A microbial biofilm community was established over 971 days within gravel in an aquarium so as to model biofouling of an aquifer. When the water was allowed to evaporate slowly, white crystalline deposits, containing several carbonate and sulphate minerals including nesquehonite (MgCO3.3H2O), were seen at the highest points on the surface of the biofouled gravel. No such deposits occurred in regions lacking biofilms. These crystals appeared to originate from evaporation of dissolved salts which had migrated through the biofilm. Surfaceadherent microbial biofilms may conceivably provide a conduit for solute transport in porous media such as soils and aquifers.  相似文献

One of the first symptoms expressed by declining trees is reduced growth in stem diameter and length increment. The possibility of a relationship between length increment and crown thinning in beech (Fagus sylvatica L.) was investigated by developing a computer model to simulate first order branching patterns of the apical 2 m of monopodially branching beech trees, 70–100 years old, for a range of length increment rates. The model was based on values for branching angle, main axis and branch length increment, number of branches produced per year and branch mortality rates for six healthy and declining trees. Shoot growth rates in the apical 2 m of the sample trees ranged from about 5 cm/year (decline class 3) to 43 cm/ year (healthy). Simulations of branching patterns in the apical 2 m of trees growing at different rates indicated that, when growth rate exceeded about 20 cm/year, total first order branch length and area explored were independent of growth rate. When growth rates fell below this value there was a reduction in total area explored and first order branch length due primarily to the formation of fewer branches. More acute branching angles contributed to a reduction in the area explored. Growth rate-related crown thinning could increase the risk of bark necrosis and secondary pathogen infection during dry and/or hot spells.  相似文献

Tree decline and deaths have been observed among 15 to 20-year-old Scots pines (Pinus sylvestris L.) in a dry heath forest in southwestern Finland. The sudden decline in height growth, the dieback of leading shoots and the yellowing of needles in young shoots in the upper part of the tree are typical symptoms of the decline of these young pines. Needle ultrastructure and chemical composition of Scots pines with or without decline and fluctuations of them in different seasons were studied. Afflicted trees were found to suffer from a deficiency in calcium and magnesium with low concentration of foliar nitrogen and phosphorus observed in all the trees studied. Ultrastructural study revealed changes characteristic of different seasons and measured nutrient status of needles. A clear reduction of membrane system in chloroplasts, especially related to Mg deficiency, was observed in most samples. The symptoms related to N deficiency, the translucent appearance of the cytoplasm and chloroplast stroma, and the elongated chloroplasts, as well as swelling of mitochondria, indicating P deficiency, were also found in the needles sampled from this forest decline area. The present study showed that it is possible to detect specific nutrient deficiency symptoms in needle ultrastructure in field samples and for use as sensitive indicators of unbalanced nutrient status.  相似文献

The responses of three different soil microbial communities to the experimental application of 2, 4-dichlorophenoxyacetic acid (2, 4-D) were evaluated with a variety of molecular genetic techniques. Two of the three soil communities had histories of prior direct exposure to 2, 4-D, and one had no prior direct application of any herbicide. Dominant 2, 4-D degrading strains isolated from these soils the previous year were screened for hybridization with three catabolic genes (tfdA, tfdAII, and tfdB) cloned from the well-studied 2, 4-D degradative plasmid, pJP4, revealing varying degrees of similarity with the three genes. Hybridization of total community DNA from the three soils with the tfd gene probes also indicated that pJP4-like tfd genes were not harboured by a significant percentage of the community. Community level response was evaluated by the comparison of different treatments by Random Amplified Polymorphic DNA (RAPD) fingerprints and by community DNA cross-hybridization. No differences between treatments within the same soil were detected in any of the RAPD fingerprints generated with 17 primers. Community DNA cross-hybridization also indicated that the application of 2, 4-D at the applied rates did not quantitatively affect the structure of the soil microbial communities present in the three soils during the time-frame studied.  相似文献

