用正交实验结果作方差分析表明:溶剂用量对试验结果有较显著的影响(a=0.10),提取温度对试验结果也有较大的影响,而提取时间对试验结果的影响不显著。喜树叶中水溶性糖的提取最优方案是A3C2B3,即溶剂用量为60ml,提取温度80℃,提取时间60min;喜树叶水溶性糖的提取最优方案是A2C2B1,即溶剂用量为40ml,提取温度80℃,提取时间20min。通过对喜树叶与枝的成对比较分析,叶与枝的水溶性糖含量无显著差异。  相似文献

The style of wheat divides into 2 branches, separated from its base and covered with a large number of slender stigmatic branches. The stigma is of dry type. The style is solid. There is no transmitting tissue differentiated in the style. Young stylar cells appear polygonal in transverse sections and elongated in longitudinal sections with an increase in length of the cells from periphery towards center. In transverse sections, mature stylar cells look extremely irregular. They are contorted and mosaicked with one another. During their development, stylar cells elongated vigorously with intrusive growth. The wall of stylar cells is thin, except at the corners where cells connect, that slight thickening of the cell wall occurs. Stylar cells start vacuolation at the earlier stages and gradually become highly vacuolated, but still remain rich in organelles, such as mitochondria, endoplasmic reticulum, dictyosomes and chloroplasts, the amount of which varied with the development stages of the style. Stigmatic branches are differentiated from the stylar epidermal cells, composed of 4 files of cells which link end to end with one another. Not long before anthesis, wall material in the intercellular corners becomes loose and porous. After pollination, pollen tubes grow along the intercellular spaces among the 4 files of cells in the stigmatic branches and then enter the style. Pollen tubes may pass through any intercellular corner throughout the 2 branches of the style, except for the lateral-outer portion which is composed of larger stylar cells. Eventually, pollen tubes enter the ovary.  相似文献

Neurons acquire their distinct shapes after passing through many transitional stages in early development. To reveal the dynamics and spatiotemporal sequence of process formation in situ, the growth of neurons in the optic tectum of live zebrafish embryos (54 to >100 h old) was monitored using time-lapse videorecordings. Neurons were labeled by injecting the fluorescent vital dye DiO into the cell-rich layer of the developing tectum in 50- to 70-h-old embryos. In phase 1, tectal neurons possess an apical “primary process” which reaches to the ventral aspect of the tectal neuropil. The primary process produces at its tip short transitory branches, some with growth cones, over a period of roughly 6 h. One of the growth cones then elongates rapidly and grows toward the caudal tectum via a route characteristic of efferent axons. After retraction of excess branches and growth cones, branching activity resumes at the tip of the primary process to form the dendritic tree (phase 2). The dendritic tree develops in the tectal neuropil through emission and retraction of many branches during a period of >20 h (our longest continuous time-lapse movie). The tectal territory “explored” in this way is larger than the area finally covered by the tree resulting from growth and loss of branches. The dynamics observed here directly are probably characteristic for dendrite formation in vertebrates. Moreover, consistent with the sequence of neuronal differentiation observed in vitro, the growth of the axon precedes that of the dendrites, although both emerge from a common primary process in this type of tectal neuron. © 1997 John Wiley & Sons, Inc. 采用HPLC法对不同季节和不同部位的八角茴香药材主成分莽草酸的含量进行分析,发现枝条、叶片、春果、秋果的莽草酸含量依次为2.64%、4.94%、8.63%、12.16%;色谱条件:Venusil HILIC(5μm,4.6 mm×250 mm)色谱柱,流动相:乙腈-0.5%三氟乙酸(95:5),流速1.0 mL·min~(-1),检测波长为210 nm。莽草酸进样量在0.001~6μg范围内线性关系良好。莽草酸的线性回归相关系数r=0.9984,平均回收率为98.4%,相对标准偏差(RSD)为1.26%。该研究为制定八角茴香药材质量检测标准提供了简便、快速、准确的方法。  相似文献

