K‐ion batteries are a potentially exciting and new energy storage technology that can combine high specific energy, cycle life, and good power capability, all while using abundant potassium resources. The discovery of novel cathodes is a critical step toward realizing K‐ion batteries (KIBs). In this work, a layered P2‐type K0.6CoO2 cathode is developed and highly reversible K ion intercalation is demonstrated. In situ X‐ray diffraction combined with electrochemical titration reveals that P2‐type K0.6CoO2 can store and release a considerable amount of K ions via a topotactic reaction. Despite the large amount of phase transitions as function of K content, the cathode operates highly reversibly and with good rate capability. The practical feasibility of KIBs is further demonstrated by constructing full cells with a graphite anode. This work highlights the potential of KIBs as viable alternatives for Li‐ion and Na‐ion batteries and provides new insights and directions for the development of next‐generation energy storage systems.  相似文献

Reproduction is energetically financed using strategies that fall along a continuum from animals that rely on stored energy acquired prior to reproduction (i.e., capital breeders) to those that rely on energy acquired during reproduction (i.e., income breeders). Energy storage incurs a metabolic cost. However, previous studies suggest that this cost may be minimal for small‐bodied ectotherms. Here I test this assumption. I use a laboratory feeding experiment with the European green crab Carcinus maenas to establish individuals with different amounts of energy storage. I then demonstrate that differences in energy storage account for 26% of the variation in basal metabolic costs. The magnitudes of these costs for any individual crab vary through time depending on the amount of energy it has stored, as well as on temperature‐dependent metabolism. I use previously established relationships between temperature‐ and mass‐dependent metabolic rates, combined with a feasible annual pattern of energy storage in the Gulf of Maine and annual sea surface temperature patterns in this region, to estimate potential annual metabolic costs expected for mature female green crabs. Results indicate that energy storage should incur an ~8% increase in metabolic costs for female crabs, relative to a hypothetical crab that did not store any energy. Translated into feeding, for a medium‐sized mature female (45 mm carapace width), this requires the consumption of an additional ~156 mussels annually to support the metabolic cost of energy storage. These results indicate, contrary to previous assumptions, that the cost of energy storage for small‐bodied ectotherms may represent a considerable portion of their basic operating energy budget. An inability to meet these additional costs of energy storage may help explain the recent decline of green crabs in the Gulf of Maine where reduced prey availability and increased consumer competition have combined to hamper green crab foraging success in recent years.  相似文献

Time correlation functions invariably suffer from random noise, especially at longer time intervals for which fewer data pairs are available. This noise is particularly of concern when calculating correlations that cannot be averaged over per-molecule contributions, such as stress in molecular simulations. In this work, a set of methods based in signal processing has been developed to reduce the inherent noise that is present in time- and frequency-domain representations of correlation functions. The stress time autocorrelation function, which leads to stress relaxation modulus and complex modulus, is used as an example. The difference between initial and final values of a time correlation function over a finite time domain is found to create so-called ‘leakage’ of noise from disallowed into harmonic frequencies during fast Fourier transformation. Decreasing this leakage effect through reflection to negative time and through applying a window function reduces noise levels significantly. Removing frequency components of insignificant magnitudes also provides significant noise reduction. Applying moving averages in the frequency and time domains also contributes to noise reduction. Specific results obtained by applying these methods to a model asphalt system enable more clear physical interpretations of the underlying relaxations after dramatic noise level reductions were attained.  相似文献

Immobilized enzymes offer different benefits such as the feasibility to be reused for reproducible bioprocesses. The challenge is to establish the appropriate storage conditions that allow the maintenance of their properties for long periods. In this study, we immobilized a recombinant leucine aminopeptidase (I-rLAP) on a siliceous support synthesized from tetraethyl orthosilicate (TEOS) activated with glutaraldehyde to evaluate its residual activity after storage in 20% v/v ethanol and sodium azide solutions at 4 and 25?°C. The characterization of the support by X-ray diffraction (XRD), diffuse reflectance infrared Fourier transform (DRIFT) and field emission probe microanalyzer (EPMA) was consistent with previous characterization reports of silica gel matrices. Particle size ≤420?μm exhibited a suitable performance that avoided high backpressure into the columns and increased the amount of immobilized enzyme. I-rLAP recovered up to 90% of the applied activity after 64 days of storage at 4 and 25?°C in 20% v/v ethanol. Conversely, no effect was observed when the insoluble enzyme was stored in sodium azide. Activity recovery of I-rLAP after storage in ethanol solution could be related to the formation of disulfide bonds as suggested by free thiol analyses. Reverse phase-ultra performance liquid chromatography (RP-UPLC) and Mass Spectrometry confirmed that the immobilized enzyme maintained its specificity to remove N-terminal methionine from a recombinant hormone. The obtained results indicate that this methodology constitutes an alternative for bioprocesses involving long-term storage of immobilized enzymes.  相似文献

