The symbiotic effectiveness of Bradyrhizobium strains isolated from three species of Parasponia and from legumes were compared on Parasponia grown in Leonard-jars. Effectiveness of each symbiotic association was estimated from dry weight and total nitrogen of shoots and nodules of plants grown on medium free of combined nitrogen. Twenty strains isolated from three species of Parasponia were found to vary in their effectiveness on P. andersonii, the least effective fixing one fifth of the nitrogen of the most effective strains. The outcome of the symbiosis was not associated with the host source of the test strain. P. andersonii, P. rugosa and P. rigida responded differently to a selection of seven strains of Parasponia Bradyrhizobium; some strains were either ineffective or fully effective on each host, while others varied in their symbiotic performance. P. andersonii fixed significantly (P < 0.001) larger quantities of nitrogen than either P. rugosa or P. rigida with p. rigida being the least effective. In contrast to Bradyrhizobium strains from Parasponia spp. which formed nodules rapidly (within 11–20 days), nine strains isolated from legumes required between 25 and 74 days to form partially effective nodules. The thre Parasponia species formed relatively large quantities of nodule tissue relative to the amount of nitrogen fixed and shoot dry matter produced. The Bradyrhizobium isolated from Parasponia plants growing in Papua New Guinea soils could be grouped together on the basis of their infection characteristics on Parasponia and legumes.  相似文献

Summary Oxygen consumption, gill ventilation, blood acid-base/ionic status and haemoglobin oxygen affinity were studied in seawater-adapted adult salmon (Salmo salar) during five weeks after transfer into fresh water. Freshwater exposure induced the following changes: Standard oxygen consumption ( ) and ventilatory flow ( ) decreased markedly during the first days after transfer, then decreased more gradually until a new steady-state was achieved at which and were about 80% and 56% of the control values, respectively. The marked increase in oxygen extraction coefficient (Ew O 2) and the marked decrease in the oxygen convection requirement ( ) were associated with a reduction in the partial pressure of carbon dioxide in arterial blood (Pa CO 2), in spite of a decrease of both ventilatory flow and water CO2 capacitance. These results suggested that transfer into fresh water induced an increase in branchial diffusive conductance. A biphasic pattern was observed in the time-course of the changes in both plasma ion concentration and acid-base status. During the first 10 days, plasma Na+, K+, and Cl concentrations fell abruptly, then more gradually. [Cl] decreased more than [Na+] resulting in a progressive increase in the [Na+]/[Cl] ratio. During the second phase of acclimation to fresh water plasma Na+, K+, and Cl concentrations progressively increased. [Cl] increased more than [Na+], so that [Na+]/[Cl] ratio decreased. Transfer into fresh water did not significantly change plasma lactate concentration. Upon exposure to fresh water, blood pH increased from 7.94±0.04 to 8.43±0.06 at day 10 and then decreased to 8.08±0.03 at day 34. The increase in blood pH induced by transfer to fresh water initially represented a mixed metabolic/respiratory alkalosis. However, after 15 days Pa CO 2 had returned to pretransfer values and the alkalosis was purely metabolic. The metabolic component of the alkalosis was associated with appropriate changes in the plasma strong ion difference (S.I.D.). Blood alkalosis moved the oxygen dissociation curve to the left, so that P50 was decreased by 30% below the value in seawater for the maximal increase in blood pH. This rise in haemoglobin affinity for O2, associated with a marked increase in blood buffer capacity, are regarded as adaptative processes allowing the salmon to cope with the markedly increased energy expenditure required for upstream migration.  相似文献

