Species establishment within a community depends on their interactions with the local environment and resident community. Such environmental and biotic filtering is frequently inferred from functional trait and phylogenetic patterns within communities; these patterns may also predict which additional species can establish. However, differentiating between environmental and biotic filtering can be challenging, which may complicate establishment predictions. Creating a habitat‐specific species pool by identifying which absent species within the region can establish in the focal habitat allows us to isolate biotic filtering by modeling dissimilarity between the observed and biotically excluded species able to pass environmental filters. Similarly, modeling the dissimilarity between the habitat‐specific species pool and the environmentally excluded species within the region can isolate local environmental filters. Combined, these models identify potentially successful phenotypes and why certain phenotypes were unsuccessful. Here, we present a framework that uses the functional dissimilarity among these groups in logistic models to predict establishment of additional species. This approach can use multivariate trait distances and phylogenetic information, but is most powerful when using individual traits and their interactions. It also requires an appropriate distance‐based dissimilarity measure, yet the two most commonly used indices, nearest neighbor (one species) and mean pairwise (all species) distances, may inaccurately predict establishment. By iteratively increasing the number of species used to measure dissimilarity, a functional neighborhood can be chosen that maximizes the detection of underlying trait patterns. We tested this framework using two seed addition experiments in calcareous grasslands. Although the functional neighborhood size that best fits the community's trait structure depended on the type of filtering considered, selecting these functional neighborhood sizes allowed our framework to predict up to 50% of the variation in actual establishment from seed. These results indicate that the proposed framework may be a powerful tool for studying and predicting species establishment.  相似文献

Abstract Plants with crassulacean acid metabolism (CAM) are increasing their abundance in drylands worldwide. The drivers and mechanisms underlying the increased dominance of CAM plants and CAM expression (i.e., nocturnal carboxylation) in facultative CAM plants, however, remain poorly understood. We investigated how nutrient and water availability affected competition between Mesembryanthemum crystallinum (a model facultative CAM species) and the invasive C3 grass Bromus mollis that co‐occur in California's coastal grasslands. Specifically we investigated the extent to which water stress, nutrients, and competition affect nocturnal carboxylation in M. crystallinum. High nutrient and low water conditions favored M. crystallinum over B. mollis, in contrast to high water conditions. While low water conditions induced nocturnal carboxylation in 9‐week‐old individuals of M. crystallinum, in these low water treatments, a 66% reduction in nutrient applied over the entire experiment did not further enhance nocturnal carboxylation. In high water conditions M. crystallinum both alone and in association with B. mollis did not perform nocturnal carboxylation, regardless of the nutrient levels. Thus, nocturnal carboxylation in M. crystallinum was restricted by strong competition with B. mollis in high water conditions. This study provides empirical evidence of the competitive advantage of facultative CAM plants over grasses in drought conditions and of the restricted ability of M. crystallinum to use their photosynthetic plasticity (i.e., ability to switch to CAM behavior) to compete with grasses in well‐watered conditions. We suggest that a high drought tolerance could explain the increased dominance of facultative CAM plants in a future environment with increased drought and nitrogen deposition, while the potential of facultative CAM plants such as M. crystallinum to expand to wet environments is expected to be limited.  相似文献

A fundamental goal of ecology is to understand the determinants of species' distributions (i.e., the set of locations where a species is present). Competition among species (i.e., interactions among species that harms each of the species involved) is common in nature and it would be tremendously useful to quantify its effects on species' distributions. An approach to studying the large‐scale effects of competition or other biotic interactions is to fit species' distributions models (SDMs) and assess the effect of competitors on the distribution and abundance of the species of interest. It is often difficult to validate the accuracy of this approach with available data. Here, we simulate virtual species that experience competition. In these simulated datasets, we can unambiguously identify the effects that competition has on a species' distribution. We then fit SDMs to the simulated datasets and test whether we can use the outputs of the SDMs to infer the true effect of competition in each simulated dataset. In our simulations, the abiotic environment influenced the effects of competition. Thus, our SDMs often inferred that the abiotic environment was a strong predictor of species abundance, even when the species' distribution was strongly affected by competition. The severity of this problem depended on whether the competitor excluded the focal species from highly suitable sites or marginally suitable sites. Our results highlight how correlations between biotic interactions and the abiotic environment make it difficult to infer the effects of competition using SDMs.  相似文献

This work addresses rapid resin selection for integrated chromatographic separations when conducted as part of a high‐throughput screening exercise during the early stages of purification process development. An optimization‐based decision support framework is proposed to process the data generated from microscale experiments to identify the best resins to maximize key performance metrics for a biopharmaceutical manufacturing process, such as yield and purity. A multiobjective mixed integer nonlinear programming model is developed and solved using the ε‐constraint method. Dinkelbach's algorithm is used to solve the resulting mixed integer linear fractional programming model. The proposed framework is successfully applied to an industrial case study of a process to purify recombinant Fc Fusion protein from low molecular weight and high molecular weight product related impurities, involving two chromatographic steps with eight and three candidate resins for each step, respectively. Bak Foong pill (BFP) is a well-known traditional Chinese medicine used for treatment of various gynaecological disorders. In addition, it exerts beneficial effects on other functional systems including the central nervous system. In the present study, we have investigated the possible neuroprotective action of BFP upon the nigrostriatal dopaminergic system by examining its effect on the expression patterns of tyrosine hydroxylase (TH) and dopamine transporter (DAT) in the 1-methyl-4-phenyl-1,2,3,6-tetrahyrdropyridine (MPTP)-induced Parkinson's disease (PD) mouse model. MPTP significantly decreased TH and DAT mRNA levels in the striatum and midbrain of both female and male C57BL/6 mice. However, with BFP pre-treatment mice showed a reduced neurotoxicity, with TH and DAT mRNA levels either not affected by MPTP or affected to a lesser extent in the midbrain and striatum when compared to vehicle treated animals. Possible anti-apoptotic activity of BFP was further studied in a dopamine-secreting neuroendocrine cell line, PC12. In this assay, MPTP elevated the expression of a pro-apoptotic gene, Bax, while this expression was reduced by BFP pre-treatment. Flow cytometry results also revealed that the effect of MPTP-induced apoptosis in PC12 cell lines was significantly reduced by BFP. The present results suggest that BFP is able to protect dopaminergic neurons from neurotoxin-induced neuronal injury with anti-apoptotic activity being one of the possible mechanisms.  相似文献

