We wanted to examine if spontaneous physical activity contributes to non-shivering thermogenesis. Ten lean, healthy male subjects wore a physical activity, micro-measurement system while the room temperature was randomly altered at two hourly intervals between thermoneutral (72 °F), cool (62 °F) and warm (82 °F) temperatures. Physical activity measured during the thermoneutral, cooling and warming periods was not significantly different. Cooling increased energy expenditure above basal and thermoneutral values 2061±344 kcal/day (p<0.01). Thus, the increase in energy expenditure associated with short-term environmental cooling in lean, healthy males does not appear to be due to increased spontaneous physical activity or fidgeting.  相似文献

Salinity interacts with many physiological functions and therefore probably influences the distribution of terrestrial fauna in tidal flooded salt marshes. The present study tests the hypothesis that the physiological tolerance of stenotopic wolf spiders for saline conditions at least partially determines their occurrence throughout salt‐marsh and nonsaline habitats. The duration of survival of three stenotopic wolf spider species (Araneae: Lycosidae) with different habitat preferences is compared in a controlled laboratory experiment. The forest‐dwelling Pardosa saltans, the salt‐marsh resident Pardosa purbeckensis and its sister species the inland‐living Pardosa agrestis are exposed to experimental conditions with different levels of salinity. Individuals (45 males and 20–45 females per treatment) are placed in individual air‐tight boxes filled with water‐saturated sand. Three levels of salinity are tested: nonsaline (0‰), medium saline (33–35‰) and highly saline (66–70‰). Contents of carbon, hydrogen and nitrogen and the molar ration carbon/nitrogen remain constant over time and do not differ among salinity treatments, indicating that starvation effects on survival, if any, are similar for all treatments. Conversely, body water significantly decreases over time and differs among salinity treatments, in accordance with patterns of survival. Conforming to their habitat preference, the survival of P. saltans and P. agrestis decreases quickly under highly saline conditions. Pardosa purbeckensis, however, has a high survival under both saline and nonsaline conditions. The duration of survival of females is significantly lower than that of males of P. saltans and P. purbeckensis. Durations of survival of ground‐living wolf spiders exposed to salinity partly match their habitat distribution but do not explain the restriction of salt‐marsh species to saline habitats.  相似文献

Cogeneration is internationally recognized as an important way to save energy and improve environment. At present, the cogeneration scale in China has been ranked at the second place all round the world. China formulated relevant laws, regulations and technology policies to actively encourage and support the development of cogeneration. This article selects 62 Northeast regional Chinese cogeneration enterprises' financial data from 2003 to 2007, and analyzes the impact of the electric power industry deregulation to cogeneration enterprises' performance by using a partial frontier nonparametric. Further more, we study whether the cogeneration enterprises actualize horizontal scope economy by comparing with the thermal power alone or heat production alone enterprises. Results show that the factory network separation and regional market trial reform can promote the cost efficiency of both five major power group cogeneration enterprises and independent cogeneration enterprises. Compared to five group enterprises, the scope economies index of the independent cogeneration enterprises is lower, the unbundling reform and northeast regional market trial reform have a negative impact on cogeneration enterprises to achieve scope economies and improve cost efficiency.  相似文献

Protein physical and chemical instability is one of the major challenges in the development of biopharmaceuticals during every step of the process, ranging from production to final delivery. This is particularly applicable to human recombinant interferon alpha-2b (rhIFN-α2b), a pleiotropic cytokine currently used worldwide for the treatment of various cancer and chronic viral diseases, which presents a poor stability in solution. In previous studies, we have demonstrated that the introduction of four N-glycosylation sites in order to construct a heavily glycosylated IFN variant (4N-IFN) resulted in a markedly prolonged plasma half-life which was reflected in an enhanced therapeutic activity in mice in comparison with the commercial non-glycosylated rhIFN-α2b (NG-IFN). Herein, we evaluated the influence of glycosylation on the in vitro stability of 4N-IFN towards different environmental conditions. Interestingly, the hyperglycosylated cytokine showed enhanced stability against thermal stress, acid pH and repetitive freeze-thawing cycles in comparison with NG-IFN. Besides, microcalorimetric analysis indicated a much higher melting temperature of 4N-IFN, also demonstrating a higher solubility of this variant as denoted by the absence of precipitation at the end of the experiment, in contrast with the NG-IFN behaviour. Furthermore, far-UV circular dichroism (CD) spectrum of 4N-IFN was virtually superimposed with that of NG-IFN, indicating that the IFN structure was not altered by the addition of carbohydrate moieties. The same conclusion could be inferred from limited proteolysis studies. Our results suggest that glycoengineering could be a useful strategy for protecting rhIFN-α2b from inactivation by various external factors and for overcoming aggregation problems during the production process and storage.  相似文献

