Low populations (200 specimens per plant) of Pratylenchus cofl''eae, Scutellonema bradys, Meloidogyne incognita, and Rotylenchulus reniformis stimulated the development of tops, roots, and tubers of Dioscorea rotundata "Guinea" yam. We demonstrated experimentally that P. coffeae was responsible for the deterioration in quality of the yam tuber in Puerto Rico, a condition known as a dry rot of yam. Initial populations of 600 P. coffeae, S. bradys, or M. incognita, and populations of 1,000 P. coffeae or S. bradys per plant were high enough to induce dry rot of the yam tubers. P. coffeae and S. bradys were pathogenic to yam cultivar Guinea, but M. incognita and R. reniformis did not cause necrosis or cracking of the tuber cortex in our experiments.  相似文献

The lesion nematode, Pratylenchus thornei, was clearly demonstrated as a parasite of wheat. It reduced plant stands and stunted plants in the field under the environmental conditions found in Sonora, Mexico. Other soil organisms also may have contributed to the problem. The nematode is widely distributed throughout the wheat-growing region, and may be a problem each growing season. Nematicides controlled the nematode and increased yields, but they were not economical. No resistance was found in existing commercial wheat cultivars. A pest management approach using variety selection, nitrogen fertilizer, planting in cool soil (15 C) and a crop rotation avoiding wheat after wheat was the most practical solution to this problem on a commercial scale.  相似文献

''Vernal'' alfalfa was grown for 30 weeks in nematode-free soil and in soil infested with Pratylenchus penetrans. Charlottetown fine sandy loam soil was used at its pH of 4.4 and at adjusted reactions of 5.2, 6.4 and 7.3. Nematode reproduction was significantly greater at pit 5.2 and 6.4 and was not related to alfalfa root production over the full pH range studied. A significant nematode infestation X soil pit interaction on forage yield was recorded. Nematode infestation significantly decreased forage yields at ptt 5.2 and 6.4 but not at pH 4.4 and 7.3.  相似文献

In a soil temperature study, population increase on ''Clark 63'' soybeatt was most rapid at 30 C in Pratylenchus alleni, P. brachyurus, P. cofleae, P. neglectus, P. scribneri, and P. zeae and at 25 C in P. penetrans and P. vulnus. The last two were the only species that reproduced at 15 C. Populations of all species increased over the range of 20-30 C, except those of P. neglectus at 20 C and P. coffeae, which was not tested below 25 C. Only P. brachyurus, P. neglectus, P. scribneri and P. zeae reproduced at 35 C. At their optimum temperatures, P. scribneri exhibited the greatest population increase, 1248-fold, and P. penetrans the least, 32-fold. This is the first report of soybean as a host for P. vulnus.  相似文献

After vessel injury, platelets adhere to the subendothelial matrix. Platelet adhesion leads to activation of the platelet integrin alpha(IIb)beta3, which then binds to fibrinogen, leading to platelet aggregation. It has been shown that a beta3-integrin binding protein, beta3-endonexin, can activate the integrin alpha(IIb)beta3 expressed in transfected CHO cells. Several isoforms of beta3-endonexin are known but it is not clear which isoforms are expressed in platelets and what role they may play during haemostasis. Here, we show that the long form of beta3-endonexin (EN-L) can be detected in platelet lysates several hours after thrombus formation, after long-term storage of platelets and after glucose deprivation. After subcellular fractionation, EN-L is found in the detergent insoluble fraction suggesting that it might be associated with the cytoskeleton. EN-L generation is temperature and Ca++ dependent and requires physiological salt concentrations. Proteolysis is responsible for the appearance of EN-L since a calpain inhibitor prevents its formation and the addition of calpain to platelet lysates induces its formation. The appearance of EN-L seems to be linked to apoptotic events occurring during long-term storage of platelets and, possibly, during late steps of haemostasis after thrombus formation.  相似文献

DNA in living cells is constantly subjected to different chemical and physical factors of the environment and to cell metabolites. Some changes altering DNA structure occur spontaneously. This raises the potential danger of harmful mutations that could be transmitted to offspring. To avoid the danger of mutations and changing genetic information, a cell is capable to switch on multiple mechanisms of DNA repair that remove damage and restore native structure. In many cases, removal of the same damage may involve several alternative pathways; this is very important for DNA repair under the most unfavorable conditions. This review summarizes data about all known mechanisms of eukaryotic DNA repair including excision repair (base excision repair and nucleotide excision repair), mismatch repair, repair of double-strand breaks, and cross-link repair. Special attention is given to the regulation of excision repair by different proteins—proliferating cell nuclear antigen (PCNA), p53, and proteasome. The initial encounter of an unrepaired DNA lesion is likely to be with a replicative DNA polymerase, and the outcome of this event determines whether an error-prone or error-free damage avoidance pathway is taken. To understand the atomic details of this critical encounter, we have determined the crystal structures of the pol alpha family RB69 DNA polymerase with DNA containing the two most prevalent, spontaneously generated premutagenic lesions, an abasic site and 2'-deoxy-7,8-dihydro-8-oxoguanosine (8-oxodG). Identification of the interactions between these damaged nucleotides and the active site provides insight into the capacity of the polymerase to incorporate a base opposite the lesion. A novel open, catalytically inactive conformation of the DNA polymerase has been identified in the complex with a primed abasic site template. This structure provides the first molecular characterization of the DNA synthesis barrier caused by an abasic site and suggests a general mechanism for polymerase fidelity. In contrast, the structure of the ternary 8-oxodG:dCTP complex is almost identical to the replicating complex containing unmodified DNA, explaining the relative ease and fidelity by which this lesion is bypassed.  相似文献

We previously established a system of in vitro regeneration and Agrobacterium-mediated transformation for hot pepper plants. The level of protection against cucumber mosaic virus in the progeny of the transgenic hot pepper plants that express cucumber mosaic virus (CMV) satellite RNA was investigated. The transgenic hot pepper plants were self-fertilized, and their progeny were tested for stable inheritance and expression of the cDNA of CMV satellite RNA. Polymerase chain reaction and RNA gel blot analyses showed that the introduced gene was stably transmitted and expressed in the progeny. Symptom attenuation in the offspring was confirmed upon inoculation with CMV-Y or CMV-Korea (CMV-Kor) strains.