An inherited deficiency of arylsulfatase A (ASA) causes the lysosomal storage disease metachromatic leukodystrophy (MLD) characterized by massive intralysosomal storage of the acidic glycosphingolipid sulfatide and progressive demyelination. Lyso-sulfatide, which differs from sulfatide by the lack of the N-linked fatty acid, also accumulates in MLD and is considered a key driver of pathology although its concentrations are far below sulfatide levels. However, the metabolic origin of lyso-sulfatide is unknown. We show here that ASA-deficient murine macrophages and microglial cells express an endo-N-deacylase that cleaves the N-linked fatty acid from sulfatide. An ASA-deficient astrocytoma cell line devoid of this activity was used to identify the enzyme by overexpressing 13 deacylases with potentially matching substrate specificities. Hydrolysis of sulfatide was detected only in cells overexpressing the enzyme fatty acid amide hydrolase (FAAH). A cell-free assay with recombinant FAAH confirmed the novel role of this enzyme in sulfatide hydrolysis. Consistent with the in vitro data, deletion of FAAH lowered lyso-sulfatide levels in a mouse model of MLD. Regardless of the established cytotoxicity of lyso-sulfatide and the anti-inflammatory effects of FAAH inhibition seen in mouse models of several neurological diseases, genetic inactivation of FAAH did not mitigate, but rather exacerbated the disease phenotype of MLD mice. This unexpected finding was reflected by worsening of rotarod performance, increase of anxiety-related exploratory activity, aggravation of peripheral neuropathy, and reduced life expectancy. Thus, we conclude that FAAH has a protective function in MLD and may represent a novel therapeutic target for treatment of this fatal condition.  相似文献

The migratory behaviour of adult cod was studied using electronic data storage tags (DSTs). Fifty-eight cod were externally tagged with LOTEK LTD-100 DSTs and released off Lowestoft in March and April 1999. Twenty-two tags were returned by March 2000. Individuals demonstrated significant shifts in behaviour patterns that were associated with migration and residence on feeding grounds. During migration, individuals were very active and utilised a large vertical range. In contrast, individuals demonstrated low activity and a much reduced vertical range whilst on summer feeding grounds. Use of the Tidal Location Method showed that individuals dispersed to feeding grounds in the central and southern North Sea. Our results demonstrate that individual-based studies of fish behaviour can provide information relevant to fisheries management.  相似文献

Surprisingly little is known about molecular mechanisms by which nitrogen (N) availability acts to modulate the growth of forest trees. To address this issue, differential display was used in conjunction with filter‐based arrays to identify 52 partial cDNA clones that were significantly regulated within days in response to limiting or luxuriant levels of NH4NO3 fertilization in Populus trichocarpa Torr. & Gray × deltoides Bartr. ex Marsh. A subset of these cDNAs also demonstrated shifts in expression patterns in stem‐girdled trees, a manipulative physiology technique that disrupts phloem transport. Stem girdling also induced changes in glutamine and asparagine pools which were correlated with the observed changes in expression profiles for these genes. The identity of these genes provides insight into biochemical processes that are altered by N availability in poplar. Carbon–nitrogen interactions appear to figure prominently in the N‐response. The gene expression data suggest that N availability modulates the partitioning of C and N resources into metabolic fates that have the potential to alter both wood quality and quantity, including synthesis of vegetative storage proteins, cell wall components, and terpenoids.  相似文献

The genetic diversity of annual wild soybeans grown in China   总被引:13,自引:0,他引:13  
Annual wild soybeans (Glycine soja), the ancestors of cultivated soybeans (G. max), are important sources of major genes for resistance to pests, diseases and environmental stresses. The study of their genetic diversity is invaluable for efficient utilization, conservation and management of germplasm collections. In this paper, the number of accessions, the variation of traits, the genetic diversity indexes (Shannon index) and the coefficient of variation were employed to study the geographical distribution of accessions, genetic diversity of characters and genetic diversity centers of annual wild soybean by statistical analysis of the database from the National Germplasm Evaluation Program of China. Most annual wild soybeans are distributed in Northeast China, and the number of accessions decreases from the Northeast to other directions in China. The genetic diversity indexes (Shannon index) were 0.49, 0.74, 0.02, 0.55, 1.45, 2.41, 1.27 and 1.89 for flower color, sootiness of seed coat, cotyledon color, pubescence color, hilum color, leaf shape, stem type and seed color, respectively. Coefficients of variation were 7.1%, 28.7%, 76.43% and 18.2% for protein content, oil content, 100-seed weight and days to maturity, respectively. Three genetic diversity centers, the Northeast, the Yellow River Valley and the Southeast Coasts of China, are proposed based on the geographical distribution of the number of accessions, genetic diversity and the multivariate variation coefficient. Based on these results and Vavilov’s theory of crop origination, two opposing possible models for the formation of the three centers are proposed, either these centers are independent of each other and the annual wild soybeans in these centers originated separately, or the Northeast center was the primary center for annual wild soybeans in China, while the Yellow River Valley center was derived from this primary center and served as the origin for the Southeast Coast center.
