Current protocols for chondrocyte expansion and chondrogenic differentiation of stem cells fail to reduce phenotypic loss and to mitigate hypertrophic tendency. To this end, cell genetic manipulation is gaining pace as a means of generating cells with stable chondrocyte phenotype. Herein, we provide an overview of candidate genes that either induce cartilage regeneration or inhibit cartilage degeneration. We further discuss in vitro, ex vivo and in vivo viral transduction and non-viral transfection strategies for targeted cells (chondrocytes, mesenchymal stem cells, induced pluripotent stem cells and synovial cells), along with the most representative results obtained in pre-clinical models and in clinical trials. Chironomid communities of mosses in a small upland stream in central Germany were highly dynamic across the year with respect to their abundance, biomass and dominant taxa. During 1988 semi-submersed mosses near a main spring and those occurring some 700 m downstream were compared with permanently submersed mosses in immediate vicinity of the downstream site. All the chironomids sampled were conspicuously small, with nearly 98% being less than 5 mm in length. A total of 65 chironomid species from 26 genera were found, with a higher diversity occurring near the source and a change in dominant taxa along the upper stream section. The mean abundance in permanently submersed mosses (250 larvae/10 cm2, n = 125) was about five times higher than in semi-submersed mosses. The maximum value of 830 larvae/10 cm2 (n = 1) is the highest chironomid density ever reported, which is explained by the sampling method used. The mean standing crop was also highest in permanently submersed mosses (1.5 mg AFDW/10 cm2 (n = 125)), even though the highest individual value was recorded in semi-submersed mosses near the spring (10.4 mg AFDW/10 cm2). The evidence suggested that the dominance of chironomid taxa depended mainly on the location of the moss along the stream, whereas abundance and biomass were determined mainly by constancy in the ambient discharge as well as the factors influenced by this (e.g. temperature, detritus deposition). Aim: This study aimed to evaluate the effect of bromelain, a cysteine protease isolated from pineapple (Ananas comosus), on growth of several agronomically important fungal pathogens. Methods and Results: Purification of bromelain from pineapple stems was carried out by chromatography techniques, and its antimicrobial activity was tested against the fungal pathogens Fusarium verticillioides, Fusarium oxysporum and Fusarium proliferatum by broth microdilution assay. A concentration of 0·3 μmol l?1 of bromelain was sufficient for 90% growth inhibition of F. verticillioides. The capability of bromelain to inhibit fungal growth is related to its proteolytic activity. Conclusions: The study demonstrates that stem bromelain exhibits a potent antifungal activity against phytopathogens and suggests its potential use as an effective agent for crop protection. Significance and Impact of the Study: The results support the use of a natural protease that accumulates at high levels in pineapple stems as alternative to the use of chemical fungicides for crop protection. 相似文献
Vaccination represents one of the greatest public health triumphs; in part due to the effect of adjuvants that have been included in vaccine preparations to boost the immune responses through different mechanisms. Although a variety of novel adjuvants have been under development, only a limited number have been approved by regulatory authorities for human vaccines. This report reflects the conclusions of a group of scientists from academia, regulatory agencies and industry who attended a conference on the current state of the art in the adjuvant field. Held at the U.S. Pharmacopeial Convention (USP) in Rockville, Maryland, USA, from 18 to 19 April 2013 and organized by the International Association for Biologicals (IABS), the conference focused particularly on the future development of effective adjuvants and adjuvanted vaccines and on overcoming major hurdles, such as safety and immunogenicity assessment, as well as regulatory scrutiny. Arsenic (As) contaminated food chains have emerged as a serious public concern for humans and animals and are known to affect the cultivation of edible crops throughout the world. Therefore, the present study was designed to investigate the individual as well as the combined effects of exogenous silicon (Si) and sodium nitroprusside (SNP), a nitric oxide (NO) donor, on plant growth, metabolites, and antioxidant defense systems of radish (Raphanus sativus L.) plants under three different concentrations of As stress, i.e., 0.3, 0.5, and 0.7 mM in a pot experiment. The results showed that As stress reduced the growth parameters of radish plants by increasing the level of oxidative stress markers, i.e., malondialdehyde and hydrogen peroxide. However, foliar application of Si (2 mM) and pretreatment with SNP (100 µM) alone as well as in combination with Si improved the plant growth parameters, i.e., root length, fresh and dry weight of plants under As stress. Furthermore, As stress also reduced protein, and metabolites contents (flavonoids, phenolic and anthocyanin). Activities of antioxidative enzymes such as catalase (CAT), ascorbate peroxidase (APX), guaiacol peroxidase (POD), and polyphenol oxidase (PPO), as well as the content of non-enzymatic antioxidants (glutathione and ascorbic acid) decreased under As stress. In most of the parameters in radish, As III concentration showed maximum reduction, as compared to As I and II concentrations. However, the individual and combined application of Si and NO significantly alleviated the As-mediated oxidative stress in radish plants by increasing the protein, and metabolites content. Enhancement in the activities of CAT, APX, POD and PPO enzymes were recorded. Contents of glutathione and ascorbic acid were also enhanced in response to co-application of Si and NO under As stress. Results obtained were more pronounced when Si and NO were applied in combination under As stress, as compared to their individual application. In short, the current study highlights that Si and NO synergistically regulate plant growth through lowering the As-mediated oxidative stress by upregulating the metabolites content, activity of antioxidative enzymes and non-enzymatic antioxidants in radish plants.
