ras原癌基因的点突变是人胃癌发生发展的重要机理之一。利用能表达c-H-ras癌基因反义RNA的质粒,导入人胃癌细胞系BGC-823,研究了ras癌基因反义RNA对人胃癌细胞生长及恶性表型的作用,结果表明,c-H-ras反义RNA可引起BGC-823生长速率及形态的变化,在半固体培养基中细胞集落形成能力减弱,部分地抑制了BGC-823在裸鼠体内的致瘤性。c-H-ras反义RNA对其RNA的过量表达呈特异性抑制作用。  相似文献

Galanin has numerous effects on gastrointestinal motility in different species; however, its cellular basis of action in mediating these effects is unclear. Dispersed gastric smooth muscle cells have been shown to possess high-affinity galanin receptors that increase cAMP and cause relaxation. Recent studies show some smooth muscle relaxants such as VIP cause relaxation by both cAMP-dependent and -independent mechanisms. It is unknown if galanin's cellular basis of relaxation is similar or different from that of VIP. To investigate galanin's relaxant effect and compare it to VIP's effect, dispersed smooth muscle cells from guinea pig stomach were prepared by collagenase digestion. The mean length in resting cells was 110 ± 2 μm and, with carbachol treatment, contracted to 89 ± 2 μm. VIP and galanin alone had no effect on cell length, but each caused a dose-dependent inhibition of carbachol-induced contraction and both had an EC50 of 3–7 nM. Galanin (1 μM) and VIP (1 μM) increased cellular cAMP from 118 ± 10 pmol/106 cells in control to 212 ± 14 and 214 ± 12 pmol/106 cells, respectively. The protein kinase A inhibitor, Rp-cAMPS, at 100 μM, completely inhibited the relaxant effect of an EC50 concentration of galanin (3 nM), but only inhibited that by VIP by 80% (p < 0.05). Adding the nitric oxide inhibitor, -NNA ( ), at 100 μM did not alter the length of resting cells or inhibit carbachol-induced contraction. However, -NNA (100 μM) decreased VIP-induced relaxation by 45%, whereas it had no effect on galanin-induced relaxation. To determine the ability of each peptide to activate nitric oxide, the incorporation of [3H]arginine into [3H]citrulline was determined. Galanin (1 μM) did not cause nitric oxide generation whereas VIP (1 μM) increased nitric oxide generation above the control by 97 ± 14% (p < 0.01). These results demonstrated that with galanin, in contrast to VIP, nitric oxide is not involved in its ability to cause gastric smooth muscle cell relaxation. The relaxant action of galanin can be accounted for completely by its ability to activate protein kinase A and therefore resembles recent results with β-adrenergic agents.  相似文献

Summary The distribution of endocrine cells in the gastrointestinal tract of the house musk shrew, Suncus murinus (Family Soricidae, Order Insectivora) was studied immunohistochemically. The hormones investigated were gastrin, cholecystokinin (CCK), somatostatin, secretin, glucagon, gastric inhibitory polypeptide (GIP), motilin and neurotensin. In the gastric mucosa, gastrin and somatostatin cells were only found in the pyloric regions, and no other hormonal cell-types were observed. In the intestinal mucosa, the largest number of endocrine cells belonged to the gastrin and glucagon/glicentin cell-types, whereas CCK-33/39 and secretin cells were the least numerous. Numbers of other cell-types were intermediate between these two groups. The gastrin and GIP cells were mostly localized in the proximal portion of the intestine, decreasing in number towards the distal portion. The motilin and CCK-33/39 cells were restricted to the proximal half. The glucagon/glicentin and neurotensin cells were most abundant in the middle portion. The somatostatin and secretin cells, although only present in small numbers, were randomly distributed throughout the intestine. This characteristic distribution of gastrointestinal endocrine cells is discussed in comparison with the distribution patterns of other mammals.Dr. Munemitsu Hoshino, who was Professor of the Department of Pathology and directed this study, passed away on May 23rd 1988  相似文献

为探讨胃液酸度对幽门螺旋菌(HP)的影响,我们对609例胃、十二指肠疾病患者进行HP检查和pH值测定,结果发现:在257例胃液pH≤2.0患者中,HP阳性率为47.5%(112/257),在311例HP阳性患者中,pH≤2.0者占39.2%(122/311);胃液pH≥4.0患者的HP阳性率高于HP<4.0者,且阳性率有显著差异(P<0.05);另外,十二指肠溃疡患者在高胃酸环境下HP阳性率较高(为75%),并对其阳性率较高的机理进行了探讨。  相似文献

Helicobacter pylori is a gram negative bacterium that infects the human stomach of approximately half of the world’s population. It produces oxidative stress, and mitochondria are one of the possible targets and the major intracellular source of free radicals. The present study was aimed at determining mitochondrial alterations in H. pylori-infected gastric epithelial cells and its relationship with oxidative stress, one of the recognized causes of apoptotic processes. Cells were treated with a strain of H. pylori for 24 h. Cellular oxidative burst, antioxidant defense analysis, mitochondrial alterations and apoptosis-related processes were measured. Our data provide evidence on how superoxide acts on mitochondria to initiate apoptotic pathways, with these changes occurring in the presence of mitochondrial depolarization and other morphological and functional changes. Treatment of infected cells with Vitamin E prevented increases in intracellular ROS and mitochondrial damage consistent with H. pylori inducing a mitochondrial ROS mediated programmed cell death pathway.  相似文献

