研究利用大型环境生长箱模拟了两种沙地优势灌木柠条和羊柴对 CO2 浓度倍增和土壤干旱交互作用的响应。 CO2 浓度倍增使柠条和羊柴的生物量分别增加了 6 2 .90 %和 5 0 .0 0 % ,使植株叶面积分别增加了 4 1.86 %和 4 5 .84 %。 CO2 浓度的倍增效应随着土壤干旱的增加而下降。 CO2 浓度倍增和土壤干旱都增加单位叶面积质量 (L MA) ,但 CO2 浓度倍增主要增加了水分充足时的 L MA。 CO2 倍增使柠条和羊柴叶片含氮量分别降低了 10 .4 0 %和 5 .0 6 %。柠条叶片含氮量在所有土壤干旱条件下均呈现出增加的趋势 ,而羊柴叶片的含氮量仅在严重干旱条件下增加。 CO2 倍增使叶片的碳氮比显著增加 ,但土壤干旱使之降低。CO2 浓度倍增降低叶肉细胞质膜的过氧化产物丙二醛 (MDA )的含量 ,干旱使之增加。叶片含氮量与 MDA呈显著正相关。研究表明 CO2 倍增有保护叶片免受严重土壤干旱的作用 ,但干旱的负面影响是 CO2 倍增效应所难以弥补的  相似文献

白藜芦醇是一种具有多种医疗保健作用的植物芪类次生代谢产物,在农业、医药、食品和化妆品等领域受到广泛的关注。白藜芦醇合酶是白藜芦醇生物合成中唯一必需的关键酶,决定植物体内白藜芦醇的合成。将中间锦鸡儿中克隆到的CiRS基因(Gen Bank登录号MF678590)转入野生型拟南芥,实验结果显示:野生型的总黄酮含量明显高于转基因株系。HPLC测得转基因拟南芥中有白藜芦醇的生成,并且含量最高达335μg/g FW。紫外照射处理后转基因植物中丙二醛的积累量明显少于野生型。转基因植物提取物DPPH自由基清除能力均高于野生型。这些结果表明,中间锦鸡儿CiRS基因异源表达后利用与黄酮类物质的共同底物合成了白藜芦醇,使得转基因植物的抗氧化性增强。  相似文献

Exploring optimum and cost-efficient medium composition for microbial growth of Candida intermedia Y–1981 yeast culture growing on whey was studied by applying a multistep response surface methodology. In the first step, Plackett–Burman (PB) design was utilized to determine the most significant fermentation medium factors on microbial growth. The medium temperature, sodium chloride and lactose concentrations were determined as the most important factors. Subsequently, the optimum combinations of the selected factors were explored by steepest ascent (SA) and central composite design (CCD). The optimum values for lactose and sodium chloride concentrations and medium temperature were found to be 18.4 g/L, 0.161 g/L, and 32.4°C, respectively. Experiments carried out at the optimum conditions revealed a maximum specific growth rate of 0.090 1/hr; 42% of total lactose removal was achieved in 24 h of fermentation time. The obtained results were finally verified with batch reactor experiments carried out under the optimum conditions evaluated.  相似文献

Summary Castrated adult male hamsters and castrated adult female rats were injected with either 0.2 g (hamsters) or 0.5 g (rats) of 3H-dihydrotestosterone (107 Ci/mmole)/100 grams body weight and killed 11/2 h later. The pituitary glands were removed and processed for both autoradiography and immunocytochemistry (hamster) or only autoradiography (rats). Localization of the androgen was found in 10–15% of the cells of the pars distalis in both species. Only cells that stained for luteinizing hormone (LH) in the hamster's pars distalis concentrated the androgen. Also cells in both the pars intermedia and pars nervosa (1–5%) concentrated the androgen in both species. Although the number of cells that concentrated the androgen in the pars intermedia and pars nervosa was small, this finding may be related to recent physiologic data that suggest that the gonadal steroids may play a role in regulating water retention and natriuresis.This study was supported by USPHS Research Career Development Award KO4NS0000164 (P.J. We have investigated the effects of EA21a and EA34 mammary carcinomas on daily PI cell proliferation in mice. Animals were divided into groups grafted with either EA34 or EA21a carcinomas (and a non-grafted control group). They were all injected intraperitoneally with 2 microg colchicine per g of body weight 4 h before sacrifice and the number of mitoses per 1000 nuclei was calculated. The mitotic index (MI) of pars-intermedia epithelial cells in control animals showed significant temporal variations. However, the MI from mice grafted with EA34 or EA21a carcinomas showed no such variation. There was no difference between the daily MIs of controls and tumor grafted groups. The absence of a 24 h mitotic activity curve in both EA21a and EA34 tumor-bearing animals demonstrates a lower level of synchronization of cells entering mitosis.  相似文献

