Local and chemical distribution of phlorotannins in brown algae   总被引:4,自引:2,他引:2  
The local and chemical distribution of phlorotannins among the Japanese Laminariaceae, Eisenia bicyclis, Ecklonia cava and Ecklonia kurome, was investigated. As a result of light microscopy observations with vanillin-HCl staining, phlorotannins were found to be accumulated within the vegetative cells of the outer cortical layer of the thalli, regardless of the species, stage of growth or organ. Crude phlorotannins comprised about 3.0% of the algal powder for each of the algae. High-performance liquid chromatography (HPLC) showed that the phlorotannins of E. bicyclis were composed of phloroglucinol (0.9%), phloroglucinol tetramer (4.4%), eckol (7.5%), phlorofucofuroeckol A (21.9%), dieckol (23.4%), and 8,8'-bieckol (24.6%), plus some other unknown phenolic compounds (17.3%). The composition of the phlorotannins differed little among the Laminariaceae, except for a significantly larger amount of the tetramer, MW 478, in E. bicyclis.  相似文献

采用SephadexG 75和HiPrep 1 6 / 6 0DEAE离子交换等方法从赤子爱胜蚓 (Eiseniafoetida)中提取到一组活性蛋白质成分 ,双向电泳分析证明它们为一组pI在 3 .0~ 4 .0之间的酸性蛋白质 ;利用体外K5 6 2、HeLa、SY5Y等肿瘤细胞抑制实验和纤维蛋白平板实验 ,跟踪测定活性 ,证明乙醇沉淀组分D2 (8)是既具肿瘤抑制 ,又具有激酶活性的蛋白质成分。同时应用非变性电泳对乙醇沉淀组分进行分离 ,并利用电洗脱、凝胶原位酶解和ESI MS等蛋白质组学方法 ,鉴定了其中 6种蛋白质的分子量、氨基酸组成、N末端序列和肽质量指纹图信息 ,其中条带 9与D2 (8)组分为同一种蛋白质 ;研究证明蚯蚓中含有既具抗肿瘤活性又具有激酶活性的蛋白质成分。采用的方法可适用于活性蛋白质成分的整体分离与鉴定  相似文献

Earthworm survival in oil contaminated soil   总被引:4,自引:0,他引:4  
Earthworms are an important component of the soil biota and their response to oil pollution needs to be better understood. Laboratory investigations were undertaken to determine the concentrations of crude oil in soil that leads to death of Lumbricus terrestris and Eisenia fetida and to determine the propensity of L. terrestris to move away from contaminated soil. Clemville sandy clay loam was amended to contain maximum oil contents of 1.5 – 2.5% depending on the particular experiment. Additionally, the ability of L. terrestristo survive in bioremediated oil-contaminated soil was evaluated. An oil content of 0.5% was not harmful to survival of earthworms for 7 d but an oil concentration of 1.5% reduced survival to less than 40%. Bioremediated soil containing 1.2% oil did not reduce survival of L. terrestrisduring 10 d. Survival of L. terrestrisin unweathered oil was improved when free movement between contaminated and uncontaminated soil was possible. Casts of earthworms exposed to oil-containing soil contained 0.2% total petroleum hydrocarbons. An allowable regulatory level of 1% oil contamination in soil may not allow for survival of earthworms.  相似文献

Vermifiltration is a new technology using earthworms to process organically polluted water. A pilot associated with a swine facility (piggery) with 66 swine was developed to treat diluted manure, produce earthworms and vermicompost, and reduce air pollution. The aim of the experiment reported here was to devise an integrated method – biological, chemical and physical – for further research and development of vermifiltration in diluted swine manure, and provide some preliminary results. The earthworm population increased by 30% in 4 weeks, indicating the acclimation of the earthworms. A reduction in ammonia emission was observed of about 50% for the whole system. Higher water (+100%), carbon (+70%), and total nitrogen (+80%) gaseous losses were observed compared to conventional breeding on a slatted floor. This methodology can be used for further studies to develop vermifiltration for earthworm and vermicompost production from diluted animal manure, without pollution transfer.  相似文献

