The ParABS system is supposed to be responsible for plasmid partitioning and chromosome segregation in bacteria. ParABS ensures a high degree of fidelity in inheritance by dividing the genetic material equally between daughter cells during cell division. However, the molecular mechanisms underlying the assembly of the partition complex, representing the core of the ParABS system, are still far from being understood. Here we demonstrate that the partition complex is formed via liquid–liquid phase separation. Assembly of the partition complex is initiated by the formation of oligomeric ParB species, which in turn are regulated by CTP-binding. Phase diagrams and in vivo analysis show how the partition complex can further be spatially regulated by parS. By investigating the phylogenetic variation in phase separation and its regulation by CTP, we find a high degree of evolutionary conservation among distantly related prokaryotes. These results advance the understanding of partition complex formation and regulation in general, by confirming and extending recently proposed models.  相似文献

Seed banking (or dormancy) is a widespread bet-hedging strategy, generating a form of population overlap, which decreases the magnitude of genetic drift. The methodological complexity of integrating this trait implies it is ignored when developing tools to detect selective sweeps. But, as dormancy lengthens the ancestral recombination graph (ARG), increasing times to fixation, it can change the genomic signatures of selection. To detect genes under positive selection in seed banking species it is important to (1) determine whether the efficacy of selection is affected, and (2) predict the patterns of nucleotide diversity at and around positively selected alleles. We present the first tree sequence-based simulation program integrating a weak seed bank to examine the dynamics and genomic footprints of beneficial alleles in a finite population. We find that seed banking does not affect the probability of fixation and confirm expectations of increased times to fixation. We also confirm earlier findings that, for strong selection, the times to fixation are not scaled by the inbreeding effective population size in the presence of seed banks, but are shorter than would be expected. As seed banking increases the effective recombination rate, footprints of sweeps appear narrower around the selected sites and due to the scaling of the ARG are detectable for longer periods of time. The developed simulation tool can be used to predict the footprints of selection and draw statistical inference of past evolutionary events in plants, invertebrates, or fungi with seed banks.  相似文献

This paper discusses the transition from heather-dominated vegetation to one dominated by grass. The dominating species involved are Calluna vulgaris and Deschampsia flexuosa. The study covered three years, 1980–82, and was done at the Hoorneboeg heathland in the Netherlands. Several factors account for this transition: damage caused by the chrysomelid heather beetle Lochmaea suturalis Thomson, soil type, and interaction between beetle damage and Deschampsia when already present underneath or in the immediate vicinity of a Calluna canopy. There was a time-lag between beetle damage and grass establishment or expansion. In fact, this transition concerns a type of vegetation succession and can be treated as such. This succession therefore involves several causative factors external to the system Calluna-Deschampsia. The successional process is conceived as a stochastic one defined by transition probabilities which depend on a time component, a space component and a logistic response component. All these components are discussed in this paper, but the discussion of yet another component, the crowding of Deschampsia itself, will be treated separately.  相似文献

Determining the probability of fixation of beneficial mutations is critically important for building predictive models of adaptive evolution. Despite considerable theoretical work, models of fixation probability have stood untested for nearly a century. However, recent advances in experimental and theoretical techniques permit the development of models with testable predictions. We developed a new model for the probability of surviving genetic drift, a major component of fixation probability, for novel beneficial mutations in the fungus Aspergillus nidulans, based on the life-history characteristics of its colony growth on a solid surface. We tested the model by measuring the probability of surviving drift in 11 adapted strains introduced into wild-type populations of different densities. We found that the probability of surviving drift increased with mutant invasion fitness, and decreased with wild-type density, as expected. The model accurately predicted the survival probability for the majority of mutants, yielding one of the first direct tests of the extinction probability of beneficial mutations.  相似文献

A Monte Carlo simulation was performed to generate probability density distributions describing hourly inhalation rates of male and female construction workers. Ranges of oxygen uptake and ventilation rates reported in the literature for various construction trades were manipulated in a simulation to generate hourly inhalation rates for male construction workers. The resulting inhalation rates can be represented by a log normal distribution with mean ± standard deviation of 1.40 ± 0.51 m3/h. Hourly inhalation rates for female construction workers were scaled from those of their male counterparts using relative awake-time inhalation rates of men and women of the general public. Female construction workers' inhalation rates can be represented by a log normal distribution with a mean ± standard deviation of 1.25 ± 0.66 m3/h. If the nature of the work is well understood, male and female construction workers' hourly inhalation rates can be scaled according to the construction trade. Construction trade-specific scaling factors were developed and range from 0.78 for electricians to 1.11 for ironworkers.  相似文献

Based on capture-mark-recapture sampling methods the problem of estimating unknown population size was considered. The sampling started with the assumption that at the beginning of the experiment all the individuals were unmarked, and the unmarked individuals caught in each sample will be marked and returned to the original population before the next sample is drawn. It is also assumed that the population is closed by birth, death, emigration and immigration. Using a general inverse sampling approach, the unknown population size N is estimated by a maximum likelihood estimator (MLE), and a simple form for approximate MLE is obtained. The probability function for S (the minimum number of samples required to be drawn to have L (L ≥ 1) samples, each of which contains at least one marked individual) and the form for E[S] are also obtained. In addition, corrections and improvements of some previous works in this field are given.  相似文献

Large anthropogenic 14C datasets are widely used to generate summed probability distributions (SPDs) as a proxy for past human population levels. However, SPDs are a poor proxy when datasets are small, bearing little relationship to true population dynamics. Instead, more robust inferences can be achieved by directly modelling the population and assessing the model likelihood given the data. We introduce the R package ADMUR which uses a continuous piecewise linear (CPL) model of population change, calculates the model likelihood given a 14C dataset, estimates credible intervals using Markov chain Monte Carlo, applies a goodness-of-fit test, and uses the Schwarz Criterion to compare CPL models. We demonstrate the efficacy of this method using toy data, showing that spurious dynamics are avoided when sample sizes are small, and true population dynamics are recovered as sample sizes increase. Finally, we use an improved 14C dataset for the South American Arid Diagonal to compare CPL modelling to current simulation methods, and identify three Holocene phases when population trajectory estimates changed from rapid initial growth of 4.15% per generation to a decline of 0.05% per generation between 10 821 and 7055 yr BP, then gently grew at 0.58% per generation until 2500 yr BP.This article is part of the theme issue ‘Cross-disciplinary approaches to prehistoric demography’.  相似文献
