Abstract

The biodegradation of the antibiotic sulfamethazine (SMT) by using different crude extracts of halophytes was investigated. For this purpose, crude water extracts of the halophytes Chenopodium quinoa, Sesuvium portulacastrum, and Tripolium pannonicum were prepared. Different amounts of SMT were added to the different extracts (final concentration of 1, 2, and 5?mg L?1) and incubated at 37?°C. Crude extracts of T. pannonicum were further used to evaluate the degradation rate over time. In order to evaluate the influence of endophytic or naturally plant-associated microorganisms on the biodegradation of SMT, extracts from plants grown in sterile and non-sterile conditions were compared. SMT was analyzed by liquid chromatography coupled to positive ion electrospray mass spectrometry (ESI LC-MS). Based on the findings, crude extracts of T. pannonicum have a high potential to biodegrade SMT with a decrease up to 85.4% (4.27?±?0.10?mg L?1) from an initial concentration of 5?mg L?1. The lowest activity was obtained using extracts of C. quinoa with degradation of 4.5%. Extracts of plants cultivated under sterile and non-sterile conditions do not have any significant difference in SMT degradation. Therefore, microorganisms and their enzymatic activities do not seem to play a significant role during this process.  相似文献

A proteolytic susceptibility index was proposed to evaluate the reaction performance of different food by-products with subtilisin. Whey, salmon muscle and feather keratine were hydrolyzed at the same peptide bond concentration at 50 °C and pH 8.0 with different subtilisin concentrations. The logarithmic equation P = 1/b ln(abt+1) was fitted to estimate the kinetic constants a and b. The a/b ratio was proposed as a proteolytic susceptibility index, which was correlated with the chemical structure and conformation of protein sources in increasing order of feather keratin, salmon muscle and whey proteins with corresponding a/b values of 0.1, 8.2 and 14.4 mM2/min, respectively. The methodology proposed in this work can be used to evaluate the proteolytic susceptibility of different protein by-products to different proteases and to evaluate the changes in proteolytic susceptibility after treatment.  相似文献

Background

In recent years, microalgae (MA) have attracted much interest considering their possible therapeutic application. They contain active natural compounds or derivatives (extracts, pure or chemically modified compounds) that have increasing applications in the pharmaceutical industry.

Methods

The present study aims to examine microalgae for new photosensitizers, with a potential to be used in the light-associated treatment of tumors. Semi-purified extracts of several microalgae strains were evaluated as photosensitizers for photodynamic therapy (PDT) applications. Four tumor cell lines (A549, LNCap, MCF-7, and MDA-MB 435) were used to assess 34 samples extracted by three methods: cellulase enzyme, lysozyme enzyme and ultra-sonication. The fluorescence measurements and the recorded images alongside the spectral intensities between 650–800 nm wavelengths provided characteristic features to some of the contents of the examined extracts.

Results

Several microalgae constituents activated by blue light (BL), red light (RL) or both (in sequence) exhibited significant effects on the viability of the tumor cell lines, decreasing it as much as 95% for certain MA constituents. Majority of the MA constituents showed a higher phototoxicity after exposure to both blue and red lights than the photo-induced toxicity when exposed to a single light source. The viability of the tumor cells exhibited the dose dependent response with the MA constituents.

Conclusion

The results clearly showed that MA constituents are potential photosensitizers that have a significant photo-damage effects on the tested cancer cells.  相似文献

The hydrolysis of polyethylene terephthalate (PET) fibers by two fungal hydrolases was investigated. The hydrolase from a newly isolated Fusarium oxysporum strain (LCH 1) was more efficient in releasing terephthalic acid from PET fibers compared to the enzyme from F. solani f. sp. pisi DSM 62420 when equal amounts of p-nitrophenyl butyrate-hydrolyzing activity were employed. PET fabrics treated under the same conditions with the enzyme from F. oxysporum LCH 1 also showed a considerably higher increase in hydrophilicity compared to fabrics treated with the enzyme from F. solani f. sp. pisi DSM 62420.  相似文献

A rational approach has been applied to design a new environmentally acceptable and industrially viable enzymatic scouring process. Owing to the substrate specificity, the selection of enzymes depends on the structure and composition of the substrate, i.e. cotton fibre. The structure and composition of the outer layers of cotton fibre has been established on the basis of thorough literature study, which identifies wax and pectin removal to be the key steps for successful scouring process. Three main issues are discussed here, i.e. benchmarking of the existing alkaline scouring process, an evaluation of several selected acidic and alkaline pectinases for scouring, and the effect of wax removal treatment on pectinase performance. It has been found that the pectinolytic capability of alkaline pectinases on cotton pectin is nearly 75% higher than that of acidic pectinases. It is concluded that an efficient wax removal prior to pectinase treatment indeed results in improved performance in terms of hydrophilicity and pectin removal. To evaluate the hydrophilicity, the structural contact angle (theta) was measured using an auto-porosimeter.  相似文献

