Molecular mechanisms of neurotrophin signaling on dendrite development and dynamics are only partly understood. To address the role of brain‐derived neurotrophic factor (BDNF) in the morphogenesis of GABAergic neurons of the main olfactory bulb, we analyzed mice lacking BDNF, mice carrying neurotrophin‐3 (NT3) in the place of BDNF, and TrkB signaling mutant mice with a receptor that can activate phospholipase Cγ (PLCγ) but is unable to recruit the adaptors Shc/Frs2. BDNF deletion yielded a compressed olfactory bulb with a significant loss of parvalbumin (PV) immunoreactivity in GABAergic interneurons of the external plexiform layer. Dendrite development of PV‐positive interneurons was selectively attenuated by BDNF since other Ca2+‐binding protein‐containing neuron populations appeared unaffected. The deficit in PV‐positive neurons could be rescued by the NT3/NT3 alleles. The degree of PV immunoreactivity was dependent on BDNF and TrkB recruitment of the adaptor proteins Shc/Frs2. In contrast, PLCγ signaling from the TrkB receptor was sufficient for dendrite growth in vivo and consistently, blocking PLCγ prevented BDNF‐dependent dendrite development in vitro. Collectively, our results provide genetic evidence that BDNF and TrkB signaling selectively regulate PV expression and dendrite growth in a subset of neurochemically‐defined GABAergic interneurons via activation of the PLCγ pathway. © 2006 Wiley Periodicals, Inc. Theoretical studies claim that if co-occurring species have very different mobilities this will result in greater small-scale species richness, but empirical evidence is still lacking. We measured horizontal vegetative mobility (VM) of 48 herbaceous understory species and estimated small-scale species richness in early and late successional boreonemoral herb-rich coniferous forests in central Estonia. VM of erosulate growth forms was significantly higher than that of hemi-rosette and rosette growth forms. Erosulate species exhibited higher mobility in young stands, but their relative and total cover was considerably higher in old stands. Local plant richness (in 1 × 1 m plots) correlated positively with the variability of VM of species in a plot—larger differences in VM resulted in a higher number of coexisting species. Our results thus suggest that species differences in VM can contribute to small-scale coexistence by providing different ways to colonise empty space. PtdIns and PtdInsP kinases from normal erythrocyte (AA) membranes and sickle cell anaemia erythrocyte (SS) membranes have been characterized. PtdIns kinase was studied in native membranes under conditions in which PtdInsP kinase and PtdInsP phosphatase do not express any activity. Kinetic analysis of the AA and SS PtdIns kinases indicate similar Km values for PtdIns and ATP but higher Vmax values for SS PtdIns kinase. PtdInsP kinase was partially purified from erythrocyte ghosts by NaCl extraction. The kinetic parameters of PtdInsP kinase determined under these conditions were similar in AA and SS NaCl extracts. These data suggest the presence of some effector of PtdIns kinase in SS cell membranes, resulting in a greater activity of the enzyme. This leads consequently, to increase the PtdInsP pool and to activate PtdInsP kinase, in agreement with our previous observations of a greater [32P]Pi incorporation in both polyphosphoinositides in SS cells relatively to AA cells.  相似文献

The ability of embryonating eggs of Ascaris lumbricoides to avoid desiccation by reducing the loss of water through the egg shell was investigated. When exposed to desiccation the eggs lost water at a rate dependent upon the relative humidity and ambient temperature, eventually resulting in the collapse of the eggs and the death of the enclosed embryo. The eggs are small with a large surface to volume ratio. A low permeability to gaseous exchange thus restricts water loss while still ensuring an adequate supply of oxygen for embryonic development. Relative humidity did not appear to affect the rate of development. In eggs exposed to desiccation at various constant temperatures, the rate of water loss increased as an exponential function of increasing temperature. When eggs were exposed to various temperatures before exposure to desiccation at 22 C, the rate of water loss increased as a function of increasing pretreatment temperature. After exposure to 63–65 C, the ability of the egg shell to slow down the loss of water was destroyed. These phenomena suggest that there is not a simple “critical” or “transition” temperature, but a gradual melting of the complex mixture of components forming the lipid layer.  相似文献

目的:研究光学相干断层成像术(OCT)在近视眼视网膜神经纤维层(RNFL)厚度测量中的应用价值。方法:选择2016年1月到2016年5月在医院就诊的近视患者73例(138眼)纳入此次研究,根据近视情况将患者分为低度近视组(-0.30D~-3.00D)共26例(48眼)、中度近视组(-3.01~-6.00D)共24例(47眼)及高度近视组(-6.00D)共23例(43眼)。另选同期在医院体检(视力正常)的健康志愿者25例(45眼)作为对照组,对比各组不同象限的RNFL厚度,屈光度及眼轴长度,分析近视眼各象限的RNFL厚度与患者屈光度和眼轴长度的相关性。结果:高度近视组的上方象限、下方象限以及鼻侧象限的RNFL厚度均明显低于对照组及中度近视组,中度近视组的下方象限及鼻侧象限的RNFL厚度均明显低于对照组,低度近视组鼻侧象限的RNFL厚度明显低于对照组,差异均有统计学意义(均P0.05)。近视组的屈光度及眼轴长度均明显大于对照组,且高度近视组均明显大于中度近视组与低度近视组,中度近视组均明显大于低度近视组,差异均有统计学意义(均P0.05)。根据Pearson法分析相关性可知,近视眼患者上象限、下象限、鼻侧象限的RNFL厚度与其屈光度及眼轴长度均呈负相关。结论:利用OCT技术检测近视眼RNFL厚度时,应考虑屈光度及眼轴长度可能造成的影响,综合进行分析判断,以获得最佳检测数值。  相似文献