A novel strategy for a more efficient synthesis of difficult sequence‐containing peptides, the S‐acyl isopeptide method, was developed and successfully applied. A model pentapeptide Ac–Val–Val–Cys–Val–Val–NH2 was synthesized via its water‐soluble S‐acyl isopeptide using an S‐acyl isodipeptide unit, Boc–Cys(Fmoc–Val)–OH. An S‐acyl isopeptide possessing excellent water solubility could be readily and quantitatively converted to the native peptide via an SN intramolecular acyl migration reaction at pH 7.4. Question: In stressful abiotic environments positive plant interaction is expected to be a frequent and an important process driving community composition and structure. In the high Andes in central Chile, the cushion plant Azorella madreporica dominates plant communities and appears to benefit the assemblage of species that grows within it. However, there are also many other species that grow outside this nurse cushion plant, which may or may not interact with this species. What is the prevailing type of spatial associations among the plant species that are not growing inside the nurse plant? What is the type of interactions between cushion plants and those species growing outside them? Location: Molina River basin (33°20'S, 70°16’ W, 3600 m a.s.l.), in the Andes of central Chile, ca. 50 km east of Santiago. Methods: Two accurate mapping plots of individual plants of different species were located at two summits (Franciscano and Tres Puntas sites). The spatial distributions and associations between species growing outside cushions and within cushions at each site were estimated by point‐pattern analyses using the univariate and bivariate transformations of Ripley's K‐functions. Results: We found both positive and, especially, negative spatial associations (8 out of 12 species in Franciscano site) between A. madreporica cushions and plants growing outside them. However, most of the species showed positive spatial associations among them. The variation in spatial association was site‐specific and also depended on the type of plants involved. Adesmia spp., the second most abundant non‐cushion species, displayed negative associations with cushions and positive associations with other species growing outside cushions. Conclusions: Our study suggests very complex interactions among species, which ranged from positive to negative, and are also affected by abiotic environmental conditions.  相似文献

BACKGROUND: Lithium (Li) has been associated with cardiac teratogenicity in the developing fetus. We took advantage of the association of therapeutic administration of Li with an increase in heart defects to gain insight into both normal and pathological heart and valve development with GSK‐3 inhibition. The objective of this study was to define whether Li mimicry of canonical Wnt/β‐catenin signaling induces cardiac valve defects. METHODS: Li was administered by a single intraperitoneal injection to the pregnant mouse on embryonic day E6.75, much earlier than heretofore analyzed. On E15.5 developing heart defects were defined by Doppler ultrasound. The embryonic hearts were analyzed for changes in patterning of active canonical Wnt expression and nuclear factor of the activated T cells‐c1 (NFATc1), both key regulators of valve development. Li‐exposed chick embryos were used to define the early cell populations during gastrulation that are susceptible to GSK‐3 inhibition and may relate to valve formation. RESULTS: Li exposure during gastrulation decreased the number of prechordal plate (PP) cells that reached the anterior intestinal portal, a region associated with valve development. Li decreased expression of Hex, an endoderm cardiac inducing molecule, normally also expressed by the PP cells, and of Sox 4 at the anterior intestinal portal and NFAT, critical factors in valvulogenesis. CONCLUSIONS: Cells existing already during gastrulation are associated with valve formation days later. The Wnt/β‐catenin signaling in PP cells is normally repressed by Wnt antagonists and Hex is up‐regulated. The antagonism occurring at the receptor level is bypassed by Li exposure by its intracellular inactivation of GSK‐3 directly to augment Wnt signaling. The fruit fly Drosophila embryo is one of the most important model organisms in genetics and developmental biology research. To better understand the biomechanical properties involved in Drosophila embryo research, this work presents a mechanical characterization of living Drosophila embryos through the stages of embryogenesis. Measurements of the mechanical forces of Drosophila embryos are implemented using a novel, in situ, and minimally invasive force sensing tool with a resolution in the range of microN. The measurements offer an essential understanding of penetration force profiles during the microinjection of Drosophila embryos. Sequentially quantitative evaluation and analysis of the mechanical properties, such as Young's modulus, stiffness, and mechanical impedance of living Drosophila embryos are performed by extracting the force measurements throughout the stages of embryogenesis. Experimental results illustrate the changing mechanical properties of Drosophila embryos during development, and thus mathematical models are proposed. The evaluation provides a critical step toward better understanding of the biomechanical properties of Drosophila embryos during embryogenesis, and could contribute to more efficient and significant genetic and embryonic development research on Drosophila.  相似文献
