大鼠皮下注射TNT,以HPLC分析其在晶状体内的代谢过程,并检测晶状体谷胱甘肽过氧化物酶、谷胱甘肽还原酶及超氧化物歧化酶的活性变化。发现在注射TNT后2h即可在晶状体内检测到为量极少的TNT及其代谢产物,第12h一氨基二硝基甲苯含量达最高峰。鼠龄较小的大鼠晶状体内TNT及其代谢产物高于鼠龄较大的大鼠.多剂量注射TNT时大鼠晶状体内一氨基二硝基甲苯于第2天达到高峰,TNT于第5天达饱和状态,第18天一氨基二硝基甲苯含量与TNT含量相近。谷胱甘肽过氧化物酶、谷胱甘肽还原酶及超氧化物歧化酶活性在注射TNT的第2天均有不同程度的升高,在第5天和第18天维持在低活性状态。实验表明TNT在晶状体内是通过硝基还原而代谢的.TNT进入晶状体后初期可诱发晶状体抗氧化相关酶活性的增高,后期则导致晶状体抗氧化相关酶活性的降低。  相似文献

Selenium is an essential element for antioxidation reactions in human and animals. In order to study its biological role in higher plants, ryegrass (Lolium perenne) was cultivated in a soil without Se or amended with increasing dosages of H2SeO4 (0.1, 1.0, 10.0 and 30.0 mg Se kg−1). Ryegrass was harvested twice and the yields were analyzed for antioxidative systems and growth parameters. Selenium exerted dual effects: At low concentrations it acted as an antioxidant, inhibiting lipid peroxidation, whereas at higher concentrations, it was a pro-oxidant, enhancing the accumulation of lipid peroxidation products. The antioxidative effect was associated with an increase in glutathione peroxidase (GSH-Px) activity, but not with superoxide dismutase (SOD) and αα-tocopherol, which was the only tocopherol detected. In the second yield, the diminished lipid peroxidation due to a proper Se addition coincided with promoted plant growth. The oxidative stress found at the Se addition level ≥ 10 mg kg−1 resulted in drastic yield losses. This result indicates that the toxicity of Se can be attributed, in addition to metabolic disturbances, to its pro-oxidative effects. Neither the growth-promoting nor the toxic effect of Se could be explained by the changes in the total chlorophyll concentration. Selenium (Se) deficiency in the experimental models,Coturnix coturnix japonica andCorcyra cephalonica, resulted in impaired mitochondrial substrate oxidations and lowered thiol levels. Studies with respiratory inhibitors confirmed reduced mitochondrial electron transport enzyme activities, especially at cytochromec oxidase (COX), the terminal segment. Enhanced mitochondrial lipid peroxidation in Se deficiency was more pronounced in the heart tissue of the quail compared to other tissues. Glutathione peroxidase (GSH-Px) activity toward H2O2 and cumene hydroperoxide were generally low in the insect muscle tissue and activity toward H2O2 was maximal in the quail heart mitochondria that was not very sensitive to Se status. Lowered COX activity in Se deficiency was more directly correlated with the increased level of lipid peroxidation than with the GSH-Px activity measured, suggestive of Se mediated protective mechanisms independent of GSH-Px. Electron microscopic observations revealed structural changes such as loss of cristae with proliferative and degenerative changes of the mitochondria in Se deficiency. Involvement of Se in maintaining structure and functional efficiency of mitochondria is evident from the present study.  相似文献

The present study was carried out to evaluate the effect of selenium (Se)-induced oxidative stress on the oxidation reduction system and the fertility status of male mice. Different levels of Se, a potent antioxidant, were fed in three separate groups for 8 wk to create the different oxidative stress in mice. A significant decrese in the glutathione peroxidase (GSH-Px) in both liver and testis was observed in the Se-deficient (0.02 ppm) group I, whereas enzyme levels in the Se-excess (1 ppm) group were comparable to the Se-adequate (0.2 ppm) group. Glutathione-S-transferase activity was enhanced in group I in comparison to group II; however, no change was seen in group III. The glutathione reductase and superoxide dismutase activities were decreased in the Se-deficient group, whereas the enzyme levels were significantly increased in the Se-excess group. The fertility status of the animals studied in terms of percentage fertility and litter size showed a significant decrease in the reproductive ability of male mice in group I when compared to group II. No changes in the fertility status of animals were observed in group III. Thus, the data clearly indicate the effect of oxidative stress generated by feeding various Se levels on the oxidation reduction system and, consequently, its effect on the reproductive ability of male mice.  相似文献