Zusammenfassung Im Laufe der Cytoplasmareduktion während der Spermiogenese von Eisenia foetida sind zwei Vorgänge zu unterscheiden: 1. Die Bildung autophagischer Vakuolen. Sie entstehen, indem Teile des Grundcytoplasmas in das Kompartiment des ER verlagert werden. Da sie keine Reaktion auf saure Phosphatase geben, sind sie als nicht lysosomale Anfangstadien der zellulären Autophagie zu betrachten. 2. Die Bildung primärer Lysosomen. Sie entstehen in Form von lytische Enzyme enthaltenden Golgivesikeln, die von einer neu im Cytoplasma entstehenden Membran zu größeren Einheiten zusammengefaßt werden: den multivesicular bodies. Autophagische Vakuolen und multivesicular bodies gelangen ins Cytophor das am Ende der Spermiogenese den Charakter eines ausgedehnten Autophagosoms annimmt. Als Struktureigentümlichkeit entstehen in ihm undulierende Tubulikörper. Der coat an den Hüllmembranen junger multivesicular bodies und am Plasmalemm der Spermatidenverbindung zum Cytophor wird in Zusammenhang mit der Membrandifferenzierung diskutiert.
Ultrastructural equivalents of cellular autophagyElectronmicroscopical observations on spermatids of Eisenia foetida during the cytoplasmic reduction
Summary During the cytoplasmic reduction phase in the spermiogenesis of Eisenia foetida two different processes may be defined: 1. The formation of autophagic vacuoles, which arise by the displacement of cytoplasmic portions into the cisternae of the endoplasmic reticulum. Since they exhibit no acid phosphatase activity they are considered to be early stages in cellular autophagy. 2. The formation of primary lysosomes. They originate in Golgi vesicles and are then enveloped by a membrane, formed in the cytoplasm de novo, which transforms them into multivesicular bodies. Autophagic vacuoles and multivesicular bodies subsequently transfer to the cytophor, which contains at the end of the spermiogenesis the characteristics of a large autophagosom, showing aggregates of undulating tubules. The outer coat of the limiting membranes in the early multivesicular bodies and of the cell membrane of the connecting piece between spermatid and cytophor appear to be associated with the membrane development.

6种重金属对赤子爱胜蚓的急性毒性效应与风险评价   总被引:3,自引:0,他引:3       下载免费PDF全文
【背景】近年来,土壤重金属污染问题日益凸显,对生态环境、食品安全和人体健康构成了严重威胁,其急性毒性尚未明确。【方法】采用滤纸接触法和人工土壤法测定了铜(Cu2+)、锌(Zn2+)、镍(Ni+)、镉(Cd2+)、铅(Pb2+)和锰(Mn2+)等6种重金属对赤子爱胜蚓的急性毒性效应,并参照欧盟指令91/414/EEC标准评价了其环境风险。【结果】滤纸接触法48h测定结果表明,6种重金属对蚯蚓的LC50为3.17(2.53~3.81)~90.42(69.45~140.47)μg.cm-2,其毒性次序为Cu2+>Zn2+>Ni+>Cd2+>Pb2+>Mn2+。人工土壤法14d测定结果表明,6种重金属对蚯蚓的LC50为1347(1236~1453)~6936(6144~8930)mg.kg-1,其毒性次序为Cu2+>Cd2+>Ni+>Zn2+>Mn2+>Pb2+。风险评价结果显示,Cu2+、Zn2+、Ni+、Cd2+、Pb2+和Mn2+等6种重金属的暴露比(toxicity/exposureratio,TER)分别为3.37、4.46、8.68、1428、13.87和5.85。【结论与意义】6种重金属对土壤动物蚯蚓均具有潜在的毒性效应。Cu2+、Zn2+、Ni+、Mn2+等4种重金属对赤子爱胜蚓存在急性毒性风险,而Cd2+和Pb2+对赤子爱胜蚓的急性毒性风险水平是可接受的。该评价结果可为我国制定基于风险的土壤环境质量标准提供依据。  相似文献

