Leptin is a key factor for the regulation of food intake and energy homeostasis in mammals, but information regarding its role in teleosts is still limited. There are large differences between mammalian and teleost leptin at both gene and protein levels, and in order to characterize the function of leptin in fish, preparation of species-specific leptin is therefore a key step. In this study, full-length cDNA coding for rainbow trout leptin was identified. In spite of low amino acid sequence similarity with other animals, leptin is highly conserved between trout and salmon (98.7%). Based on the cDNA, we produced pure recombinant trout leptin (rt-leptin) in E. coli, with a final yield of 20 mg/L culture medium. We then examined the effects of intraperitoneal (IP) injection of rt-leptin on feeding behavior and gene expression of hypothalamic NPY and POMCs (POMC A1, A2 and B) in a short-term (8 h) experiment. The rt-leptin suppressed food intake and led to transient reduction of NPY mRNA levels, while the expression of POMCs A1 and A2, was elevated compared with vehicle-injected controls. These results for rainbow trout are the first that describe a physiological role of leptin using a species-specific orthologue in teleosts, and they suggest that leptin suppresses food intake mediated by hypothalamic regulation. This anorexic effect is similar to that observed in mammals and frogs and supports that the neuroendocrine pathways that control feeding by leptin are ancient and have been conserved through evolution.  相似文献

Primary leaves of bean (Phaseolus vulgaris L.) seedlings cultivated for 14 days in a growth chamber on complete (control) and phosphate deficient (−P) Knop liquid medium were used for measurements. The −P leaves were smaller and showed an increased specific leaf area (SLA). Their inorganic phosphate (Pi) concentration was considerably lowered. They did not show any significant changes in chlorophyll (Chl) (a + b) concentration and in their net CO2 assimilation rate when it was estimated under the conditions close to those of the seedlings growth. Light response curves of photosynthetic net O2 evolution (P NO2) of the leaves for the irradiation range up to 500 μmol(photon) m−2 s−1 were determined, using the leaf-disc Clark oxygen electrode. The measurements were taken under high CO2 concentration of about 1 % and O2 concentrations of 21 % or lowered to about 3 % at the beginning of measurement. The results obtained at 21 % O2 and the irradiations close to or higher than those used during the seedlings growth revealed the phosphorus stress suppressive effect on the leaf net O2 evolution, however, no such effect was observed at lower irradiations. Other estimated parameters of P NO2 such as: apparent quantum requirement (QRA) and light compensation point (LCP) for the control and −P leaves were similar. However, with a high irradiation and lowered O2 concentration the rate of P NO2 for the −P leaves was markedly higher than that for the control, in relation to both the leaf area and leaf fresh mass. This difference also disappeared at low irradiations, but the estimated reduced QRA values indicate, under those conditions, the increased yield of photosynthetic light reaction, especially in the −P leaves. The presented results confirm the suggestion that during the initial phase of insufficient phosphate feeding the acclimations in the light phase of photosynthesis, both structural and functional appear. They correspond, probably, to the increased energy costs of carbon assimilation under phosphorus stress, e.g. connected with raised difficulties in phosphate uptake and turnover and enhanced photorespiration. Under the experimental conditions especially advantageous for the dark phase of photosynthesis (saturating CO2 and PAR, low O2 concentration), those acclimations may be manifested as an enhancement of photosynthetic net O2 evolution.  相似文献

柯氏鼠兔(Ochotona koslowi)是兔形目(Lagomorpha)鼠兔属中最古老的遗留种,也是稀有的濒危物种,自1894年命名以来,国内外对该物种的生存现状、生物学特征知之甚少.2007年10~11月,在藏北地区捕获到13只柯氏鼠兔,通过显微组织分析法对其胃及结肠内容物和收集的60份粪样进行了食性分析.结果表明,柯氏鼠兔胃、结肠内容物和粪便中镜检到可识别植物碎片属6科15种,其中豆科植物碎片占可识别植物碎片的39.44%,藜科植物碎片占36.00%,莎草科植物碎片占16.42%,禾本科、十字花科和菊科植物碎片分别占3.75%、2.67%和1.25%.食性分析表明,柯氏鼠兔偏爱豆科植物.  相似文献

