A chemically defined medium using commercially available α-MEM supplemented with HEPES, L-glutamine, D-glucose, folic acid, D-biotin and adenine supports the luxuriant growth and propagation of Leishmania donovani promastigotes. A peak parasite population of about 7.0 × 107/ml at stationary phase and a population doubling time of 11.4 h for high-subpassage promastigotes were obtained. The medium was suitable for transformation of isolated amastigotes from infected hamster spleen. Promastigotes could be detected by culturing kala-azar patients’bone-marrow aspirate or spleen puncture material in this medium. Four out of six freshly transformed isolates gradually adapted and grew well in this medium. Macroscopic colonies appeared on agar plates prepared with the medium within 16–20 days after inoculation. The cloning efficiency was increased about five-fold by glycerol supplementation. 相似文献

During exploration in the Y-Mg-Zn system for quasicrystal approximants, three new phases, YMg1−xZn1+x (0 ? x ? 0.17) adopting the hexagonal ZrNiAl structure type, have been discovered. In these structures, the elements are completely ordered to minimize both the site energies and the bond energies as calculated by tight-binding calculations. Evaluation of the electron density in YMgZn suggests that Mg-Zn and Y-Zn bonding coupled with maximizing the Zn?Zn separations is the main factor influencing the atomic arrangements. Analysis of the electronic density of states of YMgZn indicates an optimized bonding situation for eight valence electrons per formula unit, e.g., as in YMgGa. Subsequently, YMgAl, YMgGa, and YMgIn were successfully prepared and structurally characterized. Their structures show relationships to both densely-packed structures common for intermetallics as well as three-dimensional networks common for valence compounds. 相似文献

Abstract This paper aims to demonstrate how intersectionality provides an important conceptual tool to analyse practices of cultural production in ethnic minority media. In this review, we discuss changes in the regulation of gene expression in the central nervous system (CNS) associated with DNA (cytosine-5) methylation, chromatin remodeling and post-translational covalent modifications of histones. During brain development, abnormal intrinsic or extrinsic cues may compromise epigenetic processes regulating neural stem cell proliferation and differentiation and thus directly or indirectly could contribute to altered epiphenotypes leading to psychiatric disorders. These mechanisms, that include chromatin remodeling and reversible changes in promoter methylation patterns, are largely expressed by terminally differentiated cortical GABAergic neurons. These neurons are unique among various brain cell subtypes because they express high levels of DNA-methyltransferase-1 (DNMT1). Moreover, DNMT1 expression is further increased in schizophrenia (SZ) and bipolar (BP) disorder brains. To unravel how this pathological DNMT1 overexpression induces GABAergic neuronal dysfunction in SZ and in other psychoses, we report on how alterations in methylation modify the expression of susceptible vulnerability genes such as reelin or GAD67 in these neurons. The results encourage the view that promoter hypermethylation in GABAergic neurons that occurs in SZ represents a testable target for novel therapeutic strategies to treat this disorder. 相似文献

Two new 1,4-naphthoquinone derivatives, balsaminone D (1), balsaminone E (2) along with two known compounds (3 and 4) were discovered from Impatiens balsamina L. flowers. Their structures were identified with spectroscopic methods including HR–EI–MS, 1D and 2D NMR, as well as the absolute configuration was determined by ECD calculation. In addition, new compounds 1 and 2 with IC50 value of 30.54 and 40.67 μg/mL exhibited better activities against activated t-HSC/Cl-6 cells than positive control Silymarin and Fufang Biejia Ruangan Pian, of which the IC50 value were 202.34 and 231.56 μg/mL, respectively. 相似文献

The synthesis of the mixed ligand mono metallic [Ru(dpop′)(tppz)]2+ and bimetallic [(dpop′)Ru(tppz)Ru(dpop′)]4+ (dpop′ = dipyrido(2,3-a:3′,2′-j)phenazine; tppz = 2,3,5,6 tetra-(2-pyridyl)pyrazine) complexes is described. The [Ru(dpop′)(tppz)]2+ complex display an intense absorption at 518 nm which is assigned to a Ru(dπ) → dpop′ (π∗) MLCT transition, and at 447 nm which is assigned to a Ru(dπ) → tppz(π∗) MLCT transition. It undergoes emission at RT in CH3CN with λem = 722 nm. The bimetallic [(dpop′)Ru(tppz)Ru(dpop′)]4+ complex shows a low energy absorption shoulder near 635 nm assigned to a Ru(dπ) → tppz(π∗) MLCT transition and an intense peak at 542 nm due to Ru(dπ) → dpop′ (π∗) MLCT transition. The bimetallic complex also emits at RT in CH3CN with λem = 785 nm. Cyclic voltammetry shows reversible Ru+2/+3 oxidations at 1.68 V for the monometallic complex and Ru+2/+3 oxidation couples at +1.94 and +1.70 V for the bimetallic complex. 相似文献

Human capillary endothelial cells (HCEC) in normal media contain noninactivating outwardly rectifying chloride currents,
TEA-sensitive delayed rectifier K+ currents and an inward rectifier K+ current. Two additional ionic currents are induced in HCEC when the media are allowed to become conditioned: A Ca2+-activated K+ current (BKCA) that is sensitive to iberiotoxin is induced in 23.5% of the cells, a transient 4-AP-sensitive K+ current (A current) is induced in 24.7% of the cells, and in 22.3% of the cells both the transient and BKCA currents are
coinduced. The EC50 for Ca2+ activation of the BKCA current in HCEC from conditioned media is 213 nM. RNA message for BKCA (hSlo clone) is undetecable
after PCR amplification in control cells but is seen in those from conditioned cells. The induction of BKCA current is not
blocked by conditioning with inhibitors of nitric oxide synthase, cyclo-oxgenase or lypo-oxygenase pathways. Apparently the
characteristics of human endothelial cells are highly malleable and can be easily modified by their local environment.

The aim of this study was to develop novel anaerobic media using gellan gum for the isolation of previously uncultured rumen bacteria. Four anaerobic media, a basal liquid medium (BM) with agar (A‐BM), a modified BM (MBM) with agar (A‐MBM), an MBM with phytagel (P‐MBM) and an MBM with gelrite (G‐MBM) were used for the isolation of rumen bacteria and evaluated for the growth of previously uncultured rumen bacteria. Of the 214 isolates composed of 144 OTUs, 103 isolates (83 OTUs) were previously uncultured rumen bacteria. Most of the previously uncultured strains were obtained from A‐MBM, G‐MBM and P‐MBM, but the predominant cultural members, isolated from each medium, differed. A‐MBM and G‐MBM showed significantly higher numbers of different OTUs derived from isolates than A‐BM (P < 0·05). The Shannon index indicated that the isolates of A‐MBM showed the highest diversity (H′ = 3·89) compared with those of G‐MBM, P‐MBM and A‐BM (H′ = 3·59, 3·23 and 3·39, respectively). Although previously uncultured rumen bacteria were isolated from all media used, the ratio of previously uncultured bacteria to total isolates was increased in A‐MBM, P‐MBM and G‐MBM. 相似文献