This study investigated the role of predators in preventing competitive exclusion among three closely related armoured catfishes (Callichthys callichthys, Hoplosternum littorale and H. thoracatum) that occur synthopically in multi-predator freshwater swamps of Suriname, South America. The potential impact of predation on armoured catfish was determined by combining laboratory measurements of predation rates on five early developmental stages of the armoured catfish H. thoracatum for 24 aquatic predators with field studies of the density of the predators in the swamps. The contribution of a particular predator to the total predation pressure on its prey was determined to a large extent by the density of the predator in the swamp. Seemingly innocuous predators with low or moderate predation rates in the laboratory may be extremely important in the swamps due to their high abundance. Small-sized omnivorous fishes and aquatic invertebrates were major predators of early developmental stages of armoured catfish. Both qualitative and quantitative ontogenetic changes in the predation pressure on armoured catfish were observed. Major predation on eggs, larvae and juveniles of H. thoracatum resulted from a different set of predators in each developmental stage of the prey. In all developmental stages of H. thoracatum the predation pressure involved several predator species and not a single, dominant predator. The potential predation pressure of the 24 predators taken together and the number of predators that were able to prey on H. thoracatum decreased sharply with increasing age (size) of the prey. Even if egg (nest) predation is prevented by the guarding male, the potential impact of the 24 predators on the populations of armoured catfish is large. Predation may account for the high mortality of H. thoracatum observed in the swamps. The high predation pressure on callichthyid catfishes may help to explain the coexistence of three closely related and morphologically quite similar armoured catfishes in Surinamese swamps.  相似文献

Synopsis We have quantitatively analyzed a videotape of Seriola dumerili (Carangidae) displaying ram suspension-feeding behavior and ram ventilation in the field. This is the first report of facultative suspension feeding by a piscivorous carangid. The intraoral morphology of S. dumerili is not typical of ram suspension-feeding fishes in that closely-spaced, long gill rakers are lacking. While the mechanism of particle retention is not known for any ram suspension-feeding fish species, scanning electron microscopy revealed denticles on the branchial surfaces of S. dumerili that could play a role in particle entrapment.  相似文献

Synopsis In this study we investigate the effect of food availability (zooplankton biomass) on the growth of Odontesthes bonariensis (Atherinidae) larvae. The larvae were stocked in four 45 m2 outdoor tanks at relatively high densities (100 and 200 larvae m–2). Because of the high stocking densities, the zooplankton biomass was depleted in all tanks. However, the patterns of food limitation, and particularly periods of severe food shortage, differed in tanks stocked at different densities. We could therefore, observe the effect of food limitation in larvae that differed in weight and age. The effects of variables suspected to influence O. bonariensis growth rates (age and weight of larvae, available zooplankton biomass, mean individual weight of available preys, total ingested prey weight, and mean weight of ingested preys) were investigated using standard multiple regression methods, and a model assuming: (1) an allometric relationship between maximum growth rates and weight of larvae, and (2) an inverse relationship between growth depression and the available zooplankton biomass. Both methods were consistent in showing that only the weight of larvae, and the availability of zooplankton prey had significant effects on the growth of O. bonariensis. The model's results additionally suggest that, if the observed growth rates are scaled by the maximum growth rate corresponding to the larva weight, the effect of zooplankton biomass is largely independent of age and weight of larvae.  相似文献

Synopsis An annual spawning aggregation of Nassau grouper,Epinephelus striatus, observed off the southern coast of Quintana Roo, Mexico, on full moon days in December and January, was surveyed during the winters from 1991 to 1993. A fish aggregation had been appearing at one traditional site off Mahahual for more than 80 years, but currently it forms at undetermined sites far from the original site. Fish migration was northwards in the fore reef border along the 14 m isobath and isolated groups were observed. Through visual assessment fish aggregations were found to include 200 to 500 individuals. The fishing on Nassau grouper aggregations has been active since 1910 by using hook and line; however, recently fishermen used gill nets. It is necessary to continue the research on this aggregation and establish management guidelines to protect the species.  相似文献

