In order to study both the hunting efficiency and the flexibility of their predatory behavior, solitary hunters of the trap-jaw ant Odontomachus opaciventris were offered small prey (termites, fruit flies and tenebrionid larvae), presenting different morphological or defensive characteristics. The monomorphic hunters showed a moderately flexible predatory behavior characterized by short capture sequences and a noteworthy efficiency of their mandible strike (76.7-100% of prey retrievals), even when presented with Nasutitermes soldiers. Contrary to most poneromorph ants, antennal palpation of the prey before the attack was always missing, no particular targeted region of the prey's body was preferred, and no 'prudent' posture was ever exhibited. Moreover, stinging was regularly performed on bulky, fast moving fruit flies, very scarcely with sclerotized tenebrionid larvae, but never occurred with Nasutitermes workers or soldiers despite their noxious chemical defense. These results suggest that, whatever the risk linked to potentially dangerous prey, O. opaciventris predatory strategy optimizes venom use giving top priority to the swiftness and strength of the lethal trap-jaw system used by hunters as first strike weapon to subdue rapidly a variety of small prey, ranging from 0.3 to 2 times their own body size and from 0.1 to 2 times their weight. Such risk-prone predatory behavior is likely to be related to the large size of O. opaciventris colonies where the death of a forager might be of lesser vital outcome than in small colony-size species. 相似文献

Ephedra foliata Boiss. & Kotschy ex Boiss., (family – Ephedraceae), is an ecologically and economically important threatened Gymnosperm of the Indian Thar Desert. A method for micropropagation of E. foliata using nodal explant of mature female plant has been developed. Maximum bud-break (90 %) of the explant was obtained on MS medium supplemented with 1.5 mg l−1 of benzyl adenine (BA) + additives. Explant produces 5.3 ± 0.40 shoots from single node with 3.25 ± 0.29 cm length. The multiplication of shoots in culture was affected by salt composition of media, types and concentrations of plant growth regulators (PGR’s) and their interactions, time of transfer of the cultures. Maximum number of shoots (26.3 ± 0.82 per culture vessel) were regenerated on MS medium modified by reducing the concentration of nitrates to half supplemented with 200 mg l−1 ammonium sulphate {(NH4) 2SO4} (MMS3) + BA (0.25 mg l−1), Kinetin (Kin; 0.25 mg l−1), Indole-3-acetic acid (IAA; 0.1 mg l−1) and additives. The in vitro produced shoots rooted under ex vitro on soilrite moistened with one-fourth strength of MS macro salts in screw cap bottles by treating the shoot base (s) with 500 mg l−1 of Indole-3-butyric acid (IBA) for 5 min. The micropropagated plants were hardened in the green house. The described protocol can be applicable for (i) large scale plant production (ii) establishment of plants in natural habitat and (iii) germplasm conservation of this endemic Gymnosperm of arid regions. 相似文献

Procollagens are the major proteins secreted into the conditioned medium of cultured arterial smooth muscle cells. Methods for the isolation and quantification of these macromolecules have traditionally required preliminary salt precipitation of procollagens from the conditioned medium followed by cellulose ion-exchange chromatography. The method described here exploits the elongated conformation of soluble procollagens and allows the direct recovery of procollagens from culture medium by a single gel-filtration chromatographic step under nondissociating conditions. Procollagens are isolated in high yield and show minimal processing by procollagen N- or C-terminal peptidase activity. This method results in rapid recovery of highly purified procollagens, free of most proteoglycans of other products of smooth muscle cell metabolism. 相似文献

The effect of various experimental parameters upon the frequency of callus formation from cultured anthers of Oryza sativa has been investigated. Although certain medium components were found to be critical to callus formation, the concentration of these components had little effect upon the frequency of callus formation. The degree to which the callus formation frequency was influenced by cold pretreatment of the flowers was variable. Even though plants were grown under uniform conditions and flowers containing pollen in the microspore stage of development were selected for dissection, the frequency of callus formation varied nonrandomly between flowers. In experiments with populations of flowers in a physiological and developmental state favorable to callus formation 35% of plated anthers produced callus and at least 60% of these calluses gave rise to green plants. 相似文献

