【目的】蛋白样品的制备是获得良好双向凝胶电泳(2-DE)图谱的前提,建立合理的西花蓟马蛋白的双向电泳体系,获得分辨率较高、重复性较好的图谱,能够为后续的研究提供有力支撑。【方法】实验以西花蓟马成虫为实验材料,对比了饱和酚法、TCA/丙酮法和直接裂解法3种蛋白提取方法,从中选出最适宜双向电泳分析的一种蛋白提取方法。【结果】3种方法蛋白提取率差异显著,直接裂解法蛋白提取率最高,饱和酚法的蛋白提取率最低;3种方法的SDS-PAGE条带数差异不明显;TCA/丙酮法的双向凝胶图谱效果最好,蛋白点最多。【结论】TCA/丙酮法能够有效去除西花蓟马蛋白中的干扰物质,是最适合西花蓟马双向凝胶电泳的蛋白提取方法,为后续西花蓟马在蛋白组学方面的研究奠定了基础。 相似文献

The western flower thrips, Frankliniella occidentalis (Pergande) (Thysanoptera: Thripidae), is a major pest in glasshouses, where light and photoperiod are often manipulated to grow crops. The effect of five photoperiods (L0:D24, L6:D18, L12:D12, L18:D6, and L24:D0) on the behaviour of adult females was investigated in the laboratory at 25 °C. The amount of walking, pollen consumption, and oviposition all increased with increasing photophase. The three behaviours occurred at a low rate in continuous darkness, and are therefore not exclusively diurnal. Three out of 16 thrips (19%) showed weakly rhythmic walking activity in continuous darkness, with an average circadian period of 25.3 ± 1.5 h. It is proposed that the length of the photophase determines the amount of nutrient intake, which in turn determines oviposition rate and thus affects population increase. 相似文献

We studied the capacity of one species of predator to control two major pests of greenhouse crops, Western flower thrips (Frankliniella occidentalis (Pergande)) and the greenhouse whitefly (Trialeurodes vaporariorum (Westwood)). In such a one-predator–two-prey system, indirect interactions can occur between the two pest species, such as apparent competition and apparent mutualism. Whereas apparent competition is desired because it brings pest levels down, apparent mutualism is not, because it does the opposite. Because apparent competition and apparent mutualism occurs at different time scales, it is important to investigate the effects of a shared natural enemy on biological control on a time scale relevant for crop growth. We evaluated the control efficacy of the predatory mites Amblyseius swirskii (Athias-Henriot) and Euseius ovalis (Evans) in cucumber crops in greenhouse compartments with only thrips, only whiteflies or both herbivorous insects together. Each of the two predators controlled thrips, but A. swirskii reduced thrips densities the most. There was no effect of the presence of whiteflies on thrips densities. Whitefly control by each of the two predators in absence of thrips was not sufficient, yet better with E. ovalis. However, whitefly densities in presence of thrips were reduced dramatically, especially by A. swirskii. The densities of predators were up to 15 times higher in presence of both pests than in the single-pest treatments. Laboratory experiments with A. swirskii suggest that this is due to a higher juvenile survival and developmental rate on a mixed diet. Hence, better control may be achieved not only because of apparent competition, but also through a positive effect of mixed diets on predator population growth. This latter phenomenon deserves more attention in experimental and theoretical work on biological control and apparent competition. 相似文献