利用抑制神经系统的WinnerLess Competition(WLC)模型,通过数值方法分析Mulloney型龙虾胃肠神经系统神经元的电位发放,得到胃研磨囊和幽门神经系统中各个神经元的电位发放和系统的节律变化。结果表明,胃研磨系统内神经元的发放规律显示两侧牙齿和中间牙齿出现切断、挤压和研磨食物等状态,幽门系统内神经元的发放规律显示幽门节律出现依次发放的三个部分。两个神经系统的数值结果,不仅解释了龙虾胃肠神经系统中神经元电位发放与肌肉运动的关系,而且理论再现了龙虾胃肠神经系统的节律变化和实验结果。  相似文献

目的:考察辣椒素对离体大鼠胃平滑肌收缩性的影响。方法:本研究以大鼠的离体胃平滑肌条为模型,首先考察在正常钙克氏液、高钙克氏液和低钙克氏液中辣椒素对胃平滑肌收缩作用的影响。然后正常克氏液中,分别观察辣椒素对乙酰胆碱、新斯的明、阿托品分别存在下的胃平滑肌收缩性的影响。结果:辣椒素在2.5μmol/L-40μmol/L浓度范围内可剂量依赖性显著抑制高钙溶液(Ca2 终浓度5μmol/L)引起的大鼠胃平滑肌强烈收缩,在5μmol/L-40μmol/L浓度范围内可显著抑制正常克氏液中大鼠胃平滑肌条的运动,且具有明显剂量依赖性,在10μmol/L-40μmol/L浓度范围内可显著抑制低钙克氏液中大鼠胃平滑肌条的运动,且具有剂量依赖性。辣椒素(10μmol/L)可拮抗乙酰胆碱和新斯的明引起的收缩作用(P<0.01)。辣椒素(10μmol/L)的作用与阿托品具有相加作用(P<0.01)。结论:辣椒素对胃平滑肌的收缩具有明显的抑制作用。  相似文献

目的:探讨乙酰肝素酶和S-100蛋白在人胃癌组织中的表达及其意义。方法:根据胃癌的病理大体分型将40例胃癌组织分为早期组和晚期组。其中,早期组同时未伴有淋巴结转移,晚期组伴有淋巴结转移。采用光镜、透射电镜、原位杂交和免疫组化方法对这两组胃癌组织的超微结构,乙酰肝素酶和S-100蛋白表达进行检测。结果:早期组乙酰肝素酶阳性表达细胞较少,晚期组阳性细胞较多,二者数密度和面密度比较。具有统计学意义(P<0.01);早期组S-100蛋白阳性表达细胞较晚期组多,二者比较,具有统计学意义(P<0.01);电镜观察可见:在胃癌早期,淋巴细胞和树突状细胞浸润较多,树突状细胞突起与淋巴细胞相接触,基底膜基本完整。晚期,基底膜几乎消失。淋巴细胞和树突状细胞浸润较少,癌细胞穿基膜明显。结论:乙酰肝素酶和S-100蛋白的表达程度可作为判定胃癌的侵袭和转移的指标,对其预后的判断具有参考价值。  相似文献

The present study aimed to investigate the effect of knocking-down methylenetetrahydrofolate reductase (MTHFR) on the survival of the human gastric cancer cell line MKN45. Antisense and small interfering RNA (siRNA) plasmids were used to target MTHFR in MKN45. Meanwhile, we also constructed a wild-type MTHFR plasmid to assess the effect of over-expression of this protein on cell viability. The knock-down of MTHFR decreased cell survival by approximately 30% compared to the control and resulted in cell cycle arrest at the G2 phase. These cells also had lower levels of c-myc compared to control cells, while over-expression of MTHFR increased cell proliferation and induced the down-regulation of p21WAF1 and hMLH1. Inhibiting MTHFR with either antisense or siRNA decreases the viability of methionine-dependent transformed gastric cancer cells and suggests that MTHFR inhibition may be a novel anticancer approach.  相似文献

半胱胺对断奶前后仔猪胃粘膜H+-K+-ATPase表达和活性的影响   总被引:1,自引:0,他引:1  
实验选取新生仔猪18窝, 随机分为实验组和对照组各9窝, 自12日龄起, 对照组饲喂基础乳猪料, 实验组在基础饲料中添加半胱胺120 mg/kg饲料, 仔猪均于35日龄断奶。分别于断奶前1 周、断奶当天、断奶后36 h、72 h、1周以及断奶后10 d屠宰仔猪取样, 每个时间点随机选取对照组和试验组各6头仔猪, 用相对定量RT PCR方法测定不同日龄仔猪胃组织中H K ATPase mRNA 表达的相对含量, 并同时测定H K ATPase的活性。结果表明: (1) 对照组仔猪在断奶前1 周至断奶后10 d, H K ATPase mRNA相对含量和H K ATPase活性有上升趋势, 两组仔猪在断奶后10 d H K ATPase mRNA相对含量和活性均达到最高水平, 但总体不表现显著的年龄差异; (2) 半胱胺能明显提高仔猪胃粘膜中H K ATPase mRNA的表达, 在断奶前1 周、断奶当天、断奶后1周和断奶后10 d均出现显著差异。半胱胺也能明显提高H K ATPase 的活性, 在断奶当天和断奶后10 d, H K ATPase 的活性分别被提高32 3%和18 3%; (3) 观察期内对照组和实验组H K ATPase mRNA水平和H K ATPase活性之间未发现显著的相关性。以上结果说明: 断奶前后仔猪H K ATPase的mRNA相对含量和活性没有明显的发育性变化, 断奶对H K ATPase mRNA表达和活性没有影响,而半胱  相似文献
