Soybean is the most important leguminous crop in Brazil and the nitrogen required for plant growth is supplied byBradyrhizobium bacteria through the symbiotic relation established by the inoculation process. Since 1992, two new strains, CPAC 7 and CPAC 15, which have been shown to increase yields in several field experiments, have been recommended in Brazilian commercial inoculants. CPAC 15 is a natural variant of theB. elkanii SEMIA 566 strain, and was isolated after several years of adaptation to a Brazilian Cerrado soil, while CPAC 7 is a variant ofB. japonicum strain CB 1809, selected under laboratory conditions for higher nodulation and yield. The comparison between parental and variant strains, under greenhouse conditions, showed that both CPAC 15 and CPAC 7 increased N2 fixation rates in relation to the parental strains. The better performance of CPAC 15 was related to an increase in nodule efficiency (mg N2 fixed mg-1 nodule) while with CPAC 7 the higher N2 fixation rates were due to increased nodulation. Both CPAC 15 and CPAC 7 increased nodule occupancy, when co-inoculated at a ratio of 1:1 withB. elkanii 29w, in relation to their parental strains. Variant strains also differed from parental in their ability to increase numbers of root hairs (Hai phenotype) either when inoculated onto plants, or when supernatants of bacteria exposed to seed exudates were used as inoculants. This results lead to the hypothesis that a modification in some of the “common” nodulation genes had occurred. However, the increase in Hai phenotype with CPAC 7 was dependent on the soybean cultivar, indicating a possible alteration in some genotypic specific nodulation gene. Apparently, there were no differences in Nod metabolites produced by strains CPAC 15 and SEMIA 566, but a more detailed chemical analysis would be required to rule out subtle differences. On the contrary, significant differences were found between CPAC 7 and the parental strain CP 1809, in the profile of Nod metabolites. Consequently, it may be possible that diffusable molecules, responsible for Hai phenotype, would be related to nodulation ability, competiviveness, and N2 fixation, resulting in the higher yields that have been associated with CPAC 7 and CPAC 15. For the CPAC 7 strain, the increase in Hai phenotype could be atributed to the differences found in the Nod molecules. Consequently, a high degree of physiological and genetic variability can result from the adaptation of rhizobial strains to the soil. Abstract. 1.  Laboratory studies have implicated various accessory bacteria of aphids as important determinants of aphid performance, especially on certain plant species and under certain thermal regimes. One of these accessory bacteria is PABS (also known as T-type), which is distributed widely but is not universal in natural populations of the pea aphid Acyrthosiphon pisum in the U.K.
2.  To explore the impact of PABS on the performance of A. pisum , the nymphal development time and fecundity of aphids collected directly from natural populations and caged on the host plant Vicia faba in the field were quantified. Over 4 consecutive months June–September 1999, the performance of PABS-positive and PABS-negative aphids did not differ significantly.
3.  Deterministic modelling of the performance data showed that the variation in simulated population increase of PABS-positive and PABS-negative aphids would overlap substantially.
4.  Analysis of aphids colonising five host plants ( Lathyrus odoratus , Medicago sativa , Pisum sativum , Trifolium pratense , Vicia faba ) between April and September 2000 and 2001, identified no robust differences between the distribution of PABS-positive and PABS-negative aphids on different plants and with season or temperature.
5.  It is concluded that PABS is not an important factor shaping the performance or plant range of A. pisum under the field conditions tested. Reasons for the discrepancies between this study and laboratory-based studies are considered.  相似文献

This paper discusses experimental data and theoretical concepts characterizing the trophic status of symbiotic zooxanthellae and the mechanisms regulating their density in hermatypic corals. Under natural conditions, the growth of zooxanthellae is not limited by the deficiency of nitrogen. The impact of nitrate (5–15 m M , 2 to 7 days) on nitrogenase activity and nodule-oxygen limitation was investigated in nodulated, 21-day-old plants of a near-isogenic nitrate reductase-deficient pea mutant (A3171) and its wild-type parent ( Pisum sativum L. cv. Juneau). Within 2 days, 10 or 15 m M nitrate, but not 5 m M nitrate, inhibited the apparent nitrogenase activity (measured as in situ hydrogen evolution from nodules of intact plants) of wild-type plants; none of these nitrate levels inhibited the apparent nitrogenase activity of A3171 plants. Nodule-oxygen limitation, measured as the ratio of total nitrogenase activity to potential nitrogenase activity, was increased in both wild-type and A3171 plants by all nitrate treatments. By 3 to 4 days the apparent nitrogenase activity of A3171 and wild-type plants supplied with 5 m M nitrate declined to 53 to 69% of control plants not receiving nitrate. By 6 to 7 days the apparent nitrogenase activity of A3171 plants was similar to the control value whereas that of the wild-type plants continued to decline. From 3 to 7 days, no significant differences in nodule-oxygen limitation were observed between the nitrate (5 m M ) and control treatments. The results are interpreted as evidence for separate mechanisms in the initial (O2 limitation) and longer-term (nitrate metabolism) effects of nitrate on nitrogen fixation by effectively nodulated pea.  相似文献

The effect of arbuscular mycorrhizas on fructan accumulation was studied in barley ( Hordeum vulgare ) infected with Glomus mosseae . Treatments with and without fertilizer were included in order to distinguish between mere fertilizer effects and the effects of the symbiosis, and plants were harvested at two different time points, 35 and 50 d after planting. Fructan was the major storage carboyhdrate in both leaves and roots. The amounts of fructan were markedly altered in the mycorrhizal plants. In roots of non-fertilized mycorrhizal plants, fructan pools were significantly greater than in the corresponding non-mycorrhizal plants. By contrast, fertilization caused a general decrease in amounts of fructan in roots. The increase of fructan in mycorrhizal roots was correlated with a decrease of invertase activity. In leaves, fructan pools decreased or remained unchanged upon mycorrhizal infection; fertilization had a similar effect. However, when individual leaves of a plant were compared, intriguing effects of the mycorrhizal symbiosis could be observed. Whereas in non-mycorrhizal plants, the youngest leaves had the highest fructan contents and the oldest leaves the lowest (as previously reported), this gradient was markedly altered in mycorrhizal plants, indicating systemic effects of mycorrhiza on assimilate partitioning in shoots.  相似文献
