脉络丛(choroid plexus,CP)位于血液与脑脊液(cerebrospinal fluid,CSF)之间,不仅是CSF的重要来源,而且是构成血液-脑脊液屏障(blood-cerebrospinal fluid barrier,BCB)的组织基础.CP参与脑组织中一些血源性多肽的输送以及自身多肽合成的生理过程,在维持脑微环境动态平衡和调节中枢神经系统的正常功能方面起到非常重要的作用.本研究分别运用MALDI-TOF/TOF和LC-MS/MS液质联用系统分析了成年SD大鼠血液-脑脊液屏障(即脉络丛组织)中的多肽组.共鉴定到163个多肽(P0.001),这些多肽为69种蛋白质的降解肽段,其中ATP合酶(ATP synthase),细胞色素c(cytochrome c),血红蛋白(hemoglobin),NADH-辅酶Q氧化还原酶(NADH-ubiquinone oxidoreductase),β珠蛋白(beta-globin)这5种蛋白质的肽段数占总肽段数的50%以上,并且部分多肽序列相似度高,类似其前体蛋白的逐步降解片段,而这些前体蛋白质的分子量多数在10kD至20kD之间.上述研究结果为SD大鼠脉络丛组织的生理功能研究及组织多肽组学的研究方法提供了有价值的科学资料. 相似文献

Primary fatty acid amides (PFAM) are important signaling molecules in the mammalian nervous system, binding to many drug receptors and demonstrating control over sleep, locomotion, angiogenesis, and many other processes. Oleamide is the best-studied of the primary fatty acid amides, whereas the other known PFAMs are significantly less studied. Herein, quantitative assays were used to examine the endogenous amounts of a panel of PFAMs, as well as the amounts produced after incubation of mouse neuroblastoma N(18)TG(2) and sheep choroid plexus (SCP) cells with the corresponding fatty acids or N-tridecanoylethanolamine. Although five endogenous primary amides were discovered in the N(18)TG(2) and SCP cells, a different pattern of relative amounts were found between the two cell lines. Higher amounts of primary amides were found in SCP cells, and the conversion of N-tridecanoylethanolamine to tridecanamide was observed in the two cell lines. The data reported here show that the N(18)TG(2) and SCP cells are excellent model systems for the study of PFAM metabolism. Furthermore, the data support a role for the N-acylethanolamines as precursors for the PFAMs and provide valuable new kinetic results useful in modeling the metabolic flux through the pathways for PFAM biosynthesis and degradation. 相似文献

Drugs to treat African trypanosomiasis are toxic, expensive and subject to parasite resistance. New drugs are urgently being sought. Although the existing drug, eflornithine, is assumed to reach the brain in high concentrations, little is known about how it crosses the healthy and infected blood-brain barrier. This information is essential for the design of drug combinations and new drugs. This study used novel combinations of animal models to address these omissions. Eflornithine crossed the healthy blood-CNS interfaces poorly, but this could be improved by co-administering suramin, but not nifurtimox, pentamidine or melarsoprol. Work using a murine model of sleeping sickness demonstrated that Trypanosoma brucei brucei crossed the blood-CNS interfaces, which remained functional, early in the course of infection. Concentrations of brain parasites increased during the infection and this resulted in detectable blood-brain barrier, but not choroid plexus, dysfunction at day 28 post-infection with resultant increases in eflornithine brain delivery. Barrier integrity was never restored and the animals died at day 37.9 +/- 1.2. This study indicates why an intensive treatment regimen of eflornithine is required (poor blood-brain barrier penetration) and suggests a possible remedy (combining eflornithine with suramin). The blood-brain barrier retains functionality until a late, possibly terminal stage, of trypanosoma infection. 相似文献

