Abstract: Laminae and petioles from mature winter leaves of Arum italicum were studied in order to obtain information on the sun—shade intra-tissue properties of chloroplasts. This inference was based on the: (1) micro- and submicroscopic characteristics of the chloroplasts, (2) cytochemical localizations of functional PS I and PS II, (3) pigment patterns and compositions, (4) immunolocalization of Rubisco, and (5) net photosynthesis. It was inferred that all the chloroplasts across the lamina had adaptations to intermediate shade conditions, without a sun-shade dimorphism between the palisade and the spongy tissues. In the petiole, where normally-structured chloroplasts were surprisingly present in the entire thickness of the organ, a structural and chemical dimorphism was found between the outer chlorenchyma and the inner aerenchyma where intermediate shade-type and extreme shade-type chloroplasts were present, respectively. However, some anomalies in the pigment composition were noted chiefly in the inner aerenchyma (low concentrations of β-carotene and lutein, absence of zeaxanthin, presence of unusual pigments, for instance lutein epoxide, lutein cis-isomer, and chlorophyllide a). The Rubisco immunolabelling in the outer chlorenchyma of the petiole was similar to that in the lamina, while it was very scant in the inner aerenchyma. Net photosynthesis in the petiole was about 75% of that recorded in the lamina. These data suggest that the petiole of the mature winter leaf of A. italicum closely co-operates with the lamina for enhancing light capture and utilization.  相似文献

Potato (Solanum tuberosum L., cv. Sante) plantlets grown from stem node culture on medium supplemented with 90 mM sucrose accumulated lower amounts of photosynthetic pigments per mg dry weight in comparison to those grown on 30 mM sucrose. Addition of 0.1, 1 or 10 µM jasmonic acid (JA) to the medium resulted in a decrease of chlorophylls and carotenoids in the plantlets grown on either sucrose concentration. JA treatment induced de-epoxidation of violaxanthin to antheraxanthin and zeaxanthin only in those plantlets grown on a higher amount of sucrose in which hyperhydric symptoms were observed. The synergistic effect of JA and sucrose was clearly demonstrated in the plantlets grown on 90 mM sucrose and 1 µM JA. This was possibly due to overaccumulation of sucrose, the consequence of the most developed root system, and/or to stimulated water and solute transport by other mechanisms.  相似文献

Zeaxanthin, a carotenoid in the xanthophyll cycle, has been suggested to play a role in the protection against photodestruction. We have studied the importance of the parameters involved in zeaxanthin formation by comparing spinach plants grown in low light (100 to 250 mol m-2 s-1) to plants transferred to high light (950 mol m-2 s-1). Different parameters were followed for a total of 11 days. Our experiments show that violaxanthin de-epoxidase decreased between 15 and 30%, the quantity of xanthophyll cycle pigments doubled to 100 mmol (mol Chl)-1, corresponding to 27 mol m-2, and the rate of violaxanthin to zeaxanthin conversion was doubled. Lutein and neoxanthin increased from 50 to 71 mol m-2 and from 16 to 23 mol m-2, respectively. On a leaf area basis, chlorophyll and -carotene levels first decreased and then after 4 days increased. The chlorophyll a/b ratio was unchanged. The quantity of ascorbate was doubled to 2 mmol m-2, corresponding to an estimated increase in the chloroplasts from 25 to 50 mM. In view of our data, we propose that the increase in xanthophyll cycle pigments and ascorbate only partly explain the increased rate of conversion of violaxanthin to zeaxanthin, but the most probable explanation of the faster conversion is an increased accessibility of violaxanthin in the membrane.  相似文献

为筛选出山白兰(Michelia baillonii)苗木培育过程中适宜的光环境,该研究以一年生山白兰幼苗为试验材料,设置12、16 h·d-1两个光周期,配合使用红蓝复合光(8R1B、6R1B)、红蓝紫绿复合光(8R1B1P1G、6R1B1P1G)4种光质和白光(W)对照,采用双因素随机区组试验设计和隶属函数法,探讨了山白兰苗木生长、光合色素、内源激素含量对不同光质和光周期处理的响应规律。结果表明:(1)光质、光周期及其交互作用对山白兰苗高增长量、叶面积、叶绿素a、玉米素(ZR)、脱落酸(ABA)的含量、内源激素比值[IAA/ABA、(IAA+GA3+ZR)/ABA]等均有显著影响(P<0.05)。(2)16 h·d-1光周期有利于苗高增长量、叶面积、苗木质量指数、生物量、叶绿素a、生长素(IAA)、ZR的含量、内源激素比值的提高。(3)当光周期为16 h·d-1时,8R1B处理下的苗高增长量、叶面积和苗木质量指数均最大,分别为21.84 cm、158.39 cm2和2...  相似文献

Dunaliella bardawil Ben-Amotz & Avron accumulates high concentrations of β-carotene when grown under high light intensity. The β-carotene is composed mainly of 9-cis and all-trans β-carotene. Accumulation of β-carotene and an increase in the ratio of the 9-cis to the all-trans isomer are strongly dependent on the light intensity under which the algae are cultivated but are independent of light quality within the photosynthetically active radiation range. Cells grown under continuous red (>645 nm) or white light of 500 W·m?2 reach a value of about 32 pg β-carotene·cell?1 and a ratio of 9-cis to all-trans β-carotene of around 2, whereas cells grown under low red or white light intensity of 25 W·m?2 contain about 3 pg·cell?1 and a ratio of isomers of around 0.3.  相似文献