【目的】白酒可培养微生物的分离筛选是白酒行业重要的研究内容,本文旨在构建中国白酒生产环境中可培养微生物信息数据库。【方法】数据库信息主要来源于通过人工查阅和整理目前已发表的白酒微生物的相关的文献报道和菌种保藏中心相关的筛选自白酒生态系统的微生物信息。数据库主要设计3个功能:(1)白酒可培养微生物信息检索:通过菌株名称、分离位置、培养基、代谢产物等为条件检索相关的白酒可培养微生物信息,从而获取该白酒微生物详细的生理生化与分类学信息;(2)培养基信息检索:通过特定培养基成分,培养基编号和名称检索相关的培养基信息,包括培养的组成和配制方法。(3)数据更新:在线上传新的可培养微生物和培养基信息。【结果】目前数据库共收1221种白酒可培养微生物和295种培养基信息,网址为:http://cmbaijiu.i-sanger.com/。【结论】本数据库是我国白酒领域首个可培养微生物信息的数据库,将有助于白酒微生物培养的相关研究工作开展。 相似文献

Statistically based experimental designs were applied to the optimization of medium components and environmental factors for eicosapentaenoic acid (EPA) production by the diatom Nitzschia laevis in heterotrophic conditions. First, the Plackett-Burman design was used to evaluate the effects of variables including medium components and environmental factors on cell growth and EPA production. Among these variables, NaCl, CaCl(2), PI metal solution, pH, and temperature were identified to have the significant effects (with confidence level > 90 %). Subsequently, the concentrations of NaCl, CaCl(2), PI metal solution as well as the values of pH and temperature were optimized using central composite design. The cell growth and EPA production were found to respectively correlate to NaCl, CaCl(2), pH, and temperature that could be represented by second-order polynomial models. The optimal values of the four parameters were determined by response surface and numerical analyses as 8 g/L NaCl, 0.10 g/L CaCl(2), pH 8.5 and 19.8 degrees C for cell dry weight (DW), and 14 g/L NaCl, 0.10 g/L CaCl(2), pH 8.5 and 18 degrees C for EPA production, respectively. The subsequent verification experiments confirmed the validity of the models. This optimization strategy led to a DW of 9 g/L, an EPA yield of 280 mg/L and an EPA productivity of 28 mg/L/d, respectively, which were considerably higher than those obtained in the previous studies. 相似文献

Human cardiac troponin-I (cTnI) is one of the most sensitive and specific indicators, used in the diagnosis of myocardial infarction. To produce the protein efficiently, Escherichia coli and Pichia pastoris systems were used. Initial trials for the expression in E. coli were not successful, although different expression vectors with different promoters were tested. This led us to use P. pastoris for the expression. After several trials with two different expression strains of P. pastoris, it was concluded that P. pastoris was also not an optimal expression host for cTnI. Comprehensive analysis of the expression systems indicated that an efficient expression is only possible when the gene is optimized for expression in E. coli. For this purpose, the gene was optimized in-silico, but edited manually afterwards. It was synthesized and cloned into pQE-2 vector. Expression was performed using routine experimental conditions. Thus, cTnI could be efficiently expressed from the optimized gene in E. coli. The expression and purification were practical and may be used for commercial purposes since a total yield of 25µg highly pure protein per milliliter of culture could be obtained. The protein was in its ready-to-use form for many biological applications, including as a standard in diagnostic tests and an antigen for antibody production. 相似文献

研究了国产及进口LB培养基对产嗜热性木聚糖酶基因工程菌1020生长及产酶的影响,并以国产原料为基准,优化了培养基。结果表明,国产的酵母膏及蛋白胨和进口原料相比,缺少某些成分,单纯增加用量不能弥补这种差别;乳糖可以代替昂贵的IPTG起到有效的诱导作用;10g·L-1的乳糖可兼起能源及诱导剂的双重效果,菌的生物量和产酶量达到最佳;发酵8h后补加5g·L-1乳糖,生物量及酶活皆有提高。复合氮源优于单一的无机或有机氮源。优化后的酶活从进口LB培养基的270U~290U提高到1700U~1800U。 相似文献
