Human anaplasmosis is an emerging tick-borne disease in the United States, but few studies of the causative agent, Anaplasma phagocytophilum, have been conducted in southeastern states. The aim of this study was to determine if A. phagocytophilum is present in small mammals and ticks in northeast Florida. Polymerase chain reaction assays designed to amplify portions of the major surface protein 2 gene (p44), 16S rDNA, and groESL operons were used to test rodent blood and tick DNA samples for the presence of A. phagocytophilum. Positive samples were confirmed by DNA sequence analysis. Anaplasma phagocytophilum DNA was detected in less than 5% of cotton mice and 45% of cotton rats from two sites in northeast Florida. Anaplasma phagocytophilum DNA was also confirmed in 1.3% of host-seeking adult Ixodes scapularis tested and 2.7% of host-seeking adult Amblyomma americanum. This report describes the first DNA sequence data confirming strains of A. phagocytophilum in rodents and ticks in Florida. The DNA sequences of the msp2, 16S rDNA, and groESL gene fragments obtained in this study were highly similar to reference strains of human pathogenic strains of A. phagocytophilum. These findings suggest that A. phagocytophilum is present and established among some small mammal species in northeast Florida. Although the infection prevalence was low in the total number of ticks tested, the presence of A. phagocytophilum in two human biting tick species, one of which is a known competent vector, suggests that humans in this region may be at risk of granulocytic anaplasmosis caused by this pathogen. 相似文献

ABSTRACT We used nucleotide sequence data from a mitochondrial DNA fragment to characterize variation within the endangered Lower Keys marsh rabbit (Sylvilagus palustris hefneri). We observed 5 unique mitochondrial haplotypes across different sampling sites in the Lower Florida Keys, USA. Based on the frequency of these haplotypes at different geographic locations and relationships among haplotypes, we observed 2 distinct clades or groups of sampling sites (western and eastern clades). These 2 groups showed low levels of gene flow. Regardless of their origin, marsh rabbits from the Lower Florida Keys can be separated into 2 genetically distinct management units, which should be considered prior to implementation of translocations as a means of offsetting recent population declines. 相似文献

ABSTRACT. Egg volumes are most often estimated using a mathematical model that incorporates length and width measurements and a species-specific shape variable. Although adequate in many respects, this technique does not account for intraspecific variation in egg shape. We developed a computer-automated technique that uses calibrated digital photographs to render precise measurements of several egg-size parameters including length, width, volume, and surface area. The system extracts egg outlines from photographs, and divides each egg into latitudinal slices that are subsequently regarded as simple geometric shapes (cylinders or cone frustra) with volumes and surface areas that can be summed to generate size parameters for the entire egg. We tested this technique using 491 eggs from Florida Scrub-Jay ( Aphelocoma coerulescens ) nests and compared the resulting egg volumes with volumes calculated using the preeminent method of estimating volume from linear measurements. Our method was highly accurate, and differences between the volumes from our method and the alternative method were strongly associated with variation in egg shape. Advantages of our technique include decreased handling of eggs and increased accuracy. We isolated and characterized 52 novel microsatellite markers from Florida largemouth bass, Micropterus salmoides floridanus, for use in conservation, management and population genetic studies. Markers were assessed in M. s. floridanus from peninsular Florida (n = 30) and averaged eight alleles per locus with observed heterozygosity of 0.57 (range 0–0.97). Cross‐taxa amplification was successful among 88% of tested congeners. These polymorphic and potentially taxon‐diagnostic markers contribute to the limited number of microsatellites currently available for micropterids and specifically M. s. floridanus. 相似文献

Fish communities are regularly exposed to a large number of natural and synthetic toxic chemicals, whose harmful effects, both short-term and long-term, need to be studied. Mathematical simulation models are known to be useful tools in the assessment of both acute and chronic effects. A time-concentration-effect model has been developed to assess the long-term effect of PCBs on multi generations of fish communities in the Florida Everglades. The term fish is used here generically to refer to five functional fish groups (FGs), two having small and two having large adult size, and a crustacean (crayfish), each of which are further composed of several age classes. Several contaminant concentration scenarios are analyzed in the simulations, subject to mortality and hatchability failure. The model predicts the population response time series for each FG, based upon exposure concentration and duration. Model results indicate that a concentration of 0.1 gl–1 could have adverse effects on survival and reproduction success for Everglades fish communities. For comparison, the USEPA has stipulated a standard of 0.014 gl–1 for the protection of freshwater fish from chronic effects and a standard of 1.0 gl–1 from acute effects. Hopefully, the model may be further developed to assist in the formulation of policy or regulations concerning aquatic ecological risk to protect the aquatic ecosystem of the Everglades. 相似文献

Habitat loss and fragmentation can influence the genetic structure of biological populations. We studied the genetic consequences
of habitat fragmentation in Florida black bear (Ursus americanus floridanus) populations. Genetic samples were collected from 339 bears, representing nine populations. Bears were genotyped for 12 microsatellite
loci to estimate genetic variation and to characterize genetic structure. None of the nine study populations deviated from
Hardy–Weinberg equilibrium. Genetic variation, quantified by mean expected heterozygosity (H
E), ranged from 0.27 to 0.71 and was substantially lower in smaller and less connected populations. High levels of genetic
differentiation among populations (global F
ST = 0.224; global R
ST = 0.245) suggest that fragmentation of once contiguous habitat has resulted in genetically distinct populations. There was
no isolation-by-distance relationship among Florida black bear populations, likely because of barriers to gene flow created
by habitat fragmentation and other anthropogenic disturbances. These factors resulted in genetic differentiation among populations,
even those that were geographically close. Population assignment tests indicated that most individuals were genetically assigned
to the population where they were sampled. Habitat fragmentation and anthropogenic barriers to movement appear to have limited
the dispersal capabilities of the Florida black bear, thereby reducing gene flow among populations. Regional corridors or
translocation of bears may be needed to restore historical levels of genetic variation. Our results suggest that management
actions to mitigate genetic consequences of habitat fragmentation are needed to ensure long-term persistence of the Florida