The interaction of selenium with methylmercury was investigated in brain of animals labeled with 75SeO32? and CH3203Hg+. Brains were fractionated into subcellular components and the cytosol was further fractionated by chromatography on Sephadex G-150 and G-200. The main result of these studies was evidence suggesting a shift of 75Se from the cytosol to the mitochondrial fraction in brain when CH3Hg+ was given. Concurrent equimolar (10 μmoles/kg) selenite injections increased the uptake of Hg but did not alter 203Hg distribution in brain. Changing the dose of CH3Hg+ from 1 to 38 μmoles/kg had little effect on Hg uptake (% of dose per g). Gel filtrations on Sephadex G-150 and G-200 revealed that 203Hg in cytosol followed a pattern more closely related to protein (A280) than to 75Se, although a considerable portion of both isotopes eluted with proteins in the void volume. Assays of whole brain homogenates revealed a slight reduction in glutathione peroxidase activity in CH3Hg+-treated rats which was not seen when equimolar selenite was injected with the CH3Hg+.  相似文献

Salvia miltiorrhiza has strong antioxidative activity. They may have a strong potential as cardioprotective agents in ischemic–reperfusion injury. Experiments were carried out in Sprague–Dawley rats with myocardium ischemia reperfusion (IR). Myocardial injuries during IR were determined by changes in electrocardiogram analysis of arrhythmias, antioxidant enzyme activities, AST, CK-MB, lactate dehydrogenase (LDH) levels, and myocyte apoptosis. Results showed that S. miltiorrhiza aqueous extract (SAME) pre-treatment significantly decreased the ST-segment (ΣST120) and myocardium MDA, AST, CK-MB, lactate dehydrogenase (LDH) levels, increased myocardium antioxidant enzyme activities, and inhibit myocardium cell apoptosis. Furthermore, the SAME pre-treatment significantly upregulated p-JAK2 and p-STAT3 protein expression, decreased myocardium TNF-α and IL-6 concentrations in IR rats. The levels of TNF-α and IL-6 were positively correlated with the changes in myocardium p-JAK2 and p-STAT3 protein expression levels in IR rats. It can be concluded that the SAME pre-treatment has anti-ischemic and anti-apoptosis activity in heart IR rats. SAME pre-treatment protects heart against IR injury, at least in part, through its stimulating effects on injury-induced deactivation of JAK2/STAT3 signaling pathway.  相似文献

Obesity-prone (OP) and obesity-resistant (OR) rats with different responses to development of obesity in spite of the same genetic background are useful animal models for searching for markers during the development of obesity. Here, we investigated whether plasma proteins of OP and OR rats may behave in a different way in males and females. We performed a comparative proteomic analysis using 2-DE combined with MALDI-TOF/MS on proteins from OP and OR male and female rats to discover gender-specific rat plasma proteins associated with susceptibility or resistance to diet-induced obesity. A total of 29 proteins showing differential expression between the groups were identified by MALDI-TOF/MS and database searches. These proteins were classified into 4 groups according to their regulation patterns in response to diet and gender. 22 proteins showed significant differences between OP and OR rats in males and/or females (Group I, II, and III) and 7 proteins exhibited only a high fat diet (HFD)-responsive difference in male or female rats (Group IV). In conclusion, the proteins negatively (ITIH3, FGG, TUBB5, and ZAG) or positively (Hp, ITIH4, and RBP) correlated with obesity found in this study could be used for selection of new targets for gender specific-medical treatment of obesity.  相似文献

The aim of this study was to investigate the clinical, haematological, biochemical, lipid peroxidation, ultrasonographic and pathologic findings in hepatic coccidiosis induced by Eimeria stiedae in rabbits, and also to compare the treatment effects of both toltrazuril and ivermectin separately and in combination. In this study, 56 rabbits were divided into eight groups. The first group was designated as healthy control group. Rabbits were infected with 40.000 sporulated oocysts of E. stiedae. Groups 2, 3, 4, 5, 6, 7 and 8 were allocated as the infected control group, infected+toltrazuril-treated group, infected+ivermectin-treated group, infected+toltrazuril+ivermectin-treated group, non-infected+toltrazuril-treated group, non-infected+ivermectin-treated group, non-infected+toltrazuril+ivermectin-treated group, respectively. Haematocrit, Haemoglobin and MCV values as well as percentage of lymphocyte decreased in Groups 2 and 4 whereas leucocyte counts and percentage of granulocyte leucocyte increased. Serum GGT, ALT and AST activities increased but albumin value decreased. Plasma MDA concentrations increased whereas erythrocyte CAT, GSH-Px, and SOD activities decreased. Mean oocyst numbers in per gram faeces (epg values) increased in both groups during the study. Ultrasonographic examination revealed that the liver was enlarged and had hyperechogenic parenchyma. Bile ducts were dilated and hyperechogenic and the gall bladder was dilated. The livers of these animals were enlarged and typical macroscopic and microscopic findings of coccidiosis were present. Treatment with toltrazuril and toltrazuril+ivermectin combination were highly effective in reducing faecal oocyst output in infected rabbits. Haematological, biochemical and lipid peroxidation parameters and, ultrasonographic findings of the liver were close to control values for Groups 3 and 5. Necropsy of these animals showed no visible lesions related to hepatic coccidiosis although a few oocysts were detected in the bile duct epithelial cells.  相似文献