The transport stoichiometry of the electrogenic sodium-bicarbonate cotransporter (SLC4A5 or NBCe2) in mouse choroid plexus was examined. Whole-cell recording methods measured the currents carried by the NBCe2, using experimental solutions determined to minimise the contributions of the other ion conductances present. Increases in outward current were observed when 21.2 mM was added to the bath solution in the presence of Na+, but not N-methyl-d-glucamine. This -induced current was completely abolished by 500 μM 4,4′-diisothiocyanostilbene-2,2′-disulphonic acid. The reversal potential for the -induced current was −95.1 ± 7.1 mV (n = 11), a value which corresponds to a NBCe2 transport stoichiometry of 3 with 1 Na+. The NBCe2, with this stoichiometry, will mediate the efflux of and Na+ from the cell into the cerebrospinal fluid at the apical membrane of the choroid plexus. 相似文献

Abstract: In vitro , the transport of [14 C]nicotine into the isolated choroid plexus, the anatomical locus of the blood–CSF barrier, was studied. The isolated rabbit choroid plexus accumulated [14 C]nicotine by two processes: an active saturable transport process and a nonsaturable process. The [14 C]nicotine accumulation process by choroid plexus was not due to binding or intracellular metabolism of the [14 C]nicotine. The [14 C]nicotine accumulation process in isolated choroid plexus was inhibited by weak bases, including tolazoline and lidocaine, but not by the weak acid probenecid. The accumulation process was decreased 60% by iodoacetate and dinitrophenol and by low temperatures. These results are consistent with previous autoradiographic evidence showing the choroid plexus concentrated [14 C]nicotine in vivo , and suggest that the choroid plexus may transfer nicotine between blood and CSF in vivo . 相似文献

Non-steroidal anti-inflammatory drugs (NSAIDs) have a variety of potential indications that include management of pain and inflammation as well as chemoprevention and/or treatment of cancer. Furthermore, a specific form of ibuprofen, dexibuprofen or the S-(+) form, shows interesting neurological activities and has been proposed for the treatment of Alzheimer’s disease. In a continuation of our work probing the anticancer activity of small sulindac libraries, we have prepared and screened a small diversity library of α-methyl substituted sulindac amides in the profen class. Background
Spontaneous intracranial hypotension (SIH) is caused by cerebrospinal fluid (CSF) leakage. Definitive diagnosis can be difficult by clinical examinations and imaging studies.Methods
SIH was diagnosed with the following criteria: (i) evidence of CSF leakage by cranial magnetic resonance imaging (MRI) findings of intracranial hypotension and/or low CSF opening pressure; (ii) no recent history of dural puncture. We quantified CSF proteins by ELISA or Western blotting.Results
Comparing with non-SIH patients, SIH patients showed significant increase of brain-derived CSF glycoproteins such as lipocalin-type prostaglandin D synthase (L-PGDS), soluble protein fragments generated from amyloid precursor protein (sAPP) and “brain-type” transferrin (Tf). Serum-derived proteins such as albumin, immunoglobulin G, and serum Tf were also increased. A combination of L-PGDS and brain-type Tf differentiated SIH from non-SIH with sensitivity 94.7% and specificity 72.6%.Conclusion
L-PGDS and brain-type Tf can be biomarkers for diagnosing SIH.General significance

