This work studied the biodegradation of petroleum hydrocarbons (PHCs) extracted from refinery wastewater to produce industrially important by-products at different incubation periods. Two out of 13 bacterial isolates, KRD2 and KRA4 were isolated. Dichloromethane was used to extract the PHC, and gas chromatography-mass spectrometry (GC-MS) analysis revealed that the refinery wastewater PHC was successfully biodegraded using the selected bacterial isolates within 15 days of incubation. Both KRD2 and KRA4 isolates degraded all 13 initially extracted PHC compounds within 5 days, except C13BD and C9BD, which produced 6 and 4 compounds as secondary metabolites with peak area percentages of 1.58, 1.38, 0.85, 29.94, 7.59, and 11.16% and 3.55, 2.88, 52.31, and 6.14%, respectively. These metabolites have been reported in industrial and medical applications. After 10 days, only 6 and 8 compounds were degraded by both isolates, respectively, and C11PAD compound was produced, as well as C5PAD, C7PAD, and C13PAD. After 15 days, it was clear that all the initial PHC compounds have been completely degraded by both isolates. Metabolites C5PAD, C6PAD, C8PAD, and C13PAD were produced by KRD2, and metabolites C5PAD, C6PAD, C8PAD, and C9PAD were produced by KRA4 at different peak areas. The alignment revealed that the KRA4 isolate was included in the genus Chryseobacterium gambrini, while KRD2 isolate was successfully identified as Mycobacterium confluentis using the Biolog microbial identification system. The incubation period evidently affected biodegradation process by indigenous degraders. These effective bacteria were shown to be of great potential for further application in biodegradation technology of PHC contaminated refinery wastewater to produce industrially important by-products.  相似文献

Gas chromatography-mass spectrometry (GC-MS) can be applied to detect and characterize microorganisms in clinical and environmental samples, and microbial contaminants in biotechnological production cultures. With this approach, unique microbial monomeric compounds, known as chemical markers, are used as analytes. In the present article, two GC-MS-based techniques, viz. GC-ion trap tandem MS (GC-MS-MS) and conventional quadrupole GC-MS used in the selected ion monitoring mode, were compared regarding their ability to detect 3-hydroxy fatty acids, muramic acid, and ergosterol (markers for endotoxin, peptidoglycan, and fungal biomass, respectively) in complex matrices. When using GC-MS-MS, daughter ion spectra were obtained for all markers present in amounts close to the detection limit of the GC-MS. Ion-trap GC-MS-MS shows great promise as a chemical marker analysis technique for application in clinical diagnosis, occupational and public health care, and biotechnology.  相似文献

Analogs of 4-androstene-3,17-dione and testosterone were tested as substrates for the aromatizing enzyme complex of human placenta. Compounds modified in rings B, C and D were found to be aromatized via a pathway similar to that postulated for 4-androstene-3,17-dione and testosterone, in which oxidation to the 19-hydroxy and 19-oxo (or corresponding gem-diol) intermediates occurs. Gibberellins A8, A16, A24, and abscisic acid were identified by GC-MS of derivatized extracts from immature fruits of Secale cereale. Mature caryopses contained ABA and trans-ABA in a ratio 1:1 as well as 4′-dihydrophaseic acid. During milk ripeness a neutral GA conjugate was detected. Free GA, afforded by enzymatic hydrolysis of the conjugate, was chromatographically identified as GA16  相似文献

首次采用顶空固相微萃取结合气质联用技术提取并分析了贴梗海棠花蕾和花的挥发油成分。结果发现,邻苯二甲酸二(2-乙基己基)酯在花蕾开放的过程中,含量从24.67%下降到1.77%,而己二酸二(2-乙基己基)酯的含量则从0%增加到47.23%,表明在贴梗海棠花蕾开放的过程中,在植物体内多种酶的共同作用下,二者存在着生物转化,依据此结果,我们提出邻苯二甲酸二(2-乙基己基)酯与二酸二(2-乙基己基)酯在贴梗海棠开放过程中生物转化假说。  相似文献

A gas chromatography-mass spectrometry assay method for the analysis of lauric, myristic, and palmitic acids and their omega and omega(-1) hydroxylated metabolites from in vitro incubations of cytochrome P450 CYP4A1, involving solid-phase extraction and trimethysilyl derivatization, was developed. The assay was linear, precise, and accurate over the range 0.5 to 50microM for all the analytes. It has the advantages of a more rapid analysis time, an improved sensitivity, and a wider range of analytes compared with other methods. An artificial membrane system was optimized for application to purified CYP4A1 enzyme by investigating the molar ratios of cytochrome b(5) and cytochrome P450 reductase present in the incubation mixture. Using this method, the kinetics of omega and omega(-1) oxidation of lauric, myristic, and palmitic acids by CYP4A enzymes were measured and compared in rat liver microsomes and an artificial membrane system.  相似文献

