Confidentiality and informed consent are important concepts enabling the sharing of sensitive data. In a paper on this topic of this issue, Williams and Pigeot ( 2017 ) discuss that these have to be balanced with openness to ensure research standards. In this opinion paper, we give some background on how the paper by Williams and Pigeot evolved, reflect on their concepts, and provide some examples of the application of these concepts in various settings relevant to biostatisticians working in health research.  相似文献

Large stretching and un-stretching force response of adherent fibroblasts is measured by micromachined mechanical force sensors. The force sensors are composed of a probe and flexible beams. The probe, functionalized by fibronectin, is used to contact the cells. The flexible beams are the sensing element. The sensors are made of single crystal silicon and fabricated by the SCREAM process. The maximum cell stretch reached is approximately 50 microm, which is about twice of the cell initial size, and the time delay between two consecutive stretching/un-stretching steps is 75 s unless otherwise stated. We find that the force response of the cells is strongly linear, reversible, and repeatable, with a small stiffening at the initial deformation stage. Force response of single cells measured before and after cytochalasin D treatment suggests that actin filaments take almost all the cell internal forces due to stretch. These findings may shed light on the increasing understanding on the mechanical behavior of cells and provide clues for making new classes of biological materials having uncommon properties.  相似文献

Infrared, far-infrared and Raman spectra of Re2(O2CCH3)4X2 (X = Cl, Br) and Re2(O2CCD3)4Cl2 have been recorded. Assignments of the vibrational spectra of Os2(O2CCH3)4Cl2 and its deuterated derivative have been completed together with the Re complexes on the basis of normal-coordinate analysis. Force constant calculation was made for the acetate ion as well as for a four-atomic unit (with the CH3 and CD3 groups considered as point masses) using optimized masses of 16.7, 17.8, 20.5 and 21.6 for 12CH3, 13CH3, 12CD3 and 13CD3 groups, respectively. The force constants of the acetate ion have been adopted to the starting force field of the M2(O2CCH3)4X2 type complexes. The metal-halide (0.889, 0.997 and 1.286 N cm−1) and metal-metal stretching (3.32, 3.34 and 3.57 N cm−1) force constants were obtained for Re2(O2CCH3)4Cl2Re2(O2CCH3)4Br2 and Os2(O2CCH3)4Cl2 complexes, respectively. It was shown that the so-called diatomic approximation in most cases overestimates the M-M stretching force constants by 30-40%. Much better correlation has been obtained to fit these force constants, which produced values very close to those obtained by full normal-coordinate calculations. The Re-Re stretching force constants showed a reasonable correlation with the Re-Re bond distances for 18 rhenium complexes.  相似文献

The surface of hydrated cells of Staphylococcus epidermidis has been probed using an atomic force microscope. While local force measurements over the surface of bacteria reveal a heterogeneous chemical surface, with heterogeneous mechanical properties, different kinds of force curves appear with high frequency, and are thought to provide information on features contributing strongly to the overall mechanical and surface behaviour of the cell. Force curves often present two different mechanical regimes, being the first one (outer) of about 48 nm thick, and presenting a local relative elasticity of about 0.08 N/m, which is about a third of the relative elasticity of the inner part of the cell wall, harder, with a relative elasticity of about 0.24 N/m, in water. Both regimes appears as straight lines in the force versus distance curves (the ‘corresponding’ stress–strain curves in contact mechanics), but hysteresis is observed between the approach and the retraction line in the inner regime, indicating a degree of viscoelasticity. No viscoelasticity is observed in the outer regime, however, which presents quite linear and juxtaposed approach-retraction lines. These kinds of force curves do not present measurable pull-off forces nor snap-in forces, which indicates an almost null interaction between tip and bacterial surface, which could be in agreement with the measured very high hydrophobicity of this strain. Another kind of force curve has been observed recurrently, showing peaks in the retraction curves. Adhesive pull-off forces were measured giving an average of about 2 nN. Interestingly, however, these force curves appear only when quite irregular and wavy retraction curves are present, from the very beginning of its trace (maximum indentation). This leads us to think that these pull-off forces measured by our AFM do not give information on surface forces-unbinding events at the surface of the bacteria, but could be related to events at the sub-surface of the cell surface. Oscillations seen in the retraction curve in the portion corresponding to the contact with the bacteria surface could be due to rupture phenomena within the multilayered cell wall architecture expected in Gram-positive bacteria as Staphylococcus epidermidis, which could result in local irreversible deformations of the cell surface. Imaging with a sharp tip in contact mode sometimes leads to surface damage. Force curves recorded over damaged parts of the cell surface showed a completely different behaviour, in many cases with two well-defined high-adhesion peaks, and also interestingly, with snap-in forces of about 0–2 nN, which seems to indicate a completely different electrical/hydrophobicity state only a few nanometers down from the surface. Similar indentation effects can occur in the contact of a bacterial cell with a solid surface, even when showing only atomic-molecular-scale roughness, thus interacting not only with the very surface of the cell, especially when soft layers are present in the outer. Our results highlight the importance of the cell surface mechanical properties and their interplay with purely surface properties when analyzing cell–material interaction, and show the AFM as a useful method for investigating this.  相似文献