Supermatrix and supertree are two methods for constructing a phylogenetic tree by using multiple data sets. However, these methods are not a panacea, as conflicting signals between data sets can lead to misinterpret the evolutionary history of taxa. In particular, the supermatrix approach is expected to be misleading if the species-tree signal is not dominant after the combination of the data sets. Moreover, most current supertree methods suffer from two limitations: (i) they ignore or misinterpret secondary (non-dominant) phylogenetic signals of the different data sets; and (ii) the logical basis of node robustness measures is unclear.To overcome these limitations, we propose a new approach, called SuperTRI, which is based on the branch support analyses of the independent data sets, and where the reliability of the nodes is assessed using three measures: the supertree Bootstrap percentage and two other values calculated from the separate analyses: the mean branch support (mean Bootstrap percentage or mean posterior probability) and the reproducibility index.The SuperTRI approach is tested on a data matrix including seven genes for 82 taxa of the family Bovidae (Mammalia, Ruminantia), and the results are compared to those found with the supermatrix approach. The phylogenetic analyses of the supermatrix and independent data sets were done using four methods of tree reconstruction: Bayesian inference, maximum likelihood, and unweighted and weighted maximum parsimony. The results indicate, firstly, that the SuperTRI approach shows less sensitivity to the four phylogenetic methods, secondly, that it is more accurate to interpret the relationships among taxa, and thirdly, that interesting conclusions on introgression and radiation can be drawn from the comparisons between SuperTRI and supermatrix analyses. 丙酮和二甲基亚砜作为提取剂测定植物光合色素含量是最为普遍的两种方法,但是对于二者之间的可比性问题探讨很少。本文基于对19中木本植物叶片和树枝内叶绿素含量的测定结果发现如下结果:(1)二甲基亚砜测定叶绿素a、叶绿素b和叶绿素总量的结果均显著高于丙酮测定结果(而对叶绿素a/b比值的影响较小),与此相反,丙酮对类胡萝卜素提取测定结果要远高于二甲基亚砜的测定结果。(2)两种方法对叶绿素和类胡萝卜素测定结果之间均存在显著线性相关,可以应用相关线性拟合方程进行不同方法测定结果之间的转换,以减少叶绿素含量、类胡萝卜素含量比较过程中产生的测定方法误差。(3)不同单位表示方法,对两种方法测定结果的相关性存在影响,使用鲜重单位表示的相关程度普遍高于以表面积为单位的相关程度。(4)两种方法对树枝和叶片测定结果的影响存在差异:对于叶绿素含量低的树枝,两种方法测定结果的差异较小,而对于叶绿素含量高的叶片,二甲基亚砜测定结果则远高于丙酮的测定结果,可能原因在于丙酮对于叶绿素的提取效果远低于二甲基亚砜。(5)以二甲基亚砜为浸提试剂,通过不同精确度仪器(0.1和1 nm)检测发现尽管不同仪器之间相关关系达到0.99以上,但二者相比精确度为0.1 nm检测叶绿素a、总叶绿素浓度高出15%~33%,而叶绿素b浓度低4%左右。这些研究发现为不同方法之间的比较提供了依据。  相似文献

目的:以肠系膜动脉三级分支为样本,观察利用微血管技术检测血管张力功能的整个过程,以便研究各种微血管相关疾病中的微血管功能状态。方法:利用DMT张力测定仪和PowerLab数据采集系统检测微血管收缩、舒张功能,将肠系膜三级动脉血管游离,固定,标准化和激活后,通过加入血管收缩药物及舒张药物,完成对微血管张力功能的检测。结果:本文制备的肠系膜动脉三级分支血管环对血管活性药物出现良好的收缩、舒张反应,加入10^-5mol/L的去甲肾上腺素(NE)后收缩张力达19mN,之后依次加入10^-9~10^-5mol/L的乙酰胆碱(ACh)或硝普钠(SNP),血管张力呈梯度降低,ACh和SNP引起的最大舒张率分别为80%和95%。结论:利用该微血管环技术成功检测出肠系膜动脉三级分支的收缩舒张功能。  相似文献

研究了长白山北坡椴树(Tilia amurensis)枝条的分散进程,实验结果揭示,分解率是与各实验地的植被类型,海拔,生长季月平均大于5℃的积温等密切相关。其枝条分解的保存率(Y)与分解年龄(x)的相关,可用指数模型:Y=e^-kx来拟合,在红松阔叶混交林,红松外叶混交林,岳桦去冷杉林及岳桦林中椴树枝条分解常数K分别为-0.168与-0.127,-0.102和-0.094。枝条原重量50%的分解年龄在上述4个森林中分别是4,5,6和7a。实验还指出,在长白山红松阔叶混交林皆伐迹地的实验条件下适当的蔽荫和浇水对分解有正面影响。而且,枝条的分解率主要是以长白山不同海拔植被类型的温度条件而转移,特别是生长季的积温。  相似文献

Summary Shade needles of hybrid larch (Larix decidua × leptolepis) had the same rates of photosynthesis as sun needles per dry weight and nitrogen, and a similar leaf conductance under conditions of light saturation at ambient CO2 (Amax). However, on an area basis, Amax and specific leaf weight were lower in shade than in sun needles. Stomata of sun needles limited CO2 uptake at light saturation by about 20%, but under natural conditions of light in the shade crown, shade needles operated in a range of saturating internal CO2 without stomatal limitation of CO2 uptake. In both needle types, stomata responded similarly to changes in light, but shade needles were more sensitive to changes in vapor pressure deficit than sun needles. Despite a high photosynthetic capacity, the ambient light conditions reduced the mean daily (in summer) and annual carbon gain of shade needles to less than 50% of that in sun needles. In sun needles, the transpiration per carbon gain was about 220 mol mol–1 on an annual basis. The carbon budget of branches was determined from the photosynthetic rate, the needle biomass and respiration, the latter of which was (per growth and on a carbon basis) 1.6 mol mol–1 year–1 in branch and stem wood. In shade branches carbon gains exceeded carbon costs (growth + respiration) by only a factor of 1.6 compared with 3.5 in sun branches. The carbon balance of sun branches was 5 times higher per needle biomass of a branch or 9 times higher on a branch length basis than shade branches. The shade foliage (including the shaded near-stem sun foliage) only contributed approximately 23% to the total annual carbon gain of the tree.  相似文献