Question: What is the relative importance of local site effects and selected important environmental variables in determining plant species composition? How do species respond to these environmental variables? Location: Ten mesotrophic grassland sites of high conservation value in southern England. Methods: Species cover was assessed in between 10 and 25 randomly selected 1‐m2 quadrats at each site. At each quadrat degree of waterlogging (W), soil Olsen extractable phosphorus (P) and soil pH were measured. Variation partitioning was used to separate site and soil effects, and HOF (Huisman, Olff & Fresco) modelling was used to produce response curves for the major species on soil gradients, based on coenoclines derived from partial canonical correspondence analysis (pCCA). Results: Variation partitioning identified Site as the most important environmental variable (34.6%). Only 18.7% was accounted for by the three soil variables together; W (degree of waterlogging), P and pH accounted for 11.1%, 5.7% and 4.3%, respectively in raw form with 2.4% shared. However, when Site and the other soil variables were removed the variation explained reduced to 2.3% for W, 1.1% for P and 1.0% for pH. The species responses to each of these soil environmental factors could be separated into four types on each gradient. Most species were abundant at low W, low soil P and intermediate pH. Conclusions: Site‐based factors were more important than the three soil variables, which were assumed to be directly or indirectly associated with productivity. This implies that each site has unique properties that are more important than the soil variables. The three soil factors were, however, significant and the groups of the most common species, based on significant response curves, can be used as a first approximation of indicators of environmental conditions in British mesotrophic grasslands for conservation. However, W accounted for most variation, and the current reliance on soil available P and soil pH for assessing conservation/restoration potential should be viewed with caution.  相似文献

Microorganisms are everywhere - in the air, soil, and human body as well as on inanimate surfaces like laboratory benches and computer keyboards. The ubiquity of microbes creates a copious supply of potential contaminants in a laboratory. To ensure experimental success, the number of contaminants on equipment and work surfaces must be minimized. Common among many experiments in microbiology are techniques involving the measurement and transfer of cultures containing bacterial cells or viral particles. To do so without contacting non-sterile surfaces or contaminating sterile media requires (1) preparing a sterile workspace, (2) precisely setting and accurately reading instruments for aseptic transfer of liquids, and (3) properly manipulating instruments, cultures flasks, bottles and tubes within a sterile field. Learning these procedures calls for training and practice. At first, actions should be slow, deliberate, and controlled with the goal being for aseptic technique to become second nature when working at the bench. Here we present the steps for measuring volumes using serological pipettes and micropipettors within a sterile field created by a Bunsen burner. Volumes range from microliters (μl) to milliliters (ml) depending on the instrument used. Liquids commonly transferred include sterile broth or chemical solutions as well as bacterial cultures and phage stocks. By following these procedures, students should be able to: •Work within the sterile field created by the Bunsen burner flame. •Use serological pipettes without compromising instrument sterility.• Aspirate liquids with serological pipettes, precisely reading calibrated volumes by aligning the meniscus formed by the liquid to the graduation marks on the pipette. •Keep culture bottles, flasks, tubes and their respective caps sterile during liquid transfers. •Identify different applications for plastic versus glass serological pipettes. •State accuracy limitations for micropipettors. •Precisely and accurately set volumes on micropipettors. •Know how to properly use the first and second stop on a micropipettor to aspirate and transfer correct volumes.  相似文献

