A total of 177 naturally contaminated water samples were analyzed by membrane filtration according to the Standard Methods for the Examination of Water and Wastewater published by the American Public Health Association. Filters were incubated in parallel on mHPC-agar and 3M™ Petrifilm™ Aerobic Count Plates (Petrifilm™ AC plates) for heterotrophic counts. Fecal coliforms and Escherichia coli were enumerated on mFC-agar and 3M™ Petrifilm™ E. coli/Coliform Count Plates (Petrifilm™ EC plates). Typical colonies on each media type were confirmed following standard procedures. Heterotrophic counts were between 103 and 104 CFU/mL and the average log10 counts obtained on Petrifilm™ AC plates were about two-fold lower than on mHPC-agar. Counts for fecal coliforms and E. coli were between 102 and 103 CFU/mL. Average log10 counts for confirmed fecal coliforms obtained on Petrifilm™ EC plates were slightly lower than on mFC agar with a correlation coefficient of 0.949. The average log10 counts for confirmed E. coli on Petrifilm™ EC plates and on mFC agar were statistically not different (P=0.126) with a correlation coefficient of 0.879. Specificity of Petrifilm™ EC plates and mFC agar was evaluated by comparing typical colony counts with confirmed counts. On mFC agar, counts for typical colonies were by 2 log10 CFU higher than the actual confirmed counts. In contrast, on Petrifilm™ EC plates typical colony counts were almost identical to confirmed colony counts for both fecal coliforms and E. coli. This comparison illustrates the high specificity of Petrifilm™ EC plates for enumeration of both fecal coliforms and E. coli in water.  相似文献

目的探讨肠道菌群和粪便炎性标志物在炎症性肠病(IBD)活动度评估中的临床价值。方法共纳入120例IBD患者为研究组,其中溃疡性结肠炎(UC)患者68例,克罗恩病(CD)患者52例。选择30例经结肠镜检查正常的健康体检者为对照组。采集全部研究对象的新鲜粪便标本进行粪便细菌培养及炎性标志物检测,比较不同疾病活动度IBD患者的肠道菌群及粪便钙卫蛋白(FC)、乳铁蛋白(LF)、基质金属蛋白酶-9(MMP-9)、髓过氧化酶(MPO)水平的变化。结果与对照组比,UC和CD患者肠道中肠杆菌、肠球菌、拟杆菌、消化球菌及酵母菌数量均明显增加(P0.05),双歧杆菌、乳杆菌及真杆菌数量明显减少(P0.05)。UC患者梭菌数量较对照组增加(P0.05),CD患者梭菌数量较对照组减少(P0.05)。UC、CD活动期患者肠杆菌、肠球菌、拟杆菌、消化球菌及酵母菌数量明显多于缓解期患者(P0.05),双歧杆菌、乳杆菌及真杆菌数量明显少于缓解期患者(P0.05),且重度活动期患者肠道菌群改变较轻、中度活动期改变更明显(P0.05)。UC活动期患者梭菌数量明显多于缓解期(P0.05),CD活动期患者梭菌数量明显少于缓解期(P0.05)。UC和CD患者粪便中FC、LF、MMP-9及MPO水平均显著高于对照组(P0.05)。UC、CD活动期患者FC、LF、MMP-9及MPO水平显著高于缓解期患者(P0.05),且重度活动期患者高于轻、中度活动期患者(P0.05)。结论肠道菌群变化和粪便中FC、LF、MMP-9及MPO水平可作为IBD患者疾病活动性评估的辅助指标。  相似文献

目的观察粪菌移植对儿童难治性功能性便秘的临床疗效并探讨其机制。方法对4例临床确诊为难治性功能性便秘患儿给予粪菌移植治疗,观察和记录患儿临床症状改善情况并收集供体和粪菌移植治疗前后患儿粪便标本,利用Illumina MiSeq测序平台进行16SrRNA测序,并对测序结果进行生物信息学分析。结果 4例患儿接受粪菌移植治疗后排便状况明显改善,大便次数增加、粪便硬度变软、腹部胀痛明显缓解。粪菌移植后,患儿肠道菌群多样性下降,患儿与供体的肠道菌群差异先增大后逐渐缩小。菌群分析显示患儿肠道有害菌受到抑制、有益菌增加。结论粪菌移植可有效改善儿童难治性功能性便秘的临床症状,具有疗程短、治疗方便、无明显不良反应等优点。  相似文献

随着粪便DNA分析技术的不断发展与完善,其越来越多地被应用于分子生态学研究中,特别是野生动物的遗传状况评估研究。粪便DNA的获取可以在不干扰,甚至无需观察到动物本身的情况下展开,因此避免了取样活动可能给野生动物带来的干扰或伤害,极大地促进了野生动物分子生态学的研究。虽然粪便DNA分析技术在其建立伊始因DNA质量问题而受到了一定的挑战,但自其建立至今,研究者发展了多种技术来克服这一问题。现已能获得较高质量的粪便DNA,并将基因分型错误率控制在较低水平。本文将结合我们在粪便DNA分析技术上所积累的经验,从粪便样品采集、保存、DNA提取、PCR扩增以及等位基因分型等各个环节对该技术进行详细探讨,以期阐明该技术在野生动物分子生态学研究中所面临的机遇与挑战,进一步推动其在我国野生动物保护研究中的应用与发展。  相似文献

A laboratory-scale anaerobic fixed-bed reactor, operating at ambient temperature (30 to 35°C), was used to treat sewage water from tourist areas in Cuba at hydraulic retention times (HRT) ranging from 4 to 72 h. The total chemical oxygen demand (T-COD), total biological oxygen demand (T-BOD) and total suspended solids removal varied between 30 and 80%, 40 and 95% and 25 and 80%, respectively. Total and faecal coliforms were reduced by 98.1 to 99.9% and by 99.0% to 99.9% respectively, despite the marked decrease in HRT from 72 to 4 h.  相似文献