Abstract Until recently, black-pigmented Gram-negative anaerobes were classified as ‘black-pigmented Bacteroides ’. At present, 11 distinct species are recognized in this group. Because of major differences with Bacteroides fragilis , the type species of the genus Bacteroides , new genera have been proposed: Porphyromonas for three asaccharolytic species, and Prevotella for the saccharolytic species. Typing methods have been developed for some species of black-pigmented Gram-negative anaerobes. These include biotyping and serotyping, but relatively few types can be distinguished with these methods. Recently, DNA restriction endonuclease analysis has been used for typing of P. gingivalis, Pr. intermedia and P. endodontalis strains. Great heterogeneity was observed within all three species. This typing method can be useful for epidemiological studies.  相似文献

《中国古生代植物》首次将Gigantopteris这一属名限定在一些具重网状叶脉的大羽羊齿类植物,如真正的G.nicotmnaetblia等,而叶脉简单结网的一些植物则大多被归入Gigantonoclea属。这一比较简便的做法近年也开始受到质疑。Gigantopteris fukienensis Yabe et Oishi,1938也是因为叶脉简单结网而被改归于Gigantopteris属内。问题在于G.fukienensis的脉式与华北典型的Gigantonoclea迥异,它的三级脉以合轴二歧式一等二歧式分叉,细脉彼此间只是偶尔联结,而主要是借助与缝脉相交形成伸长形的网眼。加之,网眼内不具点痕,而每一网眼内有1—2个点痕则是华北典型Gigantonoclea的特征之一。根据叶脉特征,作者为一些三级脉以等二歧式一合轴二歧式分叉、细脉仅偶尔相互联结而主要借助与相邻脉束或缝脉相交而形成网眼、网眼内不具点痕的一些相关植物建立了新属Fujmnopteris gen.nov.,并对此属的4个种(包括1新种)作了种的特征描述和有关的讨论和比较。  相似文献

We previously reported daily variations in the mitotic activity of the endocrine cells in the pars intermedia of 21- and 28-day-old male mice. Since cellular proliferation might be affected by factors such as sex and age, we undertook the present experiments to study the mitotic activity of the pars intermedia from 14-, 28-, and 150-day-old female mice. Inbred C3H/S mice, grouped according to age, were housed under standard conditions (12h each of light and dark [LD 12:12]) for periodicity analysis and were killed in lots of 5-11 animals every 4h over a single 24h cycle, with each mouse receiving 2 μg/g of colchicine 4h before decapitation. Pituitaries were excised, extracted, fixed in buffered formaldehyde, embedded in celloidin-paraffin, sectioned at 5 μm, and stained with hematoxylin and eosin. We counted the total number of nuclei to estimate the total number of cells monitored and then calculated the mitotic index (metaphases/1000 nuclei). Differences were analyzed for statistical significance by the Student t test. While the 14-day-old animals manifested no significant changes in mitotic activity during the 24h cycle, the 28- and 150-day-old mice showed higher mitotic indices during the period of darkness. The average mitotic activity over the entire cycle, however, was higher in the two groups of younger animals than in the 150-day-old mice. Moreover, the averages for the 28-day-old females were higher than the corresponding values previously reported by us for male mice of the same age. 脱水素(DHNs, dehydrins)是LEA II亚家族蛋白,在种子发育后期大量积累,并受不同逆境胁迫处理诱导表达。在中间锦鸡儿干旱胁迫抑制性削减杂交文库中筛选到了一段 DHN1 序列,利用RACE技术克隆获得基因全长序列。序列比对分析表明, CiDHN1 具有开放阅读框891bp,起始密码子为ATG,终止密码子为TAG,编码297个氨基酸,含有5个Y片段和一个K片段,与CiDHN1相似性最高的为山茱萸科主教红端木(Cornus sericea),相似性为39%。系统进化分析显示CiDHN1单独聚为一支,推测该蛋白为新的功能未知蛋白。亚细胞定位结果表明CiDHN1定位在细胞质、质膜和细胞核。荧光定量PCR结果显示 CiDHN1 的表达受冷、脱水和NaCl等非生物胁迫的诱导。 CiDHN1 过量表达拟南芥后,其中表达量最强的株系对200 mmol/L的NaCl处理较为敏感。CiDHN1在中间锦鸡儿抵抗逆境胁迫中的功能有待于进一步研究。  相似文献

Summary We tested the hypothesis that acute restraint stress results in ultrastructural evidence for enhanced release of alpha-melanocyte-stimulating hormone (-MSH) and -endorphin from the intermediate lobe (IL) of the rat pituitary. Measurements of plasma -MSH-and -endorphin-immunoreactivity (ir) were used to confirm ultrastructural findings. Plasma -MSH-ir was elevated after 20 and 30 min of restraint while plasma -endorphin-ir peaked 10 min after the onset of restraint. Ultrastructural analysis revealed a decrease in the content of secretory granules within IL cells of stressed rats. Analysis of Golgi-related immature secretory granules in IL cells indicated that new peptide synthesis was not enhanced after 30 min of restraint. These results confirm previous studies showing and elevation of plasma -endorphin and -MSH-ir during acute restraint. Furthermore, these results indicate that quantitative analysis at the ultrastructural level can be used to assess peptide release from IL secretory cells during stress.  相似文献
