Versatile synthetic routes have been applied to prepare the new asymmetric phosphine-phosphite ligands 8 and 12. The chiral ligands have been designed so that the corresponding ligating groups have similar electronic properties and steric bulk, but 8 forms 6-, while 12 forms 7-membered chelate rings in their coordination compounds. The chelate size variation results in a markedly different coordination behavior towards Pt(II). In their reactions with Pt(PhCN)2Cl2 at 1:1 stoichiometry 12 forms the expected Pt(12)Cl2 complex, while 8 gives the cation quantitatively. In the kinetically controlled reaction is the major product even at a 8:Pt(PhCN)2Cl2 = 1:2 ratio. Most interestingly, at 1:1 ligand to precursor ratio, cation rearranges to Pt(8)Cl2 within one day, indicating that the neutral complex is thermodynamically more favorable.  相似文献

Histone deacetylase (HDAC) inhibitors represent a promising new avenue of therapeutic options for a range of neurological disorders. Within any particular neurological disorder, neuronal damage or death is not widespread; rather, particular brain regions are preferentially affected. Different disorders exhibit distinct focal pathologies. Hence, understanding the region-specific effects of HDAC inhibitors is essential for targeting appropriate brain areas and reducing toxicity in unaffected areas. The outcome of HDAC inhibition depends on several factors, including the diversity in the central nervous system expression of HDAC enzymes, selectivity of a given HDAC inhibitor for different HDAC enzymes, and the presence or absence of cofactors necessary for enzyme function. This review will summarize brain regions associated with various neurological disorders and factors affecting the consequences of HDAC inhibition.  相似文献

In mice, gene targeting by homologous recombination continues to play an essential role in the understanding of functional genomics. This strategy allows precise location of the site of transgene integration and is most commonly used to ablate gene expression ("knock-out"), or to introduce mutant or modified alleles at the locus of interest ("knock-in"). The efficacy of producing live, transgenic mice challenges our understanding of this complex process, and of the factors which influence germline competence of embryonic stem cell lines. Increasingly, evidence indicates that culture conditions and in vitro manipulation can affect the germline-competence of Embryonic Stem cell (ES cell) lines by accumulation of chromosome abnormalities and/or epigenetic alterations of the ES cell genome. The effectiveness of ES cell derivation is greatly strain-dependent and it may also influence the germline transmission capability. Recent technical improvements in the production of germline chimeras have been focused on means of generating ES cells lines with a higher germline potential. There are a number of options for generating chimeras from ES cells (ES chimera mice); however, each method has its advantages and disadvantages. Recent developments in induced pluripotent stem (iPS) cell technology have opened new avenues for generation of animals from genetically modified somatic cells by means of chimera technologies. The aim of this review is to give a brief account of how the factors mentioned above are influencing the germline transmission capacity and the developmental potential of mouse pluripotent stem cell lines. The most recent methods for generating specifically ES and iPS chimera mice, including the advantages and disadvantages of each method are also discussed.  相似文献

Cell lineage commitment and differentiation are governed by a complex gene regulatory network. Disruption of these processes by inappropriate regulatory signals and by mutational rewiring of the network can lead to tumorigenesis. Cancer cells often exhibit immature or embryonic traits and dysregulated developmental genes can act as oncogenes. However, the prevailing paradigm of somatic evolution and multi-step tumorigenesis, while useful in many instances, offers no logically coherent reason for why oncogenesis recapitulates ontogenesis. The formal concept of “cancer attractors”, derived from an integrative, complex systems approach to gene regulatory network may provide a natural explanation. Here we present the theory of attractors in gene network dynamics and review the concept of cell types as attractors. We argue that cancer cells are trapped in abnormal attractors and discuss this concept in the light of recent ideas in cancer biology, including cancer genomics and cancer stem cells, as well as the implications for differentiation therapy.  相似文献