Flower colours and the composition of pigments in the perianths of five cultivars of Disa orchids were analyzed. Carotenoids were major pigment components in the orange-red flowers of ‘Dawn Angel’. We identified two types of pigment composition in the red flowered cultivars: ‘San Francisco’ contained more carotenoids and less anthocyanins, while ‘Marlene’ contained more anthocyanins than carotenoids. The red-purple flowered cultivars, only contained slight amounts of carotenoids, and the red-purple colour was attributed to the relatively high density of a cyanidin-based anthocyanin. The importance of the characterization of pigments in the perianths of orchid has been discussed in both breeding for flower colour improvement and chemotaxonomy.  相似文献

The community structure and physiological characteristics of three microbial mat communities in Byers Peninsula (Livingston Island, South Shetland Islands, Antarctica) were compared. One of the mats was located at the edge of a stream and was dominated by diatoms (with a thin basal layer of oscillatorian cyanobacteria), whereas the other two mats, located over moist soil and the bottom of a pond, respectively, were dominated by cyanobacteria throughout their vertical profiles. The predominant xanthophyll was fucoxanthin in the stream mat and myxoxanthophyll in the cyanobacteria-dominated mats. The sheath pigment scytonemin was absent in the stream mat but present in the soil and pond mats. The stream mat showed significantly lower delta13C and higher delta15N values than the other two mats. Consistent with the delta15N values, N2 fixation was negligible in the stream mat. The soil mat was the physiologically most active community. It showed rates of photosynthesis three times higher than in the other mats, and had the highest rates of ammonium uptake, nitrate uptake and N2 fixation. These observations underscore the taxonomic and physiological diversity of microbial mat communities in the maritime Antarctic region.  相似文献

High levels of variability were observed in tissue cultured Gaura lindheimeri genotypes when flowered in situ. Tissue culture treatments for chromosome doubling (colchicine: 0, 0.25, 1.25 mM; trifluralin: 0, 15, 30 μM) were all highly variable for morphological traits. Experiment No. 1 tested the tissue culture protocols used. In the first control (C1), plantlets were subcultured continuously. Nodes were excised and placed on solid medium for the second control (C2). In the third control (C3), nodes were excised and put in liquid medium for 24 h at room temperature. The fourth control (C4) was the same as the third control except liquid cultures were moved to 4°C for 48 h after treatment at room temperature. Experiment No. 2 examined the stability of the variation. Representative plants with different traits were selected for clonal propagation and grown in a replicated trial in the greenhouse. Several morphological traits (flower size, leaf length:width ratios, petal length:width ratios, and flower color) were measured. All of the controls had as much variability as the treated plants. Flower size of the first flower for plant number 01G-02 was significantly different in C1 compared with C3, but not with C2 and C4. Plant number 443-1 (white) was more stable in the replicated trial for the flower size than plant number 01G-02 (pink). All traits measured for plant number 01G-02 were unstable; most flower colors and patterns reverted back to the original color of the non-tissue cultured plants. Interspecific interactions can vary within and among populations and geographical locations, and this variation can influence the nature of the interaction (e.g. mutualistic versus antagonistic) and its evolutionary stability. Globeflowers are exclusively pollinated by flies whose larvae feed only on their seeds. Here we document geographical variability in costs and benefits in globeflowers in sustaining their pollinating flies throughout the range of this arctic-alpine European plant over several years. A total of 1,710 flower heads from 38 populations were analysed for their carpel, egg and seed contents. Individual and population analyses control for the confounding influences of variation in both: (1) population traits, such as fly density and egg distribution among flower heads; and (2) individuals traits, such as carpel and egg numbers per flower head. Despite considerable variation in ecological conditions and pollinator densities across populations, large proportions (range 33–58%) of seeds are released after predation, with a benefit-to-cost ratio of 3, indicating that the mutualism is stable over the whole globeflower geographical range. The stability of the mutualistic interaction relies on density-dependent competition among larvae co-developing in a flower head. This competition is revealed by a sharp decrease in the number of seeds eaten per larva with increasing larval number, and is intensified by non-uniform egg distribution among globeflowers within a population. Carpel number is highly variable across globeflowers (range 10–69), and flies lay more eggs in large flowers. Most plants within a population contribute to the rearing of pollinators, but some pay more than others. Large globeflowers lose more seed to pollinator larvae, but also release more seed than smaller plants. The apparent alignment of interests between fly and plant (positive relationship between numbers of seeds released and destroyed) is shown to hide a conflict of interest found when flower size is controlled for.  相似文献

The aim of this work was to investigate the occurrence of phosphoenolpyruvate carboxykinase (PEPCK) in different tissues of Arabidopsis thaliana throughout its vegetative and reproductive growth. The A. thaliana genome contains two PEPCK genes (PCK1 and PCK2), and these are predicted to generate 73,404 and 72,891 Da protein products, respectively. Both genes were transcribed in a range of tissues; however, PCK1 mRNA appeared to be more abundant and was present in a wider range of tissues. PEPCK protein was present in flowers, fruit, developing seed, germinating seed, leaves, stems and roots. Two PEPCK polypeptides, of approximately 74 and approximately 73 kDa were detected by immunoblotting, and these may arise from PCK1 and PCK2, respectively. PEPCK was abundant in cotyledons during post-germinative growth, and this is consistent with its well established role in gluconeogenesis. PEPCK was also abundant in sink tissues, such as young leaves, in developing flowers, fruit and seed. Immunohistochemistry and in situ hybridization showed that PEPCK was present in the nectaries, stigma, endocarp of the fruit wall and in tissues involved in the transfer of assimilates to the developing ovules and seeds, such as the vasculature and seed coat. The potential functions of PEPCK in A. thaliana are discussed.  相似文献
