文章综述了植物2n配子发生及其遗传标记研究现状,论述涉及有关形态学标记、细胞学标记、同工酶标记乃至DNA标记等遗传标记在2n配子研究中的应用,指出通过花粉形态观察、大小孢子母细胞减数分裂行为观察、杂种后代倍性鉴定以及亲子分子标记相关分析等,对2n配子发生、2n配子遗传类型与杂合性以及2n配子在育种实践中的有效性等进行研究。分子标记技术已经成为2n配子形成相关基因分析等方面的有力工具,利用分子标记等手段,研究2n配子形成的分子机理以及应用于辅助育种等将成为今后的热点,进而推动植物育种尤其是多倍体育种蓬勃发展。 相似文献

The nature of a number of fundamental processes occurring during reproduction in sponges still remains in doubt. Among the more significant of these are: the true status of sponges described as dioecious, namely whether some are actually successive hermaphrodites; the origin of oogonia, which have recently been claimed to be derived from choanocytes; the origin and mechanism of formation of large spermatogenic masses; the specific pathway leading to fertilization taken by sperm cells within the sponge tissue of viviparous species; the role played during larval metamorphosis by somatic cells which are incorporated into embryos; the cell lineage of choanocytes which form flagellated chambers during larval metamorphosis; the specific relationship of somatic growth and dormancy to gametogenesis; the role of budding and fragmentation in population maintenance; the role, if any, of gemmules in dispersion. It is considered mandatory that new techniques be developed in order to further elucidate these and other reproductive processes and to gather definitive data concerning them. The employment of only microscopic techniques is ultimately insufficient for investigating the dynamic relationships of reproductive processes. 相似文献

Brown algae (stramenopiles) are key players in intertidal ecosystems, and represent a source of biomass with several industrial applications. Ectocarpus siliculosus is a model to study the biology of these organisms. Its genome has been sequenced and a number of post‐genomic tools have been implemented. Based on this knowledge, we report the reconstruction and analysis of a genome‐scale metabolic network for E. siliculosus, EctoGEM ( http://ectogem.irisa.fr ). This atlas of metabolic pathways consists of 1866 reactions and 2020 metabolites, and its construction was performed by means of an integrative computational approach for identifying metabolic pathways, gap filling and manual refinement. The capability of the network to produce biomass was validated by flux balance analysis. EctoGEM enabled the reannotation of 56 genes within the E. siliculosus genome, and shed light on the evolution of metabolic processes. For example, E. siliculosus has the potential to produce phenylalanine and tyrosine from prephenate and arogenate, but does not possess a phenylalanine hydroxylase, as is found in other stramenopiles. It also possesses the complete eukaryote molybdenum co‐factor biosynthesis pathway, as well as a second molybdopterin synthase that was most likely acquired via horizontal gene transfer from cyanobacteria by a common ancestor of stramenopiles. EctoGEM represents an evolving community resource to gain deeper understanding of the biology of brown algae and the diversification of physiological processes. The integrative computational method applied for its reconstruction will be valuable to set up similar approaches for other organisms distant from biological benchmark models. 相似文献

Recalling the evolutionary sequence of development first of gonad and subsequently of oviducts, ovarian endocrine regulation of all known components of oviduct physiology is reviewed. Ovaries not only influence oviducts via the systemic blood circulation, but also locally by counter‐current transfer of relatively high concentrations of steroid hormones and prostaglandins between the ovarian vein and oviduct branch of the ovarian artery. The efficiency and impact of such counter‐current transfer is greatest around the time of ovulation, the transfer process receiving further inputs from hormones present in peritoneal fluid. Classical oviduct physiology is summarised, and the potential molecular consequences of temperature gradients within the duct lumen examined. At ovulation, an oocyte‐cumulus complex is displaced in minutes from the follicular surface to the site of fertilisation at the ampullary‐isthmic junction of the oviduct. This rapid initial phase is contrasted with the subsequent slow progression of embryos to the uterus in days, still encompassed within a zona pellucida. Regarding transport of spermatozoa, the formation of a pre‐ovulatory reservoir in the caudal portion of the oviduct isthmus is noted, with suppression of motility and sperm‐head binding to epithelial organelles acting to maintain fertilising ability. Completion of capacitation is prompted shortly before ovulation, predominantly by Ca2+ influx into bound spermatozoa. A controlled release of spermatozoa coupled with their hyperactivation results in initial sperm:egg ratios at the site of fertilisation close to unity, thereby avoiding the pathological condition of polyspermy. Both the oviduct milieu and embryonic development are influenced by paracrine activity of follicular granulosa cells released at ovulation and remaining in suspension in the vicinity of the oocyte or embryo. These cells may amplify early pregnancy signals from a zygote to the endosalpinx. Beneficial effects of the oviduct on domestic animal embryos are contrasted with anomalies arising as a consequence of in vitro culture. Primate embryos do not require exposure to an oviduct for normal development, perhaps due to overlapping compositions of endosalpingeal and endometrial secretions. Additionally, primate endometrial secretions may be modified by viable gametes or an embryo in the presence of a cumulus cell suspension. 相似文献

