Summary The recombination fraction between the interstitially located gene an and interchange 303 of rye was found to be 0.244±0.038 in a test cross using the translocation as the male parent. In first metaphase translocation configurations in pollen mother cells of the same plant, the chiasma frequency between an and the translocation breakpoint was found to be significantly more than twice the recombination fraction. Recombination was concluded to be masked by a difference in the alternate frequency between configurations without interstitial chiasmata and configurations with interstitial chiasmata, the effect of the first type being of major importance. Random centromere orientation of translocation multivalents with interstitial chiasmata was concluded to be a realistic assumption. The exceptionally high recombination between an and translocation 303 is discussed. Consideration is also given to the use of interchanges in the establishment of a marker's chromosomal position, and to the use of translocation chromosomes in balanced systems for hybrid breeding purposes.  相似文献

Microtubule assembly is required for the formation of the male and female pronuclei during mouse, but not sea urchin, fertilization. In mouse oocytes, 50 μM colcemid prevents the decondensation of the maternal meiotic chromosomes and of the incorporated sperm nucleus during in vitro fertilization. Nuclear lamins do not associate with either of the parental chromatin sets although peripherin, the PI nuclear peripheral antigen, appears on both. DN A synthesis docs not occur in these fertilized, colcemid-arrested oocytes. This effect is limited to the first hours after ovulation, since colcemid added 4–6 hours later no longer prevents pronuclear development, lamin acquisition, or DNA synthesis. Neither microtubule stabilization with 10 μM taxol nor microfilament inhibition with 10 μM cytochalasin D or 2.2 μg/ml lalrunculin A prevent these pronuclear events; these drugs will inhibit the apposition of the pronuclei at the egg center. In sea urchin eggs, colcemid or griseofulvin treatment doe? not result in the same effect and the male pronucleus forms with the attendant accumulation of the nuclear lamins. The differences in the requirement for microtubule assembly during pronucleus formation may be related to the cell cycle: In mice the sperm enters a meiotic cytoplasm, whereas in sea urchin eggs it enters an interphase cytoplasm. Refertilization of mitotic sea urchin eggs was performed to test the possibility that this phenomenon is related to whether the sperm enters a meiotic/mitotic cytoplasm or one at interphase; during refertilization at first mitosis, the incorporated sperm nucleus is unable to decondense and acquire lamins. These results indicate a requirement for microtubule assembly for the progression from meiosis to first interphase during mouse fertilization and suggest that the cytoskeleton is required for changes in nuclear architecture necessary during fertilization and the cell cycle.  相似文献

A mechanism is proposed for the way in which cholesterol facilitates channel formation by polyene antibiotics and bacterial protein toxins. Callus was induced from juice vesicles of satsuma mandarin on Murashige & Skoog medium supplemented with -naphthaleneacetic acid (NAA), kinetin (K) and gibberellin (GA). Adventitious embryoids arose from the callus tissue on the medium containing 1 mgl–1 NAA alone. Sequential addition of vanadyl sulfate to a phosphate-buffered solution of H2O2 released oxygen only after the second batch of vanadyl. Ethanol added to such reaction mixtures progressively decreased oxygen release and increased oxygen consumption during oxidation of vanadyl by H2O2. Inclusion of ethanol after any of the three batches of vanadyl resulted in varying amounts of oxygen consumption, a property also shared by other alcohols (methanol, propanol and octanol). On increasing the concentration of ethanol, vanadyl sulfate or H2O2, both oxygen consumption and acetaldehyde formation increased progressively. Formation of acetaldehyde decreased with increase in the ratio of vanadyl:H2O2 above 2:1 and was undetectable with ethanol at 0.1 mM. The reaction mixture which was acidic in the absence of phosphate buffer (pH 7.0), released oxygen immediately after the first addition of vanadyl and also in presence of ethanol soon after initial rapid consumption of oxygen, with no accompanying acetaldehyde formation. The results underscore the importance of some vanadium complexes formed during vanadyl oxidation in the accompanying oxygen-transfer reactions.  相似文献