Since maize silage is an important forage in cattle nutrition, it is important to know its nutritive value. Much effort is put into breeding maize, and several new varieties are introduced on the market every year. This requires periodical analyses of the nutritive value of current maize varieties for the formulation of cattle rations. The aim of this study was to examine the nutritive value of whole crop maize silage (WCMS) from nine maize varieties in 3 consecutive years. For the analysis of nutrient composition and ruminal degradability of organic matter (OM), crude protein (CP), neutral detergent fibre (aNDFom) and non-fibre carbohydrates (NFC), varieties were harvested at three harvest dates (50%, 55% and 60% dry matter content in ear). Due to capacity limitations, the digestibility of WCMS was tested only for the middle harvest date. The CP and acid detergent fibre (ADFom) content was affected (p < 0.05) while aNDFom and NFC content was not influenced by variety. With advancing maturity, CP, aNDFom and ADFom content declined while NFC content increased. Variety influenced effective ruminal degradability (ED) of nutrients, except for CP. The ED of all examined nutrients decreased as maturity advanced from first to third harvest date. Digestibility of OM, ADFom and NFC was significantly and digestibility of aNDFom was tendentially (p = 0.064) influenced by variety. Additionally, an effect of year and a harvest date × year interaction was found for almost all examined parameters. In conclusion, variety, harvest date and year influence the nutritive value of WCMS. A comparison with earlier studies shows that current varieties have a higher fibre digestibility and a slower-ripening stover compared to older varieties. 相似文献
Anthropogenic influences have dramatically altered the environments with which primates interact. In particular, the introduction of anthropogenic food sources to primate groups has implications for feeding behaviour, social behaviour, activity budgets, demography and life history. While the incorporation of anthropogenic foods can be beneficial to primates in a variety of nutritional ways including increased energetic return, they also carry risks associated with proximity to humans, such as risk of being hunted, disease risk and risk of conflict. Given such risks, we initiated a 3‐year study where we sought to understand the underlying nutritional motivations for anthropogenic food resource use by vervet monkeys (Cercopithecus aethiops) in the humanized matrix surrounding the Nabugabo Field Station in central Uganda. Feeding effort, defined as proportion of feeding scans spent on anthropogenic food, was not associated with ripe fruit availability nor with crop availability as determined by phenological monitoring. Likewise, there was no difference in the protein, fibre, or lipid composition of crop food items compared to wild food items. Individuals spent less time feeding overall in months over the 3 years with a higher proportion of time spent feeding on crop foods, suggesting a potential benefit in terms of accessibility (reduction in the proportion of activity budget devoted to feeding). 相似文献
Arthropod crop pests are responsible for 20% of global annual crop losses, a figure predicted to increase in a changing climate where the ranges of numerous species are projected to expand. At the same time, many insect species are beneficial, acting as pollinators and predators of pest species. For thousands of years, humans have used increasingly sophisticated chemical formulations to control insect pests but, as the scale of agriculture expanded to meet the needs of the global population, concerns about the negative impacts of agricultural practices on biodiversity have grown. While biological solutions, such as biological control agents and pheromones, have previously had relatively minor roles in pest management, biotechnology has opened the door to numerous new approaches for controlling insect pests. In this review, we look at how advances in synthetic biology and biotechnology are providing new options for pest control. We discuss emerging technologies for engineering resistant crops and insect populations and examine advances in biomanufacturing that are enabling the production of new products for pest control. 相似文献