为寻找土壤低浓度多环芳烃污染分子生物标记物,采用了抑制消减双杂交的方法构建了赤子爱胜蚓在苯并[a]芘(BaP)人工土壤污染胁迫下的差异表达cDNA文库,经测序和基因比对分析后,在上调文库中分别发现2个与热休克蛋白HSP70和1个与HSP90显著匹配的cDNA克隆.经定量PCR验证了0。1 mg·kg-1和1。0 mg·kg-1 BaP对赤子爱胜蚓HSP70和HSP90的诱导作用,表明这两个新克隆到的赤子爱胜蚓热休克蛋白基因可作为土壤污染监测的备选分子生物标记物.  相似文献

Traditional neonicotinoid insecticides are used worldwide. Paichongding (IPP), as a novel neonicotinoid pesticide, has been widely used in China. However, the ecotoxicity of IPP to non-target invertebrates in soil ecosystem has not been reported yet. In this study, acute toxicity of IPP to earthworm Eisenia fetida, as well as the antioxidant response after IPP exposure, was evaluated. In the filter paper contact test, the LC50 at 24 hr and 48 hr for IPP were 14.98 μg/cm2 and 7.59 μg/cm2, respectively. In artificial soil test, the LC50 (lethal concentration) at 14 days and 28 days for IPP were 541.07 mg/kg and 238.51 mg/kg, respectively. The LC50 of IPP is much higher than that of traditional neonicotinoid insecticides. However, earthworm body weight assessment demonstrated that the growth of earthworm was inhibited by extended exposure to IPP at sublethal doses. The activities of antioxidative enzymes superoxide dismutase and catalase in earthworms were significantly induced after IPP exposure. Malondialdehyde, a biomarker of lipid peroxidation, was also increased after IPP exposure. Although the results indicated that IPP had potentially adverse effect on earthworms, its toxicity was much lower than traditional neonicotinoids.  相似文献

Bt毒素能通过转基因作物的花粉、根和残株进入土壤.为评估转基因作物对土壤动物的影响,本文模拟转基因棉的Bt毒素进入土壤的发生程度,用含不同浓度Bt毒蛋白Cry1Ac的人造土壤处理蚯蚓,测定蚯蚓存活率、重量变化及体内总蛋白含量和过氧化氢酶(CAT)、乙酰胆碱酯酶(AchE)、谷胱甘肽-S-转移酶(GST)和纤维素酶活性.结果表明,Bt毒蛋白对蚯蚓的生物量和生理水平影响均不明显,不存在急毒性和亚致死毒性影响,对蚯蚓比较安全.  相似文献

为获得一种高效,低廉的溶栓药物,从赤子爱胜蚓(Eiseniafaetida)体内分离纯化出一种可体外激活纤溶酶原从而间接降解纤维蛋白的酶(e-PA).纯化过程包括:粗品的盐析,离子交换层析,凝胶过滤层析及疏水相互作用层析.该组份是由二个亚基通过疏水相互作用维系在一起的.通过凝胶过滤层析,可测得全酶的分子量为45000;SDS电泳显示大、小亚基的分子量分别是26000与18000;而质谱法测得的大、小亚基的分子量分别为24556.7与15546.6.对大小亚基进行了氨基酸组成分析,结果显示大亚基不含Lys而小亚基不含Cys.测定了大亚基N端25个氨基酸序列:VIGGTNASPGEIPWQLSQQRQSGSW.并与部分已知蛋白质序列进行了比较.e-PA在纤维蛋白平板上表现有三种不同的纤溶活性  相似文献