Background and Aims
Patterns of ploidy variation among and within populations can provide valuable insights into the evolutionary mechanisms shaping the dynamics of plant systems showing ploidy diversity. Whereas data on majority ploidies are, by definition, often sufficiently extensive, much less is known about the incidence and evolutionary role of minority cytotypes.Methods
Ploidy and proportions of endoreplicated genome were determined using DAPI (4'',6-diamidino-2-phenylindole) flow cytometry in 6150 Gymnadenia plants (fragrant orchids) collected from 141 populations in 17 European countries. All widely recognized European species, and several taxa of less certain taxonomic status were sampled within Gymnadenia conopsea sensu lato.Key Results
Most Gymnadenia populations were taxonomically and/or ploidy heterogeneous. Two majority (2x and 4x) and three minority (3x, 5x and 6x) cytotypes were identified. Evolution largely proceeded at the diploid level, whereas tetraploids were much more geographically and taxonomically restricted. Although minority ploidies constituted <2 % of the individuals sampled, they were found in 35 % of populations across the entire area investigated. The amount of nuclear DNA, together with the level of progressively partial endoreplication, separated all Gymnadenia species currently widely recognized in Europe.Conclusions
Despite their low frequency, minority cytotypes substantially increase intraspecific and intrapopulation ploidy diversity estimates for fragrant orchids. The cytogenetic structure of Gymnadenia populations is remarkably dynamic and shaped by multiple evolutionary mechanisms, including both the ongoing production of unreduced gametes and heteroploid hybridization. Overall, it is likely that the level of ploidy heterogeneity experienced by most plant species/populations is currently underestimated; intensive sampling is necessary to obtain a holistic picture. 相似文献56.

54 localities in Japan and two localities in China, 82 candidates of other lineage(s) of cryptic clones were screened by examining
RFLP (restriction fragment length polymorphism)-PCR haplotypes in the control region of mtDNA. This analysis was performed
because triploid loaches arise from the accidental incorporation of the sperm nucleus into unreduced diploid eggs of a clone.
The categorization of members belonging to three newly identified lineages (clones 2–4) and the previously identified clonal
lineage (clone 1) was verified by evaluating the genetic identity between two or more individuals from each clonal lineage
based on RAPD (random amplified polymorphic DNA)-PCR and multilocus DNA fingerprints. We detected 75 haplotypes by observing
the nucleotide status at variable sites from the control region of mtDNA. Phylogenic trees constructed from such sequences
showed two highly diversified clades, A and B, that were beyond the level common for interspecific genetic differentiation.
That result suggests that M. anguillicaudatus in Japan is not a single species entity. Two clone-specific mtDNA sequences were included in clade A, and the loaches with
such sequences may be the maternal origin of the clones.

Abstract. Recent reports of occasional males in Austrian populations of Triops cancriformis have promoted interest in an analysis of the ultrastructure of the male gonad. It appears as a double thick and well‐defined tubular structure running along the midgut, inside the hemocoel. It is composed of two portions: the wall and the lumen. The former is made up of germ cells that are widespread and intermixed with somatic cells. The lumen is ~200 μm wide and acts as the collecting site of spermatids and sperm. The germ cells are recognizable by their rounded appearance and by the occurrence of exceptionally long synaptonemal complexes in their nuclei. Their maturation implies a volume elongation and an aggregation in a characteristic “cyst” arrangement. The cysts migrate towards the lumen, where they break open, releasing spermatids and sperm. At the end of this process the lumen is filled with maturing gametes. The sperm have very condensed chromatin nuclei and partially reduced cytoplasm where the most striking features are the axoneme and strewn microtubules together with evident pseudopodia. Our morphological data support the full functionality of males in T. cancriformis. However, further work on gametogenesis, distribution of sexes, and genetic analysis of breeding experiments are needed to reach a fuller understanding of the role of males in middle and northern European populations of this species. 相似文献

Brown algae share several important features with land plants, such as their photoautotrophic nature and their cellulose-containing wall, but the two groups are distantly related from an evolutionary point of view. The heterokont phylum, to which the brown algae belong, is a eukaryotic crown group that is phylogenetically distinct not only from the green lineage, but also from the red algae and the opisthokont phylum (fungi and animals). As a result of this independent evolutionary history, the brown algae exhibit many novel features and, moreover, have evolved complex multicellular development independently of the other major groups already mentioned. In 2004, a consortium of laboratories, including the Station Biologique in Roscoff and Genoscope, initiated a project to sequence the genome of Ectocarpus siliculosus, a small filamentous brown alga that is found in temperate, coastal environments throughout the globe. The E. siliculosus genome, which is currently being annotated, is expected to be the first completely characterized genome of a multicellular alga. In this review we look back over two centuries of work on this brown alga and highlight the advances that have led to the choice of E. siliculosus as a genomic and genetic model organism for the brown algae. 相似文献
