野生树舌多糖的脱色和脱蛋白   总被引:8,自引:0,他引:8  
从野生树舌中提取水溶性多糖,经乙醇沉淀后得多糖,并分别用活性碳,苯酚,DEAE纤维素,H2O2等4种方法进行脱色,并以酵母聚糖为标准品比较脱色效果,用紫外和可见光谱扫描法检测有色杂质的去除效果;用Zn盐,胰蛋白酶,有机溶剂,胰蛋白酶加有机溶剂等4种方法脱蛋白,以比较不同方法脱色,脱蛋白的效果。结果表明用质量分数为0.75%的活性碳脱色后,有色杂质的去除效果最好,光谱吸收峰最接近标准品;单独使用有机溶剂的脱蛋白效果最好。  相似文献

通过对灵芝菌As5.504发酵,研究不同糖源、氮源、生长因子及接种量、装液量对灵芝细胞生长和灵芝多糖生产的影响。结果表明当以葡萄糖,蛋白胨,VB5分别为碳源、氮源、生长因子及接种量、装液量分别为7.5mL、150mL时,为As5.504产多糖最佳发酵工艺条件,产多糖量最高为0.940g·L-1。  相似文献

The paper describes a mechanistic mathematical model for aroma release in the oropharynx to the nasal cavity during food consumption. The model is based on the physiology of the swallowing process and is validated with atmospheric pressure chemical ionization coupled with mass spectrometry measurements of aroma concentration in the nasal cavity of subjects eating flavored yogurt. The study is conducted on 3 aroma compounds representative for strawberry flavor (ethyl acetate, ethyl butanoate, and ethyl hexanoate) and 3 panelists. The model provides reasonably accurate time predictions of the relative aroma concentration in the nasal cavity and is able to simulate successive swallowing events as well as imperfect velopharyngeal closure. The most influent parameters are found to be the amount of the residual product in the pharynx and its contact area with the air flux, the volume of the nasal cavity, the equilibrium air/product partition coefficient of the volatile compound, the breath airflow rate, as well as the mass transfer coefficient of the aroma compound in the product, and the amount of product in the mouth. This work constitutes a first step toward computer-aided product formulation by allowing calculation of retronasal aroma intensity as a function of transfer and volatility properties of aroma compounds in food matrices and anatomophysiological characteristics of consumers.  相似文献

Abstract Invasive woody species frequently change the composition of the established vegetation and the properties of the soil under their canopies. Accordingly, invasion may well affect regenerative phases of the community, especially at the seed bank level, likely influencing community restoration. Pyracantha angustifolia (Rosaceae) is an invasive shrub in central Argentina that affects woody recruitment, particularly enhancing the recruitment of other exotic woody species. There is though no information regarding its effect on the soil seed bank within the invaded community. The present study was set up to gain further insight into the canopy effects of P. angustifolia. We aimed to assess whether the invasive shrub affects seed bank composition, richness and seed density as compared with the dominant native shrub Condalia montana (Rhamnaceae), and to relate the observed seed bank patterns with those of the established vegetation. We evaluated the composition of the germinable seed bank and the established vegetation under the canopy of 16 shrubs of P. angustifolia, 16 shrubs of C. montana, and in 16 control plots (10 m2) without shrub cover. The floristic composition of the seed bank differed among canopy treatments. However, seed bank richness did not differ significantly. There was an overall high seed density of exotic species throughout the study site, though exotic forbs showed significantly lower seed densities under the invasive shrub. Pyracantha angustifolia would not promote the incorporation of new species into the seed bank of the invaded community but rather favour the establishment of woody species that do not depend on seed banks. The absence of dominant woody species in the seed bank, the dominance of exotic forbs, and the high similarity between established exotic species and those present in the seed bank may surely affect community restoration following the main disturbances events observed in the region.  相似文献

The antiinflammatory properties of triterpenoids and steroids from both Ganoderma lucidum and Ganoderma tsugae were studied. Twelve compounds, including ergosta-7,22-dien-3beta-ol (1), ergosta-7,22-dien-3beta-yl palmitate (2), ergosta-7,22-dien-3-one (3), ergosta-7,22-dien-2beta,3alpha,9alpha-triol (4), 5alpha,8alpha-epidioxyergosta-6,22-dien-3beta-ol (5), ganoderal A (6), ganoderal B (7), ganoderic aldehyde A (8), tsugaric acid A (9), 3-oxo-5alpha-lanosta-8,24-dien-21-oic acid (10), 3alpha-acetoxy-5alpha-lanosta-8,24-dien-21-oic acid ester beta-d-glucoside (11), and tsugaric acid B (12), were assessed in vitro by determining their inhibitory effects on the chemical mediators released from mast cells, neutrophils, and macrophages. Compound 10 showed a significant inhibitory effect on the release of beta-glucuronidase from rat neutrophils stimulated with formyl-Met-Leu-Phe (fMLP)/cytochalasin B (CB) whereas compound 9 significantly inhibited superoxide anion formation in fMLP/CB-stimulated rat neutrophils. Compound 10 also exhibited a potent inhibitory effect on NO production in lipopolysaccharide (LPS)/interferon-gamma (IFN-gamma)-stimulated N9 microglial cells. Moreover, compound 9 was also able to protect human keratinocytes against damage induced by ultraviolet B (UV B) light, which indicated 9 could protect keratinocytes from photodamage.  相似文献

