Plant secondary metabolites are known to facilitate interactions with a variety of beneficial and detrimental organisms, yet the contribution of specific metabolites to interactions with fungal pathogens is poorly understood. Here we show that, with respect to aliphatic glucosinolate‐derived isothiocyanates, toxicity against the pathogenic ascomycete Sclerotinia sclerotiorum depends on side chain structure. Genes associated with the formation of the secondary metabolites camalexin and glucosinolate were induced in Arabidopsis thaliana leaves challenged with the necrotrophic pathogen S. sclerotiorum. Unlike S. sclerotiorum, the closely related ascomycete Botrytis cinerea was not identified to induce genes associated with aliphatic glucosinolate biosynthesis in pathogen‐challenged leaves. Mutant plant lines deficient in camalexin, indole, or aliphatic glucosinolate biosynthesis were hypersusceptible to S. sclerotiorum, among them the myb28 mutant, which has a regulatory defect resulting in decreased production of long‐chained aliphatic glucosinolates. The antimicrobial activity of aliphatic glucosinolate‐derived isothiocyanates was dependent on side chain elongation and modification, with 8‐methylsulfinyloctyl isothiocyanate being most toxic to S. sclerotiorum. This information is important for microbial associations with cruciferous host plants and for metabolic engineering of pathogen defenses in cruciferous plants that produce short‐chained aliphatic glucosinolates.  相似文献

Three new species of Hypomyces with KOH-negative subiculi occurring on the Aphyllophorales are described. Hypomyces pseudocorticiicola is characterized by a grayish-yellow to brown subiculum, fusiform ascospores, always with one septum, and its Cladobotryum-anamorph. Hypomyces laeticolor is characterized by a bright-colored subiculum, small apiculi on the ascospores, and well-developed verticillate conidiogenous cells. Hypomyces penicillatus forms a Trichothecium-like anamorph with well-developed verticils of conidiogenous cells.  相似文献

Dry bubble disease caused by the fungal pathogen Verticillium fungicola1 is responsible for large losses to the mushroom (Agaricus bisporus) industry. The pathogen induces various symptoms on the host, bubbles (undifferentiated spherical masses), bent and/or split stipes (blowout) and spotty caps. Inoculation of A. bisporus crops with isolates of V. fungicola var. fungicola of various degrees of aggressiveness showed that the more aggressive isolates induced higher numbers of bubbles. The production of other symptoms did not vary with the isolate of pathogen. The total weight of the crop (healthy and diseased mushrooms) was not significantly affected by the disease, but inoculation with highly aggressive isolates resulted in a significant increase in the total numbers of mushrooms. Two hypotheses are proposed to explain the effect of the pathogen on fruiting initiation in relation to aggressiveness.  相似文献

Abstract: The Lichinales are a group of lichenized ascomycetes that almost exclusively possess cyanobacteria as their primary photobiont and are hitherto separated from the Lecanorales, the major group of lichenized ascomycetes, by thallus structure, ascoma ontogeny, ascus structure and ascus function. The relationship of the two families Peltulaceae and Lichinaceae, both placed within the Lichinales, with the Heppiaceae, placed within the Lecanorales, was investigated, as well as a possible sister group relationship of the Lichinales to the Lecanorales. Phylogenetic analyses included non-molecular data as well as 18S rDNA sequence data. The monophyly of the Lichinales including the family Heppiaceae and a sister group relationship of Lichinales and Lecanorales, based on the shared presence of lecanoralean asci, are proposed in a morphological hypothesis. Parsimony and distance analyses of 18S rDNA sequence data strongly support the monophyly of the Lichinales, including all three families. Therefore, the presence of rostrate, lecanoralean asci in Peltula and part of the Lichinaceae suggests that this ascus type is an autapomorphy of the monophyletic Lichinales. Furthermore, the occurrence of prototunicate asci in the Heppiaceae and most of the Lichinaceae is autapomorphic and was gained independently by reduction of the rostrate ascus. The 18S rDNA analysis did not reject the non-molecular hypothesis of a sister group relationship of the Lichinales and the Lecanorales as based on ascus characters. The alternative placement of the Lichinales as the sister group of all inoperculate euascomycetes excluding the Sordariomycetes and most of the Leotiales in the gene tree received unsufficient bootstrap support and no support from any non-molecular data and consequently was rejected.  相似文献

Recent finds of Leightoniella zeylanensis, classified variously in the Collemataceae and Pannariaceae, enabled us to generate DNA sequence data for investigating its phylogenetic affiliation. Newly generated sequence data from the internal transcribed spacer (ITS) region and the large subunit of the nuclear ribosomal DNA (nrLSU), the small subunit of the mitochondrial ribosomal (mrSSU) DNA, and the largest subunit of the RNA polymerase II gene (RPB1) indicate that L. zeylanensis is a member of the Pannariaceae, belonging to a strongly supported clade together with Physma, Lepidocollema, and Gibbosporina (= the ‘Physma clade’). With the currently available data, however, relationships within this clade are largely impossible to reconstruct with confidence. Leightoniella zeylanensis was found to possess ellipsoid ascospores surrounded by a thick, gelatinous perispore with pointed ends, supporting a previously published hypothesis that such a perispore type is a synapomorphy for the Physma clade. A lectotype is designated for the basionym Pterygium zeylanense Leight.  相似文献

The lichenised ascomycete, Collema leptosporum Malme, was extracted with aqueous methanol to give traces of mannitol and 3-O-β- -glucopyranosyl- -mannitol (2.7% yield). The residue was consecutively extracted with hot water to give a complex uronic acid-containing polysaccharide, and then with hot aqueous alkali which provided a mixture of polysaccharides. This was fractionated with Cetavlon to give a branched galactomannan, which had the lowest content of galactose yet reported for such a lichen polysaccharide. It has a main chain of (1→6)-linked -Manp units partly substituted at O-2,4 by non-reducing end-units of Manp and Galp, shown by NMR spectroscopy to have - and β-configurations, respectively. The other polysaccharide component was unexpectedly a branched (1→3), (1→6)-linked β-glucan, which is typical of a basidiomycete, whereas those of ascomycetes contain similar linkages but in linear glucans.  相似文献

A new monotypic ascomycete genus, Dictyocyclns gen. nov. with D. hydrangeae sp. nov. as its type, is described and illustrated. It is placed in the family Parmulariaceae and it is the second genus in that family with dictyoseptate ascospores. It is compared with Aldonata the only other member of the family with similar ascospores.  相似文献