Compared with high infection areas of the world, the total HIV infection rate in China is relatively low. Nonetheless,because of China‘s vast territory and large population, the potential infection risk must be taken seriously. In the next few years, needle sharing among injection drug users will remain the most common route of transmission for the HIV/AIDS epidemic in China. Unprotected sex is gradually becoming a major route of transmission. China began to implement HAART in 1999 according to international standards. Prior to 2003, there were only about 150 HIV/AIDS patients were treated with HAART in some clinical trials and about 100 HIV/AIDS patients were treated by private sources.Results of those treatments are the scientific basis for development of the therapeutic strategies in China. In March of 2003, the Chinese government initiated China CARES program. In November of 2003, the Chinese Ministry of Health announced a national policy of free ARV treatment to all HIV Chinese citizens who were in poverty and required ARV therapy. There are total of 19,456 HIV/AIDS patients received free ARV drugs to date in 159 regions and 441 towns.Current challenges are how to follow-up and evaluate those patients in the clinical settings. The longer the therapy is postponed, the more side effects and the higher probability of drug resistance are going to occur. It remains unclear,therefore, when HAART regimen should be started in the HIV/AIDS population in China.  相似文献

Biodiversity metadata provide service to query, management and use of actual data sets. The progress of the development of metadata standards in China was analyzed, and metadata required and/or produced based on the Convention on Biological Diversity were reviewed. A biodiversity metadata standard was developed based on the characteristics of biodiversity data and in line with the framework of international metadata standards. The content of biodiversity metadata is divided into two levels. The first level consists of metadata entities and elements that are necessary to exclusively identify a biodiversity data set, and is named as Core Metadata. The second level comprises metadata entities and elements that are necessary to describe all aspects of a biodiversity data set. The standard for core biodiversity metadata is presented in this paper, which is composed of 51 elements belonging to 6 categories (entities), i.e. inventory information, collection information, information on the content of the data set, management information, access information, and metadata management information. The name, definition, condition, data type, and field length of metadata elements in these six categories (entities) are also described.  相似文献

The torque generated by a rotating joint comprises the useful force exerted by the joint on the external environment, and both the magnitude and distribution of torque through the step cycle during walking are important variables in understanding the mechanics of walking. The mechanics of the American lobster (Homarus americanus) and snow crab (Chionoecetes opilio) during walking were modelled to examine the relative roles of flexor versus extensor apodeme-muscle complexes, investigate which legs of these decapods likely contribute the greatest to locomotion, determine scaling effects of torque generation, and assess the relative roles of various model variables on torque production. Force generated along the length of the apodeme by the muscle was modelled based on apodeme surface area, muscle stress, and muscle fibre pinnation angle. Torque was then calculated from this estimated force and the corresponding moment arm. The flexor apodeme-muscle complex is calculated to generate consistently greater forces than the extensor, and generally this results in flexor torque being larger than extensor, though the snow crab does illustrate the opposite in two of its legs. This greater torque generation in flexion suggests that, in addition to the pushing of the trailing legs, the pulling action of the leading legs may play a significant role, at least during lateral walking. Leg 4 of both species appears to generate greater torques and thus provide the greatest forces for locomotion. Torque generation as a function of body size shows a second order response due to the increase in apodeme surface area. The pinnation angle of the muscle fibre is found to be insignificant in force generation, apodeme surface area (representing muscle cross sectional area) likely plays the most influential role in total force production, and moment arm controls the distribution of this force through the step cycle. Muscle stress remain a largely unknown quantity however, and may significantly affect both magnitude and distribution through step cycle of forces, and thus torque. Despite the uncertainty associated with the muscle stress parameter, the modelled results fit well with previously published force measurements.  相似文献

Drugs may interact with double stranded DNA via a variety of binding modes, each mode giving rise to a specific pharmacological function. Here we demonstrate the ability of single molecule force spectroscopy to discriminate between different interaction modes by measuring the mechanical properties of DNA and their modulation upon the binding of small molecules. Due to the unique topology of double stranded DNA and due to its base pair stacking pattern, DNA undergoes several well-characterised structural transitions upon stretching. We show that small molecule binding markedly affects these transitions in ways characteristic to the binding mode and that these effects can be detected at the level of an individual molecule. The minor groove binder berenil, the crosslinker cisplatin and the intercalator ethidium bromide are compared.  相似文献

Open resource metagenomics: a model for sharing metagenomic libraries   总被引:1,自引:0,他引:1  
Both sequence-based and activity-based exploitation of environmental DNA have provided unprecedented access to the genomic content of cultivated and uncultivated microorganisms. Although researchers deposit microbial strains in culture collections and DNA sequences in databases, activity-based metagenomic studies typically only publish sequences from the hits retrieved from specific screens. Physical metagenomic libraries, conceptually similar to entire sequence datasets, are usually not straightforward to obtain by interested parties subsequent to publication. In order to facilitate unrestricted distribution of metagenomic libraries, we propose the adoption of open resource metagenomics, in line with the trend towards open access publishing, and similar to culture- and mutant-strain collections that have been the backbone of traditional microbiology and microbial genetics. The concept of open resource metagenomics includes preparation of physical DNA libraries, preferably in versatile vectors that facilitate screening in a diversity of host organisms, and pooling of clones so that single aliquots containing complete libraries can be easily distributed upon request. Database deposition of associated metadata and sequence data for each library provides researchers with information to select the most appropriate libraries for further research projects. As a starting point, we have established the Canadian MetaMicroBiome Library (CM2BL [1]). The CM2BL is a publicly accessible collection of cosmid libraries containing environmental DNA from soils collected from across Canada, spanning multiple biomes. The libraries were constructed such that the cloned DNA can be easily transferred to Gateway® compliant vectors, facilitating functional screening in virtually any surrogate microbial host for which there are available plasmid vectors. The libraries, which we are placing in the public domain, will be distributed upon request without restriction to members of both the academic research community and industry. This article invites the scientific community to adopt this philosophy of open resource metagenomics to extend the utility of functional metagenomics beyond initial publication, circumventing the need to start from scratch with each new research project.  相似文献