Aedes aegypti and Aedes albopictus are invasive mosquito species with geographic ranges that have oscillated within Florida since their presence was first documented. Local transmission of dengue, chikungunya, and Zika viruses serves as evidence of the public health importance of these two species. It is important to have detailed knowledge of their distribution to aid in mosquito control efforts and understand the risk of arbovirus transmission to humans. Through a partnership involving the University of Florida Institute of Food and Agricultural Sciences Cooperative Extension Service and the Florida Medical Entomology Laboratory; the Florida Department of Health; and mosquito control agencies throughout Florida, a container mosquito surveillance program involving all life stages was launched in the summer of 2016 to detect the presence of Ae. aegypti and Ae. albopictus. Results from this survey were mapped to provide a picture of the current known distribution of Ae. aegypti and Ae. albopictus in Florida. Aedes aegypti and/or Ae. albopictus were detected in the 56 counties that were part of the survey. Only Aedes albopictus was detected in 26 counties, primarily in the panhandle region of Florida. The results of this work underscore the importance of maintaining container mosquito surveillance in a state where chikungunya, dengue, and Zika viruses are present and where there is continued risk for exotic arbovirus introductions. 相似文献

Eryngium cuneifolium Small. (Apiaceae) is a narrowly distributed endemic found only in Ceratiola ericoides (Florida rosemary)-dominated Florida scrub, a periodically burned, shrub-dominated habitat. Multivariate analyses using 22 ∗∗∗microhabitat characteristics indicated significant microhabitat and time-since-fire effects on survival, growth, and fecundity of 1287 individuals over a 4-yr period. Survival increased with distance to the nearest shrub, and plants in larger open patches had greater survival rates. Neighboring shrubs of Ceratiola ericoides and Calamintha ashei were associated with a higher mortality of E. cuneifolium than other neighboring shrub species. Survival was reduced by two-thirds over 4 yr (14% vs. 42%) for E. cuneifolium near C. ericoides. Sand accretion increased growth and fecundity. With greater time since fire, woody shrubs increasingly dominate and open patches shrink, significantly reducing survival, growth, and fecundity of E. cuneifolium. Effects were particularly dramatic between 2 and 7 yr postfire, when annual mortality increased from <10% to >30% (r = 0.74). This herbaceous species is dependent on an open habitat maintained by periodic fire. Belowground competition or allelopathy from shrubs probably restricts E. cuneifolium to recently burned, open patches within the most xeric parts of Florida scrub. 相似文献

Surface waters in the Florida Everglades contain high levels ofdissolved organic carbon (DOC) compounds. 13C values of DOCsamples collected from the northern Everglades indicate that less than about23%of the DOC was derived from sugarcane (the dominant agricultural crop in thearea), and the amount of DOC from sugarcane was greater during the dry period.Most of the DOC (> 50%) in the northern Everglades was in the low molecularweight (< 1000 Dalton) fraction (LMW-DOC). The relative amount of highmolecular weight DOC (HMW-DOC) was higher in the wet period than in the dryperiod. Radiocarbon ages of the DOC ranged from > modern toabout 2400 years B.P., indicating that DOC was derived from both historic peatdeposits and modern vegetation. At each site, the HMW-DOC had older radiocarbonages than the LMW-DOC, and therefore contained a greater fraction of DOCderivedfrom the historic peat deposits. It appears that at least some of the old DOCcompounds from the historic peat deposits were decomposed during theirresidencein the surface water system in the northern Everglades, and the LMW-DOC wasmoremicrobially labile than the HMW-DOC. Our analysis suggests that accelerateddecomposition of organic matter in the historic peat deposits (due to land-usechange) could be a significant source of DOC and nutrients in the northernEverglades. Our data also suggest that the radiocarbon signature of DOC couldbeused as a sensitive indicator of the overall effectiveness of a wetlandrestoration project. 相似文献

This study evaluates the bottom-up and top-down controls on epiphyte loads under low nutrient additions. Nutrients and gastropod grazers were manipulated in a field experiment conducted within a Thalassia testudinum meadow in Florida Bay, FL, USA. The effect of seagrass leaf turnover rate on epiphyte loading was also evaluated using novel seagrass short-shoot mimics that “grow,” allowing for the manipulation of leaf turnover rates. During the summer growing season and over the course of one seagrass leaf turnover period, low-level water column nutrient additions increased total epiphyte load, epiphyte chlorophyll a, and epiphyte autotrophic index. T. testudinum leaf nutrients (N and P) and leaf productivity also increased. Epiphyte loading and T. testudinum shoot biomass and productivity did not respond to a 60% mean increase in gastropod abundance. Manipulations of seagrass leaf turnover rates at minimum wintertime and maximum summertime rates resulted in a 20% difference in epiphyte loading. Despite elevated grazer abundances and increased leaf turnover rates, epiphyte loads increased with nutrient addition. These results emphasize the sensitivity of T. testudinum and associated epiphytes to low-level nutrient addition in a nutrient-limited environment such as Florida Bay. 相似文献
