The construction and use of two novel transposon(Tn)-delivery vectors is described. These vectors carry Inc.W or Inc.N broad-host-range transfer functions cloned next to the narrow-host-range replicon of pBR329. The host specificities of pSLX10 and pSLX23 both complement and extend the host specificities of existing Tn delivery vectors. Plasmids pSLX10 and pSLX23 were shown to transfer at high frequency in intergeneric matings. The lux genes which are present on each vector permit the visual monitoring of transconjugants which have retained a Tn element, but are devoid of plasmid molecules. pSLX10 and pLSX23 were efficiently used to generate a range of auxotrophic mutants in various strains of Pseudomonas as well as to clone genes from Serratia liquefaciens. These vectors may have general applicability to identify and clone genes in a wide range of Gram-negative bacteria.  相似文献

Recent years have seen the convergence of both genetic and biochemical approaches in the study of protein translocation inE. coli. The powerful combination of these approaches is exemplified in the use of anin vitro protein synthesis-protein translocaltion system to analyze the role of genetically defined components of the protein translocation machinery. We describe in this review recent results focusing on the function of thesecA, secB, andsecY gene products and the demonstration of their requirement forin vitro protein translocation. The SecA protein was recently shown to possess ATPase activity and was proposed to be a component of the translocation ATPase. We present a speculative working model whereby the translocator complex is composed of the integral membrane proteins SecY, SecD, SecE, and SecF, forming an aqueous channel in the cytoplasmic membrane, and the tightly associated peripheral membrane protein SecA functioning as the catalytic subunit of the translocator or protein-ATPase.  相似文献

Summary Effects of P fertilizers on growth of Douglas fir (Pseudotsuga menziesii var.menziesii (Mirb.) Franco.) seedlings were examined in pots and nursery beds. In pot experiments levels of P equivalent to 300 kg/ha were adequate for maximum growth over 14–18 weeks and resulted in available soil P levels of 80 ppm after 15 weeks' growth. Maximum growth in pots was obtained with shoot P concentrations of 0.18%–0.20%, with higher values at lower temperatures, but the optimum concentration for one-year old (1-0) nursery seedlings was 0.16% P. Growth of seedlings was greatly restricted at a soil temperature of 5°C and an air temperature of 12°C. At a soil temperature of 10°C and an air temperature of 14°C seedling P requirement was greater than at soil and air temperatures of 20°C.Comparison showed that monammonium phosphate was more effective than calcium superphosphate in stimulating growth in pots and nursery. Triple superphosphate was also effective in the nursery. Diammonium phosphate, potassium dihydrogen phosphate and phosphoric acid had no advantages as P sources in the nursery. Available P levels of 100–130 ppm, in the loamy sand and sandy loam nurseries studied, and needle P concentrations of 0.18%, when sampled in October, were associated with maximum growth of two-year old (2-0) seedlings.P fertilization decreased root/shoot ratio, but did not alter the allometric relationship of shoot to root. Improving P status from a low level increased root growth capacity in 2-0 seedlings and P fertilization of potted seedlings increased dry weight/height ratio. Uptakes per seed bed ha of 236 kg N, 31 kg P, 81 kg K and 73 kg Ca by 2-0 seedlings were comparable with, or greater than, uptake rates of agricultural crops. Recoveries of 6–11% of P from fertilizer were recorded in the nursery.  相似文献

Summary Field experiments were carried out using15N-labelled calcium nitrate, to investigate the relative uptake by barley of fertilizer-N and soil-N. On imperfectly drained till soils uptake of soil-N increased with increasing rate of fertilizer, but remained constant on a brown sand, possibly due to more efficient root exploration in the latter soil. In four out of five seasons, late uptake of soil-derived N was a major feature, and uptake from ploughed soil as compared with uptake from direct-drilled soil was correlated with seasonal rainfall patterns. Significantly higher quantities of both fertilizer- and soil-derived N were taken up by winter barley than by spring barley, reflecting the longer growth period and higher dry matter yield from the former crop.  相似文献

