Aspergillus niger IMI 303386 produced higher levels of intra- and extracellular -fructofuranosidase and inulinase on inulin than on sucrose. Intracellular -fructofuranosidase from sucrose medium catalysed the best transfructosylation reaction. The concentration of fructooligosaccharides (FOS) reached a maximum in 72 h with 25% (w/v) sucrose. The FOS were purified and the main products were kestose and nystose. Inulinase hydrolysed inulin in an exofashion and released mainly fructose.  相似文献

We used FOS expression, widely accepted as a marker for neuronal activation, to evaluate physiologically induced activation of gonadotropin-releasing hormone (GnRH) neurons within intraventricular preoptic area grafts in hypogonadal (hpg) female mice. Hpg mice lack endogenous GnRH due to a mutated gene, but can respond to grafted GnRH neurons with reproductive development. The purpose of this study was to determine the degree to which the host brain regulates grafted GnRH neurons. FOS expression in grafted GnRH neurons was induced in progesterone-primed female mice paired with sexually active males. The degree of sexual activity did not affect the outcome, with 40.9 ± 12.2% of the grafted GnRH cells expressing FOS when male partners performed intromissions, and 47.5 ± 10.2% when they also ejaculated. There was little or no FOS expression in the grafts of unprimed hpg mice paired with sexually active males, in unpaired mice primed with progesterone or sequential estradiol benzoate and progesterone, or in controls. The pattern of FOS expression in the brains of the female hpg mice engaged in mating behavior was similar to that reported in other species, with moderate to high expression in the medial preoptic area, ventromedial nucleus, and medial amygdala in females paired with males that ejaculated. The present results support the hypothesis that host-derived activation of grafted GnRH neurons underlies aspects of reproductive responses seen in hpg mice with grafts, and further, that at least a portion of the host-graft connectivity is steroid sensitive. © 1996 John Wiley & Sons, Inc.  相似文献

The present study proposed the idea of segment manipulation of complex cognition (SMCC), and technically made it possible the quantitative treatment and systematical manipulation on the premise diversity. The segment manipulation of complex cognition divides the previous inductive strengths judgment task into three distinct steps, attempting to particularly distinguish the psychological processes and their rules. The results in Experiment 1 showed that compared with the traditional method, the quantitative treatment and systematical manipulation of SMCC on the diversity did not change the task’s nature, and remain rational and a good measurement of inductive strength judgment. The results in Experiment 2 showed that the participants’ response rules in the triple-step task were expected from our proposal, and that in Step 2 the “feeling of surprise” (FOS), which seems implausible but predicted from the diversity premises, was measured, and its component might be the critical part that produced the diversity effect. The “feeling of surprise” may reflect the impact of emotion on cognition, representing a strong revision to premise probability principle of pure rational hypothesis proposed by Lo et al., and its roles in the diversity effect are worthy of further research. In this regards were discussed the mistakes that the premise probability principle makes when it takes posterity probability as prior probability.  相似文献

Rhythmic feeding in rabbit pups is a natural model to study food entrainment because, similar to rodents under a schedule of food restriction, these animals show food-anticipatory activity (FAA) prior to daily nursing. In rodents, several brain systems, including the orexinergic system, shift their activity to the restricted feeding schedule, and remain active when subjects are hungry. As the lamina terminalis and regions of the preoptic area participate in the control of behavioral arousal, it was hypothesized that these brain regions are also activated during FAA. Thus, the effects of daily milk ingestion on FOS protein expression in the organum vasculosum of lamina terminalis (OVLT), median preoptic nucleus (MnPO), and medial preoptic area (MPOA) were examined using immunohistochemistry before and after scheduled time of nursing in nursed and fasted subjects. Additionally, FOS expression was explored in orexin (ORX) cells in the lateral hypothalamic area and in the supraoptic nucleus (SON) because of their involvement in arousal and fluid ingestion, respectively. Pups were entrained by daily nursing, as indicated by a significant increase in locomotor behavior before scheduled time of nursing in both nursed and fasted subjects. FOS was significantly higher in the OVLT, MnPO, and MPOA at the time of nursing, and decreased 8?h later in nursed pups. In fasted subjects, this effect persisted in the OVLT, whereas in the MnPO and MPOA, values did not drop at 8?h later, but remained at the same level or higher than those at the time of scheduled nursing. In addition, FOS was significantly higher in ORX cells during FAA in nursed pups in comparison with 8?h later, but in fasted subjects it remained high during most fasting time points. Additionally, OVLT, SON, and ORX cells were activated 1.5?h after nursing. We conclude that the OVLT, MnPO, and MPOA, but not SON, may participate in FAA, as they show activation before suckling of periodic milk ingestion, and that sustained activation of the OVLT, MnPO, and MPOA by fasting may contribute to the high arousal state associated with food deprivation. In agreement with this, ORX cells also remain active after expected nursing, which is consistent with reports in other species.  相似文献