Maynard Smith and Szathmáry's analysis of the major transitions in evolution was based on changes in the way information is stored, transmitted and interpreted. With the exception of the transition to human linguistic societies, their discussion centred on changes in DNA and the genetic system. We argue that information transmitted by non-genetic means has played a key role in the major transitions, and that new and modified ways of transmitting non-DNA information resulted from them. We compare and attempt to categorise the major transitions, and suggest that the transition from RNA as both gene and enzyme to DNA as genetic material and proteins as enzymes may have been a double one. Unlike Maynard Smith and Szathmáry, we regard the emergence of the nervous system as a major transition. The evolution of a nervous system not only changed the way that information was transmitted between cells and profoundly altered the nature of the individuals in which it was present, it also led to a new type of heredity-social and cultural heredity-based on the transmission of behaviourally acquired information.  相似文献

Chromatin structure and function are regulated by families of protein-modifying enzymes that are sensitive to a variety of metabolic and environmental agents. These enzymes, and proteins that read the modifications they maintain, constitute a system by which environmental agents, such as chemical toxins and dietary components, can directly regulate patterns of gene expression. This review describes this environmental sensing system from an evolutionary perspective. It is proposed that persistent environmentally-induced changes in gene expression patterns can cause changes in phenotype that are acted upon by natural selection, and that epigenetic processes can potentially play central roles in evolution.  相似文献

Though there are multiple routes through which parents can influence their offspring, recent studies of environmentally induced epigenetic variation have highlighted the role of non-genomic pathways. In addition to the experience-dependent modification of DNA methylation that can be achieved via mother-infant interactions, there has been increasing interest in the epigenetic mechanisms through which paternal influences on offspring development can be achieved. Epidemiological and laboratory studies suggest that paternal nutritional and toxicological exposures as well as paternal age and phenotypic variation can lead to variations in offspring and, in some cases, grand-offspring development. These findings suggest a potential epigenetic germline inheritance of paternal effects. However, it may be important to consider the interplay between maternal and paternal influences as well as the experimental dissociation between experience-dependent and germline transmission when exploring the role of epigenetic variation within the germline as a mediator of these effects. In this review, we will explore these issues, with a particular focus on the potential role of paternally induced maternal investment, highlight the literature illustrating the transgenerational impact of paternal experiences, and discuss the evidence supporting the role of epigenetic mechanisms in maintaining paternal effects both within and across generations.  相似文献

The non-genomic transmission of maternal behavior from one generation to the next illustrates the pervasive influence of maternal care on offspring development and the high degree of plasticity within the developing maternal brain. Investigations of the mechanisms through which these maternal effects are achieved have demonstrated environmentally-induced changes in gene expression associated with epigenetic modifications within the promoter region of target genes. These findings raise challenging questions regarding the pathways linking experience to behavioral variation and the broader ecological/evolutionary implications of the dynamic changes in neuroendocrine function that emerge. This review will highlight studies in laboratory rodents which demonstrate plasticity in the maternal brain and the role of maternally-induced changes in DNA methylation in establishing the link between variations in maternal care and consequent developmental outcomes. The persistence of maternal effects across generations and the trade-offs in reproduction that are evident in female offspring who experience high vs. low levels of maternal care contribute to our understanding of the divergent strategies that are triggered by the quality of early-life experiences. Evolving concepts of inheritance and the interplay between genes and the environment may advance our understanding of the origins of individual differences in phenotype.  相似文献

Fuel ethanol industry presents different problems during bioreactors operation. One of them is the unexpected variation in the output ethanol concentration from the bioreactor or a drastic fall in the productivity. In this paper, a compilation of concepts and relevant results of several experimental and theoretical studies about dynamic behavior of fermentation systems for bioethanol production with Saccharomyces cerevisiae and Zymomonas mobilis is done with the purpose of understanding the stability phenomena that could affect the productivity of industries producing fuel ethanol. It is shown that the design of high scale biochemical processes for fuel ethanol production must be done based on stability studies.  相似文献
