An extensive system of neuropeptide Y (NPY) containing neurons has recently been identified in the central and peripheral nervous system. In addition, NPY and a structurally related peptide, peptide YY (PYY), containing endocrine cells have been identified in the periphery. The NPY system is of particular interest as the peptide coexists with catecholamines in the central and sympathetic nervous system and adrenal medulla. Molecular dynamics (MD) simulations are performed to study the adsorption of solute organic molecules (Ethinylestradiol (EE2) and testosterone) with different polymer membranes such as polyether sulfone (PES), polyvinylidene fluoride (PVDF). The equilibrium MD simulations results for the membrane solution interface system show that the interaction of EE2 with PES is specific and strong, whereas the interaction is weak and non-specific for PVDF. The binding free energies, the non-bonded short range interaction energies and mobility are also consistent with the interaction behaviour found in experiments. The adsorption of testosterone onto PES and PVDF is considered as control system. The result shows that binding free energies of PES and PVDF interacting with organic solute are consistent with experimental result in the order as; PES-EE2 > PES-Testosterone > PVDF-EE2 > PVDF-Testosterone. The formation hydrogen bonds and π–π interactions are observed between the EE2 and PES. In addition, adsorption of EE2 onto polyamide 6-12 (PA612) and polystyrene (PS) membranes are predicted. This simulation study provides molecular insights on the experimental observations and helps as a computational methodology to screen the membrane materials for EE2 removal from wastewater. 相似文献

The oxyntic mucosa of the mammalian stomach is rich in endocrine cells, such as ECL cells, A-like cells, somatostatin cells,
D1/P cells and, in some species, enterochromaffin cells. The various endocrine cell types can be distinguished on the basis
of their characteristic cytoplasmic granules and vesicles. The ECL cells contain numerous large secretory vesicles and relatively
few, small electron-dense granules and small clear microvesicles. We have suggested that in the rat the ECL cells contain
most of the gastric histamine with the secretory vesicles as the major histamine storage site in these cells. α-Fluoromethylhistidine
is an irreversible inhibitor of histidine decarboxylase, the histamine-forming enzyme. We have previously shown that this
enzyme inhibitor depletes histamine from the ECL cells in the rat and reduces the number of secretory vesicles in the cytoplasm.
In the present study, we have examined whether α-fluoromethylhistidine affects the ECL cells in other species and whether
it affects other types of endocrine cells in the oxyntic mucosa of the rat. Mice, rats and hamsters were treated with the
inhibitor (3 mg/kg per h) via minipumps subcutaneously for 24 h. This treatment lowered the oxyntic mucosal histamine concentration
by 65–90% and the number and volume density of the secretory vesicles by 85–95% in the ECL cells of the three species examined.
In contrast, the number and volume density of granules and microvesicles were not greatly affected. No evidence was found
for an effect of α-fluoromethylhistidine on A-like cells, somatostatin cells or D1/P cells of the rat stomach, suggesting that, unlike the ECL cells, they do not contain histamine.

The ontogeny of protein gene product 9.5 (PGP 9.5), serotonin (5-HT), calcitonin gene-related peptide (CGRP), and calcitonin (CT) immunoreactivity was evaluated in small-granule endocrine cells of hamster laryngotracheal epithelium from fetal day 11 to adulthood. Two centrifugal (proximal-to-distal) patterns of differentiation occur. The first pattern begins during fetal life. Endocrine cells, single and clustered in groups (presumptive-or protoneuroepithelial bodies, pNEBs), initially colocalize immunostaining for PGP 9.5, 5-HT, and CGRP in the larynx and proximal 2/3 of the trachea on day 12 and spread to the caudal trachea on day 13.5-HT disappears fleetingly during the 24 h preceding birth; other-wise immunoreactivity for all three substances persists into adulthood. The clusters of endocrine cells survive beyond birth but are so diluted by expansion of the nonendocrine epithelium as to become inconspicuous. Since innervation was not actually observed, these clusters may persist as pNEBs, without developing connections to afferent or efferent nerve fibers. The second pattern concerns single small-granule cells stainable for CGRP but not for 5-HT. These cells first appear in the larynx and cartilaginous part of the cranial trachea on postnatal day 3, and in the middle and caudal trachea, on day 5. The cells increase in number on day 7. In adults, they predominate among endocrine cells of the cartilaginous region. A subset of these cells begins to co-express CT proximally on postnatal day 10, reaching the caudal end of the trachea by 3 weeks. A few elements of the older 5-HT-positive population may also become immunoreactive for CT in juvenile hamsters. 相似文献

The effects of six organophosphate flame retardants (OPFRs) tris(2‐butoxyethyl) phosphate, tris(2‐chloroethyl) phosphate, tris(1‐chloro‐2‐propyl) phosphate, tris(methylphenyl) phosphate, tris(1,3‐dichloro‐2‐propyl) phosphate (TDCIPP), and triethyl phosphate on the activities of androgen receptor (AR), estrogen receptor (ER), and aryl hydrocarbon receptor (AhR) were assessed in human prostate and endometrial cancer cells. OPFRs had no effect on ER or AhR target gene activation in ECC‐1 cells. The effect of TDCIPP on mRNA and protein accumulation of AR target genes was examined further. AR‐inducible gene and protein expression were significantly altered by TDCIPP exposure and repressed PSA levels in conditioned media of prostate cancer cells. We demonstrated that TDCIPP has no affinity for the AR ligand binding domain (AR‐LBD) and exerts its antiandrogenic effects in a noncompetitive fashion. Thus, the clinical relevance of TDCIPP exposure on prostate cancer detection and progression to a therapeutically refractile state ought to be investigated further. 相似文献

Summary Atrial myoendocrine cells of rat were investigated regarding estradiol uptake. It was found that, in addition to their specific endocrine function of producing cardiac polypeptides of the cardiodilatin/atrial natriuretic peptide (CDD/ANP) family, these cells also specifically accumulate radiolabeled estradiol. The TRH secretory responsiveness of the pancreatic islet cell clusters from newborn rat in organ culture was studied. Basal TRH secretion was stable over a 9-day period. The response to various secretagogues was tested on day 4. TRH secretion was stimulated by high potassium-induced depolarization and also through both cAMP and protein kinase-C dependent pathways. Like insulin, TRH release was stimulated by glucose and arginine and inhibited by somatostatin. These data suggest the existence of a common mechanism for TRH and insulin secretion by the pancreatic β-cells. 相似文献

Summary Light-microscopic immunocytochemistry and routine staining techniques were used to localize insulin and somatostatin-immunoreactive cells within the endocrine pancreatic tissue of the lamprey, Petromyzon marinus, during various stages of the life cycle. The endocrine pancreas of larvae consists solely of follicles of insulin-immunoreactive cells surrounding the junction of oesophagus, intestine and bile duct. Somatostatin-immunoreactive cells are restricted to the intestinal epithelium.
