The effect of taurine on ATP-dependent calcium transport was examined in guinea-pig cardiac ventricle homogenates and in microsomal preparations enriched in sarcoplasmic reticulum. Taurine (5?50 mM) did not affect ATP-dependent calcium binding or uptake in either of these preparations or alter the rate of decay of calcium uptake activity. Taurine (20 mM) also did not affect the oxalate-dependent calcium uptake stimulation noted in the presence of cyclic AMP-dependent protein kinase and cyclic AMP. The mechanism by which taurine alters cardiac function remains to be elucidated. 相似文献

Synthetic TRH, crude hypothalamic extract and partially purified prolactin releasing factor stimulated prolactin and growth hormone release from isolated secretory granules. Somatostatin and partially purified prolactin release-inhibiting factor inhibited release of both hormones. 7 beta, 17-Dimethyltestosterone (17 beta-hydroxy-7 beta, 17-dimethyl-4-androsten-3-one) (I) was given to three subjects in oral doses of 400 mg per day for ten days. The initial dose contained the steroid tritiated in the 6 and 7 positions. Plasma levels and urinary excretion patterns were followed in all three subjects. Isolations were done on the urine, plasma, and stools of one patient. From the urine 7 beta, 17-dimethyl- 5 alpha-androstane-3 beta,17 beta-diol (VI) was isolated from the nonhydrolyzed fractions. Unchanged (I), 7 beta,17-dimethyl-5 beta-androstane-3 alpha,17 beta-diol (III) and 7 beta, 17-dimethyl-5 beta-androstane-3 beta,17 beta-diol (IV) were isolated from the nonhydrolyzed and enzyme-hydrolyzed fractions. 7 beta,17-dimethyl-5 alpha-androstane-3 alpha,17 beta-diol (V) was isolated from the enzymatic fractions. From the stools were isolated unchanged (I), (III), (IV), (V), and (VI). Unchanged (I) and its 5 alpha-dihydro derivative (17 beta-hydroxy-7 beta,17-dimethyl-5 alpha-androstan-3-one) (II) were identified in the plasma. The total recovery of radioactivity in the one patient on whom the isolations were done was 57%; 40% from the urine and 17% from the stools. 相似文献

Summary An organization for data-processing in phyto-sociology and plant ccology is deseribed. A good preparation by means of standard sheets for collecting and codifying information from the field is emphasized (Fig. 1). Fig. 2–5 present the arrangement and the logical sequence of the different methods of data analysis (ecological profiles, factor analysis,...) which allows the study of the relations between environmental and floristical variables, in a quick and efficient way. Cooperation with the Ecothèque méditerranéene at Montpellier is both necessary and fruitful. One example is presented using the selection e of 58 relevés of salt marsh vegetation retained by the Working Group for Data-Processing of the International Society for Vegetation Science.
Résumé Les auteurs proposent ici une organisation du traitement des données en phytosociologie et phyto-écologie. L'organisation repose sur un système bien développé pour rassembler et codifier l'information des reléves. (fig.1). Un ordinogramme (Fig. 2–5) présente l'agencement et l'enchaînement des différentes méthodes d'analyse des données (profils écologiques, analyse factorielle, ...) qui permettent d'étudier les relations entre variables du milieu et variables floristiques. La conception de l'enchaînement des fichiers et des programmes de traitement permet d'aboutir assez rapidement aux résultats, facilitant ainsi la tâche du phytosociologue et du phyto-écologue. Ce travail a été réalisé en commun entre le C.E.P.E. Louis Emberger et l'Ecothèque méditerranéenne conduisant à une coopération nécessaire et utile. Une exemple est traité utilissant la sélection e de 58 relevés de milieux salés, sélection retenue par le groupe de travail traitement des données en phytosociologie de l'Association internationale de phytosociologie.相似文献

Summary A method is described for obtaining and culturing large numbers of lung cells from normal adult male rats. The lungs were
perfused in situ to remove blood cells and then perfused via the trachea with a trypsin-collagenase solution to initiate tissue
digestion. The tissue was further digested in the enzyme solution and approximately 2×108 viable lung cells were obtained per animal. Primary cultures contained a mixed cell population. Through eight subcultures
about 70% of the cell population possessed an epithelial-like morphology, whereas the remaining 30% was fibroblast-like. Three
clones of epithelial-like cells were isolated at the fourth subculture. The mass culture lung cells and the epithelial-like
clone that was studied retained a normal karyotype and did not grow in soft agar. Both the mass culture cells and the epithelial
clone metabolized the lung carcinogen benzo(a)pyrene (BP) to water-soluble products. Furthermore, the mass culture lung cells
metabolized BP to intermediate(s) which mutated Chinese hamster V79 cells from ouabain sensitivity to ouabain resistance.
These lung cell cultures have potential use in cell transformation, mutation and carcinogen metabolism studies.