采用水蒸气蒸馏法分别从翼蓼和中华抱茎蓼中提取其挥发油,并用气相色谱-质谱联用技术(GC-MS)对挥发油中的化学成分进行分离和结构鉴定,运用气相色谱峰面积归一化法确定各成分的相对百分含量.结果表明:翼蓼中总共鉴定出31种成分,中华抱茎蓼中鉴定出21种成分,分别占到挥发油总含量的84.0%和93.8%,翼蓼挥发油中主要成分是表蓝桉醇(15.9%)、甲苯(8.6%)、反式橙花叔醇(6.9%);中华抱茎蓼挥发油主要成分为邻苯二甲酸二异丁酯(19.9%)、2,4-戊二酮(17.2%)、邻苯二甲酸二丁酯(11.7%)、3-甲基-2,3-二氢苯并呋喃(10.7%).  相似文献

采用水蒸汽蒸馏法从大戟科蝴蝶果茎中提取挥发油,用气相色谱-质谱联用技术对挥发油化学成分进行分析。分离出36个峰,鉴定出35种化合物,占总油量的98.34%,并应用面积归一化法测定各成分的相对百分含量。其主要成分为十六烷酸乙酯(13.19%)、正十六烷酸(11.11%)、十八碳烯酸乙酯(6.18%)、正十八烷(4.98%)、(Z,Z)-9,12-十八碳二烯酸(4.90%)及十八碳二烯酸乙酯(4.21%)。  相似文献

Mimivirus is one the largest DNA virus identified so far, infecting several Acanthamoeba species. Analysis of its genome revealed the presence of a nine-gene cluster containing genes potentially involved in glycan formation. All of these genes are co-expressed at late stages of infection, suggesting their role in the formation of the long fibers covering the viral surface. Among them, we identified the L136 gene as a pyridoxal phosphate-dependent sugar aminotransferase. This enzyme was shown to catalyze the formation of UDP-4-amino-4,6-dideoxy-D-glucose (UDP-viosamine) from UDP-4-keto-6-deoxy-D-glucose, a key compound involved also in the biosynthesis of L-rhamnose. This finding further supports the hypothesis that Mimivirus encodes a glycosylation system that is completely independent of the amoebal host. Viosamine, together with rhamnose, (N-acetyl)glucosamine, and glucose, was found as a major component of the viral glycans. Most of the sugars were associated with the fibers, confirming a capsular-like nature of the viral surface. Phylogenetic analysis clearly indicated that L136 was not a recent acquisition from bacteria through horizontal gene transfer, but it was acquired very early during evolution. Implications for the origin of the glycosylation machinery in giant DNA virus are also discussed.  相似文献

用水蒸气蒸馏法提取陕西汉中、贵州、福建、江西4个产地的吴茱萸和汉中产小花吴茱萸的果实挥发油,并用气相色谱-质谱法(GC-MS)共鉴定出154种化学成分,主要为萜类、酯类及酰胺类等化合物。其中陕西汉中产吴茱萸果实挥发油中鉴定出48种化学成分,未知成分2种。主要成分有反式-罗勒烯(相对含量为75.05%,下同)、顺式-罗勒烯(8.10%)、β-香叶烯(6.14%)等;贵州产吴茱萸果实挥发油中鉴定出43种化学成分,未知成分3种,其主要成分有反式-罗勒烯(67.04%)、β-香叶烯(9.66%)、顺式-罗勒烯(7.98%)等;福建产吴茱萸果实挥发油中鉴定出57种化学成分,未知成分2种,主要成分有反式-罗勒烯(73.14%)、顺式-罗勒烯(8.41%)、β-香叶烯(3.82%)等;江西产吴茱萸果实挥发油中鉴定出61种化学成分,主要种类有β-香叶烯(33.49%)、反式-罗勒烯(30.27%)、β-水芹烯(18.86%)、顺式-罗勒烯(5.23%)等;陕西汉中产小花吴茱萸果实挥发油中鉴定出化学成分83种,未知成分3个,其主要成分为反式-罗勒烯(40.21%)、顺式-罗勒烯(8.99%)、β-香叶烯(6.74%)等。结果表明,不同产地吴茱萸及小花吴茱萸果实中挥发油的主要成分种类比较接近,但各自挥发油化合物组成又都含有其特有化学成分。  相似文献
