目前尚无有效可靠的抗应激药物.传统的补益中药六味地黄汤(LW)对多种类型应激所致内分泌和免疫平衡失调具有明显调节作用,但目前已上市的LW成药缺乏可靠的质量控制方法,其临床疗效的可靠程度难以估测.本课题组在前期研究中以免疫和内分泌活性评价为导向,从LW中追踪分离并组成了质量可控且安全性好的六味地黄苷糖(LW-AFC).本文观察了悬吊应激雌性小鼠生殖内分泌及免疫系统的变化及LW-AFC的影响.结果表明,悬吊应激小鼠皮质酮升高,动情期缩短、动情间期延长,垂体LH下降,动情间期血清E2升高;脾细胞培养上清IgG含量明显降低;口服LW-AFC能显著降低血清皮质酮,升高垂体LH,且LW-AFC能明显降低动情间期血清E2,并能明显增强脾细胞IgG分泌能力.这提示,悬吊应激可导致雌性小鼠生殖内分泌功能紊乱以及免疫功能下降,口服LW-AFC具有明显调节作用,对应激所致生殖内分泌功能紊乱以及免疫功能下降具有潜在防治作用. 相似文献

Cnidarians, in contrast with bilaterians, are generally considered to exhibit radial symmetry around a single body axis (oral-aboral) throughout their life-cycles. We have investigated how the oral-aboral axis is established in the hydrozoan jellyfish Podocoryne carnea. Vital labeling experiments showed that the oral end of the blastula derives from the animal pole region of the egg as has been demonstrated for other cnidarian species. Gastrulation is restricted to the oral pole such that the oral 20% of blastula cells give rise to endoderm. Unexpectedly, bisection experiments at the 8-cell stage showed that animal regions are able to develop into normally polarized larvae, but that vegetal (aboral) blastomeres completely fail to develop endoderm or to elongate. These vegetal-derived larvae also failed to polarize, as indicated by a lack of oral-specific RFamide-positive nerve cells and a disorganized tyrosinated tubulin-positive nerve net. A different result was obtained following bisection of the late blastula stage: aboral halves still lacked the capacity to develop endoderm but retained features of axial polarity including elongation of the larva and directional swimming. These results demonstrate for the first time in a cnidarian the presence of localized determinants responsible for axis determination and endoderm formation at the animal pole of the egg. They also show that axial polarity and endoderm formation are controlled by separable pathways after the blastula stage. 相似文献

We have isolated the ten Hox genes from the pill millipede Glomeris marginata (Myriapoda:Diplopoda). All ten genes are expressed in characteristic Hox-gene-like expression patterns. The register of Hox gene expression borders is conserved and the expression profiles show that the anterior-most limb-bearing segment in arthropods (antennal/cheliceral segment) does not express any Hox gene, while the next segment (intercalary/second-antennal/premandibular/pedipalpal segment) does express Hox genes. The Hox expression patterns in this millipede thus support the conclusion that all arthropods possess a deuterocerebral segment. We find that there is an apparent posterior shift of Hox gene expression domains dorsally relative to their ventral patterns, indicating that the decoupling of dorsal and ventral segmentation is not restricted to the level of segment polarity genes but apparently includes the Hox genes. Although the mechanism for the decoupling of dorsal and ventral segmentation remains unsolved, the decoupling must be at a level higher in the hierarchy than that of the segment polarity and Hox genes. The expression patterns of Ultrabithorax and abdominal-A suggest a correlation between the function of these genes and the delayed outgrowth of posterior trunk appendages. This delay may be caused by an assumed repressor function of Ultrabithorax, which might partially repress the activation of the Distal-less gene. The Glomeris
fushi tarazu gene is expressed in a Hox-like domain and in the developing central nervous system, but not in segmental stripes such as has been reported in another myriapod species, the centipede Lithobius. The oral-aboral (OA) axis in the sea urchin is specified by the TGFβ family members Nodal and BMP2/4. Nodal promotes oral specification, whereas BMP2/4, despite being expressed in the oral territory, is required for aboral specification. This study explores the role of Chordin (Chd) during sea urchin embryogenesis. Chd is a secreted BMP inhibitor that plays an important role in axial and neural specification and patterning in Drosophila and vertebrate embryos. In Lytechinus variegatus embryos, Chd and BMP2/4 are functionally antagonistic. Both are expressed in overlapping domains in the oral territory prior to and during gastrulation. Perturbation shows that, surprisingly, Chd is not involved in OA axis specification. Instead, Chd is required both for normal patterning of the ciliary band at the OA boundary and for development of synaptotagmin B-positive (synB) neurons in a manner that is reciprocal with BMP2/4. Chd expression and synB-positive neural development are both downstream from p38 MAPK and Nodal, but not Goosecoid. These data are summarized in a model for synB neural development. 相似文献