We formulate and analyze pair-formation models for multiple groups with general pairing rates and arbitrary mixing probabilities. Under the assumption of constant recruitment rates and equal average duration of all types of partnerships, we have shown that the dynamics are relatively simple because of the monotonicity properties of the dynamical system associated with the pairing/mixing of heterogeneous populations of male and female individuals. In fact, we have shown that the corresponding asymptotic stable paired distribution is given precisely by the asymptotic values of the matrices that prescribe the mixing/contact structure. In other words, if the sizes of the mixing subpopulations of males and females are asymptotically constant and if the average durations of partnerships are about the same regardless of type, then the matrices that describe the mixing between subpopulations also characterize the distribution of paired types. Alternatively, if the distribution of the average duration of relationships between individuals has a large variance then it may be impossible to detect any relationship between the mixing/contact structure and the observed distribution of paired types. The study of models with constant per-capita recruitment rates give rise to homogeneous systems of degree one. The analysis of the dynamics of pairs for models with exponentially growing populations of singles is complicated. So far, we are only able to classify the stability of all non-strictly positive boundary exponential solutions. From our incomplete analysis, it is not possible to detect necessary and sufficient conditions for the existence and stability of strictly interior exponential solutions. We cannot rule out the possibility of oscillations. The mathematical problems associated with the stability of exponential solutions of dynamical systems of degree one are of relevance in demography, epidemiology, and population dynamics.On leave from University of Alabama in Huntsville  相似文献

Zoospores at various developmental stages in Hydrodictyon reticulatum were isolated from parent cells and cultured in Waris medium. Isolated zoospores grew to mature vegetative cells, and were able to reproduce zoo-spores that formed daughter hexagonal nets. Three types of shape appeared in cells 24 h after isolation: cylindrical, Y-shaped and 4-armed type. Protrusions of Y-shaped or 4-armed cells were formed at an angle of about 120° to the long axis of the cell. When cells were isolated at later stages, more cells became cylindrical in shape and fewer ceils became Y-shaped or 4-armed, Direction of cell growth also seemed to depend largely on the developmental stages of the zoospores. The later the isolated stages were, the more the cells elongated along the long axis of the zoospores.  相似文献

Fluorogenic peptide substrates designed to encompass the reported-secretory and amyloidogenic cleavage sites of the amyloid- precursor protein (PP) were used to analyze proteinase activities in brain extracts from control patients and those with Alzheimer's disease (AD). Activity against the secretory substrate atpH 7.5 in control and AD brains produced a major endopeptidase cleavage at the Lys687-Leu688 bond (PP770 numbering), consistent with thePP secretase cleavage. Activity in control brains against the amyloidogenic substrate atpH 7.5 produced one cleavage at the Ala673-Glu674 bond, two residues C-terminal to the amyloidogenic Met-Asp site. However, in three of four AD brains, the major cleavage was at the Asp-Ala bond, one residue from the amyloidogenic site. Both endopeptidase and carboxypeptidase activities in AD brains were lower than in control brains. Proteinase activities against the secretory substrate had a major optimum atpH 3.0–4.0 and another atpH 6.0–7.5. Proteinase activities against the amyloidogenic substrate had a major optimum at or belowpH 3.0 and another atpH 6.0. Using both substrates, activities at lowpH were higher in AD brains than in controls, while atpH above 6.5, activities in control brains were higher than in AD. We have previously shown that micromolar concentrations of GDP stimulate the GTP-mediated phosphorylation of p36, the subunit of succinyl-CoA synthetase (SCS), in lysates prepared fromDictyostelium discoideum. In this study, we report that this phenomenon represents an enhanced catalytic capacity of SCS to form the phosphoenzyme intermediate. Low concentrations of GDP stimulate phosphoenzyme formation by either GTP, or succinyl-CoA and Pi. Under these conditions GDP enhances the apparent rate of phosphoenzyme formation but does not significantly alter the fraction of phosphorylated enzyme. This effect is retained during purification of the protein and is also observed with purified pig heart SCS, indicating that GDP directly alters the enzyme to enhance its rate of phosphorylation.
