The glutathione S-transferases from the northern quahog (Mercenaria mercenaria) from control and contaminated sites were subjected to incubation with polychlorinated biphenyls in an Aroclor 1248 mixture. Subsequent solvent-solvent extraction and gas chromatographic analysis revealed that 2 of the 28 congeners present in the Aroclor 1248 mixture were affected by the presence of the GST. The first of the aforementioned congeners with a retention time of 36.7 min by gas chromatographic analysis decreased in total content. The largest decrease in the 36.7 min peak was the result of incubation with GSTs purified from quahogs taken from the Superfund site in New Bedford Harbor, New Bedford, MA. The second of the affected congeners with a retention time of 59 min showed an increase, which could result from the glutathione conjugation to the PCB congener. The Aroclor 1248 mixture (a limited number of its constituent congeners) also acts as a competitive inhibitor of the GST activity, which is indicative of a substance interacting with the free GST in solution. The ultimate result of the conjugation of the PCB congener to GSH would be the formation of a hydrophillic conjugate of an otherwise insoluble PCB. The PCB-GSH conjugating activity of the quahog GSTs may ultimately serve as a tool for PCB remediation. 相似文献

Analysis of genetic variants of the poly(ADP-ribose) polymerase-1 gene in breast cancer in French patients 总被引:1,自引:0,他引:1
Previous studies have provided evidence for an association between exposure to high levels of air pollution and increased DNA damage in human sperm. In these studies DNA damage was measured using the sperm chromatin structure assay (SCSA) wherein the percentage of sperm with abnormal chromatin/fragmented DNA is determined and expressed as % DNA fragmentation index (%DFI). Here we extend these observations to address the following hypothesis: men who are homozygous null for glutathione-S-transferase M1 (GSTM1−) are less able to detoxify reactive metabolites of carcinogenic polycyclic aromatic hydrocarbons (c-PAHs) found in air pollution. Consequently they are more susceptible to the effects of air pollution on sperm chromatin. Using a longitudinal study design in which men provided semen samples during periods of both low (baseline) and episodically high air pollution, this study revealed a statistically significant association between GSTM1 null genotype and increased SCSA-defined %DFI (beta = 0.309; 95% CI: 0.129, 0.489). Furthermore, GSTM1 null men also showed higher %DFI in response to exposure to intermittent air pollution (beta = 0.487; 95% CI: 0.243, 0.731). This study thus provides novel evidence for a gene–environment interaction between GSTM1 and air pollution (presumably c-PAHs). The significance of the findings in this study with respect to fertility status is unknown. However, it is biologically plausible that increases in %DFI induced by such exposures could impact the risk of male sub/infertility, especially in men who naturally exhibit high levels of %DFI. 相似文献

We describe here a practical system for generating selectable marker-free transgenic woody plants independent of sexual crossing. We previously reported that the GST-MAT vector system could produce marker-free transgenic tobacco plants containing a single-copy transgene at high frequency. The GST-MAT vector system consists of a DNA excision cassette of the R/RS site-specific recombination system from Zygosaccharomyces rouxii into which the isopentenyltransferase gene from Agrobacterium tumefaciens has been inserted. In this study, we applied this new GST-MAT vector to hybrid aspen (Populus Sieboldii X Populus grandidentata), a model of vegetatively propagated plant species, to produce selectable marker-free transgenic woody plants. In the new GST-MAT vector, the chimeric ipt gene fused with a light-inducible rbcS promoter efficiently produced transgenic ipt-shooty with GUS activity from 38.0% of infected stems. Upon excision of the R and ipt genes between RS sites, regulated by the inducible promoter of the maize glutathione-S-transferase (GST-II-27) gene, 3 (21.4%) transgenic hybrid aspen plants with marker-free and normal phenotype were generated from 14 ipt-shooty lines within 2 months after cutting induction. These results suggest that the MAT-vector system might be useful for removing a selectable marker gene independent of sexual crossing in vegetatively propagated species. 相似文献