We determined whether there is an association between complement factor H (CFH), high-temperature requirement A-1 (HTRA1), vascular endothelial growth factor (VEGF), and pigment epithelium-derived factor (PEDF) genotypes and the response to treatment with a single intravitreous injection of bevacizumab for age-related macular degeneration (AMD). Eighty-three patients with exudative AMD treated by bevacizumab injection were genotyped for three single nucleotide polymorphisms (SNPs; rs800292, rs1061170, rs1410996) in the CFH gene, a rs11200638-SNP in the HTRA1 gene, three SNPs (rs699947, rs1570360, rs2010963) in the VEGF gene, and four SNPs (rs12150053, rs12948385, rs9913583, rs1136287) in the PEDF gene using a TaqMan assay. The CT genotype (heterozygous) of CFH-rs1061170 was more frequently represented in nonresponders in vision than TT genotypes (nonrisk allele homozygous) at the time points of 1 and 3 months, while there was no CC genotype (risk allele homozygous) in our study cohort (p = 7.66 × 10−3, 7.83 × 10−3, respectively). VEGF-rs699947 was also associated with vision changes at 1 month and PEDF-rs1136287 at 3 months (p = 5.11 × 10−3, 2.05 × 10−2, respectively). These variants may be utilized for genetic biomarkers to estimate visual outcomes in the response to intravitreal bevacizumab treatment for AMD.  相似文献

以初始平均体重(2.02±0.02)g的牙鲆(Paralichthys olivaceus)为实验对象,进行为期70d的摄食生长实验,研究不同添加方式的植酸酶对牙鲆生长和饲料利用的影响。在5000.0g豆粕中添加2.5g植酸酶,然后用产朊假丝酵母(Candidautilis)进行发酵预处理,得到植酸酶预处理豆粕。共制作4种等氮等能(粗蛋白49.7%、总能20.9kJ/g)饲料,对照饲料主要以鱼粉为蛋白源;在对照饲料的基础上,用豆粕蛋白替代45%的鱼粉蛋白配制成豆粕组饲料;在每千克豆粕组饲料中添加1000IU植酸酶,配制成植酸酶组饲料;用植酸酶预处理豆粕蛋白替代45%的鱼粉蛋白配制成植酸酶预处理豆粕组饲料。结果表明,与对照组相比较,用豆粕蛋白替代饲料中45%的鱼粉蛋白,若不添加植酸酶则显著降低牙鲆的特定生长率(P0.01)、饲料效率、蛋白质效率和氮贮积率(P0.05);直接添加植酸酶组、植酸酶预处理豆粕组牙鲆的特定生长率、饲料效率、蛋白质效率和氮贮积率与鱼粉对照组相比较没有出现显著差异(P0.05);与不添加植酸酶的豆粕组相比较,在含豆粕饲料中添加1000IU/kg饲料的植酸酶显著提高牙鲆的特定生长率(P0.01)、氮贮积率(P0.05)和磷贮积率(P0.01),显著降低氮排放率(P0.05)和磷排放率(P0.01),但饲料效率和蛋白质效率没有显著变化(P0.05);在豆粕中添加植酸酶进行发酵预处理,降低了豆粕中植酸含量,在饲料中添加植酸酶预处理豆粕显著提高牙鲆的特定生长率(P0.01)、饲料效率、蛋白质效率和氮贮积率(P0.05),显著降低氮(P0.05)、磷和钙的排放率(P0.01)。    相似文献

Background and Aims

In temperate woody perennials, flower bud development is halted during the winter, when the buds enter dormancy. This dormant period is a prerequisite for adequate flowering, is genetically regulated, and plays a clear role in possibly adapting species and cultivars to climatic areas. However, information on the biological events underpinning dormancy is lacking. Stamen development, with clear differentiated stages, appears as a good framework to put dormancy in a developmental context. Here, stamen developmental changes are characterized in apricot (Prunus armeniaca) and are related to dormancy.

Methods

Stamen development was characterized cytochemically from the end of August to March, over 4 years. Developmental changes were related to dormancy, using the existing empirical information on chilling requirements.