Agroforestry offers proven strategies as an environmentally benign and ecologically sustainable land management practice to promote ecosystem services. In this literature review, we systematically consider the agroforestry and ecosystem services literature with the aim to identify and catalogue the knowledge field and provide the first systematic synthesis of ecosystem services research in relation to European agroforestry. We reviewed 71 scientific publications from studies conducted in farmland and forest ecosystems with various types of agroforestry management. Each publication was systematically characterized and classified by agroforestry practice and research approach in order to provide an insight into the current research state in addressing ecosystem services (including methods, indicators, and approaches). Spatial distribution of the case study sites in Europe was also explored. In addition, typical clusters of similar research approaches were identified.The results show that ecosystem service assessment of European agroforestry is currently focused on the spatially extensive wood pastures in the Mediterranean, Atlantic, and Continental agricultural mosaic landscapes. A specific emphasis has been on regulating, supporting, and provisioning services, such as provision of habitat and biodiversity, food, climate regulation, fibre, and fuel, and the consideration of cultural services has been largely limited to aesthetic value. There is a bias to biophysical and monetary research approaches. The majority of the studies focus on quantitative methods and biophysical field measurements addressing the assessment of only one or two services. Monetary approaches have been applied in less than one fifth of the studies but form a distinctive group.Our results highlight gaps and biases in the ecosystem service research agenda within agroforestry based on which we conclude that research should aim to diversify from the biophysical and monetary approaches, towards a wider variety of approaches, especially socio-cultural, and a wider coverage of ecosystem services. Stronger consideration of stakeholder participation and introduction of spatially explicit mapping are also important key actions. We make suggestions to advance the promise of ecosystem services provision from European agroforestry in decision making including various actors, stakeholders, and institutions, with strong links to policy processes, such as the EU Biodiversity Strategy and Common Agricultural Policy.  相似文献

Microbes in soils and other environments produce extracellular enzymes to depolymerize and hydrolyze organic macromolecules so that they can be assimilated for energy and nutrients. Measuring soil microbial enzyme activity is crucial in understanding soil ecosystem functional dynamics. The general concept of the fluorescence enzyme assay is that synthetic C-, N-, or P-rich substrates bound with a fluorescent dye are added to soil samples. When intact, the labeled substrates do not fluoresce. Enzyme activity is measured as the increase in fluorescence as the fluorescent dyes are cleaved from their substrates, which allows them to fluoresce. Enzyme measurements can be expressed in units of molarity or activity. To perform this assay, soil slurries are prepared by combining soil with a pH buffer. The pH buffer (typically a 50 mM sodium acetate or 50 mM Tris buffer), is chosen for the buffer''s particular acid dissociation constant (pKa) to best match the soil sample pH. The soil slurries are inoculated with a nonlimiting amount of fluorescently labeled (i.e. C-, N-, or P-rich) substrate. Using soil slurries in the assay serves to minimize limitations on enzyme and substrate diffusion. Therefore, this assay controls for differences in substrate limitation, diffusion rates, and soil pH conditions; thus detecting potential enzyme activity rates as a function of the difference in enzyme concentrations (per sample).Fluorescence enzyme assays are typically more sensitive than spectrophotometric (i.e. colorimetric) assays, but can suffer from interference caused by impurities and the instability of many fluorescent compounds when exposed to light; so caution is required when handling fluorescent substrates. Likewise, this method only assesses potential enzyme activities under laboratory conditions when substrates are not limiting. Caution should be used when interpreting the data representing cross-site comparisons with differing temperatures or soil types, as in situ soil type and temperature can influence enzyme kinetics.  相似文献

1. Global warming and biological invasions are important threats to biodiversity. Nonetheless, there is little information on how these factors influence performance or life‐history traits of invasive and native species. 2. The effects of temperature on physiological and fitness traits of two invasive alien species (Harmonia axyridis and Hippodamia variegata) and one native species (Eriopis chilensis) of coccinellid were evaluated, testing a model of eurythermality. Eggs of all species were exposed to four temperature treatments (20, 24, 30 and 33 °C). In adult F2 we measured fecundity, locomotor performance, development time (total and per life stage), survival, and preferred body temperature in a thermal gradient. 3. It was found that H. axyridis had comparatively better performance at low temperatures (i.e. 20 °C), while the performance of H. variegata and E. chilensis did not change with temperature or was better at higher temperatures (30 °C). The standardised Levins index showed that all species are eurythermic. E. chilensis had a high niche overlap with the invasive alien ladybird species, rejecting the hypothesis of greater eurythermality of invasive species than native species. 4. Although there were differences in the temperature preferences and in the response of some physiological and life‐history traits of ladybirds to temperature, both the native and invasive alien species are eurythermic, contrary to the prediction. The better performance of H. axyridis at lower temperatures may result in displacement of its current distribution, and thus not all invasive species will respond favourably to global warming.  相似文献
