Aims:  Weak acids are widely used by the food industry to prevent spoilage and to inhibit the growth of pathogenic micro-organisms. In this study the inhibitory effects of three commonly used weak acids, acetic acid, benzoic acid and sorbic acid, on the growth of Listeria monocytogenes were investigated.
Methods and Results:  In a chemically defined medium at pH 6·4 benzoic acid had the greatest inhibitory effect (50% inhibition of growth at 4 mmol l−1), while acetate was the least inhibitory (50% inhibition of growth at 50 mmol l−1). Mutants lacking either sigmaB (Δ sigB ) or two of the glutamate decarboxylase systems (Δ gadAB ) were used to investigate the contribution these systems make to weak acid tolerance in L. monocytogenes .
Conclusions:  The stress-inducible sigma factor sigmaB (σB) was not required for protection against acetate and played only a minor role in tolerating benzoate and sorbate. The glutamate decarboxylase system, which plays an important role in tolerating inorganic acids, played no significant role in the ability of L. monocytogenes to tolerate these weak acids, and neither did the presence of glutamate in the growth medium.
Significance and Impact of the Study:  These results suggest that the effectiveness of weak acid preservatives in food will not be compromised by the presence of glutamate, at least under mildly acidic conditions.  相似文献

Resistance to currently available antifungal drugs necessitates development of new drugs using rapid, robust and automated methods to test a large number of newly synthesized drugs in less time. We have compared the effect of ketoconazole, fluconazole and its synthesized analogues on Candida albicans ATCC 10231. A metabolic profile of C.albicans ATCC 10231 in presence of drugs has been compared using 1H NMR. Signals from metabolites have been monitored with time. MIC determined using conventional methods has been compared with Metabolic End Point (MEP) obtained from NMR spectroscopy. Results indicate that the activity of the fluconazole derivatives is in the order fluconazole p-methoxybenzoate > fluconazole = fluconazole benzoate > fluconazole toluate > fluconazole p-nitrobenzoate.  相似文献

Repeated (once a day for 8 days) but not single administration of estradiol benzoate (10 micrograms/kg, s.c.) induced a sevenfold increase in anterior pituitary gamma-aminobutyric acid (GABA) concentration in male rats. GABA concentration also increased in the median eminence whereas no changes or decreases were observed in other brain regions including hypothalamic arcuate nucleus, lateral septum, hippocampus, caudate nucleus, and substantia nigra. Eight-day estradiol benzoate injection also enhanced the Vmax of median eminence glutamate decarboxylase activity without affecting the Km of the enzyme for glutamic acid. Taken together, these results suggest that repeated administration of estradiol benzoate increases the activity of the tubero-infundibular GABAergic system in male rats.  相似文献

Studies were conducted to determine the potential of two avermectin compounds, abamectin and emamectin benzoate, for controlling plant-parasitic nematodes when applied by three methods: foliar spray, root dip, and pseudostem injection. Experiments were conducted against Meloidogyne incognita on tomato, M. javanica on banana, and Radopholus similis on banana. Foliar applications of both avermectins to banana and tomato were not effective for controlling any of the nematodes evaluated. Root dips of banana and tomato were moderately effective for controlling M. incognita on tomato and R. similis on banana. Injections (1 ml) of avermectins into banana pseudostems were effective for controlling M. javanica and R similis, and were comparable to control achieved with a conventional chemical nematicide, fenamiphos. Injections of 125 to 2,000 μg/plant effectively controlled one or both nematodes on banana; abamectin was more effective than emamectin benzoate for controlling nematodes.  相似文献

Insect neuropeptides are involved in many important developmental processes. Here, we cloned three neuropeptide genes (SfEH, SfETH, and SfCCAP) from Sogatella furcifera (Horváth) and analyzed the relative expression levels of these genes through Real-time Quantitative polymerase chain reaction (RT-qPCR) during different developmental stages and in different wing types of S. furcifera following exposure to sublethal concentrations (LC10 and LC25) of methylaminoavermectin benzoate. The results revealed that the three neuropeptide genes (SfEH, SfETH, and SfCCAP) of S. furcifera contained open reading frame sequences of 222, 636, and 408 bp, encoding 73, 211, and 135 amino acids, respectively. Moreover, the expression of SfEH and SfETH reached its peak on day 3 of the 5th instar nymph stage, while the SfCCAP expressed pinnacle on 40 min after eclosion. Furthermore, the relative expression levels of the three genes were in the following order: SfEH, short-winged female > long-winged female > long-winged male; SfETH, long-winged female > short-winged female > long-winged male; and SfCCAP, short-winged female > long-winged male > short-winged female. Upon calculating the relative expression of the three peptide genes after 24 and 48 h of treatment with sublethal concentrations of methylaminoavermectin benzoate, it was found that the relative mRNA expression levels of SfEH and SfETH declined upon LC10 treatment but increased upon LC25 treatment. Meanwhile, the relative mRNA expression level of SfCCAP declined upon both LC10 and LC25 treatments. This result will help us understand the effects of pesticides on insect growth and development at the molecular level.  相似文献

Female-specific serum protein(FSSP)is normally present in the sera of female fish,but it is notfound in males.However,estradiol benzoate(EB)was found to induce the appearance of FSSP inmale fish and immature female fish.Massive doses of FSSP caused a rapid increase in FSSP.Ultrastructural examination of the liver indicated an extensive proliferation of the rough endoplasmicreticulum and a decrease in glycogen and lipid droplets after EB injection.A preparative PAGE for isolating highly purified FSSP from the serum of EB-treated fish,Carassiusauratus cuvieri(Temminck & Schlegel),is described.Purified FSSP obtained from EB-induced O-crucian fish has a molecular weight of 466,000±4,000(n=10),while that in mature female serumis 480,000±40,000(n=2).FSSP appears to be a dimer,with the size of the possible monomerbeing 240,000±8,000(n=6).SDS-PAGE on gradient gels indicated that sera from males given multiple(12)injections of EBcontain a main band with a molecular weight of 147,000±6,000(n=6).However,the same serumsamples provided three bands of protein on the PAGE gels.Antiserum was raised against the electrophoretically purified FSSP.The resulting antibody formeda single,continuous precipitation line with sera from EB-treated males and vitellogenic females,butnot with that from normal males.lmmunocytochemistry(PAP method)was used to locate FSSP in the liver and ovary of maturefemales and the liver of EB-treated males.Strongly positive particles were found clustered in groupsaround liver cell nuclei under light microscopy,and the yolk granules in the oocytes were also filledwith positive particles.  相似文献

Cinnamyl benzoate specifically inhibited the growth of yeast-phase cells of the pathogenic fungus Sporothrix schenckii. A commercially available antimycotic agent, miconazole nitrate, released large amounts of K+ and Pi from S. schenckii probably due to it damaging the cell membrane, but no such release was observed with cinnamyl benzoate or with another commercial antimycotic agent, Tolnaftate. The sterol content of cells treated with cinnamyl benzoate and Tolnaftate was decreased and large amounts of squalene accumulated in the cells.