Abstract: Many studies of interactions between exotic and native ungulates have not had temporal and spatial controls nor have they considered the types of competitive interactions that would allow coexistence. For exotic axis deer (Axis axis) and native white-tailed deer (Odocoileus virginianus) to coexist one species should be superior at interference competition and the other species should be superior at exploitative competition. We generated and tested predictions, based on body size and diet breadth, about habitat selection by white-tailed deer in the presence and absence of axis deer, dominance relationships, and time at sites provisioned with high quality forage. We conducted our study in treatment (axis and white-tailed deer) and control (white-tailed deer only) areas when both species were present and after axis deer were removed. We conducted vehicle surveys to determine habitat use of both species. At provisioned feeding sites we recorded aggressive behaviors and amount of time species spent at feeding sites alone and together. In the treatment area white-tailed deer selection for wooded habitat increased 2.1 times after axis deer were removed, whereas habitat selection by white-tailed deer was constant in the control area over the same time. At feeding sites axis deer were dominant to white-tailed deer; both species spent a significantly greater amount of time alone than at feeders together, but amount of time that individuals of each species spent at feeders did not differ. Axis deer were superior at interference competition, but white-tailed deer were not superior at exploitative competition; thus, species coexistence is unlikely. Whether white-tailed deer are negatively impacted by axis deer at spatial scales larger than our experiment probably depends on abundance of axis deer at larger spatial scales. Experiments of species interactions with temporal and spatial controls that consider types of competitive interactions increase a manager's understanding of when and how native ungulates may be negatively impacted by exotic ungulates. 相似文献

Insect embryogenesis is best understood in the fruit fly Drosophila. However, Drosophila embryogenesis shows evolutionary-derived features: anterior patterning is controlled by a highly derived Hox gene bicoid, the body segments form almost simultaneously and appendages develop from imaginal discs. In contrast, embryogenesis of the
red flour beetle Tribolium castaneum displays typical features in anterior patterning, axis and limb formation shared with most insects, other arthropods as well
as with vertebrates. Anterior patterning depends on the conserved homeobox gene orthodenticle, the main body axis elongates sequentially and limbs grow continuously starting from an appendage bud. Thus, by analysing
developmental processes in the beetle at the molecular and cellular level, inferences can be made for similar processes in
other arthropods. With the completion of sequencing the Tribolium genome, the door is now open for post-genomic studies such as RNA expression profiling, proteomics and functional genomics

The common ivy (Hedera helix L.) remains juvenileat its northern, eastern and altitudinal distribution limits although juvenileparts are largely killed by severe frost spells. In order to explain thisdiscrepancy we investigated the seasonal course of frost resistance in variousorgans of juvenile and adult parts of the same H. helixplants. Maximum frost resistance of leaves (LT50–25°C) and axis (xylem parenchyma:LT50 –29°C; cambium: LT50–35°C) was quite the same in juvenile and adultparts. Thus, H. helix is able to acquire full frostresistance in its juvenile phase. However, hardening of leaves was slower anddehardening of axis was faster in juvenile parts. Leaves of juvenile partsremain 2 to 4 K less resistant than those of adult parts untilattaining the maximum resistance. This explains why mainly leaves of thejuvenile parts were damaged following severe frost episodes with temperaturesbelow –20°C. The occurrence of H.helix in its juvenile phase at the frost-caused distribution limitsmay be explained as follows: Leaves of juvenile plants may occasionally bekilled by severe frosts, but regeneration from dormant eyes enables survival.Loss of leaves may impede the change to the adult phase, but even if the plantsbecame adult frost killing of rest buds (2 to 3 K less resistantthan leaves) would induce rejuvenation. 相似文献