Using primary cultures of gill pavement cells from freshwater rainbow trout, a method is described for achieving confluent monolayers of the cells on glass coverslips. A continuous record of intracellular pH was obtained by loading the cells with the pH-sensitive flourescent dye 2,7-bis(2-carboxyethyl)-5(6)-carboxyfluorescein and mounting the coverslips in the flowthrough cuvette of a spectrofluorimeter. Experiments were performed in HEPES-buffered media nominally free of HCO3. Resting intracellular pH (7.43 at extracellular pH=7.70) was insensitive to the removal of Cl or the application of 4-acetamido-4-isothiocyanatostilbene-2,2-disulfonic acid (0.1 mmol·l–1), but fell by about 0.3 units when Na+ was removed or in the presence of amiloride (0.2 mmol·l–1). Exposure to elevated ammonia (ammonia prepulse; 30 mmol·l–1 as NH4Cl for 6–9 min) produced an increase in intracellular pH (to about 8.1) followed by a slow decay, and washout of the pulse caused intracellular pH to fall to about 6.5. Intracellular non-HCO 3 buffer capacity was about 13.4 slykes. Rapid recovery of intracellular pH from intracellular acidosis induced by ammonia prepulse was inhibited more than 80% in Na+-free conditions or in the presence of amiloride (0.2 mmol·l–1). Neither bafilomycin A1 (3 mol·l–1) nor Cl removal altered the intracellular pH recovery rate. The K m for Na+ of the intracellular pH recovery mechanism was 8.3 mmol·l–1, and the rate constant at V max was 0.008·s–1 (equivalent to 5.60 mmol H+·l–1 cell water·min–1), which was achieved at external Na+ levels from 25 to 140 mmol·l–1. Cathepsin B (EC 3.4.22.1) was purified from buffalo liver. The enzyme activity against-benzoyl-dl-arginine-naphthylamme (BANA) was substantially reduced by heat (above 37C) and by nondenaturing concentrations of urea (3 M) and guanidine hydrochloride (1 M). Cathepsin B was significantly activated by 1.5 mM EDTA alone. The activation of the enzyme was further enhanced in the presence of thiol compounds, e.g., cysteine thioglycolic acid, 2,3-dimercapto-1-propenol, and dithioerythritol (DTE). The minimum concentration of the thiol compound required for optimal activation of cathepsin B was found to be lowest (0.2 mM) for DTE. The BANA hydrolyzing activity of cathepsin B was substantially reduced by Cu2+ (20–200M) and Ca2+ (30–250 mM) as well as by thiol blocking reagents, e.g., iodoacetate, 5,5-dithiobis(2-nitro-benzoic acid) (DTNB), andp-hydroxymercuribenzoate (pHMB). The enzyme activity was completely abolished when the molar ratio of the reagent: cathepsin B was close to 1. The number of free sulfhydryl groups in cathepsin B was determined to be 2 by titration against DTNB and pHMB. Modification of one free thiol group of cathepsin B resulted in complete loss of BANA hydrolyzing activity.  相似文献

Gilthead were fed three diets. Diet A was the control diet, whereas diets B and C were supplemented with 300 and 900 mg Zn/kg, respectively. Fish fed with diet C, at the end of the experiment, showed the lowest weight. Zinc concentrations presented the higher values in gills, liver, and kidney. Muscle and brain had the lower mean values and showed a tight control of zinc levels. These results reinforce the hypothesis that zinc in the CNS should be strictly controlled in order to maintain the functional role of the metal. Significant differences in tissue zinc concentrations were obtained between fish fed different amounts of zinc, the metal concentrations being higher in tissues of fish fed diet C. The tissue decrease of zinc, found at the end of the experiment, may depend on a lower feed consumption or on different zinc requirements during the cold season. These changes, even if not univocal among the three diets, may be associated with the life cycle of fish. Furthermore, copper concentrations were little affected by the different concentrations of zinc in the three diets; liver and kidney presented the highest concentrations; liver showed a significant decrease in copper content at the end of the experiment. We conclude that: zinc concentrations of the diet may affect the gilthead weights and the tissual metal content; and zinc concentrations in the diets, depending on the growth rate, may be varied depending on the season.  相似文献

Thirteen F2 families of faba bean (Vicia faba L.), descended from plants trisomic for chromosomes 3, 4, 5 and 6, have been analyzed for morphological, isozyme and RAPD markers. This allowed the establishment of linkage relationships among these markers as well as the assignment of some markers and/or linkage groups to their respective chromosomes. The linkage analysis of partially overlapping sets of informative genetic markers for the data pooled from 13 F2 families has revealed 48 linkage groups, six of which have been precisely assigned to specific chromosomes. A statistical procedure to analyze the data of joint segregation analysis in families derived from trisomic plants has been developed.  相似文献
