Properties of partially purified NADP-malic enzyme (EC 1.1.1.40) from glumes of developing wheat grains were examined. The pH optimum for enzyme activity was influenced by malate and shifted from 7.3 to 7.6 when the concentration of malate was increased from 2 to 10 mM. The Km values, at pH 7.3, for various substrates were: malate, 0.76 mM; NADP, 20 μM and Mn2+, 0.06 mM. The requirement of Mn2+ cation for enzyme activity could be partially replaced by Mg2+ or Co2+. Mn2+ dependent enzyme activity was inhibited by Pb2+, Ni2+, Hg2+, Zn2+, Cd2+, Al3+ and Fe3+. During the reaction, substrate molecules (malate and NADP) reacted with enzyme sequentially. Activity of malic enzyme was inhibited by products of the reaction viz pyruvate, HCO3? and NADPH2. At a limiting fixed concentration of NADP, these products induced a positive cooperative response to increasing concentrations of malate.  相似文献

The conformational and binding properties towards Cu(II) and Ni(II) ions of Gly-Gly-His derivatives of poly(l-lysine) have been investigated mainly using circular dichroism (c.d.) spectroscopy. These derivatized polymers can be considered macromolecular analogues of the Cu(II) and Ni(II) binding site of human serum albumin. It has been shown that modification up to 53% of the ε-amino groups of lysine side chains by covalent binding of the tripeptide unit Gly-Gly-His does not induce appreciable alteration of the α-helix forming tendency of the polylysine backbone. The derivatized polymers exhibit strong affinity towards Cu(II) and Ni(II) ions. At neutral pH, complexes are formed in which each tripeptide chelating unit is linked to one metal ion. The spectral characteristics in the visible absorption region are consistent with a square planar geometry of the complexes, with deprotonated peptide groups and one imidazole nitrogen in the coordination sphere of the ion. C.d. measurements in the far u.v. indicate that complex formation in the side chains causes an increase of ordered structure of the peptide backbone at neutral pH. This fact is interpreted in terms of a reduced electrostatic repulsion among side chains due to charge neutralization in the tripeptide units linked to metal ions.  相似文献

植物可以吸附大气颗粒物,对降低大气颗粒物污染具有重要作用,能有效改善空气质量,植被对颗粒物的吸附功能研究已成为当前城市生态学和环境科学研究的热点。本文从植物叶片吸附大气颗粒物的方式和途径出发,概述了植物叶片对颗粒物的吸附特征及其时空变化规律。以此为基础,从植物叶表面属性、植物种类及其物理属性、植物群落特征等植物内部因素,颗粒物的粒径组成和污染程度等外界环境因素,大风、降水、温湿度等气象因素这些方面来阐述植物对不同粒径颗粒物吸附能力的影响因素。最后,文章指出今后应在植物吸附大气颗粒物的机制、定量化分析以及更加科学系统的测定方法等方面加强研究,以期为能高效吸附颗粒物的植物种类和植物配置方式的选择提供参考依据。  相似文献

Skin is a biological material the mechanical properties of which are dependent on the constituents from which it is assembled. Skin, the outer covering of animals is made up of collagen fibers arranged in more or less ordered arrays. Pufferfish skin provides a rigid framework to support the body contents and a flexible covering to allow whatever changes are necessary for the remarkable inflation mechanism. Here, we describe the structure and tensile properties of the dorsal and ventral skin of the pufferfish, Lagocephalus gloveri Abe and Tabeta, 1983. The ultimate tensile strength of ventral skin was found to be around two times higher than that of the dorsal skin. It was observed that the dorsal skin could resist more deformation than the ventral skin. The collagen fibers were arranged in different ordered arrays for ventral and dorsal skin and the concentration of fibers was found to be more in ventral than dorsal skin. This provides more stiffness to ventral skin. Scanning electron microscopy studies of the ventral skin showed a unidirectional arrangement of the collagen fibers, which provides more stretching capacity. Dorsal skin, on the other hand, has an orthogonal arrangement of fibers. The present study thus showed that the mechanical behavior of the skin of L. gloveri is strongly influenced by the concentration and arrangement of collagen fibers.  相似文献

Studies of nematode assemblages in natural ecosystems can contribute to better understanding of the occurrence, relevance, and ecology of plant-parasitic and other soil nematodes. Nematode assemblages and environmental parameters (organic matter, water content (WC), bulk density (BD), total porosity (Po), soil respiration, and soil texture) were investigated in two seasons (rainy and dry) in two forest areas of the Zona da Mata, Pernambuco State. The aim of our research was to evaluate the heterogeneity between two locations and seasons in the Brazilian Atlantic Forest. Structure and composition of the nematode assemblages differed between areas and across time. Rhabditidae dominated the rainy season in both forest soils. Rarefaction curves (RC) suggest that sampling to detect more nematode taxa should be more intensive in the rainy season. The forest soils have complex, stable soil food webs with high connectance and decomposition channels dominated by bacteria. The predator–prey relationships were not affected by changes in soil properties that fluctuate with time.  相似文献