Key Results

Stamen development continued during the autumn, and the flower buds entered dormancy with a fully developed sporogenous tissue. Although no anatomical changes were observed during dormancy, breaking of dormancy occurred following a clear sequence of events. Starch accumulated in particular places, pre-empting further development in those areas. Vascular bundles developed and pollen mother cells underwent meiosis followed by microspore development.

Conclusions

Dormancy appears to mark a boundary between the development of the sporogenous tissue and the occurrence of meiosis for further microspore development. Breaking of dormancy occurs following a clear sequence of events, providing a developmental context in which to study winter dormancy and to evaluate differences in chilling requirements among genotypes.  相似文献

采用3种需冷量估算模型(≤7.2℃模型、0~7.2℃模型和犹他模型)和2种需热量估算模型(生长度小时模型和有效积温模型)分别测定了14个设施葡萄品种的需冷量和需热量,并分析了两者的相互关系。结果表明,不同葡萄品种的需冷量和需热量存在一定差异,需冷量值介于754~1489h(≤7_2℃模型)、497-757h(0~7.2℃模型)或192~755.5CU(犹他模型),需热量值为18491~24070GDH℃(生长度小时模型)或120~377D℃(有效积温模型),且欧亚种品种需热量普遍高于欧美杂种品种。另外,本研究还表明,用生长度小时模型估算出来的需热量值与需冷量值之间呈负相关关系,而有效积温模型估算出来的需热量值与需冷量值之间则呈正相关关系。  相似文献

Sea beets grown from seeds collected in 1989 and 2009 along the coasts of France and adjacent regions were compared for flowering date under controlled conditions. Seeds from both collection years were sown simultaneously and cultivated under the same glasshouse conditions. Date of flowering onset and year of first flowering were recorded. There was an overall northward shift in flowering time of about 0.35° latitude (i.e. 39 km) over the 20‐year period. The southern portion of the latitudinal gradient – that is, from 44.7°N to 47.28°N – flowered significantly later by a mean of 1.78 days, equivalent to a 43.2‐km northward shift of phenotypes. In the northern latitudes between 48.6°N and 52°N, flowering date was significantly earlier by a mean of 4.04 days, corresponding to a mean northward shift of 104.9 km, and this shift was apparently due to a diminished requirement of exposure to cold temperatures (i.e. vernalization), for which we found direct and indirect evidence. As all plants were grown from seed under identical conditions, we conclude that genetic changes occurred in the sensitivity to environmental cues that mediate the onset of flowering in both the northern and the southern latitudes of the gradient. Microevolution and gene flow may have contributed to this change. There was no significant change in the frequency of plants that flowered without vernalization. The lack of vernalization requirement may be associated with environmental instability rather than with climate conditions.  相似文献

试验采用单因子梯度设计。在试验中,维生素E设6个梯度,分别为Ⅰ0mg/kg、Ⅱ50mg/kg、Ⅲ125mg/kg、Ⅳ200mg/kg、Ⅴ275mg/kg、Ⅵ350mg/kg,前后进行175d饲养试验,以雌鳝性腺系数、产卵力、孵化率以及各组织中SOD活性、MDA含量等指标作为判据,研究了VE对雌鳝繁殖性能的影响。结果表明,随饲料中VE含量的增加,Ⅰ、Ⅱ、Ⅲ、Ⅳ组雌鳝性腺系数、产卵力和孵化率显著提高(p0.05),Ⅴ、Ⅵ组则提高不显著(p0.05),卵中VE的含量反映了饲料中VE的含量。雌鳝卵巢中SOD活性,Ⅳ、Ⅴ和Ⅵ组比Ⅰ和Ⅲ组显著低(p0.05),相应地,MDA含量Ⅳ、Ⅴ和Ⅵ组显著低于Ⅰ、Ⅱ和Ⅲ组(p0.05)Ⅰ、Ⅱ和Ⅲ组雌鱔,虽然其卵巢中SOD活性较高,但因饲料中抗氧化性的VE缺乏或不足,引发卵子脂质过氧化,MDA含量显著升高(p0.05),导致卵质低下,从而影响了卵子孵化。综上所述,雌鳝饲料中添加维生素E,能有效地改善雌鳝的繁殖性能,其最适添加量为200mg/kg。    相似文献
