为了研制戊型肝炎新型基因工程疫苗,利用汉逊酵母表达系统表达重组戊型肝炎病毒样颗粒,成功构建了重组戊型肝炎疫苗工程菌株HP/HEV2.3,对该菌株的发酵条件和纯化工艺进行了研究。先将工作种子批进行发酵培养,收集发酵后的细胞培养物;对其先后进行细胞破碎、澄清和超滤、硅胶吸附和解吸附、超滤浓缩换液、色谱纯化及除菌过滤,制得重组汉逊酵母戊型肝炎病毒样颗粒,纯化收率为33%,纯度达99%;电镜观察显示该重组汉逊酵母戊型肝炎病毒样颗粒与天然戊型肝炎病毒颗粒理论大小一致,为32 nm;基因序列与理论一致;SDS-PAGE分析结果表明其表达的外源蛋白质分子量与预期的目的蛋白质分子量大小一致,均为56 k Da,表达量占细胞总蛋白的26%,表达水平为1.0 g/L发酵液;Western blotting、ELISA活性检测及小鼠免疫接种效力试验ED_(50)结果表明,此重组汉逊酵母戊型肝炎病毒样颗粒具有良好的抗原性和免疫原性,可用于制造戊型肝炎新型基因工程疫苗。 相似文献

We investigated if (1) dissolved compounds excreted by Phaeocystis globosa and (2) transparent exopolymer particles (TEP) formed from carbohydrates excreted into the water affect the feeding of nauplii and females of the calanoid copepod Temora longicornis during a P. globosa bloom. Copepod grazing on the diatom Thalassiosira weissflogii in the presence of these possible grazing deterrents was measured during three successive weeks of a mesocosm study, simulating the development of a P. globosa bloom. Our results demonstrate no indication for the presence of feeding deterrents in the dissolved phase, but a strong inhibitory effect of transparent exopolymer particles (TEP) on the consumption of algae by both nauplii and adult copepods. The inhibitory effect of TEP was connected to the accumulation of DOM during the progress of the bloom. We suggest that a reduction in the grazing pressure of zooplankton may increase the survival of the liberated single cells during disruption of colonies and allow seeding populations to persist. Furthermore, P. globosa reduces the trophic efficiency of the food web not only by withdrawal of its colonies from grazing but also by a relaxation of the grazing pressure on co-occurring phytoplankton and by alteration of the food web structure via TEP production. 相似文献

猪O型口蹄疫病毒细菌样颗粒疫苗的制备与免疫原性鉴定 总被引:1,自引:0,他引:1
验证基于革兰氏阳性增强基质(Gram-positive enhancer matrix,GEM)展示口蹄疫病毒细菌样颗粒(Bacteria-like particles,BLP)疫苗的可行性。按照大肠杆菌偏好性密码子优化合成基于猪口蹄疫病毒Mya98株序列的3种抗原基因设计,并将其插入到含有锚钩蛋白基因的原核表达载体p QZ-PA,鉴定阳性后转入Escherichia coli BL21,进行诱导表达。利用SDS-PAGE与Western blotting对目的基因表达及产物的可溶性进行分析。利用GEM颗粒纯化目的蛋白,制备细菌样颗粒疫苗抗原;利用BCA试剂盒测定重组蛋白的浓度,将重组蛋白与白油佐剂乳化,制备疫苗,免疫5周龄小鼠,同时设商品化多肽苗对照与空白对照,免疫后不同时间采集试验小鼠血清,利用口蹄疫病毒多肽ELISA抗体检测试剂盒和O型口蹄疫抗体液相阻断酶联免疫(Enzyme-linked immunosorbent assay,ELISA)检测试剂盒检测免疫小鼠血清的抗体水平;利用噻唑蓝比色法(Methylthiazolyldiphenyl-tetrazolium bromide,MTT)测定淋巴细胞增殖情况;利用荧光定量PCR方法检测相关细胞因子表达,评价细胞免疫水平。SDS-PAGE结果表明,设计在大肠杆菌中的3种口蹄疫病毒抗原基因均以可溶形式获得高效表达;Western blotting结果显示,表达的重组蛋白能够与口蹄疫病毒阳性血清发生反应,利用GEM颗粒能够实现重组蛋白的一步离心纯化,制备BLP疫苗抗原;免疫试验结果表明,设计的重组抗原B(T1BT2)4B不但能够刺激免疫小鼠产生更高水平的多肽特异性ELISA抗体与口蹄疫特异性液相阻断抗体,而且产生了更高水平的脾淋巴细胞增殖及Th1型的细胞因子分泌。初步实验结果表明,本研究制备的BLP疫苗GEM-B(T1BT2)4B具有良好的免疫原性,为研究口蹄疫病毒基因工程亚单位疫苗开辟了一条新的思路。 相似文献

将大葱种子干燥成1.8%-10.5%的不同含水量后。在50℃、35℃、20℃和-18℃条件下密闭贮存16个月。通过对棕榈酸、硬脂酸、油酸、亚油酸、亚麻酸含量的研究,结果表明,随着贮存温度的升高,种子中棕榈酸、硬脂酸、油酸、亚油酸、亚麻酸含量不断升高,但在20℃和-18℃的差异不明显。在50℃下含水量为1.8%~6.6%的种子。5种脂肪酸含量明显大于含水量为7.4%-10.5%的种子。 相似文献