The aim of this study was to describe the vocal repertoire of the Ponto‐Caspian goby Neogobius fluviatilis and to compare the acoustic properties of this species with those of other soniferous Mediterranean gobies belonging to the Gobius lineage. Vocalizations and associated behaviours were recorded under controlled aquarium conditions in female and male N. fluviatilis. Sound emission was elicited by means of ‘intruder tests’, using an individual of the same or opposite sex as an intruder, and recording sounds using a hydrophone placed 20 cm from the shelter used as a nest for the resident fish. Five acoustic properties, including spectral and temporal properties, were measured from 13 individuals. The vocal repertoire of the species consisted of sequences of short vocalizations during both agonistic and reproductive intraspecific interactions. The wave form of each sound resolved in a pure sine wave composed of rapidly repeated pulses. Sounds lasted about 200 ms, showing an average fundamental frequency of about 80 Hz. Sound properties did not differ between reproductive and the aggressive contexts, and the general structure of sounds was highly stereotyped. The individual means of three acoustic independent traits characterizing the sounds of seven species of the Gobius lineage, including N. fluviatilis, were then entered in a discriminant function analysis to assess how well species could be differentiated on the basis of acoustics, and their degree of affinities. The results suggested that the pulse repetition rate of the sounds, i.e. the relative tonal/pulsatile nature of the sounds, was the most important property in differentiating the species, and that this trait may contain a high level of phylogenetic signal, as the species producing tonal sounds clustered together, in line with the results of recent molecular phylogenic studies. The results were discussed in light of the geological and phylogeographical events believed to have driven the diversification of European gobies.  相似文献

Mycobacterium tuberculosis dTDP-d-glucose 4,6-dehydratase (RmlB) is the second enzyme for the biosynthesis of dTDP-l-rhamnose, which is a sugar donor to the synthesis of the cell wall linker, d-N-acetylglucosamine-l-rhamnose. RmlB is essential to mycobacterial growth and is not found in humans; therefore, it is a potential target for developing new anti-tuberculosis drugs. So far, there has been no suitable method for high-throughput screening of RmlB inhibitors. Here, the recombinant M. tuberculosis RmlB was purified and an absorbance-based microtiter plate assay was developed for RmlB activity. It could be used for high-throughput screening of RmlB inhibitors. The kinetic properties of M. tuberculosis RmlB, including optimal pH, optimal temperature, the effect of metal ions, and the kinetic parameters, were determined with this assay. The inhibitory effects of dTTP and dTDP on M. tuberculosis RmlB were also studied with the assay.  相似文献

We have developed a novel enzymatic cycling method that uses creatine kinase (CK) to measure creatine. The method takes advantage of the reversibility of the CK reaction in which the forward (creatine phosphate forming) and reverse reactions are catalyzed in the presence of an excess amount of ATP and IDP, respectively. Real-time detection was accomplished using ADP-dependent glucokinase (ADP–GK) together with glucose-6-phosphate dehydrogenase. ADP, one of the cycling reaction products, was distinguished from IDP by using the nucleotide selectivity of the ADP–GK. The increasing level of ADP was measured from the level of reduced NADP at 340 nm. The method is appropriate for an assay that requires high sensitivity because the rate of increase in absorbance at 340 nm is proportional to the amount of CK present in the reaction mix. We reasoned that the method with CK in combination with creatinine amidohydrolase could be used to assay creatinine, an important marker of kidney function. Our results confirmed the quantitative capability of the assay.  相似文献

Using a hydrothermal method, Ce3+/Tb3+ non‐/single‐/co‐doped K‐Lu‐F materials have been synthesized. The X‐ray diffraction (XRD) results suggest that the Ce3+ and/or Tb3+ doping had great effects on the crystalline phases of the final samples. The field emission scanning electron microscopy (FE‐SEM) images indicated that the samples were in hexagonal disk or polyhedron morphologies in addition to some nanoparticles, which also indicated that the doping also had great effects on the sizes and the morphologies of the samples. The energy‐dispersive spectroscopy (EDS) patterns illustrated the constituents of different samples. The enhanced emissions of Tb3+ were observed in the Ce3+/Tb3+ co‐doped K‐Lu‐F materials. The energy transfer (ET) efficiency ηT were calculated based on the fluorescence yield. The ET mechanism from Ce3+ to Tb3+ was confirmed to be the dipole–quadrupole interaction inferred from the theoretical analysis and the experimental data.