The subject of this study was an analysis of the role of active site residues in the phosphoryl transfer reaction catalyzed by 4-methyl-5-β-hydroxyethylthiazole kinase (ThiK). The ThiK-catalyzed reaction is of special interest due to the lack of a highly conserved aspartate residue serving as a catalytic base. ONIOM(B3LYP:PM3) models of stationary points along the reaction pathway consisted of reactants, two magnesium ions and several highly conserved ThiK active site residues. The results indicate that an SN2-like mechanism of ThiK, with γ-phosphate acting as an alcohol-activating base is reasonable. Geometries of substrates, transition state and products were utilized in the non-empirical analysis of the physical nature of catalytic interactions taking place in the ThiK active site. The role of particular residues was investigated in terms of their ability to preferentially stabilize the transition state relative to substrates (differential transition state stabilization, DTSS) or products (differential product stabilization, DPS). It seems that Mg2, Glu126 and Cys198 play a major catalytic role, whereas Mg1 and the same Cys198 are responsible for product release. It is remarkable that no dominant role of an electrostatic term in the interactions involved in catalytic activity is observed for product release. Determination of catalytic fields expressing differential electrostatic potential of the transition state with respect to substrates revealed the optimal electrostatic features of an ideal catalyst for the studied reaction. The predicted catalytic environment is in agreement with experimental data showing increased catalytic activity of ThiK upon mutation of Cys198 to aspartate. Food waste (FW) management is a global conundrum because of the rapid population growth and growing economic activity. Currently, incineration and landfill are still the main means for FW management, while their environmental sustainability and economic viability have been in question. Recently, the biological processes including anaerobic digestion, aerobic composting, bioethanol fermentation, feed fermentation etc. have attracted increasing interest with the aims for energy and resource recovery from FW. However, these biological approaches have inherent drawbacks, and cannot provide a comprehensive solution for future FW management. Therefore, this review attempts to offer a critical and holistic analysis of current biotechnologies for FW management with the focus on the challenges and solutions forward. The biological approaches towards future FW management should be able to achieve both environmental sustainability and economic viability. In this instance, the concept of zero solid discharge-driven resource recovery has thus been put forward. According to which, several innovative biological processes for FW management are further elucidated with critical analysis on their engineering feasibility and environmental sustainability. It turns out that is an urgent need for turning current single task-orientated bioprocess to an integrated biological process with multiple tasks of concurrent recovery of water, resource and energy together with zero-solid discharge.  相似文献

Abstract To study the effect of plasmids on the arbitrary primer-polymerase chain reaction fingerprint of bacterial strains, the Escherichia coli strains DH5, Top10, and W3110 were transformed with plasmids of different sizes: respectively, pUC19, pCEP and two clinically important plasmids carrying resistance to several antibiotics. Total DNA, i.e. both chromosomal and plasmid DNA, was prepared from transformed cells by boiling the cell suspensions and by phenol-chloroform extraction; chromosomal DNA was prepared by the same methods from the non-transformed, plasmid-free strains; plasmid DNA of pUC19 was purchased; plasmid DNA of pCEP was purified from the transformed strains by caesium chloride density gradient centrifugation. Arbitrarily primed polymerase chain reaction was carried out for all of these preparations. Amplification carried out independently with three different primers resulted in similar patterns for the chromosomal preparations whether or not plasmid was present. Amplification of plasmid DNA gave different patterns, characterized by fragments larger than those obtained when total or chromosomal DNA were used as the target. These data illustrate that the plasmids studied here do not influence the chromosomal arbitrarily primed PCR fingerprint, although plasmids alone are amplified in the absence of chromosomal DNA. Experiments comparing different relative concentrations of plasmid and chromosomal DNA indicate that under natural conditions the amount of chromosomal DNA per cell is sufficient to inhibit observable amplification of the plasmid(s) present.  相似文献

Biogenic amines (BAs) are a class of harmful compounds often be found in high protein foods, especially naturally fermented foods. BAs derive from free amino acid decarboxylation through microbial activities and can cause toxic effects (headache, heart palpitations, vomiting) on humans, depending on individual sensitivity. Indigenous amine-degrading strains or strains producing amine-degrading enzymes (ADEs) have drawn great attention since they play an important role in affecting BA accumulation, and enzymes/genes involved in the biosynthetic mechanisms. They also help maintain the sensory quality of the final products. Besides, due to ADEs’ harmless catalytic products, they can be further utilized in fermented foods and beverages to reduce BAs. This review describes in detail the mechanisms of BAs formation, as well as the diversity of ADEs able to degrade BAs in a model or real food systems. A deeper knowledge of this issue is crucial because ADEs’ activities are often associated with strains rather than species or genera. Moreover, this information can help to improve the selection and characterization of strains for further applications as starters or bioprotective cultures, to obtain high-quality foods with reduced BAs contents.  相似文献

We review recent evidence illustrating the fundamental difference between cytoplasmic and test tube biochemical kinetics and thermodynamics, and showing the breakdown of the law of mass action and power-law approximation in in vivo conditions. Simulations of biochemical reactions in non-homogeneous media show that as a result of anomalous diffusion and mixing of the biochemical species, reactions follow a fractal-like kinetics. Consequently, the conventional equations for biochemical pathways fail to describe the reactions in in vivo conditions. We present a modification to fractal-like kinetics following the Zipf–Mandelbrot distribution which will enable the modelling and analysis of biochemical reactions occurring in crowded intracellular environments.  相似文献