By employing in situ reduction of metal precursor and metal‐assisted carbon etching process, this study achieves a series of ultrafine transition metal‐based nanoparticles (Ni–Fe, Ni–Mo) embedded in N‐doped carbon, which are found efficient catalysts for electrolytic water splitting. The as‐prepared hybrid materials demonstrate outstanding catalytic activities as non‐noble metal electrodes rendered by the synergistic effect of bimetal elements and N‐dopants, the improved electrical conductivity, and hydrophilism. Ni/Mo2C@N‐doped porous carbon (NiMo‐polyvinylpyrrolidone (PVP)) and NiFe@N‐doped carbon (NiFe‐PVP) produce low overpotentials of 130 and 297 mV at a current density of 10 mA cm?2 as catalysts for hydrogen evolution reaction and oxygen evolution reaction, respectively. In addition, these binder‐free electrodes show long‐term stability. Overall water splitting is also demonstrated based on the couple of NiMo‐PVP||NiFe‐PVP catalyzer. Magnetization measurements of the European eel Anguilla anguilla demonstrated the presence of magnetic material concentrated in the region of the mandibular canals of the lateral line system. The data suggest that the material is magnetite, has a size suitable for magnetoreception and is of biogenic origin. The presence of magnetic particles in the lateral line system is discussed in relation to their possible role in allowing the fish to orientate with respect to the geomagnetic field during their extensive oceanic spawning migrations. 相似文献

为研究重组腺病毒载体作为HPV16预防性疫苗的可行性,构建了含密码子优化型HPV 16 L1基因的重组腺病毒,并对优化基因在哺乳动物细胞中的表达进行研究。首先按照哺乳动物密码子偏好对野生型HPV16 L1基因进行改造并合成优化基因,命名为mod.HPV16L1。将mod.HPV16L1基因克隆到穿梭质粒PDC316上,与骨架质粒共转染293细胞,在细胞内包装重组腺病毒rAd-mod.HPV16L1。用免疫印迹法检测病毒感染的293T细胞中HPV16L1蛋白的表达。通过Optiprep密度梯度超速离心法纯化HPV16 L1病毒样颗粒(VLPs)。用磷钨酸负染,在电子显微镜下观察HPV16 L1蛋白自我装配形成的VLPs。结果显示,重组腺病毒载体可介导mod.HPV16 L1基因在哺乳动物细胞内的高效表达,L1蛋白可自我装配形成VLPs。 相似文献

Summary The cytoplasmic content and the distribution of intramembrane particles (IMPs) of the plasma membrane of isolated sperm cells of perennial ryegrass (Lolium perenne L.) have been characterized using flow cytometry, transmission electron microscopy, confocal scanning laser microscopy and freeze-fracture studies. The isolated haploid sperm cells contain a variety of cell organelles with the exception of microtubules. Proplastids and plastids with starch were observed, although only rarely. Vacuoles containing remnants of organelles and stacked lamellae of endoplasmic reticulum with cytoplasmic inclusions were observed frequently, indicating that autophagy takes place. The number of mitochondria varies from 11 to 26 with an average of 17. Generally, the nucleus has a lobed shape and displays various interphasic stages of chromatin condensation. The analysis of the number of mitochondria and the nuclear state did not show evidence of sperm cell dimorphism. The cytological variability observed, could be explained by differences in developmental stages already present in vivo at the moment of isolation. No correlation between the number of mitochondria and the nuclear cross-sectioned area and/or the condensation state of the chromatin could be found. The density of intramembrane particles of the plasma membrane on the exoplasmic fracture face is more than twice that on the protoplasmic fracture face. That is the opposite of what was found for sporophytic cells of perennial ryegrass. These results are discussed in relation to the potential use of these cells for biotechnology and developmental studies. 相似文献

Summary. Hepatitis B core antigen is one of the most promising protein carriers of foreign epitopes of various human and animal pathogens.
Chimeric HBcAg particles can be used as effective artificial immunogenes. Unfortunately, not all chimeric proteins are able
to be particulated. The dependence of correct or incorrect folding of chimeric proteins on physical and chemical properties
of inserts was studied with the help of ProAnalyst, SALIX and QSARPro computer programs. We have found that insertion of amino
acids with high hydrophobicity, large volume, and high β-strand index prevent self-assembling chimeric proteins. These factors are most important for the C-termini of inserts. Recommendations
for obtaining correct folding of chimeric HBcAg particles have been given.

Nucleic acids from 41 strains of Metarhizium anisopliae, obtained from different parts of the world were extracted and examined by electrophoresis. Strong bands of double‐stranded RNA (dsRNA) were detected in two isolates from Brazil, V215 and V291, which had, respectively, 13 and 9 distinct bands ranging in size from ca. 0.75 to 3.5 kb. Icosahedral virus‐like particles (VLPs) (ca. 33 nm in diameter) were observed by transmission electron microscopy in extracts of these isolates. The VLPs and dsRNA were both absent from a clone of the isolate V291 which had been subcultured successively on solid medium. Bioassays against the aphid Myzus persicae showed no detectable difference in virulence between the clone of V291 which contained dsRNA and the clone that did not. 相似文献