Secretary activities associated with the rostellum of adult Echinococcus granulosus were studied using histological, histochemical, and ultrastructural techniques, following rapid fixation of the cestodes in situ in the small intestine of the anaesthetised dog. Studies concentrated on the host-parasite interface from 30 to 35 days postinfection. At this time, contraction of the muscular rostellar pad appeared to be associated with extension of the apical rostellum into a crypt of Lieberkühn. Crypt invasion by the apical rostellum coincided with morphological changes and secretory activity in a group of modified tegumental cells, previously referred to as the rostellar gland. Secretory material, a cystine-rich protein, was observed in the nuclei and cytoplasm of the rostellar gland cells. Release of this material into the interface was seen only following crypt invasion by the apical rostellum. Although the mechanism of release is not clear, it may be analagous to holocrine secretory mechanisms, since apparent degeneration of the rostellar gland region was associated with secretion. Possible functional activities of the secretion associated with hook formation, nutrition, regulation, adhesion, and protection are discussed. 相似文献

Kinetic studies of the reduction of ferrioxamine B (Fe(Hdesf)+) by Cr(H2O)62+, V(H2O)62+, and dithionite have been performed. For Cr(H2O)62+ and V(H2O)62+, the rate is ?d[Fe(Hdesf)+]/dt = k[Fe(Hdesf)+][M2+]. For Cr(H2O)62+, k = 1.19 × 104 M?1 sec?1 at 25°C and μ = 0.4 M, and k is independent of pH from 2.6 to 3.5. For V(H2O)62+, k = 6.30 × 102 M?1 sec?1 at 25°C, μ = 1.0 M, and pH = 2.2. The rate is nearly independent of pH from 2.2 to 4.0. For Cr(H2O)62+ and V(H2O)62+, the activation parameters are ΔH≠ = 8.2 kcal mol?1, ΔS≠ ?12 eu and ΔH≠ = 1.7 kcal mol?1, ΔS≠ = ?40 eu (at pH 2.2) respectively. Reduction by Cr(H2O)62+ is inner-sphere, while reduction by V(H2O)62+ is outer-sphere. Reduction by dithionite follows the rate law ?d[Fe(Hdesf)+]/dt =kK[Fe(Hdesf)+][S2O42?] where K is the equilibrium constant for dissociation of S2O42? into SO2? radicals. The value of k at 25°C and μ = 0.5 is 2.7 × 103 M?1 sec?1 at pH 5.8, 3.5 × 103 M?1 sec?1 at pH 6.8, and 4.6 × 103 M?1 sec?1 at pH 7.8, and ΔH≠ = 6.8 kcal mol?1 and ΔS≠ = ?19 eu at pH 7.8. 相似文献

Somatic DNA rearrangement generates functional rat immunoglobulin kappa chain genes: the J kappa gene cluster is longer in rat than in mouse 总被引:5,自引:0,他引:5
The kappa immunoglobulin (Ig) genes from rat kidney and from rat myeloma cells were cloned and analyzed. In kidney DNA one C kappa species is observed by Southern blotting and cloning in phage vectors; this gene most likely represents the embryonic configuration. In the IR52 myeloma DNA two C kappa species are observed: one in the same configuration seen in kidney and one which has undergone a rearrangement. This somatic rearrangement has brought the expressed V region to within 2.7 kb 5' of the C kappa coding region; the rearrangement site is within the J kappa cluster which we have mapped. The rat somatic Ig rearrangement, therefore, closely resembles that seen in mouse Ig genes. In the rat embryonic fragment two J kappa segments were mapped at 2 and 4.3 kb 5' from the C kappa coding region. Therefore, the rat J kappa cluster extends over about 2.3 kb, a region much longer than the 1.4 kb of the mouse and human J kappa clusters. In the region between C kappa and the expressed J kappa of IR52 myeloma DNA, and XbaI site present in the embryonic kappa gene has been lost. A somatic mutation has therefore occurred in the intervening sequence DNA approx. 0.7 kb 3' from the V/J recombination site. Southern blots of rat kidney DNA hybridized with different rat V kappa probes showed non-overlapping sets of bands which correspond to different subgroups, each composed of 8-10 closely related V kappa genes. 相似文献

Pronounced strand breakage of DNA analyzed by alkaline elution techniques was produced in intact Chinese hamster ovary cells by 25 μM HgCl2 within 1 hr or 100 μM HgCl2 within 15 min. HgCl2-induced strand breakage was directly proportional to concentration up to 100 μM and to time within 1 hr. Levels of reduced glutathione decreased following HgCl2 in parallel with the induction of DNA strand breakage. Evidence is presented that this rapid and pronounced induction of DNA strand breaks and other cytotoxic responses following acute exposure to HgCl2 resembles the cellular effects of X-rays. 相似文献