Fructansucrases (FSs) catalyze a transfructosylation reaction with sucrose as substrate to produce fructo-oligosaccharides and fructan polymers that contain either β-2,1 glycosidic linkages (inulin) or β-2,6 linkages (levan). Levan-synthesizing FSs (levansucrases) have been most extensively investigated, while detailed information on inulosucrases is limited. Importantly, the molecular basis of the different product specificities of levansucrases and inulosucrases is poorly understood.We have elucidated the three-dimensional structure of a truncated active bacterial GH68 inulosucrase, InuJ of Lactobacillus johnsonii NCC533 (residues 145-708), in its apo form, with a bound substrate (sucrose), and with a transfructosylation product. The sucrose binding pocket and the sucrose binding mode are virtually identical with those of GH68 levansucrases, confirming that both enzyme types use the same fully conserved structural framework for the binding and cleavage of the donor substrate sucrose in the active site. The binding mode of the first transfructosylation product 1-kestose (Fru-β(2-1)-Fru-α(2-1)-Glc, where Fru = fructose and Glc = glucose) in subsites − 1 to + 2 shows for the first time how inulin-type fructo-oligosaccharide bind in GH68 FS and how an inulin-type linkage can be formed. Surprisingly, observed interactions with the sugar in subsites + 1 and + 2 are provided by residues that are also present in levansucrases. The binding mode of 1-kestose and the presence of a more distant sucrose binding site suggest that residues beyond the + 2 subsite, in particular residues from the nonconserved 1B-1C loop, determine product linkage type specificity in GH68 FSs.  相似文献

The present study investigated the effects of prebiotic fructooligosaccharide (FOS) on the innate immune response, stress resistance, digestive enzyme activities, growth factors and survival of Caspian Roach (Rutilus rutilus) fry. After acclimation, fish (0.67 ± 0.03 g) were allocated into 12 tanks (50 fish per tank) and triplicate groups were fed a control diet or diets containing 1%, 2% or 3% FOS. At the end of the trial (7 weeks), humoral innate immune parameters (serum Ig levels, lysozyme activity and alternative complement activity (ACH50)), resistance to salinity stress (150 g L−1), digestive enzyme activities (amylase, lipase and protease) and growth factors (final weight, weight gain, specific growth rate (SGR), food conversion ratio (FCR), and condition factor) were assessed. At the end of the study the innate immune responses (Ig levels, lysozyme activity and ACH50) were significantly higher in 2% and 3% FOS fed fish (P < 0.05), whereas, 1% dietary FOS only elevated serum lysozyme activity. All dietary FOS levels significantly increased resistance to a salinity stress challenge (P < 0.05) and highest survival was observed in the 3% FOS group. Similarly, digestive enzyme activities were significantly elevated with increasing levels of dietary FOS (P < 0.05). Subsequently, elevated growth performance (final weight, SGR and FCR) was observed in roach fed 2% and 3% FOS compared to the control group (P < 0.05). These results indicate that FOS can be considered as a beneficial dietary supplement for improving the immune response, stress resistance, digestive enzyme activities and growth performance of Caspian roach fry.  相似文献

本实验利用免疫组织化学方法,以FOS阳性反应作为神经元活动的标志,研究了棕色田鼠在受到同性和异性尿液刺激后主嗅球和副嗅球的神经元活动,表明两大嗅觉系统均有感知社会性化学信号的功能.通过比较棕色田鼠在同性和异性尿液刺激后副嗅球和主嗅球的嗅小球细胞层(GL)、僧帽细胞层(MIT)、颗粒细胞层(GRL)中FOS阳性神经元数量,发现不同性别尿液刺激后棕色田鼠的副嗅球各细胞层FOS阳性神经元数量比对照组明显增加;棕色田鼠受到异性尿液刺激后其副嗅球各细胞层的FOS阳性神经元数量均多于同性尿液刺激组.不同性别尿液刺激后棕色田鼠的主嗅球各细胞层FOS阳性神经元数量相较于对照组有增加或增加显著;异性尿液刺激组主嗅球各细胞层的FOS阳性神经元数量均多于同性尿液刺激组.说明棕色田鼠的副嗅球和主嗅球均参与了通过尿液介导的性别个体的识别.  相似文献