Zusammenfassung An der lichtmikroskopischen Struktur des plexus chorioideus ventriculi III von Ateles, Cebus, Macaca und Pan fallen deutliche speziesbedingte Unterschiede auf. Bei Macaca wurde die Ultrastruktur der Plexus chorioidei ventriculi III und IV sowie der Plexus chorioidei der Seitenventrikel verglichen. Kuppenzellen überwiegen im elektronen-mikroskopischen Bild der Plexusepithelien des III. und IV. Ventrikels, während unter den Epithelien der Seitenventrikel flachere Zellen dominieren. Diese strukturellen Besonderheiten könnten auf funktionelle Unterschiede in der Liquorbildung hinweisen. Das Problem der extrachorioidalen Liquorproduktion (Curl and Pollay, 1968; Milhorat u. Mitarb., 1971) wird im Zusammenhang mit den regionalen Unterschieden in der Wandstruktur des Aquaeductus cerebri (Sylvii) diskutiert (Merker, 1970). Im Stroma und im Interstitium aller Plexus chorioidei des Rhesusaffen sind makrophagenartige Wanderzellen zu beobachten, deren Strukturbild dem der Epiplexuszellen gleicht; sie werden als ausgewanderte Monozyten gedeutet. Wahrscheinlich gelangen diese Zellen in den Ventrikel und werden dort zu Epiplexuszellen. Einschlüsse vom Typ der Biondi-Körper wurden in den Plexusepithelien der untersuchten Affen nicht beobachtet; die untersuchten Tiere hatten aber noch kein hohes Lebensalter erreicht.
The fine structure of the choroid plexuses in monkeys
Summary The light microscopic structure of the choroid plexus of the IIIrd ventricle was studied in Ateles, Cebus, Macaca and Pan. These choroid plexuses display distinct species differences. The ultrastructure of the choroid plexuses of the IIIrd, IVth and lateral ventricles was compared in Macaca. Cuboidal cells with dome-like protrusions predominate in the epithelium of the choroid plexuses of the IIIrd and the IVth ventricles, while flattened cells are characteristic of the choroid epithelium of the lateral ventricle. These structural peculiarities may reflect functional differences in the production of the cerebrospinal fluid. The problem of extrachoroidal production of the cerebrospinal fluid (Curl and Pollay, 1968; Milhorat et al., 1971) is discussed in connection with regional differences in the structure of the aqueduct wall (Merker, 1970). Macrophage-like cells which morphologically resemble epiplexus cells and which are very abundant within the connective tissue stroma and also between the epithelial cells of all choroid plexuses of the rhesus monkey, are thought to be monocytes. Probably these cells migrate into the ventricle where they become epiplexus cells. Epithelial inclusions of the type of Biondi bodies were not observed in the simian choroid plexuses. However, the animals investigated were not old.

Zusammenfassung In der Kleinhirnrinde des Menschen, des Rhesusaffen und der Katze formen die rückläufigen Kollateralen der Purkinjezellen im unteren Drittel der Molekularschicht einen markhaltigen Plexus, den Plexus supraganglionaris. Dieser Plexus ist nicht in allen Abschnitten der Kleinhirnrinde gleichmäßig entfaltet, sondern zeigt starke regionale Unterschiede in seiner Dichte.Im Lobus nodulofloccularis, dem ältesten Kleinhirnabschnitt, ist der Plexus im Wurmanteil nur spärlich entwickelt, im dazugehörigen Hemisphärenanteil dagegen etwas stärker ausgebildet. Im Lobus posterior des Corpus cerebelli ist er im Wurm und in den Hemisphären nur sehr diskret angelegt. Seine stärkste Entfaltung erfährt der Plexus supraganglionaris im Lobus anterior des Corpus cerebelli. Hier nehmen im Wurmabschnitt die rückläufigen Kollateralen beinahe das untere Drittel der Molekularschicht ein, während der Plexus in den Hemisphärenanteilen nur schwach entwickelt ist. Die mögliche Bedeutung der regional unterschiedlichen Dichte des Plexus supraganglionaris für den intracorticalen Erregungsablauf wird diskutiert.

Summary In the cerebellar cortex of man, rhesus monkey and cat the recurrent collaterals of the Purkinje cells form a myelinated plexus, the plexus supraganglionaris, in the lower third of the molecular layer. This plexus does not exhibit a uniform distribution in all parts of the cerebellar cortex but its density varies considerably in different regions.In the eldest part of the cerebellum, in the lobus nodulofloccularis, the plexus is developed sparsely in the region of the vermis, somewhat more, however, in the corresponding part of the hemispheres. In the lobus posterior of the corpus cerebelli there are only few collaterals in the vermis as well as in the hemispheres, while the plexus supraganglionaris is best developed in the lobus anterior. Here in the region of the vermis rather the lower third of the molecular layer is occupied by recurrent collaterals while in the hemispheres only a sparsely developed plexus can be observed.The possible importance of the regional differences in density of the plexus supraganglionaris in regard to the transmission of excitation is discussed.