The ureic herbicide linuron [3-(3,4-dichlorophenyl)-1-methoxy-1-methylurea] (CAS 330-55-2) was investigated for genotoxicity in a series of in vivo experiments. Since human exposure to herbicides is not only to the active principles, but also to all the chemicals present in the commercial formulation, we tested both pure and commercial linuron. Groups of rats were treated with gavage containing different doses of the herbicide (pure compound or commercial formulation) for 14 days. The doses were 150, 300 and 450 mg/kg b.wt. for the pure compound and 315.8, 631.6 and 947.4 mg/kg b.wt. for the commercial formulation (47.5% of linuron). Faeces and urine were collected at regular intervals. Urine specimens were analysed for their mutagenic metabolites, thioethers and d-glucaric acid content. Faeces extracts were tested for mutagenicity. Linuron's ability to cause DNA damage and cytogenetic effects was also investigated after treating groups of rats once with different doses of pure or commercial linuron. DNA single-strand breaks were assessed in rat liver using the alkaline elution technique and the single-cell microgel electrophoresis assay (SCGE: `comet' assay), and in rat testes cells with the SCGE assay. Micronuclei induction was analysed in rat bone marrow erythrocytes. Results obtained were mainly negative when the excretion of mutagenic metabolites in urine and faeces of animals treated with the pure compound or with the linuron-based commercial formulation were monitored, whereas an increase in the urinary excretion of thioethers and d-glucaric acid was observed in rats treated with the commercial formulation. No increase in the frequency of micronucleated polychromatic erythrocytes was observed in the treated animals. However, linuron affected the viability of hepatocytes isolated from animals treated with higher doses. This cytotoxicity was accompanied by the induction of DNA single-strand breaks in the liver, as seen by the alkaline elution assay. The potential of pure linuron to induce in vivo DNA damage was confirmed with the microgel electrophoresis technique (`comet' assay). Cytotoxicity was also seen in rat testes cells. Background

The N-Myc Downstream-Regulated Gene (NDRG) family comprises four members that function in cellular processes like proliferation and differentiation. While NDRG1 and NDRG2 are extensively studied, knowledge regarding NDRG3 and NDRG4, despite its recognition as a well-established early-detection marker for colorectal cancer (Cologuard®), is sparse.

Scope of review

To summarize expression, biomarker potential and functional mechanisms of the NDRGs in the developing, mature and cancerous gut, we combine current literature and in silico analyses from the TCGA-database, GTEX Project, E14.5 mouse intestine and enteric neural crest cells, and an RNA-sequencing time-series of human embryonic colonic samples.

Major conclusions

This study reveals that all members display a differential expression pattern in the gut and that NDRG1, NDRG2 and NDRG4 (1) can serve as biomarker for colorectal cancer and (2) have tumor suppressive properties mainly affecting cell proliferation and epithelial-mesenchymal transition.

General significance

Similar effects of the NDRGs on the key-hallmarks of cancer, could implicate analogous functions in other tissue/cancer types.  相似文献

Summary Early observations of the feeding habits of Angolosaurus skoogi (Andersson), a 50–120 g lizard inhabiting the almost vegetationless dunes of the northern Namib Desert, indicated that this species was herbivorous. Less than two percent of living lizard species are predominantly herbivorous and these species tend to have a high body mass (>100 g). We therefore analysed fecal pellets of A. skoogi to obtain dietary details of this relatively rare lizard. Eighteen food taxa were identified: 81 percent by mass was composed of vegetable matter, of which 56 percent was an endemic, perennial cucurbit Acanthosicyos horrida. Both spatial and temporal variation in dietary composition were apparent. Ready access to water in the form of A. horrida is postulated to have allowed Angolosaurus to occupy this extreme habitat and to forage on components of the windblown detritus, a food source that would probably not be usable in other circumstances.  相似文献

Synopsis Fecal production was monitored to observe the effects of meal size on retention time of food in the digestive tracts of lemon sharks, Negaprion brevirostris. Initial appearance of feces occurred more rapidly when ration level was increased. The onset of fecal production was negatively correlated with rate of intake. Production of feces continued for a longer period of time when meal size was increased. Retention time of food was directly related to feeding rate, suggesting that the rate of digestion was constant. The correlation between retention time and intake on a percentage body weight basis was greater than the correlation between retention time and intake on an energy density basis. The use of agar to bind food may have delayed digestion and prolonged food passage for sharks fed an experimental diet.  相似文献

目的:探讨粪菌移植(FMT)联合聚乙二醇治疗顽固性功能性便秘的临床疗效及对患者生活质量及心理状态的影响。方法:选择第四军医大学西京消化病院2013年11月至2015年5月门诊顽固性功能性便秘患者35例,所有患者均行FMT治疗,同时联合聚乙二醇电解质散剂,疗程4周,采用便秘患者症状评分量表(KESS)、生存质量量表(PAC-QOL)、Zung焦虑自评量表(SAS)及抑郁自评量表(SDS)观察治疗前后及随访3个月便秘患者症状、生活质量及心理状态的改变。结果:治疗后及随访3个月时,患者的临床治愈率分别为37.1%、34.3%,改善率分别为77.1%、71.4%,KESS、PAC-QOL、SAS、SDS总评分均较治疗前明显减低(P0.05)。治疗及随访期间均未发生严重不良反应。结论:FMT联合聚乙二醇能安全、有效的改善顽固性功能性便秘患者的相关症状,提高生活质量,改善异常心理状态,值得临床推广。  相似文献
