree peonies and all wild species from the shrubby section Moutan of the genus Paeonia based on sequences of 14 fast-evolved chloroplast regions and 25 presumably single-copy nuclear markers identified from RNA-seq data. The phylogeny of the wild species inferred from the nuclear markers was fully resolved and largely congruent with morphology and classification. The incongruence between the nuclear and chloroplast trees suggested that there had been gene flow between the wild species. The comparison of nuclear and chloroplast phylogenies including cultivars showed that the cultivated tree peonies originated from homoploid hybridization among five wild species. Since the origin, thousands of cultivated varieties have spread worldwide, whereas four parental species are currently endangered or on the verge of extinction. The documentation of extensive homoploid hybridization involved in tree peony domestication provides new insights into the mechanisms underlying the origins of garden ornamentals and the way of preserving natural genetic resources through domestication.  相似文献

Aim We combine evidence from palaeoniche modelling studies of several tree species to estimate the extent of Central American forest during the Last Glacial Maximum (LGM). In particular, we ask whether the distributions of these species are likely to have changed since the LGM, and whether LGM distributions coincide with previously proposed Pleistocene refugia in this area. Location Central American wet and seasonally dry forests. Methods We developed ecological niche models using two simulations of Pleistocene climate and occurrence data for 15 Neotropical plant species. We focused on palaeodistribution models of three ‘focal’ tree species that occur in wet and seasonally dry Central American forests, where recent phylogeographic data suggest Pleistocene differentiation coincident with previously proposed refugia. We added predictions from six wet‐forest and six seasonally dry‐forest obligate plant species to gauge whether Pleistocene range shifts were specific to habitat type. Correlation analyses were performed between projected LGM and present distributions, LGM distributions and previously proposed refugia. We also asked whether modelled palaeodistributions were smaller than their current extents. Results According to our models, the ranges of the study species were not reduced during the LGM, and did not correlate with refugial models, regardless of habitat type. Relative range sizes between present and LGM distributions did not indicate significant range changes since the LGM. However, relative range sizes differed overall between the two palaeoclimate models. Main conclusions Many of the modelled palaeodistributions of study species were not restricted to refugia during the LGM, regardless of forest type. While constrained from higher elevations, most species found suitable habitat at coastal margins and on newly exposed land due to lowered sea levels during the LGM. These results offer no corroboration for Pleistocene climate change as a driver of genetic differentiation in the ‘focal’ species. We offer alternative explanations for genetic differentiation found in plant species in this area.  相似文献