草本层是古尔班通古特沙漠植物群落下层层片的构建者, 冬季积雪提供了其生长发育所需要的主要水分, 积雪的增加或减少对草本植物数量和生物量会产生显著的影响。该研究利用人工增减积雪的方法, 在古尔班通古特沙漠南缘设置了5个不同厚度的积雪处理: 0积雪、50%积雪、100%积雪、150%积雪和200%积雪, 其中100%积雪为自然积雪。采用1 m × 1 m的样方, 对草本层片的物种数、盖度、密度、高度进行了调查, 还采用收获法测定了草本层片的地上生物量和优势种小花荆芥(Nepeta micrantha)的单株地上生物量。对研究区内13个科29种草本植物的研究表明: 1)单位面积出土幼苗数量跟积雪厚度呈显著正相关关系, 草本层片的盖度、密度对积雪的变化响应显著, 随着积雪增加, 草本层片的密度和盖度呈递增趋势, 而草本层片的平均高度呈递减趋势, 但不同积雪处理间的物种数和总地上生物量没有显著差异; 2)积雪厚度与优势种的株高和地上生物量呈显著负相关关系, 积雪的增加导致优势种的单株生物量和株高显著降低; 3)积雪厚度的变化主要影响了草本层片植物种子萌发的数量, 但对物种数量没有显著影响。这表明: 虽然积雪是草本植物的主要水分来源之一, 但荒漠植物群落的草本植物对积雪的变化具有很强的缓冲能力, 即使积雪很少, 草本层片的物种构成也不会发生显著变化, 草本层片的净初级生产力也保持相对稳定。  相似文献

The impact of the micropore width, external surface area, and meso‐/macropore size on the charging performance of electrochemical double‐layer capacitor (EDLC) electrodes is systematically investigated. Nonactivated carbon xerogels are used as model electrodes in aqueous and organic electrolytes. Monolithic porous model carbons with different structural parameters are prepared using a resorcinol‐formaldehyde‐based sol–gel process and subsequent pyrolysis of the organic precursors. Electrochemical properties are characterized by utilizing them as EDLC half‐cells operated in aqueous and organic electrolytes, respectively. Experimental data derived for organic electrolytes reveals that the respective ions cannot enter the micropores within the skeleton of the meso‐ and macroporous carbons. Therefore the total capacitance is limited by the external surface formed by the interface between the meso‐/macropores and the microporous carbon particles forming the xerogel skeleton. In contrast, for aqueous electrolytes the total capacitance solely depends on the total surface area, including interfaces at the micropore scale. For both types of electrolytes the charging rate of the electrodes is systematically enhanced when increasing the diameter of the carbon xerogel particles from 10 to 75 nm and the meso‐/macropore size from 10 to 121 nm.  相似文献

Addition of 15L-hydroperoxy-5,8,11,13-eicosatetraenoic acid (15-HPETE) to human leukocytes led to the formation of a novel series of compounds containing four conjugated double bonds. The yield of tetraenes was increased approx. 100-fold when ionophore A23187 (5 μM) was added simultaneously with 15-HPETE. The structure of the major tetraene was established by physical methods as well as by chemical degradation and found to be 5,6,15L-trihydroxy-7,9,11,13-eicosatetraenoic acid.  相似文献

Nymphs of Rhipicephalus appendiculatus and Hyalomma anatolicum anatolicum were fed on rabbits and maintained in the laboratory to allow moulting and further development. At specified time intervals from 0 to 500 days after engorgement, samples of ticks were ground individually in distilled water within the wells of an agglutination plate. A 0.1 ml aliquot was removed from each and levels of haemin and protein assessed from optical density values at specific wavelengths in a spectrophotometer.

Both protein and haemin levels showed an initial rapid decrease after engorgement; values did not fall to zero in either case but showed marked fluctuation throughout the study period. These fluctuations combined with the high standard errors of the results, made assessments of the physiological age of individual ticks impossible.

Such fluctuating values, however, suggested the possibility that haem biosynthesis might be taking place, and this was tested by injecting the radioactively-labelled haem precursor [4-14C]δ-amino laevulinic acid into engorged nymphs, immediately following their detachment. Both tick species revealed an incorporation of this compound into their haematin content, although neither incorporated the non-haem precursor [1-14C]2-amino isobutyric acid. These results indicate an ability of ticks to synthesize haem in vivo, although the underlying reasons for such a mechanism remain unknown.  相似文献