In this study, an attempt was made to increase the survival rate of bifidobacteria entrapped in alginate in the gastrointestinal tract, and to investigate the potential industrial applications, for example lyophilized capsules and yogurt. First, the protective effect of various food additives on bifidobacterial survivability was determined after exposure to simulated gastric juices and bile salts. The additives used in this study were skim milk (SM), poly dextrose (PD), soy fiber (SF), yeast extract (YE), chitosan (CS), κ-carageenan (κ-C) and whey, which were added at 0.6% concentration (w/v) to 3% alginate-bifidobacterial solution. In the simulated gastric juices and bile salts, the protective effect of 0.6% skim milk-3% alginate (SM-A) beads on the survival rate of bifidobacteria proved to be higher than the other additives. Second, the hydrogen ion permeation was detected through SM-A vessel without bifidobacterial cells at different SM concentrations (0.2%, 0.4%, 0.6%, 0.8%, and 1.0%). There were no differences in terms of the pH decrease in SM-A vessels at 0.6%, 0.8%, and 1.0% (w/v) SM concentrations. The survival rate of bifidobacteria in SM-A beads would appear to be related to the SM buffering capacity against hydrogen ions and its tendency to reduce the pore size of bead. In this experiment, the survival rate of bifidobacteria entrapped in beads containing 0.6% SM showed the highest viability after exposure to simulated gastric juices for 3 h, thereby indicating that 0.6% SM is the optimum concentration for 3% alginate bead preparation. Third, the effect of SM-A beads on the freeze-drying and yogurt storage for 10 days was investigated. SM-A beads were found to be more efficient for freeze drying and yogurt storage than untrapped cells and the alginate bead. Consequently, the survival rate of bifidobacteria entrapped in SM-A beads was increased in simulated gastric juices, bile salts and probiotic products such as lyophilized capsules and yogurt, SM-A beads can be expected to produce high value probiotic products.  相似文献

本文报道了基于香草醛-高氯酸显色反应的分光光度法定量测定灵芝三萜的修正方法,并对该方法应用进行了探讨和优化。采用此方法检测了灵芝子实体中含量较高的几种三萜酸,结果表明若采用齐墩果酸为标准品检测灵芝三萜,检测结果远低于真实值。在光谱分析上,研究表明对紫外-可见光扫描吸收峰进行面积积分,获得的标准曲线的线性关系更优。  相似文献

以灵芝为材料,在前期研究基础上,以不同酵母粉作为氮源,研究复合有机氮源对灵芝三萜液态深层发酵的影响。首先,由单因素实验考察3种不同的酵母粉对灵芝菌丝体合成灵芝三萜的影响,确定酵母粉的最适宜浓度范围。在此基础上,根据中心组合实验设计,对3种酵母粉分别采用2种复合和3种复合的方式,优化复合有机氮源的最佳组合配比。结果表明:当基础培养基中添加6.6g/L的酵母粉N-1与6.6g/L的酵母粉N-2时,灵芝三萜产量可达0.478g/L(理论产量为0.485g/L),比添加单一酵母粉N-1、N-2、N-3分别提高了21%、139%、103%,其氮源用量为两种组合时最低。当基础培养基中酵母粉N-1、N-2与N-3添加量分别为5.07g/L、3.78g/L、7.63g/L时,灵芝三萜产量达0.514g/L(理论产量为0.510g/L),比单因素对照组分别提高了30%、157%和74%。本研究优化的复合氮源添加方式可明显提高灵芝菌丝体液态深层发酵生产灵芝三萜的产量,为其规模化液态深层发酵的生产提供科学数据。  相似文献

运用建立的高效液相色谱条件,首次实现在完成灵芝孢子粉中麦角甾醇含量测定的同时,对灵芝孢子粉中脂溶性成分的指纹图谱进行研究。结果表明,该方法测定麦角甾醇准确、稳定、特异性好,脂溶性成分指纹图谱基线平直、色谱峰丰富且具备较好的分离度,该方法适合在分析测定灵芝孢子粉中麦角甾醇(282nm)含量的同时,对其脂溶性成分(245nm)的指纹图谱进行分析,为整体评价灵芝孢子粉及其产品的质量提供依据。运用建立的方法对收集的各孢子粉样品进行分析,表明破壁与否及破壁时间长短对灵芝孢子粉中麦角甾醇及脂溶性成分的溶出有显著的影响,实验认为最佳的破壁时间为20-30min;低温冻藏可以有效地保护样品中的成分;随着产粉时间的延长,孢子粉中麦角甾醇的含量变化不大,脂溶性成分总体上有所增加;不同原料来源孢子粉中麦角甾醇的含量和脂溶性成分指纹图谱的差异都很大,且麦角甾醇的含量与指纹图谱色谱峰的丰富程度没有一定的相关性,因此不能仅以麦角甾醇的含量来表征灵芝孢子粉的质量,二者的综合分析对灵芝孢子粉质量的评价才更有意义。  相似文献
