The conformational change of myoglobin (Mb) during guanidine hydrochloride (GuHCl)-induced protein unfolding in the presence of various ionic liquids (ILs) in phosphate buffer was investigated using both the Soret band absorption and the fluorescence of tryptophan measurements. The GuHCl-induced denaturation midpoints of Mb derived from the absorption and fluorescence spectra were almost similar in the presence of 150 mM ILs with the same cation 1-butyl-3-methylimidazolium (Bmim+) but different anions (BF4, NO3, Cl, and Br) in phosphate buffer. In addition, the denaturation midpoints of Mb in the presence of ILs were little lower than those in the absence of ILs in phosphate buffer. For the sake of clarity and comparison, we also measured the GuHCl-induced denaturation midpoints of Mb in the presence of 150 mM sodium salts with different anions (BF4, NO3, Cl, and Br) in phosphate buffer and found that their corresponding denaturation midpoints of Mb were almost similar to those observed in the absence of sodium salts in phosphate buffer. These experimental data indicate that Bmim+ cation can promote the unfolding of Mb. Further experiments revealed that the denaturation ability of ILs increases with increasing alkyl chain length of imidazolium cation of ILs and that hydroxyl-substituted imidazolium cation could also promote the unfolding of Mb.  相似文献

An accurate and selective method using micellar liquid chromatography was developed to determine metformin hydrochloride both in its pharmaceutical dosage forms and human plasma. Separation was conducted using a Zorbax SB‐Phenyl (250 × 4.6 mm id) stainless steel column at ambient temperature after pre‐column derivatization with 9,10‐phenanthraquinone. A mobile phase composed of 0.1 M sodium dodecyl sulfate, 10% 1‐propanol and triethylamine (0.3%) in 0.02 M phosphoric acid, adjusted to pH 2.5, was used at a flow rate of 1 ml/min with fluorimetric detection at 450 nm after excitation at 306 nm. The proposed method showed high sensitivity with limit of quantification of 0.35 μg/ml and limit of detection of 0.23 μg/ml, being linear from 0.5 to 3.0 μg/ml. Being highly sensitive, the method could be applied to spiked human plasma, and also to follow the pharmacokinetic parameters of the studied drug in healthy volunteers after administration of both its immediate and sustained release tablet formulations. Such procedures were carried out without any extraction steps, which improves the accuracy and precision of the proposed method when applied to human plasma. Detailed validation procedures were also carried out giving results in accordance with the comparison method. The proposed method has also the advantage of being environmentally safe, where the use of organic solvents is highly limited in comparison with other traditional chromatographic separation methods that depend mainly on a high proportion of organic modifiers. This concept, in turn, emphasizes the application of green chemistry in the analysis of pharmaceutical products. The simplicity, relatively low cost and short analysis time of the suggested method makes it a candidate for routine quality control work.  相似文献

This work presents the optimization of analytical procedures for the determination of two antibiotics, oxolinic acid (OA) and flumequine (FL), in bed sediment, river water, and soil samples. Three extraction methods (microwave-assisted extraction (MAE), ultrasonication, and reflux) were tested, and the highest recoveries were obtained with MAE (94 ± 3% and 95 ± 3% for OA and FL, respectively). A solid-phase extraction (SPE) clean-up step was optimized by comparing two polymeric sorbents: Oasis HLB and Oasis MAX. The final extracts were analyzed by liquid chromatography with fluorimetric detection. Limits of detection (LOD) obtained for OA and FL in soil and sediment ranged from 0.3 to 0.5 µg kg?1. Meanwhile, a novel SPE procedure was also implemented for OA and FL determination in river water samples. It also relied on the use of Oasis MAX, and recovery rates were in the range 90–94%; LODs were 2 ng L?1 for both OA and FL. These methods were applied for the analysis of samples taken in the Seine River basin (France). The obtained results demonstrated the widespread occurrence of OA and FL, at ng L?1 and µg kg?1 levels in water and sediment/soil, respectively, and their persistence in the environment.  相似文献

为调查马凡综合征(Marfan syndrome, MFS)患者的原纤维蛋白-1(Fibrillin-1, FBN1)基因突变情况, 应用聚合酶链反应(PCR)和变性高效液相色谱法(Denaturing high-performance liquid chromatography, DHPLC)对MFS患者的FBN1基因进行突变筛查, 对DHPLC初筛异常的DNA片段进行测序分析。结果在两个MFS家系中发现FBN1基因两种新的突变: 一种为复合突变包含第55号外显子的缺失突变c.6862_6871delGGCTGTGTAG (p.Gly2288MetfsX109)、同义突变c.6861A>G和内含子的突变c.[6871+1_6871+11delGTAAGAGGATC; 6871+34dupCATCAGAAGTGACAGTGGACA]; 另一种为第20号外显子的错义突变c.2462G>A(p.Cys821Tyr)。研究表明, FBN1基因的缺失突变c.[6862_6871delGGCTGTGTAG; 6871+1_6871+11delGTAAGAGGATC] (p.Gly2288MetfsX109)和错义突变c.2462G>A(p.Cys821Tyr)可能分别是这两个家系患者的致病原因。  相似文献

不同沼液用量对夏玉米源库代谢关键酶及产量的影响   总被引:3,自引:0,他引:3  
以玉米杂交种郑单958为材料,研究了3个不同沼液用量(15000、22500、30000 kg·hm-2)对夏玉米源库代谢关键酶和产量构成因素的影响.结果表明:与对照和施氮肥相比,田间浇灌沼液不仅可提高夏玉米地上部干物质量、叶面积指数和叶绿素含量,而且可提高叶片中硝酸还原酶(NR)、谷氨酰胺合成酶(GS)、蔗糖磷酸合成酶(SPS)和籽粒中蔗糖合成酶(SS)活性,从而显著增加了玉米穗粗、穗长、穗行数、穗粒数、穗粒重、千粒重和单位面积产量.在3个沼液用量中,以22500 kg·hm-2处理(拔节期、大口期、抽雄期各追施7500 kg·hm-2)对提高玉米酶活性和产量的效果最好,其产量可达14006.7 kg·hm-2,比对照增产40.7%.  相似文献