Synopsis Oxygen uptake (VO2) during graded hypoxia, rate of hypoxia acclimation, breathing frequency (fR), breath volume (VS, R) and gill ventilation (VG) were measured in Hoplias malabaricus. Normoxia and hypoxia acclimated fish had similar and constant VO2 and VG in a range of water PO2 from 150 to 25 mmHg. Hypoxia acclimated fish showed significantly higher VO2 in severe hypoxia (PO2 <15 mmHg). Normoxia acclimated fish showed symptoms similar to hypoxic coma after 1 h of exposure to water PO2 of 10 mmHg whereas the same symptoms were observed only at PO2 of 5 mmHg for fish acclimated to hypoxia. Fish required 14 days to achieve full acclimation to hypoxia (PO2 ≥25 mmHg). Lowering of water PO2 from 150 to 25 mmHg resulted in normoxic fish showing a 3–2 fold increase in VG. The increase was the result of an elevation in VS, R rather than fR. Among normoxia acclimated specimens, small fish showed a higher VG per unit weight than the large ones in both normoxia (PO2 =150 mmHg) and hypoxia (PO2 = 15 mmHg). A decrease in the ventilatory requirement (VG/VO2) with increased body weight was recorded in hypoxia (PO2 = 15 mmHg).  相似文献

The maximum biomass in iron-limited photosynthetic batch cultures of chlorella increased as the logarithm of the iron concentration. The growth yield from iron (Y x/Fe) showed a marked inverse relation to the specific growth rate. The maximum biomass yield, g dry biomass/g iron consumed, was 7.5x103 with specific growth rate 0.108 h-1; the minimum was 0.79×103 with specific growth rate 0.145 h-1. The maximum specific growth rate in the exponential phase of Fe limited cultures varied as the initial Fe concentration. Thermophilic Humicola lanuginosa, Penicillium duponti, Sporotrichum thermophile and Mucor pusillus required succinate in addition to glucose for optimal growth. The requirement for succinate was concentration-dependent and the concentration needed for one half of the maximal growth was 6.14 mM. In the presence of succinate, glucose utilization from the medium was markedly increased and this was associated with increased levels of the enzymes of the glycolytic and Krebs cycle pathways. Addition of succinate to cultures growing in glucose at any stage of growth stimulated the growth with the resulting rate of growth remaining high if the addition was made within 3 days of inoculation. Cycloheximide (71.4 M) prevented the succinate-mediated derepression of the enzymes suggesting that succinate may remove the catabolite repression in the presence of glucose.A preliminary part of this work was presented at the 17th annual meeting of the Association of Microbiologists of India at Manipal (India) held from Dec. 13 to 15, 1976  相似文献

Summary Bulk soil samples were collected from the top 15 cm of untreated areas adjacent to field fertilizer trials at 2 locations. Amounts of N, P, K, and lime equivalent to the field treatments were mixed with the soil in 15-cm diameter pots. Paper birch (Betula papyrifera March.) trees were grown from seed for a greenhouse bioassay. Height and dry weight of the bioassay seedlings were significantly correlated with 3-year volume growth of 10-year-old paper birch seedlings in the field. Correlation coefficients were 0.88 for height growth and 0.91 for dry weight growth on one site, and 0.72 and 0.63 on the other. With further refinements and observations on a larger number of sites, this bioassay technique should be a valuable tool for estimating potential response to fertilizer by young paper birch in the field, and for ranking the relative productivity of different soils.  相似文献

Summary Greenhouse experiments were done with two purposes: (1) to identify strains of rhizobia effective and acid-tolerant in symbiosis withLablab purpureus, and (2) to determine whether soil acidity or the symbiotic condition increased the phosphate requirement for growth.Five rhizobial strains were tested in one neutral soil, two acid soils, and the two acid soils limed to pH 6.6. In the neutral and limed soils, three of the strains were effective (CB1024, CB756, TAL169), but only two strains (CB756, TAL169) remained effective in acid soil.Strain CB756 and plus-N treatments were further compared in a factorial trial involving combinations of five levels of P with lime, no lime and CaCl2 treatments, applied to an acid soil. Some of the treatments were also applied to plants inoculated with CB1024. Between the N-fertilized and CB756 treatments there was no clear difference in growth response to applied P, and the critical internal concentration of P for 95% of maximal growth was the same (0.22% shoot dry weight). Increasing P beyond levels needed for maximal growth increased nodulation and N concentration in plants inoculated with CB756. It lowered N concentration in N-fertilized plants. There was evidence suggesting that the P requirement of symbiotic plants increased if the soil was acid, or if CB756 were replaced by CB1024 as microsymbiont; but the critical statistical interactions were not significant.  相似文献