Highly polymorphic microsatellite markers can provide important demographic information on founder events and range expansion following initial introduction of invasive insect species. Six microsatellite loci were isolated from an enriched DNA library in order to study the invasion patterns of the western flower thrips, Frankliniella occidentalis. All loci tested were found to be polymorphic and successfully amplified in all individuals. The number of alleles per locus ranged from five to nine and heterozygosity ranged from 45 to 73%. Improved laboratory methods are described in detail for mass rearing of various thrips species, such as Frankliniella occidentalis, Frankliniella intonsa, Thrips palmi, Thrips tabaci (Thysanoptera: Thripidae) and a thrips parasitoid, Ceranisus menes (Hymenoptera: Eulophidae), using various foods. In one method, plant pollen and honey solution are used as food sources. In a second method, germinated broad bean seeds are used. Eggs, produced in large numbers in water, are collected by a suction funnel onto a filter paper and incubated in a Petri dish. Large numbers of larvae that hatch are collected by using food traps (plant pollen). Larvae can be reared on pollen or on germinated broad bean seeds until adult emergence without additional water and food. This method has been found useful for producing even-aged thrips at different densities (up to 500 larvae in a cage of 80 mm diameter) with relatively low mortality rates. Evaluation of this rearing method for F. intonsa, shows that during 2 weeks at 20 °C per 100 females more than 4000 females could be produced in the next generation. About 5 min per day is required to achieve this productivity of mass production. Western flower thrips, Frankliniella occidentalis (Pergande) (Thysanoptera: Thripidae), cause very large economic damage on a variety of field and greenhouse crops. In this study, plant resistance against thrips was introduced into transgenic potato plants through the expression of novel, custom-made, multidomain protease inhibitors. Representative classes of inhibitors of cysteine and aspartic proteases [kininogen domain 3 (K), stefin A (A), cystatin C (C), potato cystatin (P) and equistatin (EIM)] were fused into reading frames consisting of four (K-A-C-P) to five (EIM-K-A-C-P) proteins, and were shown to fold into functional inhibitors in the yeast Pichia pastoris. The multidomain proteins were expressed in potato and found to be more resistant to degradation by plant proteases than the individual domains. In a time span of 14-16 days, transgenic potato plants expressing EIMKACP and KACP at a similar concentration reduced the number of larvae and adults to less than 20% of the control. Leaf damage on protected plants was minimal. Engineered multidomain cysteine protease inhibitors thus provide a novel way of controlling western flower thrips in greenhouse and field crops, and open up possibilities for novel insect resistance applications in transgenic crops. 相似文献

In this study, the effects of the accumulation of cysteine protease inhibitors on the food preferences of adult female western flower thrips, Frankliniella occidentalis (Pergande), were investigated. Representative members of the cystatin and thyropin gene families (stefin A, cystatin C, kininogen domain 3 and equistatin) were expressed in potato (Solanum tuberosum) cv. Impala, Kondor and Line V plants. In choice assays, a strong time- and concentration-dependent deterrence from plants expressing stefin A and equistatin was observed. Cystatin C and kininogen domain 3 were not found to be active. All tested inhibitors were equally or more active than stefin A at inhibiting the proteolytic activity of thrips, but, in contrast with stefin A, they were all expressed in potato as partially degraded proteins. The resistance of cysteine protease inhibitors against degradation in planta by endogenous plant proteases may therefore be relevant in explaining the observed differences in the deterrence of thrips. The results demonstrate that, when given a choice, western flower thrips will select plants with low levels of certain cysteine protease inhibitors. The novel implications of the defensive role of plant cysteine protease inhibitors as both deterrents and antimetabolic proteins are discussed. 相似文献

Western flower thrips, Frankliniella occidentalis (Pergande) causes damage to plants when they are feeding. Also, this thrips species transmits Tomato spotted wilt virus (TSWV) during stylet penetration. We investigated the penetration behaviour (probing) of thrips on pepper leaves and on liquid diet by electrical penetration graph (EPG, DC-system) recording. In addition, we used high-magnification video observations to correlate EPG waveforms with the insect's posture, head movements, and muscle contractions. Also, EPGs were correlated with probing on liquid diets containing radio-active tracers to distinguish and quantify ingestion waveforms. The previously described waveforms P, Q, and R were distinguished and additionally, a new waveform 'S' was distinguished. Waveform P could be linked with mandibular leaf penetration, waveform Q presumably with insertion of the maxillary stylets, and waveform R with ingestion of cell contents, whereas waveform S could not be correlated with any behavioural activity. Histology of the feeding damage in pepper leaves shows that thrips ingests the contents of multiple cells per probe. 相似文献

Frankliniella occidentalis is an economically important invasive pest worldwide, which can damage various horticultural crops and ornamental plants. F. occidentalis was first intercepted in Kunming, Yunnan province in 2000, and first reported to establish a population in Beijing, China in 2003. Since then, this pest is currently distributed across tens of provinces in mainland China and cause increasingly serious damage and loss. To control this pest, invasion biology, monitoring, and integrated pest management have been generally and intensively studied for 15 years in China. Furthermore, western flower thrips (WFT) as an important invasive insect pest, the research achievements on WFT has contributed to the promotion of technological innovation and development for invasive alien species management strategies and techniques in China. This review provides an overview for research on the biology, ecology, prevention, and management of this pest during 15 years in China. Meanwhile, China's “4E action” strategy on F. occidentalis is also discussed in this review. 相似文献