Summary In a greenhouse study the influence of alternate flooded and nonflooded conditions on the N2-ase activity of rice rhizosphere soil was investigated by C2H2 reduction assay. The soil fraction attached to roots represent the rhizosphere soil. Soil submergence always accelerated N2-ase and this effect was more pronounced in planted system. Moreover, rice plant exhibited phase-dependent N2-ase with a maximum activity at 60 days after transplanting. The alternate flooded and nonflooded regimes resulted in alterations of the N2-ase activity. Thus, the N2-ase activity increased following a shift from nonflooded to flooded conditions, but the activity decreased when the flooded soil was returned to nonflooded condition by draining. However, the differential influence of the water regime on N2-ase was not marked in prolonged flooded-nonflooded cycles. Microbial analysis indicated the stimulation of different groups of free-living and associative N2-fixing microorganisms depending on the water regime.  相似文献

Abstract Symbiotic experiments in glasshouse, controlled environment cabinet, and field were conducted with four lines of sulla ( Hedysarum coronarium ) and 15 strains of Rhizobium spp. This plant is highly Rhizobium -specific and appropriate strains are most unlikely to occur naturally in Australia. Under several sets of experimental conditions, H. coronarium nodulated abundantly and effectively with homologous rhizobia introduced from Spain and Italy. The optimum temperature for nitrogen fixation was relatively low (approx. 21°C) but significant interactions between line of host, strain of rhizobia, and growth temperature were frequent. Summary Rooted-cuttings and saplings of green ash (Fraxinus lanceolata) and hybrid poplar (Populus spp) were planted on a former municipal refuse landfill and on a nearby nonlandfill control plot. The root systems of four trees of each species and size were excavated on the landfill plot-two growing in an area where the concentrations of anaerobic landfill gases were relatively high and two in a relatively low-gas area. Two trees of each species and size were also excavated on the control. The root systems of both species were significantly shallower on the landfill plot than on the control. Green ash appeared to avoid the adverse gas environment of the deeper soil layers on the landfill by producing adventitious roots. Hybrid poplar became adapted in a different manner, by redirecting root growth from the deepter soil layers to the soil surface.  相似文献

Nematodes of three genera (Acrobeloides sp., Aphelenchus avenae, and Scutellonema brachyurum) were induced to coil and enter anhydrobiosis in drying soil of two types: sandy loam and loamy sand. Coiling was studied in relationship to soil moisture characteristics. Coiling and the physiological state of anhydrobiosis occurred before the water in sandy soils reached a water potential of -15 bars. Coiling was maximum at 3-6 bars, depending on the soil type and nematode species. It appeared that induction of coiling and anhydrohiosis were determined by the physical forces exerted by the water film surrounding the nematode, which, for these three species, was 6-9 monomolecular layers of water, rather than the % moisture and relative humidity of the soil per se.  相似文献

Summary N2-fixation by algae on the Broadbalk continuous wheat experiment was measured over a two year period using the acetylene reduction technique. The plots studied receive spring fertilizer treatments including farmyard manure and combinations of nitrochalk and Na, P, K and Mg which have remained much the same since the experiment started in 1843.Nitrogen applied at 196 kg ha–1 in spring suppressed algal N2-fixation until late in the season but at lower levels (48 kg N ha–1) the denser plant canopy increased both surface moisture and fixation. Herbicide treatment decreased fixation on plots of moderate nutritional status early in the season but had little effect on unfertilised plots where weed cover was sparse. On plots where weed and crop cover was very dense herbicide treatment increased fixation in August.Algal N2-ase activity, assayed by C2H2 reduction, continued throughout the night at a rate which averaged 33% of the midday value. Laboratory experiments indicate that dark fixation is very temperature sensitive and this value may represent a maximum. Algal crust in the field dried to 4.5–6.8% H2O content became active 3 1/2 h after rewetting and reached a steady state after 7 h which represented only 6–22% of that at the previous maximum suggesting that many cells had been killed.In a year with average rainfall algae on plots receiving 48 kg N ha–1 were estimated to fix 25–28 kg N ha–1 and plots without fertiliser 13–19 kg N ha–1. Algal fixation appeared to make a substantial contribution to the continuing fertility of unfertilised plots.  相似文献
